A novel control strategy: Suppressing histone acetyltransferase ClHAC703 with curcumin to prevent germination of the invasive weed Cenchrus longispinus.

The invasive alien plant Cenchrus longispinus has caused severe ecological and agricultural impacts in China. Taking into consideration its seed germination characteristics, clustered community formation, and localized distribution, the present study proposes reducing population replenishment by inhibiting seed germination in heavily infested soils. This study demonstrated that 1 mM curcumin treatment could completely inhibit the germination of C. longispinus seeds. Curcumin directly binds to histone acetyltransferase ClHAC703 and suppresses the activity of histone acetyltransferases in C. lon

Sunlight to Plasma: Mimicking nature's light for smarter agriculture and crop production.

BACKGROUND: Over Earth's history, the Sun has continuously released coronal mass ejections of plasma that have flowed toward Earth, generating the need for reactive oxygen and nitrogen species (RONS) for plants. On the other hand, engineered plasma interacts with air, generating RONS. Thus, the natural process is simulated using engineered plasma (hereafter, plasma), which acts as an artificial Sun. Plasma agriculture offers a sustainable alternative by leveraging plasma-generated RONS to stimulate plant growth and generate nitrogen fertilizer through eco-friendly methods. AIM OF REVIEW: The

The pseudoenzyme β-amylase9 from Arabidopsis binds to and enhances the activity of α-amylase3: A possible mechanism to promote stress-induced starch degradation.

Starch accumulation in plant tissues provides an important carbon source at night and for regrowth after periods of dormancy and in times of stress. Both ɑ- and β-amylases (AMYs and BAMs, respectively) catalyze starch hydrolysis, but their functional roles are unclear. Moreover, the presence of catalytically inactive amylases that show starch excess phenotypes when deleted presents an interesting series of questions on how starch degradation is regulated. Plants lacking one of these catalytically inactive β-amylases, BAM9, were shown to have enhanced starch accumulation when combined with muta

Micropropagation of Dipcadi montanum (Dalz.) Baker (Asparagaceae): a rare scapigerous herb.

Dipcadi montanum (Dalz.) Baker (Asparagaceae) is a rare scapigerous herb endemic to the Western Ghats, a global biodiversity hotspot running parallel to the western coast of India. This study reports the development of a reproducible protocol for mass propagation of this underutilized geophyte using bulb scale and immature leaf base explants. Miniature bulblets were successfully induced from both types of explants after 4 and 8 weeks of culture on full-strength semisolid MS basal medium fortified with 3% sucrose and varying levels of BAP (4.4-17.7 μM) and TDZ (4.5-18.1 μM). The addition of 2.7

On the stabilization of plant lipid droplets: Dynamic interplay between oleosins and phospholipids.

As central organelles for lipid and energy homeostasis in cells, lipid droplets (oleosomes) consist of a neutral lipid core - mainly triacylglycerols and sterol esters. Crucial to the droplet stability and dynamics, the droplet boundary is decorated by a dense phospholipid monolayer enriched in phosphatidylcholines (PCs) as well as surfactant-like proteins, which in plant cells, are predominantly oleosins (OLs). The monolayer keeps plant lipid droplets stable for years throughout seed dormancy yet delicately destabilizes them during germination to ensure the targeted release of their stored li

Parasitic Plant-Host Interactions: Molecular Mechanisms and Agricultural Resistance Strategies.

Obligate parasitic plants, particularly members of the Orobanchaceae family, including Striga and Orobanche, greatly devastate crop production. Here, we synthesize recent advances in understanding the molecular and ecological dynamics underlying parasitic plant-host interactions, focusing on critical stages of parasitism: germination, host detection, haustorium formation, and resource extraction. Orobanchaceous parasites exploit host-derived strigolactones (SLs) to break seed dormancy, whereas Cuscuta species do not rely on SLs for germination. Instead, chemotropic responses to host-exuded com

Optimising Olive Leaf Phenolic Compounds: Cultivar and Temporal Interactions.

All olive (Olea europaea L.) plant tissues have a high phenolic content. However, the effects of the cultivar and sampling period on the tissue phenolic content remain almost unknown; in addition, the interactions between nutrient uptake and leaf phenol concentrations have not been clarified. This study sampled olive leaves to explore how the cultivar, sampling period, and their interaction affect leaf phenol and nutrient concentrations. Leaves were collected from six cultivars during three seasonal periods: harvest (October; SP1), dormancy (January; SP2), and pruning (March; SP3). Five were I

Dormant bud cryopreservation of Rubus idaeus L.

Cryopreservation of dormant buds offers a reliable and cost-effective method for the long-term preservation of plant genetic resources, particularly for woody plants like raspberry (Rubus idaeus L.). This study aimed to develop an optimized protocol for the cryopreservation of dormant raspberry buds by investigating the role of dehydration and freezing conditions in maintaining bud viability. Dormant canes of two raspberry varieties, 'Sanibelle' and 'Willamette', were collected and prepared by dehydrating single-node cane segments at -4 °C until the desired water content was achieved. The stud

Factors influencing the variability of antioxidative phenolic glycosides in Salix species.

Phenolic glycosides, especially the salicylates, are important secondary metabolites in the bark of willows (Salix spp.). Because of their anti-inflammatory, analgesic, and fever-reducing properties, they are of particular interest to society. Compared to the fabrication of synthetic salicylacetylic acid, the commercial production of willow bark extracts with adequate amounts of salicylate is very difficult due to several biological and technical reasons. Therefore, one of the objectives was to identify salicylate-rich clones from three species, Salix daphnoides , Salix purpurea , and Salix pe

Caseinolytic Protease P: A Therapeutic Nexus in Infection, Inflammation, and Immunity.

Caseinolytic protease P (ClpP) is a highly conserved serine protease that plays a pivotal role in protein homeostasis and quality control in bacteria, mitochondria of mammalian cells, and plant chloroplasts. As the proteolytic core of the ATP-dependent Clp protease complex, ClpP partners with regulatory ATPases (e.g., ClpX, ClpA) to degrade misfolded, damaged, or regulatory proteins. In bacteria, ClpP is crucial for survival under host-imposed stresses and modulates virulence through degradation of transcriptional regulators and signaling proteins, contributing to immune evasion, dormancy, and

The virulome of Streptomyces scabiei in response to cello-oligosaccharide elicitors.

The development of spots or lesions symptomatic of common scab on root and tuber crops is caused by few pathogenic Streptomyces with Streptomyces scabiei 87-22 as the model species. Thaxtomin phytotoxins are the primary virulence determinants, mainly acting by impairing cellulose synthesis, and their production in S. scabiei is in turn boosted by cello-oligosaccharides released from host plants. In this work we aimed to determine which molecules and which biosynthetic gene clusters (BGCs) of the specialized metabolism of S. scabiei 87-22 show a production and/or a transcriptional response to c

Breeding Utilization of the White-Grained Pre-Harvest Sprouting-Resistant Wheat Landrace Tuotuomai Through Marker-Assisted Selection.

Improving pre-harvest sprouting (PHS) resistance in white-grained wheat is challenging because reduced grain pigmentation is often associated with weaker seed dormancy. Here, we evaluated the breeding value of the white-grained, PHS-resistant landrace Tuotuomai (TTM) and developed a molecular marker for tracking TTM-derived material during breeding. Based on phenotypic segregation and sequence polymorphism analysis, a KASP marker targeting a polymorphic site in the TaMyb10-D1 region was developed. The marker co-segregated with white grain color in the TTM × CS population, and the TTM-type mark

Long-term saline-alkaline selection rewires the growth-survival trade-off in Priestia megaterium.

Saline-alkaline soils impose persistent osmotic, ionic, pH, and nutrient stress on soil microorganisms, but the evolutionary routes by which beneficial bacteria adapt to such conditions remain poorly resolved. We performed adaptive laboratory evolution to examine the adaption of the plant growth-promoting rhizobacterium Priestia megaterium HA22 to long-term oligotrophic saline-alkaline selection. After 175 serial transfers, the evolved lineage proliferated stably at 40 g L-1 Na2SO4 at pH 10.0, whereas the wild-type strain failed to proliferate. Genome resequencing and allelic replacement revea

Synthesis of tricyclic butenolides and comparison their effects with known smoke-butenolide, KAR1.

Plant-derived smoke - butenolide, called at present karrikin 1 (KAR1) is known as an important inductor of seed germination and seedling growth. In this study, tricyclic butenolides were synthesized and their effects on germination of dormant and non-dormant Avena fatua caryopses were compared, as were also their effects versus those of KAR1 on seedling growth. KAR1 was found to be most effective and to completely remove dormancy. Butenolides, rac-8 and (S)-8a, showed a low stimulatory effect on germination of dormant caryopses, visible only when applied at very high concentrations. These comp

Epigenomic Landscape of Oak (Quercus robur) across Season and Generation.

Seasonal fluctuations strongly shape the physiology of long-lived trees by coordinating growth, dormancy, and stress responses. Increasing evidence points to epigenetic mechanisms, particularly DNA methylation, as regulators of these processes, yet their role in long-lived trees across seasons, developmental stages and generations remains poorly understood. We generated single-base resolution maps of cytosine methylation exploring the epigenetic landscape of 180 year-old mature oak (Quercus robur) trees (genetically homogeneous) along spring, summer and autumn, and in their progeny. The volatile sesquiterpene β-caryophyllene generated by the Phoebe bournei terpene synthase PbTPSa21 regulates seed germination by mediating ROS signaling.

Phoebe bournei is an evergreen tree with high economic and social value because of its fine timber and pleasant aroma, which is predominantly produced by terpenoids, including β-caryophyllene. However, the natural reproduction and artificial cultivation of P. bournei are severely hindered by long seed dormancy times and poor seed fragility. Terpenoids are secondary metabolites with vast commercial application potential in medicine, agriculture, and gardening, but their role in seed germination remains elusive. Here, we identified a novel P. bournei β-caryophyllene synthase gene, PbTPSa21, whos

Uptake of Neonicotinoid Insecticides by Water-Foraging Honey Bees (Hymenoptera: Apidae) Through Guttation Fluid of Winter Oilseed Rape.

The water-foraging activity of honey bees (Apis mellifera L.) on guttation fluid of seed-coated crops, such as winter oilseed rape (WOR; Brassica napus L.), has not yet been evaluated. We analyzed the uptake of active substances (a.s.) in guttation fluid by evaluating residues of honey-sac contents. In autumn, insecticide residues of up to 130 µg a.s. per liter were released in WOR guttation fluid; this concentration is noticeably lower than levels reported in guttation fluid of seed-coated maize. Until winter dormancy, the concentrations declined to <30 µg a.s. per liter. In spring, residues

Advancing grapevine phenology predictions: a comparison of regression-based statistical learning approach and conventional models in mediterranean environment.

Climate conditions can significantly affect plant dormancy and growing season, impacting crop development and its productivity. Temperature is considered the primary driver of grapevine phenology, with endodormancy and ecodormancy largely regulated by chilling and forcing accumulations, respectively. Nevertheless, additional climatic factors, such as precipitation, radiation and atmospheric circulation patterns, may also contribute and regulate variability of different phenological phases. As hotspot of climate change, Mediterranean regions are witnessing negative compounding effects by enhanc

Temperature during Aspergillus fumigatus conidiophore development primes spore transcriptome for asexual, parasexual or sexual development.

UNLABELLED: Aspergillus fumigatus is a thermotolerant saprobe found in soils and plant debris worldwide and an important pathogen of humans causing two million deaths annually. A. fumigatus makes abundant asexual spores (conidia) which are widely distributed by wind and can be inhaled from the environment. In susceptible individuals inhaled conidia break dormancy, germinate and grow in the lung leading to serious disease. Recent work has shown that conidia made at 37°C and 50°C have different morphologies and germination kinetics. While the asexual cycle is well-characterized at 37°C, much les
