Fluid, electrolyte, and acid-base balances in three-day, combined-training horses.

Horses competing in 3-day, combined-training events develop a metabolic acidosis that is partially compensated for by a respiratory alkalosis immediately after phases B and D. By the end of phase C and 30 minutes to 2 hours after phase D, the acidosis is resolved by the oxidation of lactate, and a metabolic alkalosis prevails. A reduction in TBW and cation content occurs, which often is not replenished 12 to 24 hours after the event, even though the serum or plasma concentration of various constituents may be within normal limits. Hypochloremia and hypocalcemia, however, may persist 12 or more

Different outcomes of endurance and resistance exercise in skeletal muscles of Oculopharyngeal muscular dystrophy.

BACKGROUND: Exercise is widely considered to have beneficial impact on skeletal muscle aging. In addition, there are also several studies demonstrating a positive effect of exercise on muscular dystrophies. Oculopharyngeal muscular dystrophy (OPMD) is a late-onset autosomal dominant inherited neuromuscular disorder caused by mutations in the PAPBN1 gene. These mutations consist in short (1-8) and meiotically stable GCN trinucleotide repeat expansions in its coding region responsible for the formation of PAPBN1 intranuclear aggregates. This study aims to characterize the effects of two types of

Intermittent hypobaric hypoxia combined with aerobic exercise improves muscle morphofunctional recovery after eccentric exercise to exhaustion in trained rats.

Unaccustomed eccentric exercise leads to muscle morphological and functional alterations, including microvasculature damage, the repair of which is modulated by hypoxia. We present the effects of intermittent hypobaric hypoxia and exercise on recovery from eccentric exercise-induced muscle damage (EEIMD). Soleus muscles from trained rats were excised before (CTRL) and 1, 3, 7, and 14 days after a double session of EEIMD protocol. A recovery treatment consisting of one of the following protocols was applied 1 day after the EEIMD: passive normobaric recovery (PNR), a 4-h daily exposure to passiv

Magnesium sulfate enhances exercise performance and manipulates dynamic changes in peripheral glucose utilization.

The effect of magnesium supplementation on exercise performance remains controversial. In the present study, the effects of magnesium sulfate on exercise performance and blood glucose metabolism were examined. In order to provide a non-invasive measure of continuous exercise, we developed an auto-blood sampling system was coupled to a microdialysis analyzer to detect the dynamic changes in glucose metabolism in conscious and freely moving gerbils subjected to forced swimming. Gerbils were pretreated with saline or magnesium sulfate (90 mg kg(-1), ip) 30 min before exercise. The duration times

Influence of voluntary exercise on hypothalamic norepinephrine.

We combined hypothalamic tissue and plasma determinations of norepinephrine, dihydroxyphenylalanine, and dihydroxyphenylglycol with measurements of abdominal fat in voluntary running rats to examine the relationship among exercise training, hypothalamic and sympathetic nervous function, and body fat stores. The hypothalamic concentrations of norepinephrine, dihydroxyphenylalanine, and dihydroxyphenylglycol were reduced after exercise training (P < 0.01), with the amount of norepinephrine being strongly associated with the plasma norepinephrine (r = 0.58, P < 0.05) and dihydroxyphenylglycol (r

Immunology of Physical Exercise: Is Equus caballus an Appropriate Animal Model for Human Athletes?

Domestic horses routinely participate in vigorous and various athletic activities. This enables the horse to serve as a model for studying athletic physiology and immunology in other species, including humans. For instance, as a model of physical efforts, such as endurance rides (long-distance running/aerobic exercise) and races (anaerobic exercise), the horse can be useful in evaluating post-exercise response. Currently, there has been significant interest in finding biomarkers, which characterize the advancement of training and adaptation to physical exercise in the horse. The parallels in c

The effects of short-term consumption of a Western diet on aerobic exercise performance in bank voles with inherently distinct metabolic rates.

Because obesity and related diseases are partly attributed to unrestricted consumption of a high-fat, high-sugar Western diet (WD), several studies have examined its adverse effects on health and physical fitness. However, short-term WD supplementation has received little attention. We asked whether such supplementation can improve the aerobic exercise performance of animals with inherently distinct aerobic capacity. We used bank voles [Clethrionomys (= Myodes) glareolus] from lines selected for high swim-induced aerobic metabolism (A) and unselected control lines (C). In a crossover design, w

Short-term exercise counteracts accelerated ageing impacts on physical performance and liver health in mice.

Senescence impairs liver physiology, mitochondrial function and circadian regulation, resulting in systemic metabolic dysregulation. Given the limited research on the effects of combined exercise on an ageing liver, this study aimed to evaluate its impact on liver metabolism, circadian rhythms and mitochondrial function in senescence-accelerated mouse-prone 8 (SAMP8) and senescence-accelerated mouse-resistant 1 (SAMR1) mice. Histological, reverse transcription quantitative polymerase chain reaction (RT-qPCR) and immunoblotting analyses were conducted, supplemented by transcriptomic data sets a

Impaired muscle force production and higher fatigability in a mouse model of sickle cell disease.

Skeletal muscle function has been scarcely investigated in sickle cell disease (SCD) so that the corresponding impact of sickle hemoglobin is still a matter of debate. The purpose of this study was to investigate muscle force production and fatigability in SCD and to identify whether exercise intensity could have a modulatory effect. Ten homozygous sickle cell (HbSS), ten control (HbAA) and ten heterozygous (HbAS) mice were submitted to two stimulation protocols (moderate and intense) to assess force production and fatigability. We showed that specific maximal tetanic force was lower in HbSS m

Chronic exercise prevents repeated restraint stress-provoked enhancement of immobility in forced swimming test in ovariectomized mice.

We assessed whether chronic treadmill exercise attenuated the depressive phenotype induced by restraint stress in ovariectomized mice (OVX). Immobility of OVX in the forced swimming test was comparable to that of sham mice (CON) regardless of the postoperative time. Immobility was also no difference between restrained mice (exposure to periodic restraint for 21 days; RST) and control mice (CON) on post-exposure 2nd and 9th day, but not 15th day. In contrast, the immobility of ovariectomized mice with repeated stress (OVX + RST) was profoundly enhanced compared to ovariectomized mice-alone (OVX

Vagus nerve stimulation during rehabilitative training enhances recovery of forelimb function after ischemic stroke in aged rats.

Advanced age is associated with a higher incidence of stroke and worse functional outcomes. Vagus nerve stimulation (VNS) paired with rehabilitative training has emerged as a potential method to improve recovery after brain injury but to date has only been evaluated in young rats. Here, we evaluated whether VNS paired with rehabilitative training would improve recovery of forelimb function after ischemic lesion of the motor cortex in rats 18 months of age. Rats were trained to perform the isometric pull task, an automated, quantitative measure of volitional forelimb strength. Once proficient,

Animal Models of Exercise and Cardiometabolic Disease.

Cardiometabolic diseases, encompassing cardiovascular and metabolic disorders, represent a significant global health challenge, driven in part by the rising prevalence of obesity and type 2 diabetes. Exercise offers profound benefits in mitigating the progression and impact of cardiometabolic diseases by improving glucose homeostasis, body composition, and cardiovascular function. However, gaps remain in our understanding of the molecular mechanisms underlying these benefits and how to optimize exercise prescriptions for cardiometabolic disease. Animal models are indispensable tools for unrave

Don't fence me in: managing psychological well being for elite performance horses.

This article posits that stereotypical behavior patterns and the overall psychological well being of today's performance horse could be substantially enhanced with care that acknowledges the relationship between domesticated horses and their forerunners. Feral horses typically roam in stable, social groups over large grazing territories, spending 16-20 hr per day foraging on mid- to poor-quality roughage. In contrast, today's elite show horses live in relatively small stalls, eat a limited-but rich-diet at specific feedings, and typically live in social isolation. Although the horse has been d

Thoroughbred horses susceptible to recurrent exertional rhabdomyolysis have elevated skeletal muscle mitochondrial capacities.

Recurrent exertional rhabdomyolysis (RER) is a skeletal muscle disease that can intermittently impede careers of equine athletes. Manifesting as episodes of muscular stiffness and degeneration, RER causes lost training and competition days. RER is associated with abnormal intracellular calcium (Ca2+) regulation. Calcium released from the sarcoplasmic reticulum to initiate contraction may also be transported into mitochondria, which impacts energy production. RER-susceptible horses exhibit downregulated mitochondrial proteins but upregulated mitochondrial genes. Thus, mitochondrial function may

Metabolomics Investigation of a Multi-Ingredient Supplements-Modulated Changes in Metabolism in Mice After Endurance Exercise.

Energy metabolism manipulation strategy, enhancing fat metabolism while reducing the dependence on glucose, is beneficial to improve exercise performance. The multi-ingredient supplements (MIS) mixed with valine, isoleucine, leucine, β-alanine, creatine, L-carnitine, quercetin, and betaine were found to be able to improve the endurance performance of mice, which was associated with a shift of energy substrates from glucose to fatty acids. Thus, we hypothesized that the MIS regulating lipid metabolism contributes to enhancing exercise endurance in mice. The present study aimed to explore the al

The effects of endurance trainability phenotype, sex, and interval running training on bone collagen synthesis in adult rats.

Bone is influenced by many factors such as genetics and mechanical loading, but the short-term physiological effects of these factors on bone (re)modelling are not well characterised. This study investigated the effects of endurance trainability phenotype, sex, and interval running training (7-week intervention) on bone collagen formation in rats using a deuterium oxide stable isotope tracer method. Bone samples of the femur diaphysis, proximal tibia, mid-shaft tibia, and distal tibia were collected after necropsy from forty-six 9 ± 3-month male and female rats selectively bred for yielding lo

Pre-race and race management impacts serum muscle enzyme activity in Australian endurance horses.

BACKGROUND: Marked increases in serum muscle enzyme activity can occur in endurance horses but the diagnostic certainty in predicting cases of myopathy is unclear. Improved understanding of horse management effects on serum muscle enzyme activity as markers of muscle health would assist interpretation of serum muscle enzyme activity and guide management to reduce myopathy risk. OBJECTIVES: To investigate associations between serum muscle enzyme activity and management factors in endurance horses. STUDY DESIGN: Cross-sectional study. METHODS: One hundred endurance horses competing in four en
