Comparative analysis of Phytophthora genes encoding secreted proteins reveals conserved synteny and lineage-specific gene duplications and deletions.

Comparative analysis of two Phytophthora genomes revealed overall colinearity in four genomic regions consisting of a 1.5-Mb sequence of Phytophthora sojae and a 0.9-Mb sequence of P. ramorum. In these regions with conserved synteny, the gene order is largely similar; however, genome rearrangements also have occurred. Deletions and duplications often were found in association with genes encoding secreted proteins, including effectors that are important for interaction with host plants. Among secreted protein genes, different evolutionary patterns were found. Elicitin genes that code for a comp

Transcriptome reveals BCAAs biosynthesis pathway is influenced by lovastatin and can act as a potential control target in Phytophthora sojae.

AIMS: Lovastatin has been indicated to impair growth and development of Phytophthora sojae. Therefore, this study was performed to understand the inhibitory mechanism of lovastatin and investigate the metabolic pathway potentially served as a new control target for this plant pathogen. METHODS AND RESULTS: Whole transcriptome analysis of lovastatin-treated P. sojae was performed by RNA-sequencing. The results revealed that 84 genes were upregulated and 58 were downregulated with more than fourfold changes under treatment. Kyoto Encyclopaedia of Genes and Genomes analysis indicated that the br

AtRTP5 negatively regulates plant resistance to Phytophthora pathogens by modulating the biosynthesis of endogenous jasmonic acid and salicylic acid.

Plants have evolved powerful immune systems to recognize pathogens and avoid invasions, but the genetic basis of plant susceptibility is less well-studied, especially to oomycetes, which cause disastrous diseases in many ornamental plants and food crops. In this research, we identified a negative regulator of plant immunity to the oomycete Phytophthora parasitica, AtRTP5 (Arabidopsis thaliana Resistant to Phytophthora 5), which encodes a WD40 repeat domain-containing protein. The AtRTP5 protein, which was tagged with green fluorescent protein (GFP), is localized in the nucleus and plasma membr

Mining Microbial Niches: Sources of Bacteria for Enhancing Plant Growth and Resilience to pH, Salinity, Drought and Phytophthora infestans.

Enhancing plant resilience is crucial for sustainable agriculture and ecosystem health in an era of climate change and the global spread of plant pathogens. The strategic exploitation of diverse ecological niches to isolate microorganisms that promote plant growth, suppress pathogens and tolerate abiotic stress conditions is paramount. Here, 21 bacterial strains were isolated from three distinct sources: digestate produced from straw feedstock, Quercus ilex roots mycorrhised with Tuber aestivum, and a peperino stone fountain. These isolates were assigned to eight genera, Bacillus, Pseudomonas,

The expression of the NPR1-dependent defense response pathway genes in Persea americana (Mill.) following infection with Phytophthora cinnamomi.

A plant's defense against pathogens involves an extensive set of phytohormone regulated defense signaling pathways. The salicylic acid (SA)-signaling pathway is one of the most well-studied in plant defense. The bulk of SA-related defense gene expression and the subsequent establishment of systemic acquired resistance (SAR) is dependent on the nonexpressor of pathogenesis-related genes 1 (NPR1). Therefore, understanding the NPR1 pathway and all its associations has the potential to provide valuable insights into defense against pathogens. The causal agent of Phytophthora root rot (PRR), Phytop

An RLP23/Cf-9TM-IC Chimeric Receptor Enhances nlp24-Triggered Immunity and Resistance to Phytophthora nicotianae in Nicotiana benthamiana.

Receptor-like protein AtRLP23 from Arabidopsis thaliana perceives the conserved microbial peptide nlp24 derived from necrosis- and ethylene-inducing peptide 1-like proteins (NLPs), but immune outputs are often weak in heterologous systems. Here, we engineered RLP23/Cf-9 chimeric receptors by fusing the extracellular domain of AtRLP23 to either the Cf-9 intracellular region (IC) or the Cf-9 transmembrane-intracellular region (TM-IC), and assessed their activity in Nicotiana benthamiana. In transient expression assays, AtRLP23 alone did not trigger hypersensitive cell death after infiltration wi

Reduced soil microbial diversity and altered co-occurrence networks associated with Phytophthora nicotianae infection in flue-cured tobacco fields.

Phytophthora nicotianae is a persistent and highly destructive soilborne oomycete that causes black shank of tobacco and continues to threaten its production despite integrated management. Although soil microbiomes are increasingly recognized as important components of disease outcomes, little is known about how soil microbial communities vary with P. nicotianae DNA concentrations and black shank pressure under field conditions. We examined relationships among 17 soil physicochemical properties, microbial community features, P. nicotianae DNA levels, and black shank outcomes in three flue-cure

From sea to farm: Repurposing invasive macroalgae into biocidal and defense-enhancing tools against tomato late blight (Phytophthora infestans).

Marine seaweeds are increasingly recognized as sources of bioactive compounds with potential applications in sustainable agriculture, including as biocontrol agents. The selection of invasive species such as Asparagopsis armata and Sargassum muticum offers a dual benefit: obtaining bioactive metabolites from a low-cost resource while simultaneously mitigating their ecological impacts using this harmful biomass. In the present study, crude extracts from A. armata and S. muticum were applied to tomato plants and were effective as biocontrol agents against Phytophthora infestans. Treatments modul

The potato StMKK5-StSIPK module enhances resistance to Phytophthora pathogens through activating the salicylic acid and ethylene signalling pathways.

Mitogen-activated protein kinase (MAPK) cascades play pivotal roles in plant responses to both biotic and abiotic stress. A screen of a Nicotiana benthamiana cDNA virus-induced gene silencing (VIGS) library for altered plant responses to inoculation with Phytophthora infestans previously identified an NbMKK gene, encoding a clade D MAPKK that we renamed as NbMKK5, which is involved in immunity to P. infestans. To study the role of the potato orthologous gene, referred to as StMKK5, in the response to P. infestans, we transiently overexpressed StMKK5 in N. benthamiana and observed that cell dea

First Report of Late Blight Caused by Phytophthora infestans Clonal Lineage US-24 on Potato (Solanum tuberosum) in Wisconsin.

Potato (Solanum tuberosum) crops are grown on over 25,090 ha in Wisconsin annually. Late blight, caused by Phytophthora infestans (Mont.) deBary, is a potentially devastating disease that affects tomato and potato crops in Wisconsin every few years when inoculum is introduced and weather conditions favor disease. Incidence and severity of late blight are highly variable in these few years due to differences in pathogen clonal lineages, their timing and means of introduction, and weather conditions. Prevention of this disease through prophylactic fungicide application can cost producers million

Computationally and Target-Guided Discovery of Trifluoromethyl-Pyrazole Derivatives Targeting the a-c Subunit Interface of V-ATPase in Phytophthora capsici.

Vacuolar H+-ATPase (V-ATPase) is essential for vacuolar acidification, ion homeostasis, and development in various organisms, including plant pathogens. The V-ATPase a-subunit has been recently identified as the target protein of the oomycete inhibitor fluopicolide, highlighting the favorable druggability of V-ATPase. In this study, we propose a structure-based virtual screening approach to identify novel bioactive compounds targeting the a-c subunit interface of V-ATPase. Following multistep docking and molecular mechanics/generalized born surface area (MM/GBSA) analysis, compound a10 was ide

A quantitative Real Time PCR based method for the detection of Phytophthora infestans causing Late blight of potato, in infested soil.

A fast and simple polymerase chain reaction method has been developed for detection of Phytophthora infestans oospores, the causal agent of Late blight of Potato in soil. The method involves the disruption of oospores by grinding dry soil, using abrasive properties, in the presence of glass powder and skimmed milk powder within a short time. The latter prevents loss of DNA by adsorption to soil particles or by degradation and reduces the co-extraction of PCR inhibitors with the DNA. After phenol/chloroform extraction; the DNA is suitable for direct PCR amplification without a precipitation ste

Down-regulation of defense genes and resource allocation into infected roots as factors for compatibility between Fagus sylvatica and Phytophthora citricola.

Phytophthora citricola is a wide spread and highly aggressive pathogen of Fagus sylvatica. The hemibiotrophic oomycete infects the roots and establishes a compatible interaction with F. sylvatica. To investigate the transcriptional changes associated with P. citricola infection, 68 custom oligo-microarray measurements were conducted. Hierarchical as well as non-hierarchical clustering was carried out to analyze the expression profiles. Experimental setup includes a time scale covering the biotrophic and necrotrophic stages of interaction as well as comparative analyses of the local and systemi

Species-specific divergence in Phytophthora infestans resistance between Nicotiana benthamiana and potato mediated by 1-aminocyclopropane-1-carboxylic acid pretreatment.

BACKGROUND: Late blight, caused by Phytophthora infestans, is a devastating oomycete disease threatening global potato (Solanum tuberosum) production. 1-Aminocyclopropane-1-carboxylic acid (ACC), has also been proposed to act as a bioactive signal in plant stress responses, but its contribution to plant immunity remains elusive. RESULTS: Here, we show that ACC pretreatment exerts opposite effects on P. infestans resistance in Nicotiana benthamiana and the potato cultivar 'Favorita', revealing strong species-specific transcriptional reprogramming. In N. benthamiana, differentially expressed ge

Mesoporous Silica Nanoparticles Induce Intracellular Peroxidation Damage of Phytophthora infestans: A New Type of Green Fungicide for Late Blight Control.

Nanopesticides are considered to be a promising alternative strategy for enhancing bioactivity and delaying the development of pathogen resistance to pesticides. Here, a new type of nanosilica fungicide was proposed and demonstrated to control late blight by inducing intracellular peroxidation damage to Phytophthora infestans, the pathogen associated with potato late blight. Results indicated that the structural features of different silica nanoparticles were largely responsible for their antimicrobial activities. Mesoporous silica nanoparticles (MSNs) exhibited the highest antimicrobial activ

Overexpression of Camellia sinensis thaumatin-like protein, CsTLP in potato confers enhanced resistance to Macrophomina phaseolina and Phytophthora infestans infection.

Thaumatin-like proteins (TLPs), a class of pathogenesis related proteins are induced in response to pathogens and exhibit antifungal property when overexpressed in transgenic plants. In the present study, we have raised transgenic potato plants overexpressing TLP gene of Camellia sinensis (CsTLP). Fungal resistance assays of transgenic potato elucidated the potential role of CsTLP in imparting tolerance to fungal pathogens, Macrophomina phaseolina (necrotrophic) and Phytophthora infestans (hemi-biotrophic). Transgenic tubers with higher resistance to M. phaseolina, showed a concomitant and sig
