Neuronal pentraxin 1: a novel mediator of hypoxic-ischemic injury in neonatal brain.

Neonatal hypoxic-ischemic brain injury is a major cause of neurological disability and mortality. Its therapy will likely require a greater understanding of the discrete neurotoxic molecular mechanism(s) triggered by hypoxia-ischemia (HI). Here, we investigated the role of neuronal pentraxin 1 (NP1), a member of a newly recognized subfamily of "long pentraxins," in the HI injury cascade. Neonatal brains developed marked infarcts in the ipsilateral cerebral hemisphere at 24 hr and showed significant loss of ipsilateral striatal, cortical, and hippocampal volumes at 7 d after HI compared with th

Neuronal pentraxin, a secreted protein with homology to acute phase proteins of the immune system.

We have identified, by affinity chromatography, a binding protein for the snake venom toxin taipoxin. The sequence of this 47 kDa protein is unique, is characteristic of a secreted protein, and has homology to the acute phase proteins serum amyloid P protein and C-reactive protein of the pentraxin family. We have named this protein neuronal pentraxin (NP), as Northern analysis and in situ hybridization demonstrate high message levels in neurons of cerebellum, hippocampus, and cerebral cortex. Because NP may be released synaptically and has homology to immune proteins potentially involved in up

Human neuronal pentraxin II (NPTX2): conservation, genomic structure, and chromosomal localization.

We have previously identified a novel rat neuronal member of the pentraxin family (neuronal pentraxin) that may mediate the uptake of synaptic material and the presynaptic snake venom toxin, taipoxin. Here we report human cDNA and genomic sequences of a second neuronal pentraxin. This pentraxin, which we propose to name neuronal pentraxin II (NPII; gene symbol NPTX2), shows 54% amino acid identity to rat neuronal pentraxin (NPI) with 69% identity over the carboxyl-terminal half of NPI and is 88% identical to a newly identified sperm acrosomal pentraxin p50/apexin. Northern blot analysis reveal

Pentraxin-3, a complementary biomarker in hepatocellular carcinoma: Systematic review and Meta-analysis.

BACKGROUND: Alpha-fetoprotein (AFP) is the most widely used biomarker for hepatocellular carcinoma (HCC) diagnosis; however, its sensitivity is limited, especially for early-stage disease and in AFP-negative HCC cases. Pentraxin-3 (PTX3), a marker of local inflammation and angiogenesis, may outperform AFP in detection by reflecting distinct biological pathways involved in hepatocarcinogenesis. METHODS: PubMed, Embase, Web of Science, Cochrane, and ClinicalTrials.gov were searched through December 2025 following PRISMA guidelines. Eligible studies included adults with chronic liver disease eva

Targeting of Pentraxin 3 Alleviated Cardiac Hypertrophy in Mice Induced by Angiotensin II.

Pentraxin 3 (PTX3) responds rapidly to inflammatory signals. This study investigated the functional involvement of PTX3 in angiotensin II (Ang II)-mediated cardiac hypertrophy pathogenesis, with particular focus on its potential regulation through inflammatory pathways and oxidative stress mechanisms. In vivo, chronic Ang II infusion over 4 weeks was used to induce hypertrophic cardiomyopathy in mice. In parallel, an in vitro model of cardiomyocyte hypertrophy was established by treating neonatal rat cardiomyocytes (NRCMs) with Ang II for 24 h. Ang II treatment significantly increased PTX3 exp

The long pentraxin 3: the invisible loom weaving the warp and weft of tumor destiny.

Pentraxins, which constitute a family of evolutionarily conserved pattern recognition molecules, are categorized into short and long branches. The long pentraxin 3 (PTX3) is a key member of the long pentraxin subfamily, while the C-reactive protein and serum amyloid P represent the short pentraxins. All pentraxins share a highly conserved C-terminal motif, an 8-amino acid sequence known as the pentraxin signature. PTX3 can be produced by a wide range of cell types, including immune cells such as dendritic cells, monocytes, and macrophages, as well as various non-immune cells, underscoring its

Serum pentraxin-3 expression varies according to polycystic ovary syndrome phenotypes.

Pentraxin-3 (PTX-3) is a multibiological protein involved in cumulus cell expansion, fertilization, and implantation. This study was designed to analyze how circulating PTX-3 levels change in women with polycystic ovary syndrome (PCOS). A total of 50 Turkish participants, 35 of whom had PCOS and 15 of whom were fertile, were included in the study. Patients in the PCOS group were divided into 4 different phenotypes according to the NIH criteria (phenotypes A-D). The number of patients in phenotype A was the highest 13 (37.1%). In the calculations made without phenotyping, the serum ptx3 levels

Analyzing the potential of neuronal pentraxin 2 as a biomarker in neurological disorders: A literature review.

Neuronal pentraxin 2 (NP2) plays a significant role in synaptic plasticity, neuronal survival, and excitatory synapse regulation. Emerging research suggests that NP2 is implicated in the pathogenesis of various neurological disorders, including neurodegenerative diseases, neuropsychiatric disorders, and neuropathies. This literature review extensively analyzes NP2's role in these conditions, thereby highlighting its contributions to synaptic dysfunction, neuroinflammation, and neurotoxic protein aggregation. In Alzheimer's and Parkinson's diseases, NP2 is linked to amyloid-beta aggregation and

Pentraxin 3 Facilitates Shrimp-Allergic Responses in IgE-Activated Mast Cells.

BACKGROUND: Since food avoidance is currently the only way to prevent allergic reactions to shrimp, a better understanding of molecular events in the induction and progression of allergy, including food allergy, is needed for developing strategies to inhibit allergic responses. Pentraxin 3 (PTX3) is rapidly produced directly from inflammatory or damaged tissues and is involved in acute immunoinflammatory responses. However, the role of PTX3 in the development of immediate IgE-mediated shrimp allergy remains unknown. METHODS: Wild-type BALB/c mice were immunized intraperitoneally and were chal

Serum pentraxin 3 concentration correlates with disease severity in patients with myasthenia gravis.

OBJECTIVE: Myasthenia gravis (MG) is an antibody-mediated inflammatory disease affecting post-synaptic membranes of neuromuscular junctions, and objective biomarkers of MG disease activity are lacking. Pentraxin 3 (PTX3) is an acute-phase inflammatory glycoprotein in the same family as C-reactive protein that is associated with disease activity in several autoimmune disorders. Thus, we investigated whether circulating PTX3 is a useful biomarker of MG activity. METHODS: Serum PTX3 was measured in 40 patients with MG who were positive for anti-acetylcholine receptor antibody, and in 30 healthy

Blockade of the pentraxin 3/CD44 interaction attenuates lung injury-induced fibrosis.

BACKGROUND: Fibrosing interstitial lung diseases (fILD) are potentially fatal with limited therapeutic options and no effective strategies to reverse fibrogenesis. Myofibroblasts are chief effector cells in fibrosis that excessively deposit collagen in the pulmonary interstitium and lead to progressive impairment of gaseous exchange. METHODS: Plasma and lung specimens from patients with fILD were applied for detecting pentraxin 3 (PTX3) abundance by ELISA and Immunohistochemistry. Masson's trichrome and Sirius red stains and hydroxyproline assay were performed for assessing collagen accumulat

Efficacy of procalcitonin and pentraxin-3 as early biomarkers for differential diagnosis of pleural effusions.

OBJECTIVES: Pleural effusion, defined as an abnormal accumulation of fluid in pleural space, can be of two types: transudative and exudative. The primary aim of the study was to assess the predictive accuracy of procalcitonin (PCT) and pentraxin-3 (PTX-3) in comparison to other biochemical markers such as C-reactive protein (CRP), and adenosine deaminase (ADA) in the differential diagnosis of pleural effusions. METHODS: A cross-sectional analytical study was conducted on patients with pleural effusion. Multiple comparisons and receiver-operating characteristics (ROC) analyses were made to eva

Pentraxin-3, Interleukin-6, and Acute Appendicitis: Biomarkers That Need Further Exploration.

Acute appendicitis is one of the most common abdominal emergencies that present in the hospital. With acute appendicitis, there is always a possibility of complications of perforation and peritonitis. Even though medicine has advanced substantially in different investigational modalities, appendicitis is confirmed clinically with the aid of a clinical approach (i.e., Alvarado Score), laboratory, and investigational modalities. However, biomarkers such as pentraxin-3 and interleukin-6 have been recently researched to assess the possibility of confirming the diagnosis of acute appendicitis in bo

Postmortem plasma pentraxin 3 is a useful marker of fatal acute coronary syndrome.

Pentraxin 3 (PTX3) is an acute-phase protein that belongs to the pentraxin superfamily. Recently, many clinical studies have demonstrated that plasma PTX3 concentrations rapidly increase in patients with the acute coronary syndrome (ACS). The aim of this study was to evaluate the forensic utility of postmortem plasma PTX3 as a marker of fatal ACS. We compared the plasma PTX3 concentration in cadavers with suspected fatal ACS to that exhibited in control cases (e.g., asphyxia and immediate death due to a fatal injury). The ACS groups included a coronary stenosis group, which exhibited apparent

Acute phase reactant, Pentraxin 3, as a novel marker for the diagnosis of rheumatoid arthritis.

INTRODUCTION: Pentraxins are a group of highly conserved acute-phase reactant proteins and play crucial role as modulators of inflammatory processes. Pentraxin 3 (PTX3) is primarily produced and released by vascular cell wall, hence, we attempt to establish the role of PTX3 as a biomarker for Rheumatoid Arthritis (RA) compared to CRP. METHODS: Thirty patients having active RA as cases and 30 osteoarthritis (OA) patients as controls were recruited. Paired serum and synovial fluid samples were analysed for concentrations of both PTX3 and CRP by using high sensitivity ELISA kit and ROC curve was

Pentraxin-3 modulates lipopolysaccharide-induced inflammatory response and attenuates liver injury.

UNLABELLED: Acute-on-chronic liver injury is characterized by an important inflammatory response frequently associated with endotoxemia. In this context, acute-phase proteins such as Pentraxin-3 (PTX3) are released; however, little is known about their role in chronic liver disease. The aim of this study was to elucidate the role of PTX3 in liver injury. The role of PTX3 was evaluated in cultured human cells, liver tissue slices, and mice with acute-on-chronic liver injury. PTX3 expression was assessed in tissue and serum samples from 54 patients with alcoholic hepatitis. PTX3 expression was u
