Gallic acid ameliorates calcium oxalate crystal-induced renal injury via upregulation of Nrf2/HO-1 in the mouse model of stone formation.

BACKGROUND: High prevalence and reoccurrence rate of nephrolithiasis bring about serious socioeconomic and healthcare burden, necessitating the need of effective therapeutic agents. Previous study revealed that gallic acid (GAL) alters the nucleation pathway of calcium oxalate (CaOx). On the other hand, it appears protective role against oxidative injury. Whether GAL could protect against crystal-induced lesion in vivo, and its underlying mechanism is yet unsolved. PURPOSE: This study aims to investigate the protective effects of GAL on the crystal-induced renal injury and its underlying mech

Observation and comparison of gas formation during holmium:YAG laser lithotripsy of cystine, uric acid, and calcium oxalate stones: a chromatographic and electron microscopic analysis.

The primary aim of the present in vitro study is to analyze the chemical content of the bubbles occurring during the fragmentation of cystine stones with both the high-power and low-power holmium:YAG (Ho:YAG) lasers. The secondary aim is to discuss their clinical importance. Three types of human renal calculi calcium oxalate monohydrate (COM), cystine, and uric acid were fragmented with both low-power and high-power Ho:YAG lasers in separate experimental setups at room temperature, during which time it was observed whether gas was produced. After laser lithotripsy, a cloudy white gas was obtai

The comparative effects of 1,2-dibromo-3-chloropropane (DBCP) and its metabolites, 3-chloro-1,2-propaneoxide (epichlorohydrin), 3-chloro-1,2-propanediol (alphachlorohydrin), and oxalic acid, on the urogenital system of male rats.

Reported similarities in the acute toxic effects of 1,2-dibromo-3-chloropropane (DBCP), 3-chloro-1,2-propaneoxide (epichlorohydrin, ECH), 3-chloro-1,2-propanediol (alphachlorohydrin, ACH), and oxalic acid (OA) have been suggested as presumptive evidence that the metabolism of DBCP to OA, via ECH and ACH, is the cause of the resulting injuries to the kidney and, perhaps, to the epididymis and testis. To test this hypothesis, the comparative toxicities of these four chemicals were studied in male rats after single subcutaneous (sc) injections of maximally tolerated (nonlethal) doses. Kidney, tes

SIRT5-Mediated Desuccinylation of ACAA2 Protects Against Calcium Oxalate-Induced Kidney Injury by Regulating Fatty Acid Oxidation to Suppress Mitochondrial Oxidative Injury and Ferroptosis.

Renal tubular epithelial cells (RTECs) are highly metabolically active and vulnerable to pathological stress. Calcium oxalate (CaOx) crystals disrupt lipid metabolism and damage RTECs, thereby promoting nephrolithiasis. We found that SIRT5, a highly expressed desuccinylase in RTECs, was markedly reduced in kidneys from patients with CaOx stones and in mouse models. Multi-omics analyses integrating succinylomics, lipidomics, and proteomics showed that SIRT5 maintains lipid metabolic homeostasis by desuccinylating key fatty acid oxidation (FAO) enzymes. SIRT5 deficiency impaired FAO, causing pol

Chlorogenic acid mitigates calcium oxalate monohydrate crystal-induced epithelial-to-mesenchymal transition in renal epithelial cells via suppression of HSP60 expression and p38 signaling.

BACKGROUND: Epithelial-to-mesenchymal transition (EMT) is a prominent process involved in kidney fibrosis. Calcium oxalate monohydrate (COM), a crystalline composition predominantly found in renal stones, is one of the known stimuli that can trigger the EMT process. Chlorogenic acid (CGA), a phenolic hydroxycinnamate naturally found in several plants (especially coffee beans), has several health benefits, including antifibrotic properties. However, the mechanisms underlying its preventive effects against COM-induced EMT in renal epithelial cells remain unclear. METHODS: Herein, Madin-Darby ca

Oxalate-degrading activity in Bifidobacterium animalis subsp. lactis: impact of acidic conditions on the transcriptional levels of the oxalyl coenzyme A (CoA) decarboxylase and formyl-CoA transferase genes.

Oxalic acid occurs extensively in nature and plays diverse roles, especially in pathological processes. Due to its highly oxidizing effects, hyperabsorption or abnormal synthesis of oxalate can cause serious acute disorders in mammals and can be lethal in extreme cases. Intestinal oxalate-degrading bacteria could therefore be pivotal in maintaining oxalate homeostasis and reducing the risk of kidney stone development. In this study, the oxalate-degrading activities of 14 bifidobacterial strains were measured by a capillary electrophoresis technique. The oxc gene, encoding oxalyl-coenzyme A (Co

An electrochemiluminescent biosensor for uric acid based on the electrochemiluminescence of bis-[3,4,6-trichloro-2-(pentyloxycarbonyl)-phenyl] oxalate on an ITO electrode modified by an electropolymerized nickel phthalocyanine film.

Nickel(ii) tetrasulfophthalo-cyanine (NiTSPc) could be electrodeposited onto an indium tin oxide (ITO) electrode to form an electropolymerization film of NiTSPc. The electrochemiluminescent (ECL) behavior of bis-[3,4,6-trichloro-2-(pentyloxy-carbonyl)-phenyl] oxalate (BTPPO) on this modified electrode was investigated in detail. The emission of ECL of BTPPO can be greatly enhanced by hydrogen peroxide at this modified electrode. Thus, a new ECL biosensor for uric acid was developed based on the enzymatic reaction of uric acid in the presence of uricase to produce H(2)O(2). The developed sensor

Investigation of gaseous end products produced by thulium fiber laser lithotripsy of cystine, uric acid, and calcium oxalate monohydrate stones: A gas chromatographic and electron microscopic analysis.

Laser lithotripsy mechanisms can cause the chemical decomposition of stone components and the emergence of different end products. However, the potentially toxic end products formed during thulium fiber laser (TFL) lithotripsy of cystine stones have not been sufficiently investigated. The aim of our in vitro study is to analyze the chemical content of the gas products formed during the fragmentation of cystine stone with TFL. Human renal calculi consisting of 100% pure cystine, calcium oxalate monohydrate, or uric acid were fragmented separately with TFL in experimental setups and observed for

Trace metal-citric acid complexes as inhibitors of calcification and crystal growth. II. Effects of Fe(III), Cr(III) and Al(III) complexes on calcium oxalate crystal growth.

The high molecular weight Fe(III)-citric acid complex, formed at low ratios of citrate:iron in solution, has been shown to be an effective inhibitor of calcium oxalate crystal growth. This appears to be a specific reaction, since 2 other similar metal ions. Al(III) and Cr(III), exhibit no synergism with citric acid in this system. Because the low molecular weight Fe(III)-citric acid complex, formed at high ratios of citrate:iron, inhibits calcium phosphate precipitation but has no effect on calcium oxalate, the Fe(III)-citric acid system might have a selective biologic role in regulating the g

Improvement of chemical analysis of antibiotics. V. A simple method for the analysis of tetracyclines using reversed-phase high-performance liquid chromatography.

An isocratic high-performance liquid chromatographic method for the determination of tetracyclines is described using a mobile phase containing oxalic acid and C8- and C18-modified silica gel columns. For good separations of tetracyclines, oxalic acid concentrations of above 0.01 and 0.2 M respectively for parent tetracyclines (group I) and impurities in tetracycline (group II) are required. The optimum pH of the aqueous oxalic acids solution in the mobile phase is 2.0. The combinations of the C8-modified silica gel column with methanol-acetonitrile-0.01 M aqueous oxalic acid solution pH 2.0 (

Oxidative photodegradation of herbicide fenuron in aqueous solution by natural iron oxide α-Fe2O3, influence of polycarboxylic acids.

The photodegradation of the herbicide fenuron (1,1-dimethyl-3-phenylurea) by using a natural iron oxide (NIO), α-Fe2O3, in aqueous solution at acidic pH has been undertaken. The NIO was characterized by the Raman spectroscopy method. The degradation pathways and the formation of degradation products were studied. A high-pressure mercury lamp and sunlight were employed as light source. Fenuron photodegradation using NIO with oxalic acid followed the pseudo-first-order kinetics, the optimal experimental conditions were [oxalic acid]0 = 10(-3) M and [NIO] = 0.1 g L(-1) at pH 3. Organic acids inhibit the formation of pyromorphite and Zn-phosphate in phosphorous amended Pb- and Zn-contaminated soil.

Pyromorphite (PY) and some zinc phosphates (Zn-P) are very sparingly soluble minerals and hence can immobilize Pb and Zn in contaminated soils. However, mechanisms leading to the poor efficiency of PY and Zn-P formation in contaminated soils amended with P still remain unclear. We studied the influence of two low molecular weight organic acids (LMWOA) - oxalic acid and citric acid and diethylene triamine pentaacetic acid (DTPA) - in PY and Zn-P formation in a P-amended contaminated soil. Despite the high levels of metals (∼4% Pb and 21% Zn) in the study soil, the addition of up to 1% inorganic

A study of P release from Fe-P and Ca-P via the organic acids secreted by Aspergillus niger.

Phosphate solubilizing fungi (PSF) have been widely applied to dissolve insoluble phosphates (IPs). However, the PSF usually demonstrates a different phosphate solubilizing capacity for various IPs. This study explored the mechanisms of Aspergillus niger for the dissolution of ferric phosphate (FePO4, Fe-P), and tricalcium phosphate (Ca3[PO4]2, Ca-P) regarding the tricarboxylic acid (TCA) cycle. Aspergillus niger has higher phosphorus (P) content released from Ca-P, reached the maximum value of 861 mg/L after seven days of incubation, compared with the 169 mg/L from Fe-P. Oxalic acid promoted

Effect of succinimide on hyperoxaluria in the rat estimated value of the different dosing methods of oxaluria.

In the female rat intoxicated with ethylene glycol the oxaluria increases with the degree of intoxication. The increase is less in the animals treated with succinimide. The comparative study of the results of the dosages made with gas-liquid-chromatography and by various colorimetric methods show that this later gives varying results and underestimates high concentrations of oxalic acid. The result is that any study based on results of dosages of urinary oxalic acid made by colorimetry must be taken with some reserve, and this on whether the oxalic lithiasis is experimentally induced or human,

Bioleaching of nickel and cobalt from lateritic chromite overburden using the culture filtrate of Aspergillus niger.

Extraction of metals (Ni, Co) from chromite overburden of Sukinda mines of Orissa, India, with the culture filtrate of Aspergillus niger was studied. Results showed that the amounts of metals leached varied directly with reaction temperature and period of fermentation. The culture filtrate was analyzed for citric and oxalic acids, and contained only oxalic acid-the concentration of which increased with time. Although this acid played the major role in leaching of metals, other unidentified metabolites present in the culture filtrate influenced the dissolution of the metals significantly. Maxim

Determination of tetracycline residues in animal tissues by liquid chromatography.

A simple liquid chromatographic (LC) method was developed for the determination of tetracyclines (oxy-tetracycline, tetracycline and chlortetracycline) in animal tissues. Isolation of tetracyclines from biological matrices was performed with oxalic buffer followed by dechelation and deproteinization with oxalic acid-acetonitrile solution. For clean-up solid phase extraction with a SDBI (styrene-divinylbenzene) cartridge was used. LC analysis was performed on a polymeric analytical column (PLRP-S 5 microns, 150 x 4.6 mm) and using an oxalic acid mobile phase (0.01 M oxalic acid - acetonitrile 7
