Micropropagation of Ginger (Zingiber officinale Roscoe) 'Bentong' and Evaluation of Its Secondary Metabolites and Antioxidant Activities Compared with the Conventionally Propagated Plant.

'Bentong' ginger is the most popular variety of Zingiber officinale in Malaysia. It is vegetatively propagated and requires a high proportion of rhizomes as starting planting materials. Besides, ginger vegetative propagation using its rhizomes is accompanied by several types of soil-borne diseases. Plant tissue culture techniques have been applied in many plant species to produce their disease-free planting materials. As 'Bentong' ginger is less known for its micropropagation, this study was conducted to investigate the effects of Clorox (5.25% sodium hypochlorite (NaOCl)) on explant surface s

Fine-tuning by strigolactones of root response to low phosphate.

Strigolactones are plant hormones that regulate the development of different plant parts. In the shoot, they regulate axillary bud outgrowth and in the root, root architecture and root-hair length and density. Strigolactones are also involved with communication in the rhizosphere, including enhancement of hyphal branching of arbuscular mycorrhizal fungi. Here we present the role and activity of strigolactones under conditions of phosphate deprivation. Under these conditions, their levels of biosynthesis and exudation increase, leading to changes in shoot and root development. At least for the

IBA Delivery Technique and Media Salts Affected In Vitro Rooting and Acclimatization of Eight Prunus Genotypes.

Difficult-to-root plants often perform poorly during acclimatization and in vitro rooting can increase the survival and quality of plants. The influence of auxin application and mineral nutrition on in vitro rooting and subsequent effects on plant quality in eight Prunus genotypes were investigated. Microshoots were rooted in vitro on Murashige and Skoog (MS), ½ MS, Driver and Kuniyuki (DKW), or New Prunus Medium (NPM) media formulations in combination with 15 µM indole-3-butyric acid (IBA), 4-day 15 µM IBA pulse, 1 mM 30 s quick-dip, or IBA-free treatments. Shoots were observed pre- and post-

Differential partitioning of seed-inhabiting methylotrophs in the endosphere of wheat plants.

BACKGROUND: Both endophytes, microorganisms that reside within plants, and methylotrophs, which grow using methanol produced from plant leaves, play key roles in protecting plants against biotic and abiotic stresses. However, the source of endophytes and the mechanisms of their selection in plants are poorly understood. Therefore, experiments were carried out to identify wheat seed methylotrophic endophytes and evaluate their partitioning in root, stem and leaf of aseptic-controlled plants cultivated from surface-sterilized seeds. RESULTS: The counts of endophytic methanol utilizers were high

Enhanced arsenic tolerance of transgenic eastern cottonwood plants expressing gamma-glutamylcysteine synthetase.

Arsenic is a metalloid that occurs naturally at parts per million (ppm) levels in the earth's crust. Natural and human activities have contributed to arsenic mobilization and increased concentration in the environment, such that World Health Organization guidelines for arsenic levels in drinking water are exceeded at many locations, worldwide. This translates into an increased risk of arsenic-related illnesses for millions of people. Recent studies demonstrate that increasing thiol-sinks in transgenic plants by overexpressing the bacterial gamma-glutamylcysteine synthetase (ECS) gene results i

Differential alterations of antioxidant defenses as bioindicators of mercury and cadmium toxicity in alfalfa.

Several physiological parameters related to oxidative stress, which is a characteristic of plants exposed to toxic metals, were studied in 3-week-old alfalfa plants treated with cadmium (Cd) or mercury (Hg) at doses of 0, 3, 10 and 30 microM for 7d. The concentrations of biothiols, glutathione (GSH), homoglutathione (hGSH) and phytochelatins (PCs) increased dramatically in metals-treated plants, in particular in the presence of Cd. This was accompanied by a remarkable up-regulation of gamma-glutamyl cysteine synthetase gene, probably in response to the higher demand for GSH|hGSH needed for PC

Transgenic expression of DwMYB2 impairs iron transport from root to shoot in Arabidopsis thaliana.

In plant, iron uptake and homeostasis are tightly regulated to ensure its absorption from soil and to avoid excess iron in the cell. Many genes involved in this process have been identified during past several years, but there are many problems remain unsolved in the genetic regulation of whole plant iron trafficking and allocation. MYB transcription factors contain tandem repeats of a approximately 50 amino acid DNA-binding motif (R) and are involved in the regulation of many aspects of plant development, hormone signaling and metabolism. Here, we report that the ectopic expression of orchid

Formation of methyl mercury in an aquatic macrophyte.

In the nature, inorganic forms of mercury (Hg) may be transformed to the organic, very toxic, methyl-Hg. Occasionally methyl-Hg has been detected in plants, also so in the aquatic macrophyte water spinach (Ipomoea aquatica), which is a popular vegetable in tropical regions. The objectives of this study were to investigate if methyl-Hg is formed and/or degraded in water spinach. Water spinach plants were exposed to inorganic Hg via spiked soil or spiked nutrient solution. Tests were performed in a climate chamber and in experimental units, one for each individual plant, that were equipped with

The Establishment of a Highly Efficient In Vitro Regeneration System for Viburnum opulus L. 'Roseum'.

Viburnum opulus L. 'Roseum' is a highly valuable ornamental plant for landscaping, but it has a long propagation cycle and low propagation coefficient. In this study, stem segments with axillary buds from Viburnum opulus L. 'Roseum' were used as explants. We systematically analyzed the use of sodium hypochlorite for the sterilization of explants, as well as the effects of different plant growth regulator combinations and concentrations on shoot bud induction, shoot proliferation, the rooting of tissue-cultured shoots, and the transplanting of the tissue-cultured shoots. Mitigating copper toxicity in Carthamus tinctorius L. through 6-benzylaminopurine during seed germination and vegetative growth.

Cytokinins are essential regulators of plant growth and development and are actively involved in plant responses to both biotic and abiotic stresses. In the present study, the role of 6-Benzylaminopurine (BAP) in modulating growth performance and copper (Cu) tolerance in Carthamus tinctorius L. was examined. Treatments consisted of four CuSO4 levels and three BAP levels in a factorial design. High Cu levels significantly reduced seed germination parameters, growth characteristics, photosynthetic pigments, root and shoot protein content, and peroxidase (POX) activity, while increasing soluble c

A rapid and efficient protocol for in vitro multiplication of genetically uniform Stevia rebaudiana (Bertoni).

Stevia rebaudiana (Bertoni), commonly called candy leaf or sweet leaf, endemic to South America, is an important medicinal plant. As a source of low calorie natural sweetener 'stevoside', it is used in obesity, diabetes, treatment of heartburn and tooth decay, and also serves as a food supplement. Large scale commercial propagation of S. rebaudiana demands a suitable protocol. Here, we propose an improved protocol for in vitro multiplication of S. rebaudiana from nodal explants. In this protocol, the effect of laboratory grade urea on multiple shoot induction from nodal explants was studied. T

Australian native plant species Carpobrotus rossii (Haw.) Schwantes shows the potential of cadmium phytoremediation.

Many polluted sites are typically characterized by contamination with multiple heavy metals, drought, salinity, and nutrient deficiencies. Here, an Australian native succulent halophytic plant species, Carpobrotus rossii (Haw.) Schwantes (Aizoaceae) was investigated to assess its tolerance and phytoextraction potential of Cd, Zn, and the combination of Cd and Zn, when plants were grown in soils spiked with various concentrations of Cd (20-320 mg kg(-1) Cd), Zn (150-2,400 mg kg(-1) Zn) or Cd + Zn (20 + 150, 40 + 300, 80 + 600 mg kg(-1)). The concentration of Cd in plant parts followed the order

Mn tolerance in rice is mediated by MTP8.1, a member of the cation diffusion facilitator family.

Manganese (Mn) is an essential micronutrient for plants, but is toxic when present in excess. The rice plant (Oryza sativa L.) accumulates high concentrations of Mn in the aerial parts; however, the molecular basis for Mn tolerance is poorly understood. In the present study, genes encoding Mn tolerance were screened for by expressing cDNAs of genes from rice shoots in Saccharomyces cerevisiae. A gene encoding a cation diffusion facilitator (CDF) family member, OsMTP8.1, was isolated, and its expression was found to enhance Mn accumulation and tolerance in S. cerevisiae. In plants, OsMTP8.1 and

Crosstalk between above- and belowground herbivores is mediated by minute metabolic responses of the host Arabidopsis thaliana.

Plants are frequently under attack by multiple herbivores and can be infested at their shoots as well as their roots. As a consequence, plant metabolites are readily induced, mediated by phytohormones such as salicylic acid and jasmonic acid. Thereby, cross-talk between signal transduction pathways may occur if different herbivores attack the plant simultaneously. In turn, modifications in the plant metabolic pattern can affect herbivores infesting local and systemic tissue. Here, an integrative approach combining metabolomics and performance experiments was used to study the induction of plan

Modulation of metal transporters, oxidative stress and cell abnormalities by synergistic application of silicon and titanium oxide nanoparticles: A strategy for cadmium tolerance in rice.

Heavy metals, especially cadmium (Cd), cause severe toxicity symptoms in crop plants. Applying nanoparticles (NPs) as nano-fertilizers is a novel approach to mitigating plants' Cd stress. However, knowledge about the combinational use of silicon (Si) and titanium dioxide (TiO2) NPs to mitigate Cd stress, especially in rice, must be highlighted. TiO2-NPs (15 mg L-1) and Si-NPs (2.5 mM) were applied alone and in combination to rice plants grown without (control; no Cd stress) and with (100 μM) Cd concentration. Results revealed that compared to the control plants, root length, shoot length, shoo

High-frequency shoot regeneration, assessment of genetic fidelity, and histochemical analysis of forskolin production in Coleus forskohlii Briq.

Forskolin, a diterpenoid found in the roots of Coleus forskohlii, has generated significant interest in the medical field due to its various therapeutic uses. This study aimed to establish an effective system for regenerating C. forskohlii plants, ensuring a year-round supply of plant material and forskolin production. We tested different concentrations of cytokinins, either alone or combined with auxin, to see their impact on shoot multiplication and growth. We found that a medium supplemented with 1.5 mg L-1 of meta-topolin (mT) resulted in the highest number of shoots (~ 12.66) and leaves (

What is often overlooked is how results from controlled conditions translate back to field situations. A meta-analysis showed that lab-grown plants had faster growth rates, higher nitrogen concentrations and different morphology. They remained smaller, however, because the lab plants had grown for a much shorter time. We compared glassh
