Probiotic Bacilli Inhibit Salmonella Biofilm Formation Without Killing Planktonic Cells.

Salmonellosis is a foodborne infection caused by Salmonella. Domestic poultry species are one of the main reservoirs of Salmonella, which causes the foodborne infection salmonellosis, and are responsible for many cases of animal-to-human transmission. Keeping backyard chickens is now a growing trend, increasing the frequency of direct contact with the flock and, by consequence, the incidence of Salmonella infections. Bacillus subtilis KATMIRA1933 and Bacillus amyloliquefaciens B-1895 are probiotic bacilli that produce the bacteriocins subtilosin A and subtilin, respectively. The antimicrobial

Antibacterial Effects of Phage Lysin LysGH15 on Planktonic Cells and Biofilms of Diverse Staphylococci.

Treatment of infections caused by staphylococci has become more difficult because of the emergence of multidrug-resistant strains as well as biofilm formation. In this study, we observed the ability of the phage lysin LysGH15 to eliminate staphylococcal planktonic cells and biofilms formed by Staphylococcus aureus, Staphylococcus epidermidis, Staphylococcus haemolyticus, and Staphylococcus hominis All these strains were sensitive to LysGH15, showing reductions in bacterial counts of approximately 4 log units within 30 min after treatment with 20 μg/ml of LysGH15, and the MICs ranged from 8 μg/

The Legionella pneumophila GIG operon responds to gold and copper in planktonic and biofilm cultures.

Legionella pneumophila contaminates man-made water systems and creates numerous exposure risks for Legionnaires' Disease. Because copper/silver ionization is commonly used to control L. pneumophila, its mechanisms of metal response and detoxification are of significant interest. Here we describe an L. pneumophila operon with significant similarity to the GIG operon of Cupriavidus metallidurans. The Legionella GIG operon is present in a subset of strains and has been acquired as part of the ICE-βox 65-kB integrative conjugative element. We assessed GIG promoter activity following exposure of L.

Investigation of mercury methylation pathways in biofilm versus planktonic cultures of Desulfovibrio desulfuricans.

Biofilms can methylate mercury (Hg) at higher rates than unattached bacteria and are increasingly recognized as important Hg methylation sites in the environment. Our previous study showed that methylation rates in biofilm cultures were up to 1 order of magnitude greater than those in planktonic cultures of a sulfate-reducing bacterium. To probe whether the differential Hg methylation rates resulted from metabolic differences between these two cultures, Hg methylation assays following molybdate or chloroform inhibition (a specific inhibitor of the acetyl-CoA pathway) were conducted on biofilm

Influence of the diadinoxanthin pool size on photoprotection in the marine planktonic diatom Phaeodactylum tricornutum.

The pool size of the xanthophyll cycle pigment diadinoxanthin (DD) in the diatom Phaeodactylum tricornutum depends on illumination conditions during culture. Intermittent light caused a doubling of the DD pool without significant change in other pigment contents and photosynthetic parameters, including the photosystem II (PSII) antenna size. On exposure to high-light intensity, extensive de-epoxidation of DD to diatoxanthin (DT) rapidly caused a very strong quenching of the maximum chlorophyll fluorescence yield (F(m), PSII reaction centers closed), which was fully reversed in the dark. The no

Plankton-mediated coupling of phthalate esters across seawater, biota, and sediments in the northern Persian Gulf: Industrial vs urban coasts.

Phthalate esters (PAEs) are widely used plastic additives continuously released into marine environments, yet biologically mediated partitioning and sediment-biota coupling in semi-enclosed industrial basins remain insufficiently quantified. This study presents an integrated multi-matrix assessment of six priority PAEs (DMP, DEP, DBP, BBP, DEHP, DnOP) in seawater, plankton, and surface sediments from the northern Persian Gulf, comparing a petrochemical-dominated coastline (Assaluyeh) with an urban shoreline (Bushehr) across cold and warm seasons. Total PAE concentrations (ΣPAEs) followed a con

Mercury bioaccumulation and assimilation in marine plankton in meltwater influenced fjords and shelf waters along the east coast of Greenland.

The rapid melting of the Arctic cryosphere due to climate change will result in significant freshwater input into Arctic marine ecosystems. This might also cause the release of legacy mercury (Hg) stored in the cryosphere, increasing Hg concentration and its subsequent effects on the marine biota. However, there is scarce knowledge on the concentration of Hg in the lower trophic level organisms at the base of the Arctic pelagic food web. This is particularly important since these organisms modulate the transfer of Hg to higher trophic levels, including fish and marine mammals. We quantified th

Total and methyl mercury distribution in water, sediment, plankton and fish along the Tapajós River basin in the Brazilian Amazon.

Mercury (Hg) is known as one of the major contaminants in the Amazon. The Tapajós River basin, in the Brazilian Amazon, has diverse anthropogenic activities which increase Hg concentrations in the aquatic ecosystem. Moreover, high concentrations of Hg are naturally found in this basin. Distribution of total (THg) and methyl (MeHg) mercury were assessed in unfiltered water (n = 47), suspended particulate matter (SPM, n = 30), superficial sediment (BS, n = 29), plankton (n = 28) and fishes (n = 129) from the Tapajós River basin. Suspended particles were the main carrier of Hg in the water column

Evaluation of enrichment media for improved PCR-based detection of V. cholerae and V. vulnificus from estuarine water and plankton.

AIMS: Pathogenic Vibrio spp., including V. cholerae and V. vulnificus, are commonly found along the estuaries of the south-east United States; however, it is often difficult to recover these species directly from environmental samples. Pre-enrichment assays are commonly used to improve the detection of pathogenic vibrios from environmental sources. Here, we evaluated a novel enrichment procedure using freshly collected and autoclaved natural estuarine water amended with 1% peptone (designated as estuarine peptone water, EPW) and compared it to traditional alkaline peptone water (APW) for detec

Aquatic reservoir of Vibrio cholerae in an African Great Lake assessed by large scale plankton sampling and ultrasensitive molecular methods.

The significance of large tropical lakes as environmental reservoirs of Vibrio cholerae in cholera endemic countries has yet to be established. By combining large scale plankton sampling, microbial culture and ultrasensitive molecular methods, namely Droplet Digital PCR (ddPCR) and targeted genomics, the presence of Vibrio cholerae was investigated in a 96,600 L volume of surface water collected on a 322 nautical mile (596 km) transect in Lake Tanganyika. V. cholerae was detected and identified in a large area of the lake. In contrast, toxigenic strains of V. cholerae O1 or O139 were not detec

Morphological diversity and phylogeny of the diatom genus Entomoneis (Bacillariophyta) in marine plankton: six new species from the Adriatic Sea.

The diatom genus Entomoneis is known from the benthos and plankton of marine, brackish, and freshwaters. Entomoneis includes diatoms with a bilobate keel elevated above the valve surface, a sigmoid canal raphe, and numerous girdle bands. Owing mostly to the scarcity of molecular data for a diverse set of species, the phylogeny of Entomoneis has not been investigated in depth. The few previous studies that included Entomoneis were focused on broader questions and the available data were from a small number of either unidentified Entomoneis or well-known species (e.g., E. paludosa). Since the fi

Influence of E-Liquids and Oral Commensal Bacteria on the Growth of Porphyromonas gingivalis Planktonically and in Biofilms.

Background: The increasing use of electronic cigarettes (ECIGs), especially among youth, has raised concerns about the impact of vaping on oral health. While ECIGs are often marketed as a safer alternative, the existing literature suggests that their use may have detrimental effects on the pulmonary and cardiovascular systems. The oral cavity is the first point of contact for ECIG aerosol, and new reports link vaping to the onset of periodontal disease. It is critical to understand the potential effects of vaping on the oral microbiome, which affects systemic health. This study investigates ho

Biofilm and planktonic Staphylococcus aureus exhibit distinct gene expression patterns in response to cinnamaldehyde.

Staphylococcus aureus forms biofilms in the context many infections, including endocarditis, lung infection, and the colonization of implants. How antimicrobials specifically affect S. aureus biofilms as opposed to planktonic S. aureus is an important consideration in the development of treatments of these infections. It is well known that bacteria in biofilms are more resistant to antimicrobials, and the degree and nature of the responses is crucial to understanding the basis of this resistance. While certain antimicrobials such as antibiotics have specific mechanisms that induce pathways rel

Innovative Approaches Against Planktonic and Biofilm-Producing Clostridium perfringens Associated with Necrotic Enteritis.

Necrotic enteritis (NE), caused by the Gram-positive, anaerobic bacterium Clostridium perfringens (C. perfringens; CP), is one of the most economically important diseases affecting the poultry industry worldwide. The pathogenicity of C. perfringens is attributed to the production of more than twenty extracellular toxins and enzymes, with strains classified into seven toxinotypes (A-G) based on their major toxin genes. While the alpha-toxin gene (cpa) is present across all C. perfringens toxinotypes, netB is recognized as the primary virulence determinant of avian NE. However, toxin-gene carria

Silver Nanoparticles Functionalized with Polymeric Substances to Reduce the Growth of Planktonic and Biofilm Opportunistic Pathogens.

The global rise in antimicrobial resistance, particularly among ESKAPE pathogens, has intensified the demand for alternative therapeutic strategies. Silver nanoparticles (AgNPs) have exhibited broad-spectrum antimicrobial activity and represent a promising approach to combat multidrug-resistant infections. This study aimed to synthesize and functionalize AgNPs using various polymeric agents-ethylene glycol (EG), polyethylene glycol (PEG), polyvinylpyrrolidone (PVP), and their combinations-and to evaluate their antimicrobial and antibiofilm efficacy against clinically relevant bacterial strains

Characterizing species-specific metabolic signatures in vaginal microbiota across planktonic and biofilm states.

Bacterial vaginosis (BV) affects about 29 % of U.S. women, with higher rates in some groups and up to 50-69 % recurrence within a year. It increases the risk of STIs, pregnancy complications, and can cause significant discomfort and reduced quality of life. Prior studies mapped vaginal metabolomes, but individual microbial metabolite signatures remain poorly understood. Given that biofilms exhibit distinct metabolic requirements compared to planktonic cultures, analyzing biofilm vs. planktonic culture metabolites with their unique metabolic needs may uncover novel treatment targets. This study

Rifabutin: a repurposed antibiotic with high potential against planktonic and biofilm staphylococcal clinical isolates.

Staphylococcus aureus poses a significant threat as an opportunistic pathogen in humans, and animal medicine, particularly in the context of hospital-acquired infections (HAIs). Effective treatment is a significant challenge, contributing substantially to the global health burden. While antibiotic therapy remains the primary approach for staphylococcal infections, its efficacy is often compromised by the emergence of resistant strains and biofilm formation. The anticipated solution is the discovery and development of new antibacterial agents. However, this is a time consuming and expensive pro

Quantification of Polyphosphate in Environmental Planktonic Samples Using a Novel Fluorescence Dye JC-D7.

Polyphosphate (polyP) is found in plankton of diverse aquatic ecosystems and is important for plankton ecology and biogeochemical cycling. However, our knowledge of polyP in aquatic environments is hindered by a lack of data due to the limitations of quantification methods. The estimate of polyP in model organisms using phenol-chloroform extraction followed by enzymatic hydrolysis is complicated and fails for environmental samples. The commonly used 4',6-diamidino-2-phenylindole (DAPI) fluorescence method for environmental studies, on the contrary, severely overestimates polyP due to interfere

Design, synthesis and antibacterial activity of chalcones against MSSA and MRSA planktonic cells and biofilms.

Staphylococcus aureus is the one of the most successful modern pathogens. The same bacterium that lives as a skin and mucosal commensal can be transmitted in health-care and community-settings and causes severe infections. Thus, there is a great challenge for a discovery of novel anti-Staphylococcus aureus compounds, which should act against resistant strains. Herein, we designed and synthesized a series of 17 chalcones, substituted by amino group on ring A, which were evaluated against methicillin-susceptible S. aureus (MSSA) and methicillin-resistant S. aureus MRSA planktonic cells. The anti
