Feeding responses of sheep and steers were compared following intraventricular injection of alpha-adrenergic agonists and antagonists in 2-hr. tests. In sheep, injection of the alpha-agonist 1-norepinephrine (1-NE) (140-1,120 nmoles) increased feed intakes 288% and 388% compared with intakes following synthetic cerebrospinal fluid. This response was blocked by the alpha-antagonist phenoxybenzamine, whereas the antagonist alone decreased feed intake. The beta-adrenergic antagonist propranolol did not modify the 1-NE response. A purer but weaker alpha-agonist, 1-phenylephrine, also resulted in i

Introduction Two federal drug policies, the Orphan Drug Act of 1983 (ODA) and the Unapproved Drug Initiative of 2006 (UDI), created market exclusivity and arbitrage opportunities in pharmaceutical pricing for two rare diseases, namely, pheochromocytoma and paraganglioma (PPGL). Historically, phenoxybenzamine and nitroprusside, amongst other medications, have been the first-line drugs for the perioperative and intraoperative management of patients undergoing PPGL resection. We aimed to assess the prescription behavior in perioperative pheochromocytoma management at an institution where provider

The effects of conditioning stimulation at various positions in the pons and the medulla on the lumbar spinal monosynaptic reflex (MSR) were examined in rats. The conditioning stimulation facilitated the MSR. Facilitation with long latency was completely blocked by phenoxybenzamine, chlorpromazine or haloperidol. Phenoxybenzamine and chlorpromazine inhibited the MSR. Haloperidol, on the other hand, augmented the MSR, and this effect became more potent after the brain stem had been transected at the upper part of the medulla oblongata. These results suggest that pontine noradreneric systems hav

1. The effect on rectal temperature of adrenoceptor blocking agents, injected through a cannula chronically implanted into a lateral cerebral ventricle, was examined in unanaesthetized rabbits, cats and rats, kept at room temperature (19-22 degrees C).2. In rabbits, the alpha-adrenoceptor blocking agent phenoxybenzamine (50 or 100 mug) produced marked hypothermia when injected intraventricularly but not when injected intravenously. In some rabbits as little as 1 mug was effective on intraventricular injection. Phentolamine and ergotamine, the other alpha-adrenoceptor blocking agents examined,

Participation of the autonomic nervous system in cerebellar autoregulation during supratentorial cerebral ischemia induced by bilateral carotid ligation was studied using 23 spontaneously hypertensive rats. Cerebral and cerebellar blood flows measured by a hydrogen clearance method were evaluated under stepwise hemorrhagic hypotension before and 30 minutes after ligation and after a 30-minute recirculation period following 1 hour of ligation. alpha-Adrenergic blockade with phenoxybenzamine, beta-adrenergic blockade with propranolol, and muscarinic cholinergic blockade with atropine were select

Effects of norepinephrine, epinephrine and tyramine on the carotid body chemoreceptor in the rabbit were studied by measuring reflex ventilatory responses. The basic pattern of the responses was transient inhibition with a decrease in tidal volume followed by sustained excitation, even though the predominant feature was excitation in the epinephrine-induced response. The initial inhibition which was either unaffected or slightly potentiated after phenoxybenzamine was completely blocked by haloperidol. The late excitation was enhanced after mecamylamine and haloperidol treatments and was abolis

The changes in the synthesis and utilization or noradrenaline cranial and caudal to an acute section of the rat spinal cord have been used to investigate the importance of nerve impulses for these processes. 1. Cranial to a lesion of the spinal cord, the alpha-methyltyrosine-induced disappearance of noradrenaline was accelerated by the alpha-adrenoreceptor blocking agents yohimbine (10 mg/kg), piperoxan (60 mg/kg) and tolazoline (50 mg/kg). In the absence of nerve impulses caudal to a lesion of the spinal cord, this disappearance was decelerated as compared to that cranial to the lesion and it

PURPOSE: Surgery is the major treatment option for pheochromocytoma but carries potential risks, including hemorrhage and hemodynamic instability. Even with laparoscopic adrenalectomy, intraoperative blood transfusion happens from time to time, but few studies have investigated risk factors. For the first time we develop and validate a nomogram for prediction of red blood cell transfusion in pheochromocytoma surgery.
METHODS: There were 246 patients in our center and 56 patients in Peking Union Medical College Hospital, who underwent pheochromocytoma surgery, enrolled in the study. We incorpo

1. Dopamine produced vasoconstriction followed by secondary vasodilatation in the perfused cat hindquarters and splanchnic region. Low doses produced only vasodilatation in the splanchnic region.2. Phenoxybenzamine abolished, while despiramine and cocaine potentiated, the vasoconstrictor and secondary dilator actions of dopamine.3. After phenoxybenzamine, dopamine produced only vasodilatation in both the hindquarters and splanchnic region, being more effective in the latter.4. Haloperidol, but not propranolol, atropine or mepyramine, antagonized dopamine-induced vasodilatation.5. Dopamine diff

The effects of dopamine administered in graded intravenous bolus injections (0.1 to 51.2 micrograms.kg-1) were studied simultaneously in a number of splanchnic vessels at organ level in anesthetized dogs with and without preceding administration of phenoxybenzamine. Hemodynamic data are presented for each artery as conductance, which were obtained by dividing mean flow by mean arterial pressure. The data were analyzed by two different means: 1) the response to 12.8 micrograms of dopamine during one minute, and 2) by dose-response curves. Early and late effects during the one minute post inject

1 Drugs which release or modify the response to catecholamines were examined for their effect on the permeability of the mouse peritoneal vascular bed to circulating plasma albumin, labelled with Evans blue.2 Phenoxybenzamine, phentolamine, piperoxane, yohimbine or cocaine reduced the extravasation of Evans blue into the peritoneum, an effect which was antagonized by beta-adrenoceptor blocking drugs. The inhibitory effect of desipramine on the extravasation of Evans blue was less completely antagonized by beta-adrenoceptor blockade.3 Inhibition of catecholamine biosynthesis, ganglion blockade

Rolling mouse Nagoya (RMN), an ataxia mutant mouse, is adequate to checking up the efficacy of drugs on the ataxic movement disorders. The effects of some drugs on the ataxia of RMN were investigated by open field study. The drugs administered to RMN were as follows: TRH, FLA-63, DL-threo-DOPS, phenoxybenzamine and propranolol. Ataxic gait of RMN were remarkably improved by TRH, but remained unchanged by other drugs. The pretreatments with FLA-63, DOPS and phenoxybenzamine were not influenced to the antiataxic effects of TRH, while the pretreatment with propranolol increased the antiataxic eff

To twenty six patients with angina at rest (including 13 patients with Prinzmetal's variant angina), propranolol 40--50 mg, dilitiazem 60--90 mg, dipyridamole 50 mg, atropine sulfate 0.6 mg and phenoxybenzamine 10--20 mg were given at 9:00 p.m. and 2:00 a.m. (except phenoxybenzamine which was given only at 9:00 p.m.) to examine the effect of these drugs on the attack. Propranolol was not only ineffective in suppressing the attack but rather tended to aggravate it in all the cases of Prinzmetal's variant angina. It was effective to some degree in 5 of 13 cases of angina at rest other than Prinz

1 Intravenous injections of urotensin I regularly caused a long-lasting, dose-related, lowering of blood pressure and an increase in heart rate in conscious rats, or a reduction in perfusion pressure in the isolated hind limb of the rat.2 After subcutaneous administration, the hypotensive effect of urotensin I was greater in extent and in duration (> 24 hours).3 Anaesthesia with ether, chloralose, pentobarbitone and thiobarbitone caused a decrease in blood pressure and only slightly diminished the hypotensive effect of urotensin.4 Mecamylamine, hexamethonium, atropine, phenoxybenzamine, propra

1. Bupropion (12.5-75 mg kg-1) was given intraperitoneally to rats and was found to decrease the food consumption of the animals dose-dependently. While phenoxybenzamine, propranolol and methergoline failed to antagonize the anorectic effect of the drug; pimozide a dopamine receptor blocker decreased anorexia induced by bupropion. 2. Bupropion (12.5-50 mg kg-1) also caused a marked increase in locomotor activity of the rats. The increase in locomotion produced by bupropion was completely antagonized by pretreatment of the animals with pimozide and reserpine plus a-methyl-p-tyrosine, but not by

Electroconvulsive shock (ECS) applied immediately after passive avoidance learning in rats caused partial amnesia. This could be prevented by administering r-ANP into the lateral brain ventricle. The effects of pre-treatment with different receptor blockers: (haloperidol, atropine, phenoxybenzamine, propranolol, naloxone, bicuculline and methysergide) on the ANP-induced antiamnesia were investigated. The receptor blockers per se in the doses selected had no influence on the ECS-induced amnesia. Haloperidol, atropine and propranolol blocked the antiamnestic action of the peptide, while phenoxyb

OBJECTIVE: Prolonged hypotension is a common complication after resection of pheochromocytoma (PCC) or paraganglioma (PGL). The objective of our study was to identify preoperative or intraoperative clinical factors that can predict prolonged hypotension after PCC/PGL resection.
PATIENTS AND METHODS: A total of 414 patients who underwent resection of PCC or PGL at our institution between January 2013 and January 2020 were included. Patients were divided into two groups according to whether or not vasopressor support was required postoperatively. Associations between preoperative and intraopera

Urocortin 3 (Ucn 3) was tested for anxiolytic action in mice an elevated plus maze. For detection of the possible involvement of neurotransmitters, the mice were pretreated with receptor blockers: haloperidol, phenoxybenzamine, propranolol, atropine, methysergide, bicuculline or naloxone. The peptide was administered into the lateral brain ventricle; the receptor blockers were applied intra- peritoneally. Ucn 3 alone elicited dose-dependent bell-shaped anxiolytic action. The most effective dose was 0.5 μg. In the combined testing a 0.5 μg dose was used. Haloperidol, propranolol, atropine, meth

Acetazolamide, a carbonic anhydrase inhibitor, induced right forelimb ectrodactyly in rat fetuses when the mothers were treated on late day 10 and early day 11 of gestation. Coadministration of the selective alpha-1-adrenergic agonist phenylephrine significantly increased the incidence of acetazolamide-induced right forelimb ectrodactyly while failing to induce the lesion when administered alone. Pretreatment with the alpha-adrenergic antagonists phenoxybenzamine and prazosin prevented the phenylephrine-induced increase in right forelimb ectrodactyly. In addition, treatment with either phenoxy

Effects of carotid body chemoreceptor stimulation by dopamine (DA) on ventilation were studied by injecting it into the external carotid artery of rabbits. DA (3 micrograms--300 micrograms) induced dose-dependent ventilatory depression which was preceded by a transient increase in ventilatory rate in 65% of the experiments at higher doses. The DA-induced depression remained unaffected by prior administration of atropine, mecamylamine or phenoxybenzamine, while it was blocked or suppressed after haloperidol or reserpinization, indicating that the DA-induced ventilatory depression was due to its
