Four-Octyl Itaconate Alleviates Enterococcus faecalis-Induced Apical Periodontitis by Inhibiting Macrophage M1 Polarisation.

AIM: Enterococcus faecalis is strongly associated with persistent inflammation of the periapical tissue. 4-Octyl itaconate (4-OI), a derivative of itaconate that can permeate the cell membrane, has potent immunomodulatory effects on macrophage polarisation. However, the effect of 4-OI on apical periodontitis induced by the gram-positive bacterium E. faecalis has not yet been reported. This study explored whether 4-OI could alleviate E. faecalis-induced apical periodontitis by regulating macrophage polarisation. METHODOLOGY: A model of apical periodontitis was established in mice by pulp expos

[Reattachment or new attachment in apical-marginal periodontal bone loss. A clinical-histological discussion].

Loss of marginal alveolar bone of the tooth can be caused by periodontal, pulpal-periapical, or a combined pulpal and periodontal disease. The regeneration is supported by the formation of reattachment or new attachment. Reattachment is probably the usual healing process in periapical surgery of the tooth with apicomarginal bone loss caused by pulpal-periapical disease. In animal experiments (rats, dogs) ist was well documented, that the cells which repopulate the periodontal wound adjacent to the root surface determine the nature of the attachment formed.

Healing assessment of osseous defects of periapical lesions associated with failed endodontically treated teeth with use of freeze-dried bone allograft.

Freeze-dried bone allograft (FDBA) has been used extensively in periodontal surgery and in the treatment of pocket osseous defects resulting from inflammatory periodontal disease. No other endodontic investigation has used this type of graft material in the treatment of bony resorptive defects of the periapical lesions associated with failed endodontic therapy. Therefore this study was conducted with the use of FDBA, to assess healing ability of osseous defects after removal of periapical lesions associated with failed endodontically treated teeth. In all patients periapical lesions were surgi

The expression of cyclic GMP-AMP synthase in human apical periodontitis: A laboratory investigation.

AIM: As a key DNA sensor, cyclic GMP-AMP synthase (cGAS) has emerged as a major mediator of innate immunity and inflammation. Human apical periodontitis has yet to be studied for the presence of cGAS. This investigation was conducted to determine if cGAS is involved in the pathological process of human apical periodontitis. METHODOLOGY: Sixty four human periapical lesions, comprising 20 periapical granulomas and 44 radicular cysts, were employed in this investigation. Healthy gingiva (n = 6), dental pulp (n = 3), and apical papilla (n = 3) were used as control samples. The expression of cGAS

Association between Experimental Bacterial Meningitis and Periapical Lesion.

INTRODUCTION: Mortality and morbidity from bacterial meningitis in African adults is significantly higher than those in better resourced settings. At the same time, the periodontal diseases are highly prevalent and can affect up to 90% of the population. Dental caries in Uganda was recorded in 40% and 62.5% of the children and adults, respectively. We hypothesize that pneumococcal meningitis could interfere in the development of periapical lesion. The aim of this study was to evaluate periapical lesion in Wistar rats subjected to pneumococcal meningitis. MATERIALS AND METHODS: The animals wer

The role of smoking as a risk indicator for apical periodontitis and endodontic status: a cross-sectional study of a portuguese adult sample.

This study aimed to determine whether smoking acts as an independent predictor of apical periodontitis and endodontic treatment in a Portuguese adult population. This retrospective cross-sectional study was conducted in accordance with the PROBE (Preferred Reporting items for OBservational studies in Endodontics) guidelines. A total of 1841 first-time patient records were analyzed and met the inclusion criteria. Smoking status and systemic conditions were obtained from self-reported medical histories, and periapical status was assessed on panoramic radiographs using the Periapical Index (PAI).

Hypercitrullination and anti-citrullinated protein antibodies in chronic apical periodontitis, a laboratory investigation. Does autoimmunity contribute to the pathogenesis?

AIM: The presence of Gram-negative anaerobic bacteria, in particular, Porphyromonas gingivalis (P. gingivalis) in periapical granulomas predicts the generation of citrullinated proteins in the lesion. Citrullination of proteins may lead to the formation of anti-citrullinated autoantibodies (ACPA-s) initiating the formation of an autoimmune loop which may contribute to the perpetuation of inflammatory reactions and tissue damage in chronic apical periodontitis. The objective of this study was to demonstrate the formation of citrullinated proteins in chronic apical periodontitis and whether they

Источник пометил публикацию как отозванную

BACKGROUND/AIMS: Periapical periodontitis is a common oral disease caused by bacterial invasion of the tooth pulp, which usually leads to local release of pro-inflammatory cytokines and osteolytic lesion. This study is intended to examine the effect of TNF-α on BMP9-induced osteogenic differentiation of the stem cells of dental apical papilla (SCAPs). METHODS: Rat model of periapical periodontitis was established. TNF-α expression was assessed. Osteogenic markers and ectopic bone formation in iSCAPs were analyzed upon BMP9 and TNF-α treatment. RESULTS: Periapical periodontitis was successful

Apical periodontitis: preliminary assessment of microbiota by 16S rRNA high throughput amplicon target sequencing.

BACKGROUND: Apical periodontitis includes periapical granulomas and radicular cysts, which are histologically distinguished by the absence and the presence of an epithelial lining, respectively. The main cause of apical periodontitis is the bacterial colonization of the root canal space. This research aimed at assessing whether and how periapical granulomas and radicular cysts differ in terms of microbiota using high throughput amplicon target sequencing (HTS) techniques. METHODS: This study included 5 cases of Periapical Granulomas (PGs) and 5 cases of Radicular Cysts (RCs) selected on the b

Effects of melatonin against experimentally induced apical periodontitis in rats.

Apical periodontitis is an inflammatory condition resulting from microbial invasion in the root canal system, causing periapical tissue destruction and bone resorption. This study investigated melatonin's effects, known for its antioxidant and anti-inflammatory properties, on experimentally induced apical periodontitis in rats. Three groups of rats were studied: control, apical periodontitis and apical periodontitis with melatonin treatment. Proinflammatory cytokines and enzyme levels in blood serum were measured, and micro-CT analysis assessed bone resorption. Results showed significantly ele

Preexisting Periapical Inflammatory Condition Exacerbates Tooth Extraction-induced Bisphosphonate-related Osteonecrosis of the Jaw Lesions in Mice.

INTRODUCTION: Surgical interventions such as tooth extraction increase the chances of developing osteonecrosis of the jaw in patients receiving bisphosphonates (BPs) for the treatment of bone-related diseases. Tooth extraction is often performed to eliminate preexisting pathological inflammatory conditions that make the tooth unsalvageable; however, the role of such conditions on bisphosphonate-related osteonecrosis of the jaw (BRONJ) development after tooth extraction is not clearly defined. Here, we examined the effects of periapical periodontitis on tooth extraction-induced BRONJ developmen

Exploring the Influence of Genetic Single-Nucleotide Polymorphism (SNPs) on Endodontic Pathologies: A Comprehensive Review.

A considerable portion of the global population is affected by pulpitis and periapical lesions. While the impact of infections caused by various microbes and host effector molecules in pulpal and periapical diseases is widely recognized, disease susceptibility and progression are also influenced by the dynamic interaction between host genetic factors and environmental influences. Apical periodontitis occurs as an inflammatory response to microorganisms present in the root canals of infected teeth. Initially functioning as the body's defense mechanism, this response often progresses to chronic
