Exploring the transcriptomic changes underlying recombinant vaccine efficacy against Teladorsagia circumcincta in 3-month-old lambs.

Teladorsagia circumcincta is an abomasal parasitic nematode that can cause serious issues in small ruminant production, which are aggravated by drug resistance. Vaccines have been suggested as a feasible, long-lasting alternative for control since adaptation to the host's immune mechanisms by helminths develops at a much slower pace than anthelmintic resistance. Recently, a T. circumcincta recombinant subunit vaccine yielded over a 60% reduction in egg excretion and worm burden and induced strong humoral and cellular anti-helminth responses in vaccinated 3-month-old Canaria Hair Breed (CHB) la

Anthelmintic resistance in gastrointestinal nematodes of dairy cattle in the Macalister Irrigation District of Victoria.

OBJECTIVE: To report anthelmintic resistance in gastrointestinal nematode parasites of cattle on commercial dairy farms in the Macalister Irrigation District of Gippsland, Victoria. METHODS: Faecal egg count reduction tests (FECRTs) were used to assess anthelmintic resistance on 20 Macalister Irrigation District dairy farms between May 2013 and June 2014. All three currently available anthelmintic classes for cattle nematodes in Australia were tested. Faecal samples were collected 10-14 days post-treatment for individual faecal egg counts (FEC) and larval differentiation. The arithmetic mean

Epidemiology and field efficacy of anthelmintic drugs associated with gastrointestinal nematodes of sheep in Nejo district, Oromia, Ethiopia.

Gastrointestinal nematodes (GINs) are major constraints to health and productivity of small ruminants. Methods of their control relies mainly on anthelmintic drugs; however, the indiscriminate use of these drugs could lead to the development of anthelmintic resistance (AR). This study aimed to investigate the epidemiology of GINs infection, and field evaluation of anthelmintic efficacy in sheep. The epidemiological data were collected using a cross-sectional study design while a farm-based field study design was employed for the evaluation of anthelminthic efficacy. Furthermore, standard paras

Modelling the consequences of targeted selective treatment strategies on performance and emergence of anthelmintic resistance amongst grazing calves.

The development of anthelmintic resistance by helminths can be slowed by maintaining refugia on pasture or in untreated hosts. Targeted selective treatments (TST) may achieve this through the treatment only of individuals that would benefit most from anthelmintic, according to certain criteria. However TST consequences on cattle are uncertain, mainly due to difficulties of comparison between alternative strategies. We developed a mathematical model to compare: 1) the most 'beneficial' indicator for treatment selection and 2) the method of selection of calves exposed to Ostertagia ostertagi, i.

The identification of small molecule inhibitors with anthelmintic activities that target conserved proteins among ruminant gastrointestinal nematodes.

Gastrointestinal nematode (GIN) infections are a major concern for the ruminant industry worldwide and result in significant production losses. Naturally occurring polyparasitism and increasing drug resistance that potentiate disease outcomes are observed among the most prevalent GINs of veterinary importance. Within the five major taxonomic clades, clade Va represents a group of GINs that predominantly affect the abomasum or small intestine of ruminants. However, the development of effective broad-spectrum anthelmintics against ruminant clade Va GINs has been challenged by a lack of comprehen

Haemonchus contortus and other trichostrongylid infections in parturient, lactating and dry ewes.

From autumn (April) to spring (November), groups of pregnant and dry Merino ewes grazed pasture contaminated with infective larvae of Haemonchus contortus, Ostertagia circumcincta and Trichostrongylus colubriformis. A periparturient rise in faecal egg counts occurred in the pregnant ewes, following the maturation in early spring of arrested fourth-stage larvae of H. contortus and failure of pregnant ewes to expel the resulting adult worms. Peak egg counts were seen in September, just before lambing. In dry ewes, egg counts were slightly elevated at the time of the rise in the pregnant ewes, bu

Internal parasites of Washington cattle.

The occurrence of internal parasites in Washinton cattle was estimated. During the survey period in 1972 and 1973, more than 3,700 fecal samples from 29 herds and 4 feedlots, and viscera from 55 cattle were examined. The percentage of fecal samples with parasite eggs or oocysts from herds was as follows: oocysts, 77%; "strongyline" eggs, 44%; Nematodirus eggs, 3%; Moniezia eggs, 3%; Fasciola eggs, 2%; Trichuris eggs, 2%; and Dictyocaulus larvae, 1%. The fecal samples from feedlots contained principally strongyline eggs which varied markedly in numbers among groups of cattle in the lots. The pr

Influence of chemoprophylaxis on protective immunity to nematodes in cattle: a two-year study comparing four control strategies.

A two-year study was conducted in three sequential phases. Initially, four matched groups of nine calves grazing similar pastures were subjected to different chemoprophylactic control programmes while a fifth group acted as unprotected controls. After being housed in the autumn, three calves from each group and two parasite-naive controls were challenged experimentally with Ostertagia ostertagi, Cooperia oncophora and Dictyocaulus viviparus. Post mortem worm counts revealed gradations of protective immunity, with the field controls showing the highest level and the naive controls the lowest. I

In vivo passage through calves of nematophagous fungi selected for biocontrol of parasitic nematodes.

The experiment was designed to test the survival and performance of stress selected nematophagous fungi after passage through the gastro-intestinal tract of cattle. Ruminating calves were fed daily with a fixed amount of fungal material grown on barley grains. The excreted dung was collected on days four and five after the start of the feeding experiment. Barley grains were washed out of the excreted dung and incoculated on water-agar plates. After incubation for one week, nine of ten fungal isolates were re-isolated from these plates. The predatory capacity of the fungi in the excreted faeces

Overwintering of bovine gastrointestinal nematodes in southwestern Ontario.

Several steers, reared in isolation until approximately six months of age, were placed on a small isolated enclosed pasture from late spring to late fall of 1970, 1971 and 1972. The pasture was left vacant and unused during the winters and early springs. The pasture had been used in previous years by cattle, and in the late spring of 1970 was grazed by cattle know to be passing gastrointestinal nematode eggs in their feces. The steers were slaughtered periodically, and the prevalence of nematode species in the abomasum, small intestine, and large intestine, was determined from random samples o

Reduced efficacy of moxidectin and abamectin in young red deer (Cervus elaphus) after 20 years of moxidectin pour-on use on a New Zealand deer farm.

A study was undertaken on weaned 4-5 month old farmed red deer to test the efficacy of moxidectin and abamectin anthelmintics, given by three different routes of administration, compared with an untreated control. Faecal samples were collected on days 0, 7 and 14 for a faecal egg count reduction test (FECRT), blood samples were collected on days 0, 0.5, 1, 2, 3, 5, 7, 10 and 14 for pharmacokinetics, and the deer were killed on days 14 or 15 for total nematode count. The control group averaged 1264 adult Ostertagia-type nematode parasite species and treatment efficacy was 77.4% for moxidectin i

GLOWORM-META: modelling gastrointestinal nematode metapopulation dynamics to inform cattle biosecurity research.

Gastrointestinal nematode (GIN) parasite infections in grazing livestock cause significant disease, and are responsible for estimated annual losses of over €1.8 billion in Europe alone. The management of GINs in cattle is threatened by anthelmintic drug resistance (AR). Immediate action is needed to slow the progression of AR in cattle GINs and avoid the increasingly common scenario of multiple drug resistance seen in sheep. Although AR can arise independently on multiple farms, it may also be spread between holdings via purchased cattle. Therefore, effective biosecurity measures on cattle ent

Teladorsagia circumcincta resistance to moxidectin and multiple anthelmintic groups in ewes following use of the persistent drug before lambing.

This paper describes the nematode control strategy adopted by a Scottish hill sheep farmer. It provides an example of the limited use of anthelmintic drugs, targeted towards control of the periparturient rise in faecal nematode egg output in ewes and exploitation of grazing management, resulting in limited exposure of naive sheep to infective larvae on pasture. Resistance to benzimidazole, imidazothiazole and macrocyclic lactone anthelmintics, and to a combination of a macrocyclic lactone and imidazothiazole anthelmintic drugs was diagnosed. The targeted use of a persistent anthelmintic drug t

Development of Bovine Gastric Organoids as a Novel In Vitro Model to Study Host-Parasite Interactions in Gastrointestinal Nematode Infections.

Gastro-intestinal nematode (GIN) parasites are a major cause of production losses in grazing cattle, primarily through reduced growth rates in young animals. Control of these parasites relies heavily on anthelmintic drugs; however, with growing reports of resistance to currently available anthelmintics, alternative methods of control are required. A major hurdle in this work has been the lack of physiologically relevant in vitro infection models that has made studying precise interactions between the host and the GINs difficult. Such mechanistic insights into the infection process will be valu

First report of cattle farms with gastrointestinal nematodes resistant to levamisole in Mexico.

The objectives of the present study were: (1) to report the percentage of cattle farms with gastrointestinal nematodes (GINs) resistant to levamisole in Veracruz, Mexico, (2) to identify the genera of GINs involved in resistance, and (3) to identify factors associated with these resistances. The faecal egg count reduction test (McMaster technique) was used to detect the presence of resistant GINs. A questionnaire was given to owners to understand the history of anthelmintic use. The percentage of cattle farms with GINs resistant to levamisole was 36.4% (4/11). The percentage of faecal egg coun

Gastrointestinal nematode infection and performance of weaned stocker calves in response to anthelmintic control strategies.

Gastrointestinal nematode (GIN) parasite control recommendations are in a state of flux because of the increase in anthelmintic resistant cattle parasites, such as Cooperia spp. In addition, Cooperia spp. infection is typically high in warm-season grass pastures and can affect growth performance of grazing stocker calves in the Gulf Coast Region. This study evaluated the effects of moxidectin pour-on, oxfendazole oral suspension, or a combination of the two given at separate times on infection and performance of weaned beef calves grazing summer forages. Steers (n=42) and heifers (n=31) were s

Genetic markers for strongylid nematodes of livestock defined by PCR-based restriction analysis of spacer rDNA.

Twenty-four species of parasitic nematode (order Strongylida) from sheep, goats, cattle or pigs were characterised using a polymerase chain reaction-linked restriction fragment length polymorphism technique (PCR-RFLP). The ribosomal (r)DNA region spanning the first internal transcribed spacer (ITS-1), 5.8S rRNA gene and the second internal transcribed spacer (ITS-2) (designated ITS) was amplified from genomic DNA by polymerase chain reaction (PCR), digested separately with four restriction endonucleases (RsaI, HinfI, DraI or NlaIII) and the fragments separated by agarose gel electrophoresis. The efficacy of an ivermectin/closantel injection against experimentally induced infections and field infections with gastrointestinal nematodes and liver fluke in cattle.

Three studies were performed to test the efficacy of an ivermectin/closantel injection (200 microg/kg(-1) ivermectin and 5 mg/kg(-1) closantel) in cattle. Two were experimentally induced infections of Ostertagia ostertagi, Cooperia oncophora and Fasciola hepatica in calves, and the third had natural field infections in cattle with several species of gastrointestinal nematodes and F. hepatica. In the two studies with artificial infections, four groups of 8 calves were used. All calves were infected with metacercariae on Day 0. Infection with the nematodes took place on Day 33 in groups 1 and 2

Widespread resistance to macrocyclic lactones in cattle nematodes in Ecuador.

The aim of the present study was to assess the resistance status of bovine gastrointestinal nematodes (GINs) against ivermectin (IVM) and fenbendazole (FBZ) in Ecuador. The study involved five cattle farms located in different topographic zones of the country. Anthelmintic efficacy was assessed by calculating the percentage of fecal egg counts reduction (FECR) after treatment. Additionally, DNA from pooled larval cultures was screened to ascertain benzimidazole resistance alleles. For animals treated with IVM, FECR percentages ranged from 0 to 68%, indicating the presence of highly resistant w
