Metabolomic analysis reveals novel ethylated hydroxytyrosol metabolites in colon cancer cells.

Plants produce a diverse array of secondary metabolites, with hydroxytyrosol (HT) and its derivatives distinguished by their antioxidant and chemopreventive properties. Upon dietary consumption, native HT undergoes extensive biotransformation, initially facilitated by the gut microbiota. Increasing evidence suggests that several of these downstream products, rather than the parent molecule, are responsible for the most significant biological effects in mammalian cells. The identification of novel metabolites may reveal unknown metabolic pathways, more particularly in distinct pathological cont

Soft-Sensing-Guided Glucose/l-Phenylalanine Cofeeding Strategy and Transcriptomic Analysis of the Optimal l-Phenylalanine Supply Range for Efficient β-Phenylethanol Biosynthesis in Saccharomyces cerevisiae.

β-Phenylethanol (2-PE) is a natural aroma compound used in food and cosmetic applications, and its efficient microbial production requires the coordinated control of glucose and precursor availability. Here, Raman and near-infrared (NIR) spectroscopy were implemented as soft sensors to guide glucose/l-phenylalanine (l-Phe) cofeeding during 2-PE biosynthesis by Saccharomyces cerevisiae. Raman-Artificial Neural Network and NIR-Partial Least Squares models showed the best performance (external validation R2 up to 0.99). A soft-sensing-guided glucose-feeding strategy maintained glucose below 20 g/

Hydroxytyrosyl Eicosapentaenoate as a Potential Antioxidant for Omega-3 Fatty Acids: Improved Synthesis and Comparative Evaluation with Other Natural Antioxidants.

Hydroxytyrosol (HT), the primary phenolic compound in virgin olive oil, has notable cardiovascular benefits, particularly in preventing low-density lipoprotein (LDL) oxidation. However, its hydrophilicity limits its solubility and integration into lipid-based formulations. This study aimed to enhance its lipophilicity by synthesizing hydroxytyrosyl eicosapentaenoate (HT-EPA), a derivative of HT and eicosapentaenoic acid (EPA), using a one-step enzymatic catalysis with lipase B from Candida antarctica (CALB). The reaction, performed as a suspension of HT in ethyl eicosapentaenoate (Et-EPA) (1:9

A Nutraceutical Compound Containing a Low Dose of Monacolin K, Polymethoxyflavones, Phenolic Acids, Flavonoids, and Hydroxytyrosol Improves HDL Functionality.

BACKGROUND: In earlier studies, it has been observed that 8-week treatment with a novel nutraceutical compound (NC) containing low monacolin K dose, polymethoxyflavones, phenolic acids, flavonoids, and hydroxytyrosol improves lipid profile and endothelial function and reduces the level of oxidized low-density lipoprotein (oxLDL). We hypothesize that this effect might be, at least in part, explained by positive modulation exerted by the NC on the atheroprotective function of high-density lipoprotein (HDL). AIM: This study aimed to evaluate whether the NC could influence determinants of HDL fun

Hydroxytyrosol effectively improves the quality of pig sperm at 17 °C.

Artificial insemination (AI) is a proven breeding technology which has been widely used in pig reproduction. Low temperature can cause very serious damage to pig sperm below 15 °C and the situation is even more serious at lower temperature. Besides, the preservation of pig sperm is mainly carried out at 17 °C because of its outstanding performance in pig reproduction. However, the accumulation of a large amount of reactive oxygen species (ROS) during the preservation process is the main reason for the deterioration of sperm quality. In our research, by adding different concentrations of hydrox

Oleil Hydroxytyrosol (HTOL) Exerts Anti-Myeloma Activity by Antagonizing Key Survival Pathways in Malignant Plasma Cells.

Polyphenols from olive oil are endowed with several biological activities. Chemical modifications have been recently applied to these compounds to improve their therapeutic activity in different pathological settings, including cancer. Herein, we describe the in vitro effects on multiple myeloma (MM) cells of oleil hydroxytyrosol (HTOL), a synthetic fatty ester of natural hydroxytyrosol with oleic acid. HTOL reduced the viability of various human MM cell lines (HMCLs), even when co-cultured with bone marrow stromal cells, triggering ER stress, UPR and apoptosis, while it was not cytotoxic agai

Caffeic acid phenethyl ester as a protective agent against nephrotoxicity and/or oxidative kidney damage: a detailed systematic review.

Caffeic acid phenethyl ester (CAPE), an active component of propolis, has been attracting the attention of different medical and pharmaceutical disciplines in recent years because of its antioxidant, anti-inflammatory, antiproliferative, cytotoxic, antiviral, antifungal, and antineoplastic properties. One of the most studied organs for the effects of CAPE is the kidney, particularly in the capacity of this ester to decrease the nephrotoxicity induced by several drugs and the oxidative injury after ischemia/reperfusion (I/R). In this review, we summarized and critically evaluated the current kn

Testing for phytochemical synergism: arthropod community responses to induced plant volatile blends across crops.

Using herbivore-induced plant volatiles (HIPVs) to attract specific natural enemies in the field has proven challenging, partly because of a poor understanding of: (i) which compound(s) to manipulate to attract specific taxa, and (ii) the ecological conditions over which HIPVs are effective. To address these issues, we quantified the response of a complex arthropod community to three common HIPVs (methyl salicylate, cis-3-hexen-1-ol, and phenylethyl alcohol) as individual compounds and equal part blends in corn and soybean fields. Of 119 arthropod taxa surveyed, we found significant responses

Hydroxytyrosol-based natural deep eutectic solvents (NADESs): Hydrogen bond network formation, anti-freezing properties, and the regulation of starch in freeze-thaw systems.

This study employed hydroxytyrosol as a hydrogen donor to construct five natural deep eutectic solvent (NADESs) systems. A systematic investigation was conducted on the changes of functional groups within the NADESs, their freezing point depression, low-temperature rheological behavior, ice recrystallization inhibition capacity, and their applications in freeze-thaw starch systems. The hydroxyl stretching peak (3700-3000 cm-1) showed significant enhancement and broadening, indicating a hydrogen bond network formation. The freezing points of 20% NADESs ranged from -24.47 °C to -17.43 °C, demons

Hypoxia-Driven Functional Conversion of CAPE: From Anti-Inflammatory to Pro-Tumorigenic Action in the Human Astrocytoma Cell Line CCF-SSTG1.

The glioblastoma multiforme (GBM) microenvironment, characterized by hypoxia and inflammation, is a principal driver of therapeutic resistance. Although natural compounds such as Caffeic Acid Phenethyl Ester (CAPE) are investigated for their anti-neoplastic properties, their bioactivity within the distinct metabolic landscape of the tumor core remains to be fully elucidated. Taking advantage of the recognized immunomodulatory properties of CAPE and its ability to cross the blood-brain barrier, we hypothesized that hypoxia is a key factor determining its effect on glioma-associated inflammation

Caffeic Acid Phenethyl Ester (CAPE) Inhibits Arginase Activity and Growth of Leishmania amazonensis Promastigotes and Intracellular Amastigotes.

Caffeic acid phenethyl ester (CAPE) is a polyphenol produced by many plants and is found in red and green propolis. Here, we evaluated the antileishmanial activity of this natural product against Leishmania amazonensis. CAPE exhibited IC50 values of 8.07 µM (95% CI, 6.79-9.62 µM) and 13.51 µM (95% CI, 10.71-17.16 µM) against L. amazonensis promastigotes and intracellular amastigotes, respectively. Additionally, CAPE inhibited L. amazonensis arginase in a non-competitive manner with a Ki value of 1.51 ± 0.04 µM. These results highlight the potential of CAPE as a promising lead compound for deve

Pro-inflammatory protein S100A9 targeted by a natural molecule to prevent neurodegeneration onset.

Accumulation of the pro-inflammatory protein S100A9 has been implicated in neuroinflammatory cascades in neurodegenerative diseases (NDs) such as Alzheimer's disease (AD) and Parkinson's disease (PD). S100A9 co-aggregates with other proteins such as α-synuclein in PD and Aβ in AD, contributing to amyloid plaque formation and neurotoxicity. The amyloidogenic nature of this protein and its role in chronic neuroinflammation suggest that it may play a key role in the pathophysiology of these diseases. Research into molecules targeting S100A9 could be a potential therapeutic strategy to prevent its

The potential of hydroxytyrosol fatty acid esters to enhance oral bioavailabilities of hydroxytyrosol and fatty acids: Continuous and slow-release ability in small intestine and blood.

HPLC-UV analysis in rat everted gut sac and in vitro simulated digestion models indicated that hydroxytyrosol fatty acid esters (HTy-Es) could be hydrolyzed by pancreatic lipase to slow-release of free fatty acids (FAs) and HTy. Meanwhile, the HTy-Es, the liberated FAs and the HTy could cross the membrane and were transported into blood circulation. HTy-Es were further hydrolyzed by carboxylesterase in in vitro rat plasma hydrolysis model, which also showed slow-release of FAs (C1-C4) and HTy. Especially, the rates of hydrolysis and transport initially increased and then decreased with the inc

Structure, antioxidative potency and potential scavenging of OH and OOH of phenylethyl-3,4-dihydroxyhydrocinnamate in protic and aprotic media: DFT study.

DFT methods including B3LYP, B3PW91 and M05-2x associated to 6-31+G(d,p) were used for the structural and antioxidant potency studies of phenylethyl-3,4-dihydroxy-hydrocinnamate (PDH). Solvents were employed according to their protric and aprotic character. So, calculated structures agree with the experimental data. O4H4 is propitious to scavenge radicals whatever the medium except in water where O3H3 and O4H4 are competitive. The explicit solvents of dichloromethane (DCM) and water present a disparity of OH bond dissociation enthalpy and free energy (BDE and BDFE). These parameters are low in

Integrated analysis reveals the effects of substituting pea with mung bean on microbial succession and metabolite profiles during medium-temperature Daqu fermentation.

Daqu serves as the saccharifying and fermenting starter in Baijiu production, yet the effects of raw material variation on community succession and metabolite formation during fermentation remain unclear. In this study, amplicon sequencing and metabolomic profiling were applied to compare mung bean Daqu (MBQ), produced by substituting pea with mung bean, with traditional medium-temperature Daqu. Microbial succession patterns were largely conserved after mung bean substitution, whereas stage-specific differences were observed in community composition, environmental associations, and microbial c
