Rice bran extract attenuates cognitive impairment by enhancing pancreatic β-cell insulin secretion in STZ-induced diabetic rats targeting the PPARγ/PDX1 pathway.

Type I diabetes (T1D), also known as juvenile diabetes, is an autoimmune disease that causes gradual destruction of pancreatic cells and leads to intellectual disability, neuropathy, cognitive impairment, and impaired learning ability in children. Despite standard treatment with synthetic human insulin, T1D patients can maintain up to 40% of their insulin-producing islets. PPARγ receptor activation research that aims to restore β-cell biology could help reverse the loss of pancreatic mass that comes with getting older and improve β-cell function. Palliation of pain in chronic pancreatitis. Use of enzymes.

According to the concept of negative feedback regulation of pancreatic enzyme secretion by proteases, treatment with pancreatic extracts has been proposed to lower pain in chronic pancreatitis by decreasing pancreatic duct pressure. The author, however, has demonstrated in healthy volunteers that intraduodenal application of porcine pancreatic extracts does not inhibit but rather stimulates pancreatic enzyme secretion. This is probably because of the high-protein content of porcine pancreatic extracts that may overwhelm a potential inhibitory effect of proteases. In a prospective placebo-contr

Syzygium jambos extract mitigates pancreatic oxidative stress, inflammation and apoptosis and modulates hepatic IRS-2/AKT/GLUT4 signaling pathway in streptozotocin-induced diabetic rats.

The protective effect of Syzygium jambos (SJ) bark extract against streptozotocin-induced diabetes was tested in rats. Animals were treated with 100 or 200 mg/kg of the extract or glibenclamide, 0.5 mg/kg per os, once daily: started 2 days before streptozotocin (STZ) injection and lasted for 14 days after STZ injection. The effect of the extract was also evaluated on normal rats in comparison with glibenclamide. Diabetic animals showed an elevated blood glucose level, positive glycosuria, elevated fructosamine, pancreatic malondialdehyde, pancreatic TNF-a, and pancreatic caspase-3 levels and d

Exploring aryl hydrocarbon receptor (AhR) as a target for Bisphenol-A (BPA)-induced pancreatic islet toxicity and impaired glucose homeostasis: Protective efficacy of ethanol extract of Centella asiatica.

The estrogenic impact of Bisphenol-A (BPA), a widely recognized endocrine disruptor, causes disruption of pancreatic β-cell function through estrogen receptors (ERs). While BPA's binding affinity for ERs is significantly lower than that of its natural counterpart, estrogen, recent observations of BPA's affinity for aryl hydrocarbon receptor (AhR) in specific cellular contexts have sparked a specific question: does AhR play a role in BPA's toxicological effects within the endocrine pancreas? To explore this question, we investigated BPA's (10 and 100 μg/ kg body weight/day for 21 days) potentia

Biologically active human islet amyloid polypeptide/amylin in transgenic mice.

OBJECTIVE: Human islet amyloid polypeptide (hIAPP), also named amylin, is a pancreatic beta cell protein implicated in the pathogenesis of pancreatic islet amyloid formation and type 2 diabetes mellitus. To study the (patho)physiological roles of hIAPP, we have generated transgenic mice that overexpress hIAPP mRNA, in relation to endogenous mouse IAPP (mIAPP) mRNA, in pancreatic beta cells. The biological activity of human and mouse IAPP derived from pancreatic extracts was determined. METHODS: Pancreatic and plasma extracts of transgenic and control mice were analyzed by reversed-phase high-

Purification and characterization of a pancreas cancer-related antigen.

A pancreatic cancer-related antigen (PCRA) has been identified, isolated and characterized from liver metastases of pancreatic carcinoma. The purified PCRA was homogeneous by polyacrylamide disc gel electrophoresis, had an Mr of about 500,000, consisted of 30% carbohydrate and 70% protein, and migrated in the alpha 2-beta region in electrophoresis. In gel immunoprecipitation the antiserum against PCRA reacted with extracts of pancreatic cancer and spleen, but neither with normal human serum nor with normal pancreas, liver, lung, large and small intestine extracts. It clearly differed from othe

Chronic pancreatitis: maldigestion, intestinal ecology and intestinal inflammation.

Exocrine pancreatic insufficiency caused by chronic pancreatitis results from various factors which regulate digestion and absorption of nutrients. Pancreatic function has been extensively studied over the last 40 years, even if some aspects of secretion and gastrointestinal adaptation are not completely understood. The main clinical manifestations of exocrine pancreatic insufficiency are fat malabsorption, known as steatorrhea, which consists of fecal excretion of more than 6 g of fat per day, weight loss, abdominal discomfort and abdominal swelling sensation. Fat malabsorption also results i

Chicoric acid-rich extract of Cichorium intybus seeds (CAREOCI) preserves pancreatic β-cell function under glucotoxic stress through modulation of MAFA, DYRK1A, cell cycle, apoptosis, and mitochondrial dysfunction.

INTRODUCTION: Progressive decline in functional pancreatic β-cell mass is a key pathological feature of type 2 diabetes mellitus (T2DM), highlighting the need for therapies that preserve and regenerate β-cells. This study investigated the preservative effects of a chicoric acid-rich aqueous extract of Cichorium intybus seeds (CAREOCI) under glucotoxic conditions (25 mM glucose) in pancreatic β-cells. METHODS: Phytochemical standardisation by HPTLC, HPLC, and LC-MS/MS confirmed chicoric acid is the principal constituent present in CAREOCI. A molecular docking study identified DYRK1A is a one o

Magnetic resonance imaging in the assessment of pancreatic cancer with quantitative parameter extraction by means of dynamic contrast-enhanced magnetic resonance imaging, diffusion kurtosis imaging and intravoxel incoherent motion diffusion-weighted imaging.

BACKGROUND: Despite great technical advances in imaging, such as multidetector computed tomography and magnetic resonance imaging (MRI), diagnosing pancreatic solid lesions correctly remains challenging, due to overlapping imaging features with benign lesions. We wanted to evaluate functional MRI to differentiate pancreatic tumors, peritumoral inflammatory tissue, and normal pancreatic parenchyma by means of dynamic contrast-enhanced MRI (DCE-MRI)-, diffusion kurtosis imaging (DKI)-, and intravoxel incoherent motion model (IVIM) diffusion-weighted imaging (DWI)-derived parameters. METHODS: We

Sequential extracts of red honeybush (Cyclopia genistoides) tea: Chemical characterization, antioxidant potentials, and anti-hyperglycemic activities.

The antioxidant, antidiabetic, and anti-obesogenic potentials of different extracts (dichloromethane, ethyl acetate, ethanol, and aqueous) of the red honeybush (Cyclopia genistoides) tea were investigated in vitro and ex vivo. All extracts exhibited significant scavenging and reducing power activities, with the aqueous and ethyl acetate extracts being the most potent. In vitro antidiabetic analysis revealed the extracts to be potent inhibitors of α-glucosidase and lipase activities. All extracts increased catalase and SOD activities, and glutathione level in oxidative pancreatic injury. GC-MS

Immunoreactive adrenocorticotropin in the gastrointestinal tract and pancreatic islets of the rat.

Radioimmunoassayable ACTH is detectable in the gastrointestinal tract and pancreatic islets of the rat by using a midportion ACTH antiserum. Portions of the gastrointestinal tract and isolated pancreatic islets were extracted with 0.1 N HCl. Serial dilutions of the extracts resulted in inhibition curves parallel with human alpha ACTH-(1-39). The highest concentrations were in the isolated pancreatic islets (20 pg/ml protein) and the gastric antrum-pylorus (17.9 +/- 1.2 pg/ml protein). Chromatographic characterization of the stomach extract showed a main peak, with an elution constant identical
