Metabolomics technology combined with O2PLS analysis reveals the active components of Paeoniae Radix Alba in preventing renal damage in rheumatoid arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by symmetrical, erosive polyarthritis and multi-organ involvement, with renal complications posing a life-threatening risk. Paeoniae Radix Alba, known as BaiShao (BS) in Chinese, is a widely used herbal medicine for clinical RA treatment, yet its potential in preventing RA-associated renal damage and its underlying active ingredients remain elusive. To address this knowledge gap, we employed a type II collagen-induced arthritis (CIA) rat model. Biochemical assays and histopathological analyses confirmed that BS exerted ro

Bioactive metabolites from Paeonia lactiflora protect against heat-induced male infertility in Drosophila melanogaster by modulating Vasa: integrating in vivo and computational analyses.

This study investigates the potential of isolated compounds from Paeonia lactiflora to mitigate heat stress-induced male infertility in Drosophila melanogaster, with egg-hatching rates as a quantitative fertility indicator. Exposure to thermal stress (27.5 °C) significantly impaired male fertility, resulting in egg viability declining to 16.18-23.08%. Supplementation with 10 µM of paeoniflorin (1), benzoic acid (2), and albiflorin (4) significantly restored egg-hatching rates to 55.17-93.48%, demonstrating protective effects against heat stress-induced reproductive impairment. Total glucosides of paeony (Paeonia lactiflora) alleviates blood-brain barrier disruption and cerebral ischemia/reperfusion injury in rats via suppressing inflammation and apoptosis.

At present it remains uncertain as to whether total glucosides of paeony (TGP) are able to mediate neuroprotection in the context of cerebral ischemia/reperfusion (CIR) injury, and if so, what mechanisms underlie such protection. We employed a rat model of middle cerebral artery occlusion and reperfusion, and then Evans blue (EB), hematoxylin and eosin, Nissl staining, TUNEL staining, ELISAs and immunohistochemistry were used. We observed marked reductions in infarct volume, neurological deficits, and CIR-associated histopathological changes following TGP treatment. We further found that TGP w

Wine processing as a chemical driver for reprogramming the metabolomic profile and color phenotype of Radix Paeoniae Rubra: an integrative molecular networking and thermochemical study.

Wine processing is a long-established strategy in herbal food manufacturing, yet the molecular rules by which rice wine reshapes complex herbal matrices and translates chemical remodeling into visible quality traits remain poorly defined. Here, Radix Paeoniae Rubra (Rpr), a representative medicinal-edible herb, was used to construct an integrated "processing-metabolism-phenotype-mechanism" framework by combining a four-factor, multi-level processing design with color phenotyping, UPLC-Q-TOF-MS-based metabolomics, multivariate statistics, machine-learning-assisted feature selection, feature-bas
