The hippocampal dentate gyrus (DG) is thought to orthogonalize inputs from the entorhinal cortex (pattern separation) and relay this information to the CA3 region. In turn, attractor dynamics in CA3 perform a pattern completion or error correction operation before sending its output to CA1. In a mouse model of congenital hypoplasia of the DG, a deficiency in the Wntless (Wls) gene, specifically in cells expressing Gfap-Cre, which targets neuronal progenitors, led to an almost total absence of dentate granule cells and modestly impaired performance in spatial tasks. Here, we investigated the ph

The Mecp2+/- mouse model recapitulates many phenotypes of patients with Rett syndrome (RTT), including learning and memory deficits. It is unknown, however, how the disease state alters memory circuit functions in vivo in RTT mice. Here we recorded from hippocampal place cells, which are thought to encode spatial memories, in freely moving RTT mice and littermate controls. We found that place cells in RTT mice are impaired in their experience-dependent increase of spatial information. This impairment is accompanied by an enhanced baseline firing synchrony of place cells within ripple oscillati

GABAergic medial septal neurons play a key role in regulating hippocampus-dependent spatial memory, but the underlying mechanisms are not yet fully understood. In male mice, we establish an object-place recognition impairment model using an acute sleep deprivation protocol. Here we show that parvalbumin-positive neurons in the medial septum regulate object-location memory specificity during encoding by modulating hippocampal place cell population activity. Using in vivo electrophysiology combined with optogenetics, we characterize the role of medial septal parvalbumin-positive neurons in objec

New concepts build on research on the rodent hippocampus, but go beyond that to understand how the hippocampal system operates in primates including humans. First, in rodents place cells and entorhinal cortex grid cells represent the place where the individual is located, but in primates including humans, many hippocampal system neurons respond to spatial view, to where the individual looks in space. Second, rodent navigation is described from place to place, but in primates including humans, the highly developed foveal visual system and hippocampal spatial view cells allow use of visual landm

Neurophysiological studies show that the firing of place and head-direction (HD) cells in rats can become anchored to features of the perceptible environment, suggesting that those features partially specify the rat's position and heading. In contrast, behavioral studies suggest that disoriented rats and human children rely exclusively on the shape of their surroundings, ignoring much of the information to which place and HD cells respond. This difference is explored in the current study by investigating young children's ability to locate objects in a square chamber after disorientation. Child

The encoding, storage, and updating of memories in cortical networks are poorly understood. In retrosplenial cortex (RSC), cells respond to the animal's position as it traverses a real or virtual (VR) linear track. Most position correlated cells (PCCs) in RSC require an intact hippocampus to form, but survive subsequent hippocampal damage. To examine whether RSC and hippocampal PCCs undergo remapping and spatial tuning development in parallel or sequentially, neuronal activity in RSC or CA1 was recorded using two-photon calcium imaging in mice running on VR tracks. RSC PCC activity underwent g

During pancreatic organogenesis endocrine cells arise from non self-renewing progenitors that express Ngn3. The precursors that give rise to Ngn3+ cells are presumably located within duct-like structures. However, the nature of such precursors is poorly understood. We show that, at E13-E18, the embryonic stage during which the major burst of beta-cell neogenesis takes place, pancreatic duct cells express Hnf1beta, the product of the maturity-onset diabetes of the young type 5 (MODY5) gene. Ngn3+ cells at this stage invariably cluster with mitotically competent Hnf1beta+ cells, and are often in

An animal's survival depends on finding food and the memory of food and contexts are often linked. Given that the hippocampus is required for spatial and contextual memory, it is reasonable to expect related coding of space and food stimuli in hippocampal neurons. However, relatively little is known about how the hippocampus responds to tastes, the most central sensory property of food. In this study, we examined the taste-evoked responses and spatial firing properties of single units in the dorsal CA1 hippocampal region as male rats received a battery of taste stimuli differing in both chemic

C-C chemokine receptor type 5, also known as CCR5 or CD195, is best known as a viral co-receptor that facilitates entry of HIV into cells. Evidence that CCR5 knockout mice display fewer dopamine neurons, lower striatal dopamine levels, and reduced locomotor activation compared to wild types also suggest a link between CCR5 receptors and cocaine dependence. Here, we tested the hypothesis using male Sprague-Dawley rats that cocaine-induced locomotor activation and conditioned place preference (CPP) are inhibited by a FDA-approved CCR5 antagonist (maraviroc), and that CCR5 gene expression in meso

Peptides with morphine-like efficacy, which are called endorphines and enkephalines, are formed by the nerve cells of numerous segments of the central nervous system and of the gastrointestinal canal. The efficacy of these peptides which decrease the sensation of pain is increased in certain stress (lesions, operative interventions, heat, birth). They stimulate the activity of the sympathetic nervous system, influence the secretion of certain hormones and decrease the sensitivity to pain. In acupuncture the release of the endorphines is increased. The formation of the endorphines takes place b

The purpose of this study was to investigate the applicability of flow cytometry to human placenta and to challenge the hypothesis that no cell division takes place in the last 4-6 weeks of pregnancy. Relative DNA content in individual placental cells was measured and high-resolution DNA histograms were obtained, based upon measurements of more than 10 000 cells. The fraction of cells with S-phase DNA content was lower in cases of intra-uterine growth retardation. In contrast to the earlier concept based on measurements of total organ content of DNA, this study indicates that cell division in

The localization of allergen-specific IgE synthesis in allergic diseases of the upper respiratory tract is so far unknown. It has been suggested that the IgE production takes place in the nasal mucosa itself. The present immunohistochemical studies with anti-IgE and monoclonal markers for B lymphocytes, plasma cells, antigens of the major histocompatibility complex, T helper and T suppressor cells indicate that there are no IgE-producing plasma cells in the nasal mucosa of patients with seasonal allergic rhinitis. The IgE-associated cells have been defined as two different types of mast cells.

Alzheimer's disease (AD) patients exhibited episodic memory impairments including location-object recognition in a spatial environment, which was also presented in animal models with amyloid-β (Aβ) accumulation. A potential cellular mechanism was the unstable representation of spatial information and lack of discrimination ability of novel stimulus in the hippocampal place cells. However, how the firing characteristics of different hippocampal subsets responding to diverse spatial information were interrupted by Aβ accumulation remains unclear. In this study, we observed impaired novel object-

Ductal carcinoma in situ (DCIS) refers to breast epithelial cells that have become "cancerous" but still reside in their normal place in the ducts and lobules. In this setting, cancerous means that there is an abnormal increase in the growth of the epithelial cells, which accumulate within and greatly expand the ducts and lobules. DCIS is a nonlethal type of cancer because it stays in its normal place. However, DCIS is very important because it is the immediate precursor of invasive breast cancers, which are potentially lethal. This article provides a general overview of DCIS, including histor

The primate posterior parietal cortex (PPC) withholds a unified representation of the visual space supporting visual exploration, while the hippocampus (HPC) provides a memory-based cognitive place map of the environment. To probe the interactions between these two representations, i.e. between view and place, we compared neural activity in the two regions of macaques navigating a virtual maze. We show that a large proportion of PPC neurons displayed spatial selectivity, along with the HPC. We hypothesized that such modulation by self-position might stem from visual cues processing through sac

Animals use sensory information and memory to build internal representations of space. It has been shown that such representations extend beyond the geometry of an environment and also encode rich sensory experiences usually referred to as context. In mammals, contextual inputs from sensory cortices appear to be converging on the hippocampus as a key area for spatial representations and memory. How metric and external sensory inputs (e.g., visual context) are combined into a coherent and stable place representation is not fully understood. Here, I review the evidence of attentional effects alo

A major breakthrough in cancer treatment occurred with the development of strategies that overcome T-cell tolerance toward tumor cells. These approaches enhance antitumor immunity by overcoming mechanisms that are normally in place to prevent autoimmunity but simultaneously prevent rejection of tumor cells. Although tolerance mechanisms that restrict antitumor immunity take place both in the thymus and periphery, only immunotherapies that target peripheral tolerance mechanisms occurring outside of the thymus are currently available. We review here recent gains in our understanding of how thymi

Specific interchange takes place between 131I-marked antibody combined with cellular antigens and the free unmarked antiserum antibody in three systems studied: (A) bovine cells with (i) human infectious mononucleosis serum and (ii) rabbit anti-bovine serum and (B) human Rh D-positive cells with Rh incomplete anti-D serum. The rate of interchange is much slower than the initial combination of an unmarked antibody with an unoccupied antigenic site, and also is slower in the Rh incomplete system than in the other systems studied. Rabbit antibovine red cell antiserum partly interchanges with 131I

During both sleep and awake immobility, hippocampal place cells reactivate time-compressed versions of sequences representing recently experienced trajectories in a phenomenon known as replay. Intriguingly, spontaneous sequences can also correspond to forthcoming trajectories in novel environments experienced later, in a phenomenon known as preplay. Here, we present a model showing that sequences of spikes correlated with the place fields underlying spatial trajectories in both previously experienced and future novel environments can arise spontaneously in neural circuits with random, clustere

We have examined the cellular changes taking place in rat popliteal lymph nodes undergoing a graft-versus-host (GvH) reaction. Examination of immunoperoxidase-stained lymph node sections, using a panel of mouse monoclonal antibodies directed against different rat lymphoid cell subsets, revealed a disorganization of the lymph node architecture with disappearance of the follicles, and an intermingling of T and B cells, so that no distinct T- and B-cell areas were visible any more. Since the GvH nodes showed a preferential accumulation of host B cells over host T cells (particularly over the W 3/
