Atypical porcine pestivirus (APPV) was identified and associated with congenital tremor (CT) type A-II in new born piglets and has been reported in many countries. In China, the first APPV identification in swine herds was reported in Guangdong province in 2016. To investigate the genetic characteristics of APPV in Guangxi province, 53 tissue samples from neonatal piglets with CT were collected and detected from October 2017 to May 2019. Five APPV strains which were named as GX04/2017, GX01-2018, GX02-2018, GX01-2019 and GX02-2019 were obtained. Sequence analysis revealed that all six APPV str

Self-amplifying replicon RNA (RepRNA) promotes expansion of mRNA templates encoding genes of interest through their replicative nature, thus providing increased antigen payloads. RepRNA derived from the non-cytopathogenic classical swine fever virus (CSFV) targets monocytes and dendritic cells (DCs), potentially promoting prolonged antigen expression in the DCs, contrasting with cytopathogenic RepRNA. We engineered pestivirus RepRNA constructs encoding influenza virus H5N1 (A/chicken/Yamaguchi/7/2004) nucleoprotein (Rep-NP) or hemagglutinin (Rep-HA). The inherent RNase-sensitivity of RepRNA ha

Congenital tremor (CT) type A-II in piglets is caused by an emerging atypical porcine pestivirus (APPV), which is prevalent in swine herds and a serious threat to the pig production industry. This study aimed to construct APPV E2 subunit vaccines fused with Fc fragments and evaluate their immunogenicity in piglets. Here, APPV E2Fc and E2ΔFc fusion proteins expressed in Drosophila Schneider 2 (S2) cells were demonstrated to form stable dimers in SDS-PAGE and western blotting assays. Functional analysis revealed that aE2Fc and aE2ΔFc fusion proteins could bind to FcγRI on antigen-presenting cell

Atypical porcine pestivirus (APPV) is a newly recognized member of the Flaviviridae family. This novel porcine pestivirus was first described in 2015 in the USA, where it has been associated with congenital tremor type A-II in new-born piglets. APPV is widely distributed in domestic pigs in Europe and Asia. In this study, a virological survey was performed in Northern Italy to investigate the presence of APPV using molecular methods. Testing of 360 abortion samples from pig herds revealed two APPV strains from distinct provinces in the Lombardy region and testing of 430 wild boar blood samples

BACKGROUND: Atypical porcine pestivirus (APPV), genus Pestivirus, is the causative agent of congenital tremor type A-II, a neurological disease in newborn piglets that is characterized by tremors. Although the virus can spread horizontally, clinical disease is observed only after in utero infection. While venereal transmission has been proposed as a potential route, the pathogenesis and transmission dynamics of APPV remain incompletely understood. In this study, we aimed to investigate the presence of APPV in semen from Norwegian breeding boars, its temporal shedding patterns, and the potentia

Atypical porcine pestivirus (APPV) is responsible for congenital tremor (CT) type A-II in pigs, a globally distributed neurological disease, with many unresolved questions regarding its pathogenesis and pathology. This descriptive case-control study assessed the viral load of APPV and its association with lesions in the central nervous system (CNS), as piglets born with severe clinical signs of CT recovered from clinical disease. The virus was found in all pigs with CT across 3 age groups (newborn, 3-week-old, 4- to 5-month-old CT pigs) using reverse transcription-quantitative polymerase chain

Atypical porcine pestivirus (APPV) is a novel member of the Pestivirus genus detected in association with congenital tremor (CT) type A-II outbreaks and from apparently healthy pigs, both as singular infection and as part of multi-pathogen infections. 'Classical' pestiviruses are known to cause immunosuppression of their host, which can increase susceptibility to secondary infections, severely impacting health, welfare, and production. To investigate APPV's effect on the host's immune system and characterise disease outcomes, 12 piglets from a natural APPV CT type A-II outbreak were experiment

Atypical porcine pestivirus (APPV) is one major causative agent of congenital tremor (CT) type A-II. In this study, the geographical distribution and genetic characteristics of APPV strains in China were established. To date, APPV has been detected in at least ten provinces/regions of China. Genetic variability analysis showed that NS3 genes were highly conserved among Chinese APPV strains, while NS5A-, Npro-, and Erns-encoding genes were highly variable. Phylogenetic analysis revealed that all of the reference strains could be classified into 3 genotypes (1-3) and 7 subgenotypes (1.1-1.7). Th

Classical swine fever (CSF) is one of the most important viral diseases of pigs. In many countries, the use of vaccines is restricted due to limitations of subunit vaccines with regard to efficacy and onset of protection as well as failure of live vaccines to differentiate infected from vaccinated animals (DIVA principle). Chimeric pestiviruses based on CSF virus (CSFV) and the related bovine viral diarrhea virus (BVDV) have been licensed as live marker vaccines in Europe and Asia, but cross-reactive antibodies can cause problems in DIVA application due to close antigenic relationship. Bovine viral diarrhoea virus 1 (BVDV-1) and Bovine viral diarrhoea virus 2 (BVDV-2) are two recognised bovine pestivirus species of the genus Pestivirus. Recently, a pestivirus, termed SVA/cont-08, was detected in a batch of contaminated foetal calf serum originating from South America. Comparative sequence analysis showed that the SVA/cont-08 virus shares 15-28% higher sequence identity to pestivirus D32/00_'HoBi' than to members of BVDV-1 and BVDV-2. In order to reveal the phylogenetic relationship of SVA/cont-08 with other pestiviruses, a molecular dataset of 30 pestiviruses and 1,896 chara

Class II fusion proteins encoded by tick-borne encephalitis virus (TBEV), dengue virus, and Semliki Forest virus have a fusion peptide located at the end of a rod-like molecule comprised of three antiparallel beta sheet domains. Proteomics computational analyses suggest that hepatitis C virus (HCV) envelope glycoprotein E1 and pestivirus envelope glycoprotein E2 are truncated class II fusion proteins. Similarities were also detected between the receptor-binding portion of TBEV E and HCV E2, and between TBEV small membrane protein precursor prM and pestivirus E1. The proposed models of Flavivir

This study was carried out to investigate the frequency and genetic diversity of pestiviruses in abortion cases in cattle and small ruminants in Turkey. During January 2012 and December 2017, a total of 2029 aborted foetuses (553 bovine foetuses, 1,388 sheep foetuses and 88 goat foetuses) were collected from different regions of Turkey. Real-time RT-PCR (RRT-PCR) assays were used to detect pestiviral RNA in aborted foetuses. To confirm the cause of abortion, pestivirus-positive foetuses were also examined for the presence of Brucella spp., Campylobacter spp., Chlamydophila abortus (C. abortus)

Atypical porcine pestivirus (APPV), a newly discovered member of the genus Pestivirus, is considered to be associated with congenital tremors (CT) in piglets. From June 2016 to January 2018, 440 serum and tissue samples from CT-affected piglets in Anhui, Guangdong and Guangxi provinces were collected to detect APPV. The results showed a high level of 63.4% preference for APPV in 27 swine farms and complicated co-infection cases between APPV and other 12 swine viruses. Meanwhile, 12 novel APPV genomes were screened and identified. Results showed that complete genomes, Npro and NS5A genes of the

BACKGROUND: This study aimed to characterise the RNA microbiome, including the virome of extended semen from Swedish breeding boars, with particular focus on Atypical porcine pestivirus (APPV). This neurotropic virus, associated with congenital tremor type A-II in piglets, was recently demonstrated to induce the disease through insemination with semen from infected boars.
RESULTS: From 124 Artificial Insemination (AI) doses from Swedish breeding boars, APPV was detected in one dose in addition to a sparse seminal RNA virome, characterised by retroviruses, phages, and some fecal-associated con

The pestiviruses bovine viral diarrhea virus 1 and 2 (BVDV-1 and -2, respectively) and HoBi-like pestivirus (HoBiPeV) are important pathogens of cattle, and a number of reverse-transcription PCR (RT-PCR)-based assays have been developed for their detection in clinical specimens. We evaluated a newly designed set of pan-bovine pestivirus primers (BP189-389) in a gel-based RT-PCR screening test for pestiviruses in the sera of beef calves destined for export from southern Brazil. Serum samples positive for BVDV antigens by an antigen ELISA ( n = 135) were submitted to RT-PCR assays using differen

Atypical porcine pestivirus (APPV), the causative agent of congenital tremor (CT) type A-II, is a single-stranded (+) RNA virus classified in the genus Pestivirus of the family Flaviviridae that infects domestic pigs and wild boars. While surveys and studies on APPV infection in pigs are ongoing, information on APPV in wild boars is lacking. In Japan, there have been no reports of APPV infection in wild boars. We therefore screened serum samples from 333 wild boars to determine the prevalence of APPV in Ishikawa Prefecture. APPV-RNA was detected in two samples, and the complete viral genome wa

Microfluidic technology is an advanced platform for isolating single B cells and constructing paired antibody chain libraries. In the present study, a customized immunization approach coupled with a high-throughput, microfluidic-based single B cell platform was utilized to identify and characterize a pool of monoclonal antibodies (mAbs) targeting the B/C domain of the classical swine fever virus (CSFV) E2 protein. A total of 4241 antigen-specific plasma cells were isolated and subsequently subjected to next-generation sequencing, which yielded 82 distinct naturally paired antibody sequences. B

BACKGROUND: HoBi-like pestivirus (HoBiPeV), as a newly discovered virus species in recent years, poses a serious threat to the global cattle industry due to its wide transmission and high pathogenicity. The genetic sequence of HoBiPeV exhibits significant differences when compared to the known strains of BVDV-1 (Pestivirus bovis) and BVDV-2 (Pestivirus tauri).Current detection methods are typically limited to identifying BVDV-1 and BVDV-2, and are unable to effectively differentiate HoBiPeV from the other two genotypes. To facilitate the rapid detection and differentiation of various genotypes

The genus Orthopestivirus, within the family Flaviviridae, includes important pathogens such as classical swine fever virus (CSFV), bovine viral diarrhea virus types 1 and 2 (BVDV-1 and BVDV-2), and border disease virus (BDV). Although Costa Rica is endemic for BVDV, it remains officially free of CSFV and BDV, highlighting the need for reliable molecular tools for pestivirus surveillance and differential diagnosis in swine populations. This study validated a conventional broad-range in-house RT-PCR assay targeting the 5' untranslated region (5'UTR) of pestiviruses, a genomic region that is hig

The genus Pestivirus includes several species responsible for economically important diseases in domestic and wild animals. In 2017, a novel ovine pestivirus (ovIT PeV) was isolated from aborted lamb fetuses and molecular analysis showed a genetic distinction from known Pestivirus species with the closest phylogenetic relationship to classical swine fever virus (CSFV, Pestivirus suis). This close similarity is concerning because CSF is a notifiable disease to the World Organization for Animal Health (WOAH) and is subject to eradication campaigns. Moreover, ovIT PeV can infect pigs and induce a
