Daily melatonin treatments regulate the circadian melatonin rhythm in the adult Djungarian hamster.

The present study tested the hypothesis that daily melatonin treatments influence the biological clock mechanism controlling the circadian melatonin rhythm. Adult male and female Djungarian hamsters in light:dark = 16L:8D (lights on 0300-1900 h) were administered melatonin subcutaneously (s.c.) each day (5 micrograms/0.2 ml saline) in the morning at 1000 h (AM) or late afternoon at 1700 h (PM); controls received a vehicle injection (CON). After 14 days, pineal and serum melatonin concentrations were determined at various times on the last day of treatment and the next day in constant darkness

Phenotypic differences in reentrainment behavior and sensitivity to nighttime light pulses in siberian hamsters.

Spontaneous reentrainment to phase shifts of the photocycle is a fundamental property of all circadian systems. Siberian hamsters are, however, unique in this regard because most fail to reentrain when the LD cycle (16-h light/day) is phase delayed by 5 h. In the present study, the authors compared reentrainment responses in hamsters from 2 colonies. One colony descended from animals trapped in the wild more than 30 years ago (designated "nonentrainers"), and the other colony was outbred as recently as 13 years ago (designated "entrainers"). As reported previously, only 10% of hamsters from th

Effect of various acute 60 Hz magnetic field exposures on the nocturnal melatonin rise in the adult Djungarian hamster.

Acute exposure to a 1 Gauss 60 Hz magnetic field for 15 min beginning 2 hr before darkness delays and blunts the nighttime melatonin rhythm in some but not all studies. To determine whether other exposure parameters (dose, mode, or time) influence the nocturnal melatonin rise, adult Djungarian hamsters reared in long days (16L:8D, lights off at 1000-1800 hr) were acutely exposed to a 60 Hz continuous magnetic field (15 min of 1 or 0.1 Gauss) beginning 4 hr before or 4 hr after lights off. Other hamsters were exposed to a 60 Hz intermittent magnetic field (15 or 60 min of a 1 Gauss field, 1 min

Exogenous T3 mimics long day lengths in Siberian hamsters.

Siberian hamsters (Phodopus sungorus) exhibit seasonal cycles of reproduction driven by changes in day length. Day length is encoded endogenously by the duration of nocturnal melatonin (Mel) secretion from the pineal gland. Short-duration Mel signals stimulate reproduction and long-duration signals inhibit reproduction. The mechanism by which Mel signals are decoded at the level of neural target tissues remains uncharacterized. In Siberian hamsters, exposure to short day lengths or injections of Mel in long days results in a decrease in hypothalamic expression of type 2 iodothyronine deiodinas

Do metabolic responses to solar radiation scale directly with intensity of irradiance?

Endotherms exposed to air temperatures below thermal neutrality reduce their metabolic heat production when exposed to sunlight. The physiological effects of this additional source of heat gain from the environment usually are assumed to be proportional to the intensity of irradiance if other factors are held constant. We test this assumption by measuring changes in metabolic heat production produced by exposing a small mammal, the Siberian hamster (Phodopus sungorus) to four intensities of simulated solar radiation (0 W m-2, 317 W m-2, 634 W m-2 and 950 W m-2). In the absence of solar radiati

Photoperiod control of the melatonin rhythm and reproductive maturation in the juvenile Djungarian hamster: 60-Hz magnetic field exposure effects.

Photoperiodic regulation of the melatonin rhythm was studied in juvenile Djungarian hamsters to test the hypothesis that magnetic field (MF) exposures disrupt sexual maturation. Juveniles in long or short days postweaning were exposed each day to a 1-gauss 60-Hz MF (15 min, 2 h before lights-off). At age 25 days, nighttime duration of the melatonin rise in the pineal gland and circulation of sham-treated controls (adjacent coil system but without current) was < 8 h in long-day juveniles and nearly 13 h in short-day hamsters. This is the first study in juveniles to demonstrate that the melatoni

Maternal photoperiod programs hypothalamic thyroid status via the fetal pituitary gland.

In wild mammals, offspring development must anticipate forthcoming metabolic demands and opportunities. Within species, different developmental strategies may be used, dependent on when in the year conception takes place. This phenotypic flexibility is initiated before birth and is linked to the pattern of day length (photoperiod) exposure experienced by the mother during pregnancy. This programming depends on transplacental communication via the pineal hormone melatonin. Here, we show that, in the Siberian hamster (Phodopus sungorus), the programming effect of melatonin is mediated by the par

Rapid gonadal recrudescence and body and lipid mass increases with hypothalamic lesions in photoregressed Siberian hamsters.

The effects of lesions (x) of the suprachiasmatic nucleus (SCN) or paraventricular nucleus (PVN), or pinealectomy (PINX) on gonadal recrudescence, body and fat pad weights, and food intake were examined in photoregressed male Siberian hamsters (Phodopus sungorus sungorus). Blood was sampled weekly for serum follicle-stimulating hormone (FSH) and prolactin (PRL) measurement. Lesions were classified as complete if greater than 80% of the nuclei were destroyed and designated as 'hits', whereas incomplete lesions were designated as 'misses'. Endocrine mechanisms of seasonal adaptation in small mammals: from early results to present understanding.

Seasonal adaptation is widespread among mammals of temperate and polar latitudes. The changes in physiology, morphology and behaviour are controlled by the photoneuroendocrine system that, as a first step, translates day lengths into a hormonal signal (melatonin). Decoding of the humoral melatonin signal, i.e. responses on the cellular level to slight alterations in signal duration, represents the prerequisite for appropriate timing of winter acclimatization in photoperiodic animals. Corresponding to the diversity of affected traits, several hormone systems are involved in the regulation downs

The circadian cycle of mPER clock gene products in the suprachiasmatic nucleus of the siberian hamster encodes both daily and seasonal time.

The circadian clock in the hypothalamic suprachiasmatic nuclei (SCN) regulates the pattern of melatonin secretion from the pineal gland such that the duration of release reflects the length of the night. This seasonally specific endocrine cue mediates annual timing in photoperiodic mammals. The aim of this study was to investigate how changes in photoperiod influence the cyclic expression of recently identified clock gene products (mPER and mTIM) in the SCN of a highly seasonal mammal, the Siberian hamster (Phodopus sungorus). Immunocytochemical studies indicate that the abundance of both mPER

Pineal and gonadal influences on ultradian locomotor rhythms of male Siberian hamsters.

The extent to which changes in ultradian and circadian rhythms (URs and CRs) reflect seasonal variations in pineal melatonin secretion was assessed in male Siberian hamsters transferred from long to short day lengths. The period of the locomotor activity UR increased from 2.5 h in long days to 4.5 h in short day lengths, but this and most other features of the short-day ultradian phenotype were unaffected by pinealectomy; only the short-day increase in UR amplitude was counteracted by pineal extirpation. Virtually all UR components were unaffected by gonadectomy or replacement testosterone or

Short day lengths alter stress and depressive-like responses, and hippocampal morphology in Siberian hamsters.

Many psychological disorders comprise a seasonal component. For instance, seasonal affective disorder (SAD) is characterized by depression during autumn and winter. Because hippocampal atrophy may underlie the symptoms of depression and depressive-like behaviors, one goal of this study was to determine whether short days also induce structural changes in the hippocampus using photoperiod responsive rodents--Siberian hamsters. Exposure to short days increases depressive-like responses (increased immobility in the forced swim test) in hamsters. Male hamsters were housed in either short (LD 8:16)

Analysis of adrenergic regulation of melatonin synthesis in Siberian hamster pineal emphasizes the role of HIOMT.

Ubiquitin-proteasome system (UPS) dysfunction is associated with the pathology of a wide range of human diseases, including myopathies and muscular atrophy. However, the mechanistic understanding of specific components of the regulation of protein turnover during development and disease progression in skeletal muscle is unclear. Mutations in KLHL40, an E3 ubiquitin ligase cullin3 (CUL3) substrate-specific adapter protein, result in severe congenital nemaline myopathy, but the events that initiate the pathology and the mechanism through which it becomes pervasive remain poorly understood. Establishment and persistence of photoperiodic memory in hamsters.

Long summer days unequivocally stimulate, and short winter days inhibit reproduction in Siberian hamsters. By contrast, intermediate-duration day lengths (12.5-14 h long) either accelerate reproductive development or initiate regression of the reproductive apparatus. Which of these outcomes transpires depends on an animal's photoperiodic history, suggesting that hamsters must encode a representation of prior photoperiods. The duration of nocturnal melatonin secretion is the endocrine representation of day length, but nothing is known about how long it takes to establish photoperiodic histories

Melatonin rhythm onset in the adult siberian hamster: influence of photoperiod but not 60-Hz magnetic field exposure on melatonin content in the pineal gland and in circulation.

To determine the relationship between pineal melatonin production and its appearance in circulation, the rising phase of the pineal and serum melatonin rhythm was studied in the adult Siberian hamster. Melatonin concentrations increased in the pineal gland and in serum at 1.50 and 1.75 h, respectively, relative to lights off in long days (16 h of light/day) and at 2.00 and 2.75 h, respectively, in short days (10 h of light/day). Thus, a photoperiod-dependent melatonin rise in circulation lagged production by the pineal gland by 0.50 h--a delay of 0.75 h in short-day hamsters versus 0.25 h in l

Photoperiod-driven testicular DNA methylation in gonadotropin and sex steroid receptor promoters in Siberian hamsters.

Seasonal cycles in breeding, often orchestrated by annual changes in photoperiod, are common in nature. Here, we studied how change in photoperiod affects DNA methylation in the testes of a highly seasonal breeder: the Siberian hamster (Phodopus sungorus). We hypothesized that DNA methylation in promoter regions associated with key reproductive genes such as follicle-stimulating hormone receptor in the testes is linked to breeding and non-breeding states. Using Oxford Nanopore sequencing, we identified more than 10 million (10,151,742) differentially methylated cytosine-guanine (CpG) sites in

Central angiotensin 1-7 triggers brown fat thermogenesis.

We tested the hypothesis that third ventricular (3V) injections of angiotensin 1-7 (Ang 1-7) increases thermogenesis in brown adipose tissue (BAT), and whether the Mas receptor mediates this response. First, in male Siberian hamsters (n = 18), we evaluated the effect of Ang 1-7 in the interscapular BAT (IBAT) temperature and, using selective Mas receptor antagonist A-779, the role of Mas receptor in this response. Each animal received 3V injections (200 nL), with 48 h intervals: saline; Ang 1-7 (0.03, 0.3, 3, and 30 nmol); A-779 (3 nmol); and Ang 1-7 (0.3 nmol) + A-779 (3 nmol). IBAT temperatu

Photoperiod alters testicular methyltransferase complex mRNA expression in Siberian hamsters.

Exposure to long photoperiods stimulates, whereas exposure to short photoperiods transiently inhibit testicular function in Siberian hamsters via well-described neuroendocrine mechanisms. However, less is known about the intra-testicular regulation of these photoperiod-mediated changes. N6-methyladenosine (m6A) is one of the most common mRNA modifications in eukaryotes, with alterations in m6A mRNA methylation affecting testis function and fertility. We hypothesized that genes controlling m6A methylation such as methyltransferase-like-3 (Mettl3) and -14 (Mettl14) and Wilms' tumor-1 associated

Photoperiod modulates the effects of norepinephrine on lymphocyte proliferation in Siberian hamsters.

Siberian hamsters (Phodopus sungorus) rely on photoperiod to coordinate seasonally appropriate changes in physiology, including immune function. Immunity is regulated, in part, by the sympathetic nervous system (SNS), although the precise role of the SNS in regulating photoperiodic changes in immunity remains unspecified. The goal of the present study was to examine the contributions of norepinephrine (NE), the predominant neurotransmitter of the SNS, to photoperiodic changes in lymphocyte proliferation. In experiment 1, animals were maintained in long [16:8-h light-dark cycle (16:8 LD)] or sh
