Fine mapping of the dominant female sterility gene and novel model of hybrid seed production in cabbage (Brassica oleracea L. var. capitata).

Hybrid breeding based on male sterility requires the removal of male parents, which is time- and labor-intensive; however, the use of female sterile male parent can solve this problem. In the offspring of distant hybridization between Brassica oleracea and Brassica napus, we obtained a mutant, 5GH12-279, which not only fails to generate gynoecium (thereby causing female sterility) but also has serrated leaves that could be used as a phenotypic marker in seedling screening. Genetic analysis revealed that this trait was controlled by a single dominant gene. Further analysis revealed that Bo2g005

Mre11 deficiency in Arabidopsis is associated with chromosomal instability in somatic cells and Spo11-dependent genome fragmentation during meiosis.

The Mre11/Rad50/Nbs1 complex is involved in many aspects of chromosome metabolism. Aberrant function of the complex is associated with defects in the DNA checkpoint, double-strand break repair, meiosis, and telomere maintenance. In this article, we report the consequences of Mre11 dysfunction for the stability of mitotic and meiotic chromosomes in Arabidopsis thaliana. Although plants homozygous for a T-DNA insertion in a conserved region of the MRE11 gene are viable, they exhibit growth defects and are infertile. Analysis of mitotic chromosomes prepared from the mutant plants revealed abundan

Mitochondrial gene editing and allotopic expression unveil the role of orf125 in the induction of male fertility in some Solanum spp. hybrids and in the evolution of the common potato.

Genic-cytoplasmic male sterility (CMS) due to interactions between nuclear and cytoplasmic genomes is a well-known phenomenon in some Solanum spp. hybrids, but genes involved are not known. In this study, the chondriomes of two isonuclear male-fertile and sterile somatic hybrids (SH9A and SH9B, respectively) between the common potato (S. tuberosum Group Tuberosum, tbr) and the wild species S. commersonii were sequenced and compared to those of parental species to identify mitochondrial genes involved in the expression of male sterility. A putative novel gene (orf125) was found only in tbr and

Introduction of barnase/barstar in soybean produces a rescuable male sterility system for hybrid breeding.

Hybrid breeding for increased vigour has been used for over a century to boost agricultural outputs without requiring higher inputs. While this approach has led to some of the most substantial gains in crop productivity, breeding barriers have fundamentally limited soybean (Glycine max) from reaping the benefits of hybrid vigour. Soybean flowers self-pollinate prior to opening and thus are not readily amenable to outcrossing. In this study, we demonstrate that the barnase/barstar male sterility/rescue system can be used in soybean to produce hybrid seeds. By expressing the cytotoxic ribonuclea

A tripartite genetic conflict system controls hybrid sterility in rice.

Interspecific Asian-African hybrid rice could substantially boost yield but is limited by severe hybrid sterility. We identify RHS3 as a major quantitative trait locus controlling this trait. RHS3 encodes a tripartite toxin-antidote system composed of MAO, DUN, and JIA, in which MAO acts as a toxin that aborts gametes by disrupting mitochondrial function, whereas DUN and JIA function as antidotes that neutralize MAO toxicity, conferring a transmission advantage to the African allele. We demonstrate that detoxification relies on selective autophagy through formation of a tripartite JIA-DUN-MAO

Overexpression of TCP5 or Its Dominant Repressor Form, TCP5-SRDX, Causes Male Infertility in Arabidopsis.

TCP transcription factors have long been known to play a crucial role in leaf development, but their significance in reproduction has recently been revealed. TCP5 is a member of class II of the TCP family, which predominantly regulates cell differentiation. This study used overexpression and SRDX fusion to evaluate the role of TCP5 in anther development. TCP5 overexpression resulted in lower fertility, primarily due to anther non-dehiscence. We also observed reduced lignin accumulation in the anther endothecium. In addition, TCP5 overexpression resulted in smaller anthers with fewer pollen sac

Identification and characterization of miRNAs associated with sterile flower buds in the tea plant based on small RNA sequencing.

BACKGROUND: miRNAs are a type of conserved, small RNA molecule that regulate gene expression and play an important role in the growth and development of plants. miRNAs are involved in seed germination, root development, shoot apical meristem maintenance, leaf development, and flower development by regulating various target genes. However, the role of miRNAs in the mechanism of tea plant flower sterility remains unclear. Therefore, we performed miRNA sequencing on the flowers of fertile male parents, female parents, and sterile offspring. RESULTS: A total of 55 known miRNAs and 90 unknown miRN

The WD40 repeat protein OsDPMS1 controls pistil development and female fertility in rice.

The pistil, a key female reproductive organ in flowering plants, plays a critical role in sexual reproduction. Although pistil function has been well studied in some species, the molecular mechanisms governing its development in rice (Oryza sativa) remain unclear. Here, we isolated and characterized a female-sterile rice mutant, Osdpms1, which exhibits pleiotropic reproductive defects, including greater stigma number and aberrant gametophyte development, while maintaining normal vegetative growth and male fertility. Using map-based cloning, genetic complementation, and CRISPR-Cas9 knockout exp

Impacts of cadmium on male fertility: Lessons learnt so far.

Cadmium (Cd) is one of the most dangerous heavy metals in the world. Globally, toxicities associated with cadmium and its attendant negative impact on humans and animals cannot be under-estimated. Cd is a heavy metal, and people are exposed to it through contaminated foods and smoking. Cd exerts its deleterious impacts on the testes (male reproductive system) by inducing oxidative stress, spermatogenic cells apoptosis, testicular inflammation, decreasing androgenic and sperm cell functions, disrupting ionic homeostasis, pathways and epigenetic gene regulation, damaging vascular endothelium and

From Hippocrates to Italian traditional medicine: Ethnopharmacological evidence for a potential pharmacological perspective in the management of polycystic ovary syndrome.

ETHNOPHARMACOLOGICAL RELEVANCE: Since antiquity, plant-based remedies have played a significant role in treating gynecological disorders. In Italy, from the late 19th to mid-20th century, traditional medicine documented numerous botanical treatments for conditions such as menstrual irregularities, infertility, and hirsutism-now recognized as clinical features of Polycystic Ovary Syndrome (PCOS). AIM OF THE STUDY: This study investigates the historical use of plant-based therapies in Italian traditional medicine for managing PCOS-related symptoms. It also evaluates these remedies through conte

A toxin-antidote system contributes to interspecific reproductive isolation in rice.

Breakdown of reproductive isolation facilitates flow of useful trait genes into crop plants from their wild relatives. Hybrid sterility, a major form of reproductive isolation exists between cultivated rice (Oryza sativa) and wild rice (O. meridionalis, Mer). Here, we report the cloning of qHMS1, a quantitative trait locus controlling hybrid male sterility between these two species. Like qHMS7, another locus we cloned previously, qHMS1 encodes a toxin-antidote system, but differs in the encoded proteins, their evolutionary origin, and action time point during pollen development. Meiotic instability and irregular chromosome pairing underpin heat-induced infertility in bread wheat carrying the Rht-B1b or Rht-D1b Green Revolution genes.

Mutations in the Rht-B1a and Rht-D1a genes of wheat (Triticum aestivum; resulting in Rht-B1b and Rht-D1b alleles) cause gibberellin-insensitive dwarfism and are one of the most important elements of increased yield introduced during the 'Green Revolution'. We measured the effects of a short period of heat imposed during the early reproductive stage on near-isogenic lines carrying Rht-B1b or Rht-D1b alleles, with respect to the wild-type (WT). The temperature shift caused a significant fertility loss within the ears of Rht-B1b and Rht-D1b wheats, greater than that observed for the WT. Defects i

Covariation in reproductive investment between sterile and fertile sex organs contributes to sterile stamens persistence in cryptically dioecious Actinidia.

BACKGROUND: Cryptic dioecy is a sexual system in which individuals of at least one sex are morphologically hermaphroditic but functionally unisexual. The presence of non-functional reproductive organs may represent an intermediate stage during the evolutionary transition from hermaphroditism to dioecy. The ancestral hypothesis suggests that sterile sex organs may persist as vestiges of an ancestral bisexual state, potentially due to their low resource cost or absence of significant conflict with fertile organs, yet this has rarely been tested. All Actinidia species exhibit cryptic dioecy with
