Proteomics of isolated sieve tubes from Nicotiana tabacum: sieve element-specific proteins reveal differentiation of the endomembrane system.

Symplasmicly connected cells called sieve elements form a network of tubes in the phloem of vascular plants. Sieve elements have essential functions as they provide routes for photoassimilate distribution, the exchange of developmental signals, and the coordination of defense responses. Nonetheless, they are the least understood main type of plant cells. They are extremely sensitive, possess a reduced endomembrane system without Golgi apparatus, and lack nuclei and translation machineries, so that transcriptomics and similar techniques cannot be applied. Moreover, the analysis of phloem exudat

The role of methyl salicylate in plant growth under stress conditions.

Methyl salicylate is a volatile compound, the synthesis of which takes place via the salicylic acid pathway in plants. Both compounds can be involved in the development of systemic acquired resistance and they play their role partly independently. Salicylic acid transport has an important role in long-distance signalling, but methyl salicylate has also been suggested as a phloem-based mobile signal, which can be demethylated to form salicylic acid, inducing the de-novo synthesis of salicylic acid in distal tissue. Despite the fact that salicylic acid has a protective role in abiotic stress res

Dispersal of Agrilus planipennis (Coleoptera: Buprestidae) from discrete epicenters in two outlier sites.

Emerald ash borer, Agrilus planipennis (Fairmaire) (Coleoptera: Buprestidae), a phloem-feeding beetle native to Asia, has become one of the most destructive forest pests in North America. Since it was first identified in 2002 in southeast Michigan and Windsor, Ontario, dozens of isolated A. planipennis populations have been discovered throughout Michigan and Ontario, and in 12 other states and the province of Quebec. We assessed realized A. planipennis dispersal at two discrete outlier sites that originated 1 yr and 3 yr earlier from infested nursery trees. We systematically sampled ash trees

DAPI and Confocal Laser-Scanning Microscopy for In Vivo Imaging of Phytoplasmas.

As phytoplasmas are located inside the phloem tissue, always surrounded by numerous layers of other cells, they can result difficult candidates for microscopical investigations. Moreover, the necessity to kill the plant tissues for microscopy observations causes instantaneous and irreversible modifications in the sieve elements, leading to misleading information and erroneous interpretations. Phytoplasmas were here investigated in intact Vicia faba host plants using DAPI as fluorescent probe and confocal laser scanning microscopy. The described nondestructive technique may be applied for the i

A unique virulence factor for proliferation and dwarfism in plants identified from a phytopathogenic bacterium.

One of the most important themes in agricultural science is the identification of virulence factors involved in plant disease. Here, we show that a single virulence factor, tengu-su inducer (TENGU), induces witches' broom and dwarfism and is a small secreted protein of the plant-pathogenic bacterium, phytoplasma. When tengu was expressed in Nicotiana benthamiana plants, these plants showed symptoms of witches' broom and dwarfism, which are typical of phytoplasma infection. Transgenic Arabidopsis thaliana lines expressing tengu exhibited similar symptoms, confirming the effects of tengu express

Biochemistry and molecular biology of Arabidopsis-aphid interactions.

To ensure their survival in natural habitats, plants must recognize and respond to a wide variety of insect herbivores. Aphids and other Hemiptera pose a particular challenge, because they cause relatively little direct tissue damage when inserting their slender stylets intercellularly to feed from the phloem sieve elements. Plant responses to this unusual feeding strategy almost certainly include recognition of aphid salivary components and the induction of phloem-specific defenses. Due to the excellent genetic and genomic resources that are available for Arabidopsis thaliana (Arabidopsis), t

Structural changes in the vascular bundles of light-exposed and shaded spruce needles suffering from Mg deficiency and ozone pollution.

The correlation between structural changes of the vascular bundles and needle yellowing was examined for needles of damaged spruce (Picea abies (L.) Karst.) growing at a Mg-deficient and ozone polluted mountain site in the Central Black Forest (840m a.s.l.). In the previous year's sun-exposed needles, the following sequence of events was observed: (1) rapid needle yellowing, (2) hypertrophy and anomalous divisions of cambium cells, (3) phloem collapse, and, (4) production of atypical xylem tracheids. Under defined shade (reduction of the photosynthetically active photon flux density of the amb

Jasmonate- and salicylate-induced defenses in wheat affect host preference and probing behavior but not performance of the grain aphid, Sitobion avenae.

Jasmonate- and salicylate-mediated signaling pathways play significant roles in induced plant defenses, but there is no sufficient evidence for their roles in monocots against aphids. We exogenously applied methyl jasmonate (MeJA) and salicylic acid (SA) on wheat seedlings and examined biochemical responses in wheat and effects on the grain aphid, Sitobion avenae (Fab.). Application of MeJA significantly increased levels of wheat's polyphenol oxidase, peroxidase and proteinase inhibitor 1, 2 and 6 days after treatment. In two-choice tests, adult aphids preferred control wheat leaves to MeJA- o

Overexpression of IRM1 enhances resistance to aphids in Arabidopsis thaliana.

Aphids are insects that cause direct damage to crops by the removal of phloem sap, but more importantly they spread devastating viruses. Aphids use their sophisticated mouthpart (i.e. stylet) to feed from the phloem sieve elements of the host plant. To identify genes that affect host plant resistance to aphids, we previously screened an Arabidopsis thaliana activation tag mutant collection. In such mutants, tagged genes are overexpressed by a strong 35S enhancer adjacent to the natural promoter, resulting in a dominant gain-of-function phenotype. We previously identified several of these mutan

Efficient Establishment of Interfamily Heterograft of Nicotiana benthamiana and Arabidopsis thaliana.

The grafting technique has been applied to study systemic signaling in plants, to investigate whether gene action is graft-transmissible and/or gene products such as RNAs and proteins are transported systemically. Here, we describe two protocols, stem grafting and micrografting, for an interfamily heterograft system between Nicotiana benthamiana scion plants and Arabidopsis stock plants for the identification of systemic mobile signals. Since these plants belong to evolutionally distant families and both genome databases are available, this system can be used to identify mobile substances tran

Method for Analyzing CAZyme Activity from Whole-Body and Saliva Proteome of Russian Wheat Aphid.

Advanced proteome extraction and characterization techniques for Diuraphis noxia (Russian Wheat Aphid: RWA) enable precise biochemical profiling, where high-quality proteome extraction is critical. This chapter introduces an optimized proteome extraction method for comprehensive profiling of D. noxia, enabling analysis of whole-body and saliva proteomes to advance biochemical and functional characterization. The efficacy of two optimized proteome extraction methods was compared: a nondestructive artificial saliva extraction and a destructive whole-body extraction. The proteome extract concentr

Retrospective study of methylmercury and other metal(loid)s in Madagascar unpolished rice (Oryza sativa L.).

The rice ingestion rate in Madagascar is among the highest globally; however studies concerning metal(loid) concentrations in Madagascar rice are lacking. For Madagascar unpolished rice (n = 51 landraces), levels of toxic elements (e.g., total mercury, methylmercury, arsenic and cadmium) as well as essential micronutrients (e.g., zinc and selenium) were uniformly low, indicating potentially both positive and negative health effects. Aside from manganese (Wilcoxon rank sum, p < 0.01), no significant differences in concentrations for all trace elements were observed between rice with red bran (n

Feeding guild of non-host community members affects host-foraging efficiency of a parasitic wasp.

Interactions between predator and prey, or parasitoid and host, are shaped by trait- and density-mediated processes involving other community members. Parasitoids that lay their eggs in herbivorous insects locate their hosts through infochemicals such as herbivore-induced plant volatiles (HIPVs) and host-produced kairomones. Hosts are frequently accompanied by non-host herbivores that are unsuitable for the parasitoid. These non-hosts may interfere with host location primarily through trait-mediated processes, by their own infochemicals, and their induction of the emission of plant volatiles.

Filamentous fungal associates of the alder bark beetle, Alniphagus aspericollis, including an undescribed species of Neonectria.

Bark beetles (Coleoptera: Curculionidae; Scolytinae) are tree-infesting insects that consume subcortical tissues and fungi. Species capable of killing their host trees are most commonly associated with conifers, as very few bark beetle species infest and kill hardwood hosts directly. The alder bark beetle, Alniphagus aspericollis, is a hardwood-killing bark beetle that colonizes and kills red alder, Alnus rubra. Conifer-killing bark beetles have well-known associations with symbiotic ophiostomatoid fungi that facilitate their life histories, but it is unknown whether A. aspericollis has any fu

Vascular tissue anatomy of Norway spruce needles and twigs in relation to magnesium deficiency.

Vascular tissues of needles and twigs of Norway spruce with low foliar magnesium concentrations were examined by light microscopy. Observations were made on samples from (1) apparently healthy trees, (2) trees exhibiting progressive symptoms of decline, including the yellowing and fall of needles and the death of twigs and branches in the subapical canopy, and (3) trees recovering from symptoms of decline after fertilization with magnesium sulfate. Abnormalities in cambium and phloem anatomy, which were apparent in 2-year-old needles of declining trees, were seen only in needles at least 4 yea

High-Throughput Hypocotyl Grafting and Vascular Reconnection Assessment in Arabidopsis Seedlings.

Plant grafting is a powerful tool in research and agriculture, enabling the study of long-distance signaling and improving crop performance. Hypocotyl grafting in Arabidopsis thaliana has become a key method for investigating inter-tissue communication, root-to-shoot transport, and vascular regeneration. Here, we describe a high-throughput hypocotyl grafting protocol that consistently achieves success rates above 90% while minimizing complications, such as adventitious root formation, through an optimized hypocotyl cutting position. Our streamlined approach eliminates the need for silicone tub

Methods to Estimate Carbon Export from Leaves.

In most plant species, the majority of carbon assimilation takes place in leaves. The export of carbon from leaves to the rest of the plant's organs is thus a fundamental process in plant biology. At the same time, only a few studies have estimated the rate of this process and its dynamics. In the current chapter, we will describe two simple methods for evaluating the rate of carbon export from leaves. Both methods calculate the export as the difference between carbon assimilation and carbon storage. The first method evaluates the carbon storage based on the leaf mass per area (LMA) difference

Up-regulation of abscisic acid signaling pathway facilitates aphid xylem absorption and osmoregulation under drought stress.

The activation of the abscisic acid (ABA) signaling pathway reduces water loss from plants challenged by drought stress. The effect of drought-induced ABA signaling on the defense and nutrition allocation of plants is largely unknown. We postulated that these changes can affect herbivorous insects. We studied the effects of drought on different feeding stages of pea aphids in the wild-type A17 of Medicago truncatula and ABA signaling pathway mutant sta-1. We examined the impact of drought on plant water status, induced plant defense signaling via the abscisic acid (ABA), jasmonic acid (JA), an

Influence of cell wall polymers and their modifying enzymes during plant-aphid interactions.

Aphids are a major issue for commercial crops. These pests drain phloem nutrients and transmit ~50% of the known insect-borne viral diseases. During aphid feeding, trophic structures called stylets advance toward the phloem intercellularly, disrupting cell wall polymers. It is thought that cell wall-modifying enzymes (CWMEs) present in aphid saliva facilitate stylet penetration through this intercellular polymer network. Additionally, different studies have demonstrated that host settling preference, feeding behavior, and colony performance of aphids are influenced by modulating the CWME expre

Immune activation drives citrus Huanglongbing.

Citrus Huanglongbing (HLB), caused by the phloem-limited bacterium Candidatus Liberibacter asiaticus (CLas) and considered the most destructive citrus disease worldwide, has long been regarded as a typical bacterial infection. However, recent compelling evidence shows that CLas drives HLB through chronic effector-triggered immunity, not via direct pathogenicity, thereby redefining HLB as an immune-mediated disease (Huang et al.).
