Enhanced HGF with increased receptor affinity and nitration-dysfunction resistance through interaction with lipoic acid trisulfide.

Myogenic stem cell activator HGF (hepatocyte growth factor) undergoes nitration of tyrosine residues (Y198, Y250) predominantly on fast-twitch fibers to lose its binding affinity to the signaling receptor c-met, in response to peroxynitrite (ONOO-) generation during aging. Here we show that HGF adopts an enhanced form with dynamically increased receptor-binding affinity and nitration-dysfunction resistance through interaction with lipoic acid trisulfide (LASSS) under physiological conditions. When evaluated after exposure to LASSS at a 1:8000 molar ratio to HGF and subsequent ultra-filtration

Visulization of peroxynitrite variation for accurate diagnosis and assessing treatment response of hepatic fibrosis using a Golgi-targetable ratiometric fluorescent probe.

Hepatic fibrosis (HF) is caused by persistent inflammation, which is closely associated with hepatic oxidative stress. Peroxynitrite (ONOO-) is significantly elevated in HF, which would be regarded as a potential biomarker for the diagnosis of HF. Research has shown that ONOO- in the Golgi apparatus can be overproduced in HF, and it can induce hepatocyte injury by triggering Golgi oxidative stress. Meanwhile, the ONOO- inhibitors could effectively relieve HF by inhibiting Golgi ONOO-, but as yet, no Golgi-targetable fluorescent probe available for diagnosis and assessing treatment response of

Low Intensity Electromagnetic Fields Act via Voltage-Gated Calcium Channel (VGCC) Activation to Cause Very Early Onset Alzheimer's Disease: 18 Distinct Types of Evidence.

Electronically generated electromagnetic fields (EMFs), including those used in wireless communication such as cell phones, Wi-Fi and smart meters, are coherent, producing very high electric and magnetic forces, which act on the voltage sensor of voltage-gated calcium channels to produce increases in intracellular calcium [Ca2+]i. The calcium hypothesis of Alzheimer's disease (AD) has shown that each of the important AD-specific and nonspecific causal elements is produced by excessive [Ca2+]i. [Ca2+]i acts in AD via excessive calcium signaling and the peroxynitrite/oxidative stress/inflammatio

Illuminating epilepsy: A deep NIR-I fluorogenic sensor reports peroxynitrite dynamics in vivo.

This work presents BDP-NIR-S-Fa, a deep NIR-I fluorogenic probe designed to report peroxynitrite (ONOO-) dynamics in epilepsy. The probe features a specific emission at 805 nm and responds rapidly and sensitively to ONOO-. It successfully visualized elevated endogenous ONOO- flux in both kainic acid-induced epileptic cells and a corresponding mouse model, demonstrating its potential as a diagnostic tool. Beyond detection, BDP-NIR-S-Fa was further applied to evaluate the therapeutic efficacy of antiepileptic drugs by monitoring the reduction of ONOO- levels in treated disease models. The probe

A dual-signal near-infrared fluorescent probe for concurrent detection of Hg²⁺ and ONOO⁻: Applications from environmental monitoring to imaging of Hg²⁺-induced oxidative stress.

As a highly toxic heavy metal, Hg²⁺ can accumulate in the human body through the food chain, causing multi-system damage, and induce the generation of peroxynitrite (ONOO⁻). The imbalance of ONOO⁻ is associated with various diseases, so it is necessary to accurately monitor both of them and the Hg²⁺-induced oxidative stress state in biological systems. Near-infrared (NIR) fluorescent probes are ideal tools, but most existing probes only detect Hg²⁺ or ONOO⁻ individually, and multifunctional probes are relatively scarce. In this study, a NIR fluorescent probe DCI-2a was developed. It uses dicya

Pepper catalase: a broad analysis of its modulation during fruit ripening and by nitric oxide.

Catalase is a major antioxidant enzyme located in plant peroxisomes that catalyzes the decomposition of H2O2. Based on our previous transcriptomic (RNA-Seq) and proteomic (iTRAQ) data at different stages of pepper (Capsicum annuum L.) fruit ripening and after exposure to nitric oxide (NO) enriched atmosphere, a broad analysis has allowed us to characterize the functioning of this enzyme. Three genes were identified, and their expression was differentially modulated during ripening and by NO gas treatment. A dissimilar behavior was observed in the protein expression of the encoded protein catal

[Research progress of metalloporphyrin against neurodegene-rative diseases].

Neurodegenerative diseases are a group of diseases caused by the degeneration and apoptosis of neurons in the brain and spinal cord, which seriously affect human ability of motion perception, memory and cognition. Peroxynitrite can cause oxidative damage in the brain by exhibiting neurotoxicity, and its excessive accumulation is closely related to neurodegenerative diseases. Therefore, effectively scavenging peroxynitrite may become a therapeutic strategy for neurodegenerative diseases. Due to their high peroxynitrite scavenging ability, water-soluble metalloporphyrins have recently attracted

Peroxynitrite is Involved in the Apoptotic Death of Cultured Cerebellar Granule Neurons Induced by Staurosporine, but not by Potassium Deprivation.

Nitric oxide (NO) regulates numerous physiological process and is the main source of reactive nitrogen species (RNS). NO promotes cell survival, but it also induces apoptotic death having been involved in the pathogenesis of several neurodegenerative diseases. NO and superoxide anion react to form peroxynitrite, which accounts for most of the deleterious effects of NO. The mechanisms by which these molecules regulate the apoptotic process are not well understood. In this study, we evaluated the role of NO and peroxynitrite in the apoptotic death of cultured cerebellar granule neurons (CGN), wh

The role of oxidative and nitrosative bursts caused by azoles and amphotericin B against the fungal pathogen Cryptococcus gattii.

OBJECTIVES: Although the most accepted mechanisms of action of amphotericin B and azoles are related to ergosterol, it is possible that these drugs have other effects on the fungal cell. In the present study, the role of endogenous reactive oxygen species (ROS) and peroxynitrite produced by azoles and amphotericin B in the fungus Cryptococcus gattii were examined. METHODS: We studied distinct parameters to evaluate the effect of oxidative and nitrosative stresses induced by these drugs in C. gattii cells: lipid peroxidation, ergosterol content, ROS and peroxynitrite production, enzymatic acti

Nitric oxide and peroxynitrite as new biomarkers for early diagnosis of autism.

Autism spectrum disorder, or autism, is a neurodevelopmental disorder of the developing child's brain with a genetic causality. It can be diagnosed at about three years after birth when it begins to present itself via a range of neuropsychiatric symptoms. Nitric oxide is a crucial small molecule of life synthesized within cells of our body systems, including cells of our brain. Peroxynitrite is the product of reaction between superoxide anion and nitric oxide. It normally isomerizes into harmless nitrates or nitrites. However, when excessive superoxide anion is present, the cellular concentrat

A Microenvironment-responsive small-molecule probe and application in quick acute myocardial infarction imaging.

Acute myocardial infarction (AMI) patients are at an elevated risk for life-threatening myocardial ischemia/reperfusion injury. Early-stage nonradioactive and noninvasive diagnosis of AMI is imperative for the subsequent disease treatment, yet it presents substantial challenges. After AMI, the myocardium typically exhibits elevated levels of peroxynitrite (ONOO-), constituting a distinct microenvironmental feature. In this context, the near-infrared imaging probe (BBEB) is employed to precisely delineate the boundaries of AMI lesions with a high level of sensitivity and specificity by monitori

Mercaptoethylguanidine attenuates caustic esophageal injury in rats: a role for scavenging of peroxynitrite.

INTRODUCTION: After ingestion of caustic material, tissue damage is caused by reactive oxygen species and reactive nitrogen species such as peroxynitrite. Mercaptoethylguanidine (MEG) is a well-known scavenger of peroxynitrite. This study was designed to determine whether MEG has a beneficial effect on caustic esophageal injury. MATERIALS AND METHODS: Forty-five rats were allocated into 3 groups: sham-operated, untreated, and treated groups. Caustic esophageal burn was created by instilling 15% NaOH in the distal esophagus. The rats were left untreated or treated with 10 mg/kg per day MEG int

Sites and mechanisms of aconitase inactivation by peroxynitrite: modulation by citrate and glutathione.

Aconitases are iron-sulfur cluster-containing proteins present both in mitochondria and cytosol of cells; the cubane iron-sulfur (Fe-S) cluster in the active site is essential for catalytic activity, but it also renders aconitase highly vulnerable to reactive oxygen and nitrogen species. This study examined the sites and mechanisms of aconitase inactivation by peroxynitrite (ONOO-), a strong oxidant and nitrating agent readily formed from superoxide anion and nitric oxide generated by mitochondria. ONOO- inactivated aconitase in a dose-dependent manner (half-maximal inhibition was observed wit

Chemistry of nitric oxide and related species.

Nitric oxide (NO) has essential roles in a remarkable number of diverse biological processes. The reactivity of NO depends upon its physical properties, such as its small size, high diffusion rate, and lipophilicity (resulting in its accumulation in hydrophobic regions), and also on its facile but selective chemical reactivity toward a variety of cellular targets. NO also undergoes reactions with oxygen, superoxide ions, and reducing agents to give products that themselves show distinctive reactivity toward particular targets, sometimes with the manifestation of toxic effects, such as nitrosat

Sequence-Activated Fluorescent Probe with Heteroatom Modulation Enables Dual Channel Detection of H2S and ONOO.

Hydrogen sulfide (H2S) and peroxynitrite (ONOO-) are critical redox biomarkers whose dysregulated fluctuations are closely associated with oxidative stress and related diseases. Because their interplay depends strongly on their relative levels and dynamic changes, the sequential, simultaneous, and distinguishable detection of these two species is highly important, yet it remains challenging. Herein, we employed a heteroatom substitution strategy to modulate the optical properties and sensing behavior of the probe, thereby developing a sequence-activated fluorescent probe, BSNS, for dual-channe

Nitroxidative stress in human neural progenitor cells: In situ measurement of nitric oxide/peroxynitrite imbalance using metalloporphyrin nanosensors.

Nitric oxide (NO) is an essential inorganic signaling molecule produced by constitutive NO synthase (cNOS) in the neurological system. Under pathological conditions, NO rapidly reacts with superoxide (O2•-) to generate peroxynitrite (ONOO¯). Elevated ONOO¯ concentrations induce nitroxidative stress, potentially contributing to numerous pathological processes as observed in neurodegenerative diseases including Alzheimer's disease (AD). Metalloporphyrin nanosensors, (200-300 nm diameter), were applied to quantify the NO/ONOO¯ balance produced by a single human neural progenitor cell (hNPC), in s

Simultaneous Detection of Hydrogen Sulfide and Peroxynitrite Dysregulation with a Sequentially Activated Fluorescent Probe in Ulcerative Colitis Disease.

Ulcerative colitis (UC), often referred to as "green cancer", is a chronic inflammatory bowel disease with an unclear etiology, closely associated with the imbalance of hydrogen sulfide (H2S) and peroxynitrite (ONOO-). H2S exhibits anti-inflammatory effects at physiological levels, but excessive concentrations can compromise the intestinal barrier, while ONOO- aggravates inflammation. To facilitate the molecular-level monitoring of these compounds in UC, we developed a novel fluorescent probe, BCH, capable of simultaneously detecting H2S and ONOO- via distinct fluorescent channels in a cascade

In vitro immuno-prevention of nitration/dysfunction of myogenic stem cell activator HGF, towards developing a strategy for age-related muscle atrophy.

In response to peroxynitrite (ONOO-) generation, myogenic stem satellite cell activator HGF (hepatocyte growth factor) undergoes nitration of tyrosine residues (Y198 and Y250) predominantly on fast IIa and IIx myofibers to lose its binding to the signaling receptor c-met, thereby disturbing muscle homeostasis during aging. Here we show that rat anti-HGF monoclonal antibody (mAb) 1H41C10, which was raised in-house against a synthetic peptide FTSNPEVRnitroY198EV, a site well-conserved in mammals, functions to confer resistance to nitration dysfunction on HGF. 1H41C10 was characterized by recogni

Antithyroid drugs and their analogues protect against peroxynitrite-mediated protein tyrosine nitration--a mechanistic study.

In this paper, the effect of some commonly used antithyroid drugs and their analogues on peroxynitrite-mediated nitration of proteins is described. The nitration of tyrosine residues in bovine serum albumin (BSA) and cytochrome c was studied by Western blot analysis. These studies reveal that the antithyroid drugs methimazole (MMI), 6-n-propyl-2-thiouracil (PTU), and 6-methyl-2-thiouracil (MTU), which contain thione moieties, significantly reduce the tyrosine nitration of both BSA and cytochrome c. While MMI exhibits good peroxynitrite (PN) scavenging activity, the thiouracil compounds PTU and
