Alfalfa is believed to have originated in north-western Iran and has a long history of coexistence with its bacterial symbiont Sinorhizobium in soils of Iran. However, little is known about the diversity of Sinorhizobium strains nodulating Iranian alfalfa genotypes. In this study, Sinorhizobium populations were sampled from eight different Iranian sites using three cultivars of Medicago sativa as trap host plants. A total of 982 rhizobial strains were isolated and species were identified showing a large prevalence of Sinorhizobium meliloti over Sinorhizobium medicae. Analysis of salt tolerance

The formation of nitrogen-fixing symbiotic nodules is a complex process that involves extensive transcriptional reprogramming of legume roots to enable intracellular accommodation of host compatible rhizobia. This process requires coordinated regulation of hundreds to thousands of genes, including the induction of highly expressed genes, traditionally referred to as nodulin genes. While the functions of many nodulins have been elucidated, a substantial number remained poorly characterized, or their functions are still unclear. One such gene, NOD25, was originally identified based on differenti

During host-microbe symbioses, the fitness of mutualistic microbes is determined by the interactions that concurrently occur, throughout their life cycle, with their host and other members of the surrounding microbial community. Disentangling how these multiple interactions shape the fitness of microbial symbionts is challenging but is essential to understand the diversity and functioning of mutualisms. Here, we examined the different fitness components of rhizobial symbionts of the legume plant Mimosa pudica across the multiple stages of their symbiotic life cycle. By comparing rhizobial symb

Cadmium (Cd)-contamination impairs biological nitrogen fixation in legumes (BNF), threatening global food security. Innovative strategies to enhance BNF and improve plant resistance to Cd are therefore crucial. This study investigates the effects of graphitic carbon nitride nanosheets (g-C3N4 NSs) on soybean (Glycine max L.) in Cd contaminated soil, focusing on Cd distribution, chemical forms and nitrogen (N) fixation. Soybean plants were treated with 100 mg kg-1 g-C3N4 NSs, with or without 10 mg kg-1 Cd for 4 weeks. Soil addition of g-C3N4 NSs alleviated Cd toxicity and promote soybean growth

Symbiotic nitrogen (N) fixation entails successful interaction between legume hosts and rhizobia that occur in specialized organs called nodules. N-fixing legumes have a higher demand for phosphorus (P) than legumes grown on mineral N. Medicago truncatula is an important model plant for characterization of effects of P deficiency at the molecular level. Hence, a study was carried out to address the alteration in metabolite levels of M. truncatula grown aeroponically and subjected to 4 weeks of P stress. First, GC-MS-based untargeted metabolomics initially revealed changes in the metabolic prof

The advent of endosymbiosis underlies evolutionary innovation and ecosystem function. However, whether free-living partners tend to benefit or exploit each other during the early stages of novel endosymbiosis remains a dilemma. Rhizobia soil bacteria can initiate root nodules and fix nitrogen for host plants as endosymbionts due to genes carried on mobile genetic elements such as the symbiosis island (SI). We conjugated marked SIs into the genomes of non-nodulating strains, which was sufficient to generate de novo root nodule-forming endosymbionts. Most novel endosymbionts originated as commen

Legume plants rely upon multipartite interactions between rhizobia and bacterial endophytes within root nodules to facilitate plant growth. This study aimed to isolate and identify indigenous endophytic bacteria from root nodules of Sulla aculeolata L. in Northeast Morocco. Based on their tri-calcium phosphate (TCP) solubilization capacity, five endophytes were chosen for further evaluation of their plant growth traits. All isolates were hydrogen cyanide (HCN) and siderophore producers, while only BCH24 tested positive for ACC deaminase activity. Indole-3-acetic acid (IAA) synthesis ranged fro

Legumes form a symbiosis with rhizobia in which the plant provides an energy source to the rhizobia bacteria that it uses to fix atmospheric nitrogen. This nitrogen is provided to the legume plant, allowing it to grow without the addition of nitrogen fertilizer. As part of the symbiosis, the bacteria in the infected cells of a new root organ, the nodule, are surrounded by a plant-derived membrane, the symbiosome membrane, which becomes the interface between the symbionts. Fractions containing the symbiosome membrane (SM) and material from the lumen of the symbiosome (peribacteroid space or PBS

The NIN-like protein 2 (NLP2) transcription factor is highly expressed in the infected cells of the N2 fixation zone of mature nodules in Medicago truncatula. In these cells, NLP2 directly activates the expression of leghemoglobin genes and is required for the full expression of nitrite reductase (NiR). Histological examination of nodules formed under different nitrate regimes revealed that the infected cells of nlp2 display abnormal vacuole morphology under high nitrate (5 mm KNO3). This phenotype is associated with starch accumulation, higher expression of starch biosynthesis genes, and incr

Legumes establish symbiosis with rhizobia, forming nitrogen-fixing nodules. The central role of reactive oxygen species (ROS) and reactive nitrogen species (RNS) in nodule biology has been clearly established. Recently, hydrogen sulfide (H2S) and other reactive sulfur species (RSS) have emerged as novel signaling molecules in animals and plants. A major mechanism by which ROS, RNS, and RSS fulfil their signaling role is the post-translational modification of proteins. To identify possible functions of H2S in nodule development and senescence, we used the tag-switch method to quantify changes i

Bradyrhizobia are abundant soil bacteria, which can form nitrogen-fixing symbioses with leguminous plants, including important crops such as soybean, cowpea and peanut. Many bradyrhizobia can denitrify, but studies have hitherto focused on a few model organisms. We screened 39 diverse Bradyrhizobium strains, isolated from legume nodules. Half of them were unable to reduce N2 O, making them sources of this greenhouse gas. Most others could denitrify NO3 - to N2 . Time-resolved gas kinetics and transcription analyses during transition to anaerobic respiration revealed a common regulation of nirK

Nitrous oxide (N2O) is a potent greenhouse gas, and its largest anthropogenic source is agricultural soils. In these soils, N2O emissions are produced from nitrogen fertilization and nitrogen-rich crop residues, such as soybean nodules. Previous studies demonstrated that inoculation with Bradyrhizobium diazoefficiens possessing the N2O reductase gene (nosZ) effectively mitigated N2O emissions from soybean fields at harvest. More recently, Bradyrhizobium ottawaense, which exhibits higher nosZ expression, reduced N2O emissions more effectively than B. diazoefficiens during the degradation of soy

Bambara groundnut (Vigna subterranea L. Verdc.) is an indigenous, drought-tolerant, underutilized African food legume, with the ability to fix atmospheric N2 in symbiosis with soil bacteria called rhizobia. The aim of this study was to assess the morpho-physiological, symbiotic and phylogenetic characteristics of rhizobia nodulating Bambara groundnut in Ghana, Mali and South Africa. The morpho-physiologically diverse isolates tested were also found to exhibit differences in functional efficiency and phylogenetic positions. Based on Enterobacterial Repetitive Intergenic Consensus (ERIC)-PCR ban

Legumes, which develop a symbiosis with nitrogen-fixing bacteria, have an increased demand for iron. Iron is required for the synthesis of iron-containing proteins in the host, including the highly abundant leghemoglobin, and in bacteroids for nitrogenase and cytochromes of the electron transport chain. Deficiencies in iron can affect initiation and development of the nodule. Within root cells, iron is chelated with organic acids such as citrate and nicotianamine and distributed to other parts of the plant. Transport to the nitrogen-fixing bacteroids in infected cells of nodules is more compli

Plants constantly experience partial hypoxia due to varying external oxygen availability and the existence of hypoxic niches, such as the shoot apical meristem, phloem or root nodules. Waterlogging experiments indicate that hypoxic stress at the root does not only lead to a local metabolic response, such as the accumulation of hypoxia-related metabolites, but also causes metabolic alterations in the normoxic shoot. Moreover, hypoxia-related metabolites are exported from the hypoxic root towards the normoxic shoot, where they can be recycled. Maintaining import of glycolytic substrates from the

The present study showed all the 16 strains isolated and identified from the alfalfa rhizosphere and nodules, and registered in GenBank, to be good candidates for targeted use in studies addressing the rather weak known mechanism of plant growth promotion, including that of Medicago truncatula, a molecular crop model. Based on physiological, biochemical and molecular analysis, the 16 isolates obtained were ascribed to the following five families: Bacillaceae, Rhizobiaceae, Xantomonadaceae, Enterobacteriaceae and Pseudomonadaceae, within which 9 genera and 16 species were identified. All these

Three soybean [Glycine max (L) Merr.] small RNA libraries were generated and sequenced using the Illumina platform to examine the role of miRNAs during soybean nodulation. The small RNA libraries were derived from root hairs inoculated with Bradyrhizobium japonicum (In_RH) or mock-inoculated with water (Un_RH), as well as from the comparable inoculated stripped root samples (i.e. inoculated roots with the root hairs removed). Sequencing of these libraries identified a total of 114 miRNAs, including 22 novel miRNAs. A comparison of miRNA abundance among the 114 miRNAs identified 66 miRNAs that

Low-level bioavailable cadmium (Cd) and copper (Cu) in agricultural soils poses a severe threat to soil health and food safety; however, the mechanisms by which indigenous Cu-Cd tolerant rhizobia modulate plant-microbe-soil interactions remain poorly understood. In this field trial, the effects of two Cu-Cd tolerant strains, Sinorhizobium xinjiangense YN5 (RB) and Rhizobium pusense GF4 (RD), when inoculated individually and in combination (RC), were assessed with respect to soybean growth, heavy‑metal partitioning, rhizosphere and nodule endophytic microbiomes, and soil functional genes under

The legume species Astragalus sinicus (Chinese milk vetch [CMV]) has been widely cultivated for centuries in southern China as one of the most important green manures/cover crops for improving rice productivity and preventing soil degeneration. In this study, we generated the first chromosome-scale reference genome of CMV by combining PacBio and Illumina sequencing with high-throughput chromatin conformation capture (Hi-C) technology. The CMV genome was 595.52 Mb in length, with a contig N50 size of 1.50 Mb. Long terminal repeats (LTRs) had been amplified and contributed to genome size expansi

Cadmium (Cd) contamination severely threatens peanut (Arachis hypogaea L.) production and symbiotic nitrogen fixation (SNF). Although rhizobia inoculation can alleviate heavy metal toxicity in plants, how nitrate (NO3-) regulates Cd translocation and the defense of the "rhizobia-root-nodule" system remains unclear. Here, we investigated the mechanisms by which the peanut rhizobium, Rhizobium sp. HM13, mitigates Cd toxicity and sustains SNF under varying NO3- levels (N- and N+) via field and pot trials. Field trials showed that HM13 reduced rhizosphere bioavailable Cd and decreased seed Cd accu
