Human antibody reaction against recombinant salivary proteins of Phlebotomus orientalis in Eastern Africa.

BACKGROUND: Phlebotomus orientalis is a vector of Leishmania donovani, the causative agent of life threatening visceral leishmaniasis spread in Eastern Africa. During blood-feeding, sand fly females salivate into the skin of the host. Sand fly saliva contains a large variety of proteins, some of which elicit specific antibody responses in the bitten hosts. To evaluate the exposure to sand fly bites in human populations from disease endemic areas, we tested the antibody reactions of volunteers' sera against recombinant P. orientalis salivary antigens. METHODOLOGY/PRINCIPAL FINDINGS: Recombinan

Differential expression profiles of the salivary proteins SP15 and SP44 from Phlebotomus papatasi.

BACKGROUND: Sand fly saliva has been shown to help parasite establishment and to induce immune responses in vertebrate hosts. In the current study, we investigated the pattern of expression of two Phlebotomus papatasi salivary transcripts in specific physiological and seasonal conditions at a hyperendemic area of zoonotic cutaneous leishmaniasis (ZCL) in Iran. METHODS: Sand flies were collected during 2012-2013, and grouped according to physiological stages such as unfed, fed, semi-gravid, gravid, parous, nulliparous, infected or non-infected with Leishmania major and also based on the season

Kinetics of Anti-Phlebotomus perniciosus Saliva Antibodies in Experimentally Bitten Mice and Rabbits.

BACKGROUND: Sand flies are hematophagous arthropods that act as vectors of Leishmania parasites. When hosts are bitten they develop cellular and humoral responses against sand fly saliva. A positive correlation has been observed between the number of bites and antibody levels indicating that anti-saliva antibody response can be used as marker of exposure to sand flies. Little is known about kinetics of antibodies against Phlebotomus perniciosus salivary gland homogenate (SGH) or recombinant salivary proteins (rSP). This work focused on the study of anti-P. perniciosus saliva antibodies in sera

Specificity of anti-saliva immune response in mice repeatedly bitten by Phlebotomus sergenti.

Sand flies are bloodsucking insects transmitting parasites of genus Leishmania, the causative agents of diseases in humans and dogs. Experimental hosts repeatedly exposed to sand fly saliva can control Leishmania infection. Cell-mediated anti-saliva immune response is most likely responsible for this protective effect; however, there is no study so far concerning its antigenic specificity towards different sand fly vectors. In this study, splenocytes from BALB/c mice repeatedly exposed to the bites of Phlebotomus sergenti were challenged ex vivo with salivary gland homogenates from three diffe

Impact of pyrethroid-impregnated curtains on Phlebotomus papatasi sandflies indoors at Khartoum, Sudan.

Laboratory and field investigations were made in an endemic focus of cutaneous leishmaniasis in Khartoum State, Sudan, to evaluate the effects of permethrin-impregnated curtains on the human-biting activity, nocturnal activity and resting behaviour of the vector sandfly Phlebotomus papatasi (Diptera: Psychodidae) indoors. Laboratory bioassays showed that curtains impregnated with 0.5, 1.0 or 1.5 g/m2 permethrin all gave 100% mortality within 24h of exposure of P. papatasi for 3 min. Under natural field conditions, the biting activity indoors and the resting density of P. papatasi were signific

First report of the visceral leishmaniasis vector Phlebotomus martini (Diptera: Psychodidae) in Tanzania.

Phlebotomus martini is a known vector of visceral leishmaniasis caused by Leishmania donovani in sub-Saharan Africa. The disease is known to be endemic in areas of north and south Sudan, Kenya, Ethiopia, Uganda, and Somalia but has not been reported from Tanzania. In this report we present the first documented collection of P. martini and P. vansomerenae in Tanzania. Sand flies were collected using standard dry-ice baited CDC light traps (John W. Hock Company, Gainesville, FL) from five sampling sites in the Arusha and Kilimanjaro regions from 14 to 20 July 2010. Phlebotomus martini was collec

Phlebotomus sergenti (Parrot, 1917) identified as Leishmania killicki host in Ghardaïa, south Algeria.

Since 2005, an outbreak of human cutaneous leishmaniasis (CL) in Ghardaïa, south Algeria, was studied and one output of these investigations was the identification of two Leishmania species, Leishmania major and Leishmania killicki, as the CL causative agents. In the present study, we were curious to focus on sand fly fauna present in this area and detection of Leishmania-positive sand fly females. Sand flies (3717) were collected during two seasons using sticky papers and CDC light traps in urban, rural and sylvatic sites. Twelve Phlebotomus species were identified. Phlebotomus papatasi was d

Preliminary study on the galectin molecular diversity in Moroccoan Phlebotomus papatasi sandfly population.

CONTEXT: Galactose binding protein (PpGalec) plays an important role in the specificity of Phlebotomus papatasi sandfly for Leishmania major. The molecular diversity of this ligand is currently unknown but might have some influence on the ability of PpGalec to efficiently recognize L. major in natural sandfly populations. OBJECTIVE: To explore the molecular diversity of the P. papatasi Galectin gene (PpGalec) in natural sandfly population of Morocco. RESULTS & CONCLUSIONS: Sequence variations of PpGalec was analyzed in 31 P. papatasi specimens collected from endemic and non-endemic zoonotic

Natural infection of Phlebotomus (Larroussius) langeroni (Diptera: Psychodidae) with Leishmania infantum in Tunisia.

Phlebotomine sand flies were captured from an active transmission focus of sporadic cutaneous leishmaniasis, caused by Leishmania infantum, in El Kef region, northern Tunisia. Both Phlebotomus perniciosus and P. langeroni were found. Phlebotomus langeroni females showed a statistically significant intradomiciliary dominance (P<0.01 for the 2003 and 2004 seasons) when compared to animal shelters. During the 2003 season, dissection of collected female specimens showed the presence of flagellates within the digestive tracts of two P. perniciosus among 1086 observed, but none in 232 P. langeroni.

Optimised dietary regimens for the laboratory maintenance of Phlebotomus langeroni nitzulescu (Diptera: Psychodidae).

The optimal feeding conditions for laboratory colonies of Phlebotomus langeroni, the vector of infantile visceral leishmaniasis in Egypt, were investigated. Sugarmeals were offered only before bloodmeals (the SB regimen), only after blood (BS), or both before and after blood (SBS). In each regimen, three different solutions of sucrose (10%, 30% and 50%, w/v) were tested as the source of sugarmeals. In general, the concentration of sucrose used had no significant effect on female longevity, although the longest lived females were those fed on 30% sucrose. Females on the BS regimen were the long

Laboratory infection of the sandfly Phlebotomus papatasi Scopoli (Diptera, Psychodidae) with three Phleboviruses.

Membrane feeding and intrathoracic inoculation techniques were used to infect Phlebotomus papatasi sandflies with Phlebotomus fever group viruses. Sandflies were orally infected with Pacui virus, and were inoculated with Pacui, Sicilian sandfly fever and Naples sandfly fever viruses. Pacui virus did not multiply in these insects after oral infection, but an 8 X 10(4)-fold increase in virus concentration occurred within four days of intrathoracic inoculation of this virus. The infection rate was 100%. Sicilian sandfly fever virus multiplied in P. papatasi after infection by inoculation, althoug

A possible role for Phlebotomus (Anaphlebotomus) rodhaini (Parrot, 1930) in transmission of Leishmania donovani.

BACKGROUND: Visceral leishmaniasis (VL, kala azar), caused by Leishmania donovani is a major health problem in Sudan and other East African countries. In this region the only proven vectors of L. donovani are Phlebotomus orientalis in eastern Sudan, Ethiopia and Upper Nile areas of Southern Sudan and Phlebotomus martini in Ethiopia, Kenya and Southern Sudan. In this report, we present the first evidence that Phlebotomus rodhaini may also play a role in maintaining transmission of L. donovani between animal reservoir hosts in eastern Sudan. The study was conducted in a zoonotic focus of viscera

Distribution of Leishmania major zymodemes in relation to populations of Phlebotomus papatasi sand flies.

Phlebotomus papatasi (Scopoli) (Diptera: Psychodidae) is the main vector of Leishmania major Yakimoff & Schokhor (Kinetoplastida: Trypanosomatidae), the causative agent of zoonotic cutaneous leishmaniasis in the Old World. Multilocus enzyme electrophoresis (MLEE) was extensively used to type different L. major stocks all over the world. Multilocus microsatellite typing (MLMT) has been recently used to investigate P. papatasi sand flies at population and subpopulation levels. In this article, the association between geographical distribution of L. major zymodemes and the distribution of populat

Colonization of Phlebotomus neglectus (Diptera: Psychodidae), the major vector of visceral leishmaniasis in Greece.

Colonization of Phlebotomus neglectus Tonnoir, the major vector of visceral leishmaniasis, in Greece is reported for the first time. Starting with wild-caught specimens, a small closed colony was established that was maintained for 17 mo or 10 generations. Gonotrophic discordance, stenogamic mating behavior, low fecundity, and dormancy because of low temperature were the most important findings that characterized the colony.

[Isolation of Leishmania major in Phlebotomus papatasi in Biskra (Algeria). The end of an ecoepidemiological saga].

Out of 1,167 females of sandflies dissected, one specimen of Phlebotomus papatasi captured at a transmission site near Biskra, a well known Algerian focus of zoonotic cutaneous leishmaniasis, was found naturally infected with Leishmania major zymodeme MON-25. This supports classical observations of Sergent and al. P. papatasi as vector in this focus in 1921.

Oviposition diapause and other factors affecting the egg-laying of Phlebotomus papatasi in the laboratory.

Stimuli which modulate oviposition of P. papatasi were investigated to improve insectary breeding efficiency. Oviposition and survival of gravid females were observed weekly during April-December 1987, in plastic cages at 28 +/- 1 degrees C with L:D 17:7. Oviposition of controls was subject to seasonal variation despite the relatively uniform insectary conditions. From April to mid-October (summer), mean weekly oviposition ranged from 11.6 to 18.6 eggs per fly, dropping to 1.4 eggs/fly in November (winter). Monthly yields of eggs were found to correlate with the seasonal cycle of ultraviolet (
