Merging photocatalysis with C-H insertions enabled switchable synthesis of indoles and indolines.

C-H insertion offers a powerful route to C-C bond formation, yet it has often proven challenging to integrate into cascade synthetic sequences for further molecular elaboration. This limitation stems primarily from the aggressive conditions required for carbene generation and the high sensitivity of carbene species, which lead to premature termination and incapability with downstream transformations. Although intramolecular C-H insertion provides direct access to indolines, conventional approaches tend to produce ungoverned mixtures of indolines and indoles, owing to harsh conditions. To addre

Photooxidation of Dipyrrinones: Reaction with Singlet Oxygen and Characterization of Reaction Intermediates.

Bilirubin (BR) is a water-insoluble product of heme catabolism in mammals. Elevated blood concentrations of BR, especially in the neonatal period, are treated with blue-green light phototherapy. The major mechanism of BR elimination during phototherapy is photoisomerization, while a minor, less studied mechanism of degradation is oxidation. In this work, we studied the oxidation of the bilirubin model tetramethyl-dipyrrinone (Z-13) by singlet oxygen in methanol using UV-vis and ESI-MS spectroscopy, resulting in propentdyopents as the main oxidation products. We also identified two additional i

Heterogeneous Photochemical Generation of Hydroxyl Radical in Mineral-Organics Systems: Dual Roles of Iron Oxides.

Photosensitive organic molecules and semiconducting minerals have been widely reported to produce hydroxyl radicals (•OH) in aquatic environments. However, studies employing multicomponent systems remain limited. This study investigates the role of minerals in •OH photochemical generation in the presence of organic compounds with distinct functional groups. Using model compounds to represent key moieties in dissolved organic matter (DOM), we examined the capacity of both single-component systems (organic matter and minerals) and composite systems (ferrihydrite-organic matter) to generate •OH.

A facile access to aliphatic trifluoromethyl ketones via photocatalyzed cross-coupling of bromotrifluoroacetone and alkenes.

Biological molecules incorporating trifluoromethyl ketones (TFMKs) have emerged as reversible covalent inhibitors, aiding in the management and treatment of inflammatory diseases, cancer, and respiratory conditions. TFMKs, renowned for their versatile binding properties and adaptability, are pivotal in the rational design of novel drugs for diverse diseases. The photocatalytic insertion of alkenes, abundant feedstocks, into the α-carbon of trifluoromethylacetone represents a highly effective and atom-economical method for synthesizing valuable TFMKs. However, these processes typically necessit

Photoinduced Atom Transfer Radical Addition Reaction of Olefins with α-Bromo Carbonyls.

The irradiation of halogen-bonded complexes with light leads to the homolysis of carbon-halogen bonds and the formation of the corresponding carbon radical species. However, the only methodology reported for these halogen-bonding complexes is using CBr4 as the halogen-bond donor and its applicability is of great interest. In this study, the atom transfer radical addition (ATRA) reaction of olefins using bromomalonates as halogen-bonding donors was developed. Using 4-phenylpyridine as the halogen-bonding acceptor, the desired reaction proceeded well under external irradiation of 380 nm light to

Effects of light irradiation on the complexes of cadmium and humic acids: The role of thiol groups.

In natural environments, humic substances (HS) play a key role in the control of Cd stability by forming Cd-HS complexes. In this study, we investigated the influence of the photo-irradiation on the Cd levels remaining in Leonardite humic acid (HA) standard solution after chemical precipitation. The kinetic experiment showed that after the irradiation of ultraviolet light, especially UVC, Cd-HA complexes become more stable. The coexisting Cd in the HA solution under light irradiation seemed to enhance the stability of Cd-HA complexes. Both Fourier transform infrared and X-ray photoelectron spe

Gas-Phase Photochemical Overall H2 S Splitting by UV Light Irradiation.

Splitting of hydrogen sulfide is achieved to produce value-added chemicals. Upon irradiation at 254 nm in the gas phase and in the absence of catalysts or photocatalysts at near room temperature, H2 S splits into stoichiometric amounts of H2 and S with a quantum efficiency close to 50 %. No influence of the presence of CH4 and CO2 (typical components in natural gas and biogas in which H2 S is an unwanted component) on the efficiency of overall H2 S splitting was observed. A mechanism for the H2 and S formation is proposed.

A photoredox-neutral Smiles rearrangement of 2-aryloxybenzoic acids.

We report on the use of visible light photoredox catalysis for the radical Smiles rearrangement of 2-aryloxybenzoic acids to obtain aryl salicylates. The method is free of noble metals and operationally simple and the reaction can be run under mild batch or flow conditions. Being a redox neutral process, no stoichiometric oxidants or reductants are needed.

Photophysics of a coumarin in different solvents: use of different solvatochromic models.

This study reported the photophysics of 7-(diethylamino)coumarin-3-carboxylic acid N-succinimidyl ester (7-DCCAE) in different neat solvents of varying polarity using steady-state absorption, fluorescence emission and picosecond time-resolved spectroscopy. In nonpolar solvents, the dye molecule predominantly exists in nonpolar structure and exhibits very low value of nonradiative decay rate constant (k(nr)), demonstrating the emission takes place from S(1) -LE to S(0) ground state. The fluorescence quantum yields, lifetime values of 7-DCCAE in different solvents are rationalized on the basis o

Calorimetric determination of rate constants and enthalpy changes for zero-order reactions.

Calorimetry is a general method for determination of the rates of zero-order processes, but analysis of the data for the rate constant and reaction enthalpy is difficult because these occur as a product in the rate equation so evaluation of one requires knowledge of the other. Three methods for evaluation of both parameters, without prior knowledge, are illustrated with examples and compared with literature data. Method 1 requires the reaction to be studied in two buffers with different enthalpies of ionization. Method 2 is based on calculation of reaction enthalpy from group additivity functi

Photochemically induced radical transformation of C(sp3)-H bonds to C(sp3)-CN bonds.

A general protocol for direct transformation of unreactive C(sp(3))-H bonds to C(sp(3))-CN bonds has been developed. The C-H activation was effected by photoexcited benzophenone, and the generated carbon radical was subsequently trapped with tosyl cyanide to afford the corresponding nitrile in a highly efficient manner. The present methodology is widely applicable to versatile starting materials and, thus, serves as a powerful tool for selective one-carbon elongation for construction of architecturally complex molecules.

Photoredox-Catalyzed Lysine C(sp3)-H Functionalization for Peptide Editing.

The selective chemical modification of lysine residues beyond its ε-amino group remains a long-standing challenge in peptide synthesis due to the conformational flexibility of the residue, the nucleophilicity of the amine group, and the inert C(sp3)-H bonds of the aliphatic side chain. Herein we report a photoredox-catalyzed reaction for site-selective C(sp3)-H functionalization of lysine and other primary amine residues through α-amino radical intermediates generated from trifluoroacetamide protecting groups. This transformation proceeds under mild conditions and exhibits broad tolerance to r

The Human Circadian Clock Protein Cryptochrome-1 Responds to Photochemistry by Conformational Changes.

The human clock protein cryptochrome-1 (HsCRY1) can be targeted by small molecular modulators to shift the 24 h rhythm, opening up possibilities for treating circadian disorders. GO1323 is a photopharmacological compound that selectively binds to HsCRY1 and enables light-controlled manipulation of the circadian rhythm via photoisomerization. We synthesized isotope-labeled and unlabeled GO1323 to investigate conformational changes in full-length HsCRY1 that are caused by isomerization of GO1323 using infrared difference spectroscopy. Activation of GO1323 induces specific changes in secondary st

Discovery and engineering of enzymes for new-to-nature photobiocatalysis.

Photobiocatalysis integrates enzymatic catalysis with photochemistry, enabling challenging radical transformations with high selectivity under mild conditions. Early developments in this field were largely driven by the discovery that enzyme-bound cofactors can form photoactive charge-transfer complexes with substrates, thereby initiating radical chemistry upon light irradiation. Recent advances, however, have substantially expanded the mechanistic landscape of photobiocatalysis through diverse mechanisms. This review summarizes major developments in photobiocatalysis reported since 2024. Rath

Environment-dependent photochemical behavior of adenosylcobalamin and its influence on the quenching of the 5'-deoxyadenosyl radical.

Adenosylcobalamin (AdoCbl) shows different photochemical behavior when free in solution compared to when it is bound to the photoregulatory protein CarH. In both cases, the initial photoproduct is cob(II)alamin and the 5'-deoxyadenosyl radical (Rios-Santacruz et al., Nature, 2026, 650, 1045-1052). However, in solution, the ultimate products are cob(II)alamin and the cyclic product, 5',8-cycloadenosine, while in the CarH protein, cob(I)alamin and the exocyclic methylene product 4',5'-anhydroadenosine are formed. This has led to speculation that alkene formation in CarH plays a protective role,

Photoisomerization and Kinetic Characterization of Molecular Photoswitches Using a Semi-Automated Experimental System.

Molecular photoswitches occur widely in nature, where they play key roles in vision, and are increasingly utilized in emerging technologies. Their development, however, is often limited by the labor-intensive characterization of photochemical properties. This work presents an automated flow-based platform for efficient and reproducible photochemical characterization of molecular photoswitches. To accurately measure photoisomerization quantum yields, absorption profiles, and thermal back-conversion kinetics, the setup combines in-line UV-Vis spectroscopy with a programmable LED array. Recent advances in chemoselective protein desulfurization: from VA-044 to emerging boron, photochemical, iron-catalyzed, and ultrasound methods.

Native chemical ligation (NCL) represents a powerful strategy for the chemical synthesis of tailor-made proteins that are difficult to obtain via biological expression approaches. Early desulfurization methods, particularly VA-044-based desulfurization, have profoundly advanced the field by enabling the post-ligation conversion of cysteine to alanine, thereby expanding the sequence space accessible for protein synthesis. In recent years, a variety of innovative desulfurization technologies have emerged that achieve superior reaction kinetics, broader functional group compatibility or eliminati
