Prevalence of fish parasites from Lake Hawassa, Ethiopia.

BACKGROUND: An estimated 200 species of fishes of marine and freshwater fish are known to occur in Ethiopia. Fish parasites are the main cause of the significant threat to the fish biodiversity of Ethiopia's freshwater islands. Lake Hawassa plays a central role in the local economy of the city which supports the artisanal fishing sector. The parasite of the fishes at lake Hawassa are hitherto poorly known and there are limited reports on the type of parasites across different fish hosts such as size, age, sex, condition factor, or other biological or ecological traits. METHODS: A cross-sectio

Effects of aging on resistance to Trichinella spiralis infection in rodents exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin.

Immune function, including resistance to infection, decreases as humans and rodents age. We have shown that preinfection exposure of young (9-11 weeks) mice or rats to TCDD decreased resistance to Trichinella spiralis (Ts) infection, expressed as delayed onset or completion of parasite elimination and as increased muscle burdens of larvae. It has also been shown that aged mice express lower constitutive levels of resistance to Ts infection, compared to young adult animals. This study tested the hypothesis that the age-related decrease in constitutive levels of resistance to Ts infection exacer

A nucleotide sugar transporter involved in glycosylation of the Toxoplasma tissue cyst wall is required for efficient persistence of bradyzoites.

Toxoplasma gondii is an intracellular parasite that transitions from acute infection to a chronic infective state in its intermediate host via encystation, which enables the parasite to evade immune detection and clearance. It is widely accepted that the tissue cyst perimeter is highly and specifically decorated with glycan modifications; however, the role of these modifications in the establishment and persistence of chronic infection has not been investigated. Here we identify and biochemically and biologically characterize a Toxoplasma nucleotide-sugar transporter (TgNST1) that is required

Host resistance to Trichinella spiralis infection in rats exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD).

We have previously shown decreased resistance to Trichinella spiralis (Ts) infection and reduced parasite antigen-specific responses in B6C3F1 mice exposed to TCDD before infection. The current study was done to characterize the effects of preinfection administration of 1, 10, or 30 micrograms TCDD/kg on host resistance of female F344 rats to Ts infection and to examine parasite antigen-specific responses in the spleen and mesenteric lymph nodes of infected animals. TCDD exposure did not affect adult parasite elimination from the small intestine or the numbers of encysted larvae in the muscle,

Toxoplasmic myositis as a presenting manifestation of idiopathic CD4 lymphocytopenia.

Toxoplasma gondii encysts in skeletal muscle. Although only rarely found at muscle biopsy, this parasite has previously been regarded as a possible cause of polymyositis. We report a case of biopsy-proven toxoplasmic myositis in a non-HIV-infected patient that led to recognition of idiopathic CD4 lymphocytopenia (ICL), a rare condition typically associated with opportunistic infections. Interestingly, the CD25(+) subset that corresponds to the CD4(+) regulatory T cells controlling autoimmune processes was lacking. Steroid and antiprotozoal therapy led to recovery.

Disseminated sarcocystosis in a cat with lymphosarcoma.

Cysts of the protozoan Sarcocystis sp were found in skeletal and cardiac musculature in a 1.5-year-old cat with lymphosarcoma. The cat was FeLV-positive and had grossly visible neoplastic involvement of the spinal cord, mediastinum, bone marrow, and kidneys. Ultrastructural examination of the parasitic cyst wall suggested that the species in this case was different from that described in the only other reported case. It was hypothesized that immunosuppression from FeLV infection permitted an aberrant life cycle with encystment of Sarcocystis sp in this cat.

Dynamics of amylopectin granule accumulation during the course of the chronic Toxoplasma infection is linked to intra-cyst bradyzoite replication.

The contribution of amylopectin granules (AG), comprised of a branched chain storage homopolymer of glucose, to the maintenance and progression of the chronic Toxoplasma gondii infection has remained undefined. Here we describe the role of AG in the physiology of encysted bradyzoites by using a custom developed imaging-based application AmyloQuant that permitted quantification of relative levels of AG within in vivo derived tissue cysts during the initiation and maturation of the chronic infection. Our findings establish that AG are dynamic entities, exhibiting considerable heterogeneity among

Isolated Myocysticercosis.

Cysticercosis, a parasitic disease caused by larval form of Taenia solium, is a major health concern in the developing world. The encysted larval stage can affect any part of the body, but are most frequently detected in brain, eye, skeletal muscle and subcutaneous tissue. Most muscular cysticercosis are almost always associated with central nervous system involvement or with multiple intramuscular cysts or both. Here we report an unusual case of cysticercosis in right rectus muscle which was an isolated muscle involvement without any other systemic manifestation.

Intravitreal dirofilariasis: a rare ocular infection.

Human ocular dirofilariasis is a zoonotic disease, rare in Europe, caused by filarial nematodes. The parasite is either encysted in a subcutaneous nodule or located under the bulbar conjunctiva. We report the case of a 62-year-old man with intravitreal dirofilariasis, which is a rare site of presentation of the nematode in the human eye. It was located in the fundus area and was surgically removed. The nematode was identified as Dirofilaria repens (D. conjuctiva) by two different Microbiology Departments, making this the fifth report of identified intravitreal dirofilariasis caused by D. repen

Sirtuin GdSir2.4 participates in the regulation of rRNA transcription in the Giardia duodenalis parasite.

Giardia duodenalis is a parasite of great medical interest due to the number of infections it causes worldwide each year. Although research on epigenetic mechanisms in this protist has only begun recently, epigenetic regulation has already been shown to have important roles in encystation, antigenic variation, and resistance to antibiotics in Giardia. In this work, we show that a Giardia ortholog of Sir2, GdSir2.4, is involved in the silencing of rRNA expression. Our results demonstrate that GdSir2.4 localizes to the nucleolus, and its binding to the intergenic spacer region of the rDNA is ass

Modeling effective transmission pathways and control of the world's most successful parasite.

Toxoplasma gondii(T. gondii) is a single-celled, intracellular protozoan responsible for the disease toxoplasmosis. The parasite is prevalent worldwide, and it infects all warm-blooded vertebrates. Consumption of meats in which this parasite has encysted confers risk of infection to people and other animals, as does ingestion of water or foods contaminated with environmentally resistant oocysts excreted by cats. Vertical transmission (from mother to offspring) is also possible, leading to disease risk and contributing additional means of ensuring perpetuation of transmission. In this work, we

Recent advances in the genomic and molecular biology of Giardia.

Giardia duodenalis is the most common gastrointestinal protozoan parasite of humans and a significant contributor to the global burden of both diarrheal disease and post-infectious chronic disorders. Robust tools for analyzing gene function in this parasite have been developed and a range of genetic tools are now available. These together with public databases have provided insights on the function of different genes in Giardia. In this review we provide a current perspective on different molecular aspects of Giardia related to genomics, regulation of encystation, trophozoite transcriptional r

Encysted cyathostomin larval counts: Mucosal digestion revisited.

Cyathostomins are pervasive equine parasites in horses across the world, and larval stages are known to cause the deadly disease larval cyathostominosis. The mucosal digestion technique is widely used for enumeration of encysted larval stages. Previous studies have investigated the spatial variation of encysted larvae, however current protocols lack a description of a standardized area from which to take the tissue sample. This study sought to evaluate spatial variation in encysted cyathostomin larval counts among the large intestinal organs and their subsections. Following humane euthanasia,

Incidental coenurosis: larval cestode presenting as an axillary mass.

This article describes the occurrence of a coenurus, the encysted larval stage of a cestode of the genus Multiceps, within the axilla of a 40-year-old Nigerian woman. The patient had a history of intraductal carcinoma of the breast two years prior to the finding of an ipsilateral axillary mass. Pathologic evaluation of the biopsy specimen yielded the diagnosis of coenurosis. Only four other cases of human Multiceps infestation have been reported in the United States with fewer than 100 validated cases worldwide. Surgical removal of the larval parasite is the only known effective therapy.

Albendazole versus ricobendazole (albendazole-sulphoxide) against enteral and parenteral stages of Trichinella spiralis in mice.

Comparison of the anthelmintic activity and pharmacokinetic profiles following albendazole (ABZ) and albendazole-sulphoxide (ricobendazole = RBZ) administration was made in a mouse model for helminthic infections. Swiss CD-1 mice were experimentally infected with Trichinella spiralis and treated with either ABZ or RBZ at 3 different stages of the parasite life-cycle: pre-adult (day 1 p.i.), migrating larvae (days 13, 14 and 15 p.i.) and encysted muscle larvae (days 34, 35 and 36 p.i.). Plasma concentrations of albendazole-sulphoxide (ABZSO) were measured in age matched non-infected mice by hig

Differentiation of Toxoplasma into latent forms is linked to central carbon metabolism and requires a GID/CTLH-type E3 ubiquitin ligase.

Toxoplasma and other Apicomplexan parasites, switch between different developmental stages to persist in and transmit between hosts. Toxoplasma can alternate between systemic tachyzoites and encysted bradyzoite forms found in the CNS and muscle tissues. How parasites sense these tissue types and trigger differentiation remains largely unknown. We show that Toxoplasma differentiation is induced under glucose-limiting conditions and using a CRISPR screen identify parasite genes required for growth under these conditions. From ∼25 identified genes important for differentiation we show that lactat

Characterization of proteolytic activities of Giardia lamblia with the ability to cleave His-tagged N-terminal sequences.

Giardia lamblia is one of the most common protozoan infectious agents in the world and is responsible for diarrheal disease and chronic postinfectious illness. During the host-parasite interaction, proteases are important molecules related to virulence, invasion, and colonization, not only for Giardia but also for other parasites. We aimed to characterize the cysteine protease activity detected in trophozoite lysates. This proteolytic activity showed the ability to cleave NH-terminal sequences with either a recognition sequence for a viral protease or a recognition sequence for thrombin. This

First Report of Clinostomum complanatum (Trematoda: Digenea) in European Perch (Perca fluviatilis) from an Italian Subalpine Lake: A Risk for Public Health?

Clinostomum complanatum, a digenean trematode of the Clinostomidae family, is a fish-borne zoonotic parasite responsible for Halzoun syndrome in humans and is transmitted through the consumption of raw or undercooked freshwater fish. Of the total of 112 specimens of European perch (Perca fluviatilis) sampled from a subalpine lake (Lake Endine) in North Italy in 2019, 21 (18.75%) tested positive for encysted metacercariae in the fillet. This study reports the first isolation of C. complanatum in P. fluviatilis and highlights the possible zoonotic risk for consumers, since P. fluviatilis is a fo

Evolutionarily Diverged TRAPP Components in Giardia lamblia That Lacks a Classical Golgi.

The endomembrane system of the intestinal pathogen Giardia lamblia lacks a separate Golgi compartment. Without this sorting compartment, how cargo sorting to various subcellular destinations occurs within Giardia remains an open question. While the distribution of various Golgi-associated SNAREs and Rabs has been documented in this parasite, the TRAPP (TRAnsport Protein Particle) complex, a guanine nucleotide exchange factor for Golgi-associated Rabs, remained uncharacterized. Herein, we report that Giardia expresses a minimal set of TRAPP complex components, GlBet3, GlBet5, GlTrs23, and GlTrs
