The S1 to S2 and S2 to S3 state transitions in plant photosystem II: relevance to the functional and structural heterogeneity of the water oxidizing complex.

In Photosystem II, light-induced water splitting occurs via the S state cycle of the CaMn4O5-cluster. To understand the role of various possible conformations of the CaMn4O5-cluster in this process, the temperature dependence of the S1 → S2 and S2 → S3 state transitions, induced by saturating laser flashes, was studied in spinach photosystem II membrane preparations under different conditions. The S1 → S2 transition temperature dependence was shown to be much dependent on the type of the cryoprotectant and presence of 3.5% methanol, resulting in the variation of transition half-inhibition temp

Evaluation of an oil-producing green alga Chlorella sp. C2 for biological DeNOx of industrial flue gases.

NOx, a significant portion of fossil fuel flue gases, are among the most serious environmental issues in the world and must be removed in an additional costly gas treatment step. This study evaluated the growth of the green alga Chlorella sp. C2 under a nitrite-simulated NOx environment and the removal rates of actual flue gas fixed salts (FGFSs) from Sinopec's Shijiazhuang refinery along with lipid production. The results showed that nitrite levels lower than 176.5 mM had no significant adverse effects on the cell growth and photosynthesis of Chlorella sp. C2, demonstrating that this green al

The effects of light-induced reduction of the photosystem II reaction center.

Accumulation of reduced pheophytin a (Pheo-D1) in photosystem II reaction center (PSII RC) under illumination at low redox potential is accompanied by changes in absorbance and circular dichroism spectra. The temperature dependences of these spectral changes have the potential to distinguish between changes caused by the excitonic interaction and temperature-dependent processes. We observed a conformational change in the PSII RC protein part and changes in the spatial positions of the PSII RC pigments of the active D1 branch upon reduction of Pheo-D1 only in the case of high temperature (298 K

Expression of the psbA gene during photoinhibition and recovery in Synechocystis PCC 6714: inhibition and damage of transcriptional and translational machinery prevent the restoration of photosystem II activity.

The D1 reaction center protein of the photosystem II complex is very sensitive to light. It is continuously being damaged, degraded and resynthesized. Under high light, photosystem II inactivation is observed. This is because the rate of D1 damage is faster than that of its replacement. This process can be reversed if exposure to high light is not too long. In this work we study the changes that occur in the transcriptional and translational machinery that could lead to irreversible photoinhibition in Synechocystis PCC 6714. In the first minutes of photoinhibition, high light induced an accumu

Protein film photoelectrochemistry of the water oxidation enzyme photosystem II.

Photosynthesis is responsible for the sunlight-powered conversion of carbon dioxide and water into chemical energy in the form of carbohydrates and the release of O2 as a by-product. Although many proteins are involved in photosynthesis, the fascinating machinery of Photosystem II (PSII) is at the heart of this process. This tutorial review describes an emerging technique named protein film photoelectrochemistry (PF-PEC), which allows for the light-dependent activity of PSII adsorbed onto an electrode surface to be studied. The technique is straightforward to use, does not require highly speci

Water-splitting manganese complex controls light-induced redox changes of cytochrome b559 in photosystem II.

The effect of water-splitting Mn complex on light-induced redox changes of cytochrome b(559) (cyt b(559)) was studied in spinach photosystem II (PSII) membranes. Photoreduction of the heme iron in the intact PSII membranes was completely suppressed by DCMU, whereas photoreduction and photooxidation of the heme iron in the Mn-depleted PSII membranes were unaffected by DCMU. Interestingly, photoreduction and photooxidation of the heme iron in the Mn-depleted PSII membranes were completely diminished by exogenous superoxide dismutase (SOD), whereas no effect of SOD on photoreduction of the heme i

PfsR is a key regulator of iron homeostasis in Synechocystis PCC 6803.

Iron is an essential cofactor in numerous cellular processes. The iron deficiency in the oceans affects the primary productivity of phytoplankton including cyanobacteria. In this study, we examined the function of PfsR, a TetR family transcriptional regulator, in iron homeostasis of the cyanobacterium Synechocystis PCC 6803. Compared with the wild type, the pfsR deletion mutant displayed stronger tolerance to iron limitation and accumulated significantly more chlorophyll a, carotenoid, and phycocyanin under iron-limiting conditions. The mutant also maintained more photosystem I and photosystem

Unraveling design principles of protein landscapes in photosynthetic membranes in plant chloroplasts.

The supramolecular organization of proteins within photosynthetic membranes is crucial for energy conversion in plants. Here, we introduce an analytical and computational pipeline that integrates high-resolution cryo-scanning electron microscopy, biochemical quantification, advanced Monte Carlo computer simulations, and statistical methods to elucidate the elusive protein landscapes of grana membranes in intact Arabidopsis leaves. Our integrated analysis challenges the prevailing view that particles on the exoplasmic fracture faces in freeze-fracture samples represent photosystem II exclusivel

A siphonous morphology affects light-harvesting modulation in the intertidal green macroalga Bryopsis corticulans (Ulvophyceae).

The macroalga Bryopsis corticulans relies on a sustained protective NPQ and a peculiar body architecture to efficiently adapt to the extreme light changes of intertidal shores. During low tides, intertidal algae experience prolonged high light stress. Efficient dissipation of excess light energy, measured as non-photochemical quenching (NPQ) of chlorophyll fluorescence, is therefore required to avoid photodamage. Light-harvesting regulation was studied in the intertidal macroalga Bryopsis corticulans, during high light and air exposure. Photosynthetic capacity and NPQ kinetics were assessed in

Effect of chromium oxide (III) nanoparticles on the production of reactive oxygen species and photosystem II activity in the green alga Chlamydomonas reinhardtii.

With the growth of nanotechnology and widespread use of nanomaterials, there is an increasing risk of environmental contamination by nanomaterials. However, the potential implications of such environmental contamination are hard to evaluate since the toxicity of nanomaterials if often not well characterized. The objective of this study was to evaluate the toxicity of a chromium-based nanoparticle, Cr2O3-NP, used in a wide diversity of industrial processes and commercial products, on the unicellular green alga Chlamydomonas reinhardtii. The deleterious impacts of Cr2O3-NP were characterized usi

Electrogenic reactions on the donor side of Mn-depleted photosystem II core particles in the presence of MnCl2 and synthetic trinuclear Mn-complexes.

An electrometric technique was used to investigate the generation of a photovoltage (Deltapsi) by Mn-depleted spinach photosystem II (PS II) core particles incorporated into liposomes. In the presence of MnCl2, the fast kinetically unresolvable phase of Deltapsi generation, related to electron transfer between the redox-active tyrosine YZ and the primary plastoquinone acceptor QA was followed by an additional electrogenic phase (tau approximately 20 micros, approximately 5% of the phase attributed to YZoxQA-). The latter phase was ascribed to the transfer of an electron from the Mn, bound to t

Herbicidal action of 2-hydroxy-3-alkyl-1,4-naphthoquinones.

The main mode of herbicidal activity of 2-hydroxy-3-alkyl-1,4-naphthoquinones is shown to be inhibition of photosystem II (PSII). The herbicidal and in vitro activities have been measured and correlated with their (Log)octanol/water partition coefficients (Log Ko/w). The length of the 3-n-alkyl substituent for optimal activity differed between herbicidal and in vitro activity. The maximum in vitro activity was given by the nonyl to dodecyl homologues (Log Ko/w between 6.54 and 8.12), whereas herbicidal activity peaked with the n-hexyl compound (Log Ko/w = 4.95). The effect of chain branching w

Using ΦPSII and leaf temperature as indicators of non-steady-state photosynthesis and stomatal conductance during stepwise changes in light intensity.

Quantifying the kinetics of net CO2 assimilation (A) and stomatal conductance (gs) under fluctuating light typically relies on gas exchange measurements, which are slow and thus unsuited for high-throughput phenotyping. As a result, faster, non-invasive phenotyping methods are needed to further evaluate these traits at a larger scale. However, first the relationship between non-steady-state parameters must be examined in greater detail. In this study, we aimed to determine whether variations in non-steady-state values of chlorophyll fluorescence and leaf temperature reflect differences in key

Glutamate and Glutamine Enable Rapid Acclimation of Haematococcus lacustris to High-Light During Culture Phase Shift by Pre-Activating Photoprotective Pathways.

Microalga Haematococcus lacustris is known for its ability to produce high-value product astaxanthin. The abrupt shift from weak-light in the vegetative phase to high-light in the astaxanthin induction phase triggers severe photodamage during the large-scale cultivation of H. lacustris for astaxanthin production in two phases, causing photobleaching with resultant biomass loss. Current mitigation relies primarily on physical shading. This study found that glutamate/glutamine supplementation before phase shift mitigated the photodamage caused by sudden high-light stress in H. lacustris. Glutama

Effect of different concentrations of foliar iron fertilizer on chlorophyll fluorescence characteristics of iron-deficient rice seedlings under saline sodic conditions.

The effectiveness of iron is reduced in saline conditions, which can easily lead to iron deficiency and inhibit photosynthesis in rice. In this study, 4-week-old Fe-deficient rice seedlings were treated under saline sodic stress (50 mM) to different concentrations (0, 0.2%, 0.4%, 0.8%, 1.6%, and 3.2%) of foliar iron fertilizer (FeEDDHA). Differences in prompting fluorescence and the MR820 signal of rice leaves after 7 days of treatment were probed using the JIP-test. The results show that the performances of the two rice varieties were in general agreement. Under iron deficiency and soda salin

The Effect of Abiotic Factors on Abundance and Photosynthetic Performance of Airborne Cyanobacteria and Microalgae Isolated from the Southern Baltic Sea Region.

Cyanobacteria and microalgae present in the aquatic or terrestrial environment may be emitted into the air and transported along with air masses over long distances. As a result of staying in the atmosphere, these organisms may develop a greater tolerance to stressful factors, but this topic is still relatively unknown. The main aim was to show an autecological characteristic of some airborne microalgae and cyanobacteria strains by a factorial laboratory experiment approach, including changes in irradiance, temperature, and salinity conditions. The additional purpose of this work was also to p

Changes in antenna of photosystem II induced by short-term heating.

Changes in antenna of photosystem II, induced by short-term heating, were studied using characteristics of a short-wavelength band in low-temperature fluorescence spectra (77 K) of pea chloroplasts. Heating for 5 min was carried out at 25 and 45°C in the darkness or in the presence of white light with intensity of 260 or 1,400 μmol/m(2)s. Most modes of thermal treating induced a decrease in integral intensity of the band and an increase of its half-width. The changes were more prominent at high-temperature heating. The second derivative of the contour of a short-wavelength band showed its thre

The effect of cactus spines on light interception and Photosystem II for three sympatric species of Opuntia from the Mojave Desert.

Cactus spines reduce herbivory, direct water toward roots and reduce the impacts of high- and low-temperature extremes. Yet, shading of stems by spines reduces incident photosynthetic photon flux density (PFD), photosynthesis and growth. This study compared spinescence, PFD interception, stem temperature, Photosystem II (PSII) photochemistry and xanthophyll pigment composition for three species of cacti from the Mojave Desert, CA. The species vary in spinescence: Opuntia basilaris, which has no central or radial spines; Opuntia erinacea, which is densely covered with spines; and Opuntia phaeac

Secondary quinone in photosystem II of Thermosynechococcus elongatus: semiquinone-iron EPR signals and temperature dependence of electron transfer.

The secondary quinone acceptor, Q(B), has been studied in photosystem II (PSII) isolated from Thermosynechococcus (T.) elongatus. Thermoluminescence indicated that Q(B) was present in this preparation. An EPR signal observed at low temperature at g = 1.9 was attributed to Fe2+ Q(B)- on the basis of the characteristic period-of-two variations in its intensity depending on the number of laser flashes given at 20 degrees C. When samples showing the Fe2+ Q(B)- signal were illuminated at 77 K, an EPR signal at g = 1.66 appeared with an amplitude proportional to that of the Fe2+ Q(B)- signal. This s
