Three-choice drug discrimination: phencyclidine-like stimulus effects of opioids.

To assess the commonalities and differences in the discriminative stimulus properties of phencyclidine (PCP) and psychotomimetic opioids, rats were trained to discriminate PCP (2.0 mg/kg), cyclazocine (1.0 mg/kg), and saline in a three-choice discrete-trial avoidance paradigm. Stimulus control of behavior, defined as the reliable completion of 18 trials of a 20-trial session on the appropriate choice lever after administration of PCP, cyclazocine, or saline, was established in an average of 157 sessions. In tests of stimulus generalization, SKF-10,047 and dextrorphan engendered lever choices a

Sensitization to amphetamine, but not phencyclidine, disrupts prepulse inhibition and latent inhibition.

RATIONALE: Schizophrenia has been linked to dysregulation of dopamine and glutamate transmitter systems. Attempts to model aspects of schizophrenia in animals have made use of treatments that primarily affect dopaminergic (e.g., amphetamine, Amp) and glutamatergic (e.g., phencyclidine, PCP) function. In addition to exerting short-term acute effects, these agents also induce long-term effects, as seen, for example, in neurochemical and behavioural sensitization. OBJECTIVES: The goal of this work was to compare Amp- and PCP-sensitized states on two measures of information processing that are im

Motor effects of amphetamine in rats pretreated with either dizocilpine or phencyclidine.

The aim of the present study was to examine motor effects of amphetamine (AMPH) in rats pretreated with either dizocilpine (MK-801) or phencyclidine (PCP), and to estimate possible differences in these effects. Our results showed that AMPH increases the duration of motor effects of PCP, while it does not change motor effects of MK-801. These findings may reflect different mechanisms of action of MK-801 and PCP, as well as selective influence of AMPH on metabolism of these drugs.

Phencyclidine- and diazepam-like discriminative stimulus effects of inhalants in mice.

It has been shown that abused solvents, such as 1,1,1-trichloroethane (TCE) and toluene, share certain pharmacological properties with central nervous system depressants, such as alcohol and anesthetic vapors. Several vapors were tested for diazepam (DZ)- and phencyclidine (PCP)-like discriminative stimulus effects to further explore their pharmacological specificity. In DZ-trained mice, methoxyflurane fully substituted, and TCE produced partial substitution. Fluorothyl and toluene produced no appreciable DZ-lever responding at any concentration tested. On the other hand, toluene produced conc

Effects of risperidone on phencyclidine-induced behaviors: comparison with haloperidol and ritanserin.

In this study, we investigated whether risperidone, a serotonin-S2A (5-HT2A)/dopamine-D2 (D2)-receptor antagonist, inhibits phencyclidine (PCP)-induced stereotyped behaviors in comparison with haloperidol and ritanserin. Moreover, we also attempted to investigate the effects of these antipsychotics on the contents of dopamine, serotonin (5-HT) and their metabolites in rat striatum and frontal cortex. In rats, PCP (5 mg/kg, i.p.) caused hyperlocomotion and stereotyped behaviors, including sniffing, head-weaving, backpedalling and turning. Both resperidone (0.8-2.4 mg/kg, p.o.) and haloperidol (

Electrophysiological effects of phencyclidine and the sigma agonist ditolylguanidine in the cerebellum of the rat.

The electrophysiological actions of phencyclidine (PCP) and the sigma agonist 1,3-di(2tolyl)guanidine (DTG) were examined in the cerebellum of urethane-anesthetized rats. The object of the study was to determine if PCP and sigma agonists shared a common mechanism of action. The cerebellar Purkinje neuron was chosen because it has sigma receptors but not N-methyl-D-aspartate receptors, where PCP has additional effects. Both DTG and PCP decreased the spontaneous discharge rate of cerebellar Purkinje neurons after parenteral administration. When the drugs were applied locally to single Purkinje n

Blunting of TSH response to TRH in chronic cocaine and phencyclidine abusers.

The TRH stimulation test was administered to 10 cocaine and 10 phencyclidine abusers as well as to 10 controls. No subjects had clinical evidence of depression. Significantly more blunting of the response of TSH to TRH was shown in cocaine and phencyclidine abusers compared with that seen in controls. No significant differences in blunting of response were seen between the cocaine and phencyclidine groups.

Effects of phencyclidine and ketamine on cardiovascular activity and temperature in the squirrel monkey.

Heart rate (HR), mean arterial blood pressure (BP) and core temperature (TEMP) were recorded from chair-restrained squirrel monkeys surgically prepared with chronically indwelling arterial and venous catheters to determine the effects of acute intravenous (i.v.) injections of phencyclidine and ketamine and intramuscular (i.m.) injections of ketamine. Phencyclidine (0.03-3.0 mg/kg) and ketamine (0.3-30.0 mg/kg) i.v. increased BP and decreased TEMP, and the changes in BP and in TEMP were greater in magnitude and duration after phencyclidine. Heart rate also increased monotonically after 0.03-0.3

Effects of chlorpromazine and phencyclidine on mouse C2 acetylcholine receptor kinetics.

Patch-clamp techniques were used to record acetylcholine- (ACh) activated single-channel currents in cell-attached membrane patches from myotubes of the mouse cell line, C2. The effects of the phenothiazine derivative chlorpromazine (CPZ) and of the hallucinogen phencyclidine (PCP) on ACh-activated single-channel properties were studied under conditions where both compounds are positively charged (pH 7.2). The single-channel conductance was unaffected by either CPZ or PCP at concentrations ranging from 10 to 500 nM. 10-200 nM-CPZ and PCP led to shortened mean burst times. CPZ and PCP effects o

Phencyclidine (PCP) abuse. A close-up look at a growing problem.

PCP or "angel dust" is a dissociative anesthetic agent with notoriety as an abuse substance. Numerous members of many subcultures are frequent users of this drug. It is well known in California's psychedelia, along the East Coast, and in the middle- and working-class suburbs of the Midwest. It is important that practitioners become acquainted with the drug and its effects. Persons intoxicated with PCP have murdered their own children and have even jumped out of high-rise apartment buildings. States of florid psychosis lasting for days can follow a brief encounter with PCP. Inadvertent administ

Hair analysis for detection of phencyclidine in newly admitted psychiatric patients.

The authors implemented a new procedure for analyzing phencyclidine (PCP) content in hair. They compare the results of analyses of hair with results of analyses of blood and urine in 47 patients newly hospitalized with acute psychiatric illness. Hair analysis identified 11 patients who had used PCP, and blood and urine analyses did not identify any among the sample population. In three patients, the results of hair analysis aided in establishing a diagnosis of PCP intoxication. The authors discuss interpretations of their findings and psychiatric applications of this new technique.

Quantitative electrocortical changes in the rat induced by phencyclidine and other stimulants.

An automated technique for the continuous analysis of different frequency bands of the electrocorticogram (ECoG) of the rat has been used to quantify the actions of phencyclidine (PCP) and various other stimulant drugs. It has been demonstrated that phencyclidine, etoxadrol and LY154045 produced similar changes in the individual frequency bands whereas amphetamine and apomorphine had different profiles of activity. The phencyclidine-like compounds exhibited extremely strong stimulation of the ECoG with very large increases recorded in high frequency (15-50 Hz) activity and reductions in all ot

Anaesthesia for studies of the cerebral circulation: a comparison of phencyclidine and althesin in the baboon.

The effects of phencyclidine on cerebral blood flow, cerebral metabolic rate for oxygen and carbon dioxide reactivity were compared with those of Althesin in six baboons. Under similar and stable levels (Stage 1) of electroencephalographic activity cerebral blood flow and the cerebral metabolic rate for oxygen were significantly less during the infusion of Althesin when compared with the values obtained during the infusion of phencyclidine. Reactivity to carbon dioxide was present and similar during the infusion of either drug.

Subchronic continuous phencyclidine administration potentiates amphetamine-induced frontal cortex dopamine release.

Functional dopaminergic hyperactivity is a key feature of schizophrenia. Etiology of this dopaminergic hyperactivity, however, is unknown. We have recently demonstrated that subchronic phencyclidine (PCP) treatment in rodents induces striatal dopaminergic hyperactivity similar to that observed in schizophrenia. The present study investigates the ability of PCP to potentiate amphetamine-induced dopamine release in prefrontal cortex (PFC) and nucleus accumbens (NAc) shell. Prefrontal dopaminergic hyperactivity is postulated to underlie cognitive dysfunction in schizophrenia. In contrast, the deg

Toxicological and Demographic Profiles of Phencyclidine-Impaired Driving Cases in Houston.

Phencyclidine (PCP) was first synthesized in 1926 and originally developed in 1950s as a general anesthetic agent. Abuse of PCP declined at the national level since its first illicit use in 1960s, but it continues in certain areas including Houston. This research evaluates PCP-positive cases of driving while intoxicated (DWI) in 2013-2018. The blood samples were collected from drivers, submitted by the Houston Police Department and analyzed for alcohol and drugs. Toxicological findings and demographic information were evaluated for the impaired driving cases tested positive for PCP in blood. A

Effects of hallucinogenic agents mescaline and phencyclidine on zebrafish behavior and physiology.

Mescaline and phencyclidine (PCP) are potent hallucinogenic agents affecting human and animal behavior. As their psychotropic effects remain poorly understood, further research is necessary to characterize phenotypes they evoke in various animal models. Zebrafish (Danio rerio) are rapidly emerging as a new model organism for neuroscience research. Here, we examine the effects of mescaline (5-20mg/l) and PCP (0.5-3mg/l) in several zebrafish paradigms, including the novel tank, open field and shoaling tests. Mescaline and PCP dose-dependently increased top activity in the novel tank test, also r

Differential effects of antipsychotic and glutamatergic agents on the phMRI response to phencyclidine.

Acute administration of NMDA receptor (NMDAR) antagonists such as phencyclidine (PCP) or ketamine induces symptoms that closely resemble those of schizophrenia in humans, a finding that has led to the hypothesis that a decreased NMDAR function may be a predisposing or even causative factor in schizophrenia. However, the precise neuropharmacological mechanisms underlying these effects remain to be fully elucidated. Here, we applied pharmacological MRI (phMRI) to examine the brain circuitry underlying the psychotomimetic action of PCP in the anesthetized rat, and investigated how these functiona
