Revealing structure and shaping priorities in plant and fungal cell wall architecture via solid-state NMR.

Plant and fungal cell walls are essential for growth, adaptation, and survival, with their intricate architectures dictating both resistance to stress and susceptibility to antifungal or biomass-degrading strategies. Understanding how these walls are built, remodeled, and function at the molecular level is therefore central to both clinical and biotechnological applications. Solid-state nuclear magnetic resonance (ssNMR) has emerged as a uniquely powerful tool for this purpose, as it reveals the structure, dynamics, and interactions of intact biopolymers without disrupting their native organiz

Structurally diverse abietane diterpenoids from the whole plants of Salvia przewalskii Maxim. With anti-inflammatory activity.

Using bioactivity guidance, eight previously undescribed abietane diterpenoids (1-8), were isolated from the whole plants of Salvia przewalskii Maxim. along with seven known analogues (9-15). Their structures were elucidated by extensive spectroscopic analyses, quantum chemical calculations, and single crystal X-ray diffraction. Among them, compound 1, an abietane diterpenoid bearing unique C-20 m-cymen-8-ol monoterpenoid moiety, was the first example of the adduct between a monocyclic monoterpenoid and an abietane diterpenoid connected by ester bond within the Salvia genus. The plausible bios

Deciphering the anthocyanin metabolism gene network in tea plant (Camellia sinensis) through structural equation modeling.

BACKGROUND: Tea is an important cash crop that significantly contributes to rural development, poverty reduction and food security in many developing countries. It provides livelihoods for millions of smallholder producers and aids their economic stability. Anthocyanins in tea leaves provides excellent commercial quality and germplasm exploration potential. These compounds give tea leaves vibrant colors and increase health benefits. The current understanding of the synergistic regulation mechanisms responsible for color changes in purple tea, attributed to anthocyanin degradation, remains uncl

"Mindset Matters": Perseverance, a balanced approach and structured support as facilitators of whole foods plant-based adoption.

Whole foods plant-based diets (WFPBD) show potential for preventing and addressing chronic diseases. However, concerns exist about their acceptability and feasibility. Research on firsthand WFPBD adoption experiences is limited but crucial for promoting dietary change. In a 12-week remotely delivered lifestyle modification program using an ad libitum WFPBD, twenty weight-loss seeking adults (ages 18-75) with overweight or obesity completed self-report surveys and semi-structured interviews via Zoom. The study aimed to explore: (1) experiences with WFPBD adoption; (2) factors that helped or hin

Seeing is believing: Understanding functions of NPR1 and its paralogs in plant immunity through cellular and structural analyses.

In the past 30 years, our knowledge of how nonexpressor of pathogenesis-related genes 1 (NPR1) serves as a master regulator of salicylic acid (SA)-mediated immune responses in plants has been informed largely by molecular genetic studies. Despite extensive efforts, the biochemical functions of this protein in promoting plant survival against a wide range of pathogens and abiotic stresses are not completely understood. Recent breakthroughs in cellular and structural analyses of NPR1 and its paralogs have provided a molecular framework for reinterpreting decades of genetic observations and have

Antifungal symbiotic peptide NCR044 exhibits unique structure and multifaceted mechanisms of action that confer plant protection.

In the indeterminate nodules of a model legume Medicago truncatula, ∼700 nodule-specific cysteine-rich (NCR) peptides with conserved cysteine signature are expressed. NCR peptides are highly diverse in sequence, and some of these cationic peptides exhibit antimicrobial activity in vitro and in vivo. However, there is a lack of knowledge regarding their structural architecture, antifungal activity, and modes of action against plant fungal pathogens. Here, the three-dimensional NMR structure of the 36-amino acid NCR044 peptide was solved. This unique structure was largely disordered and highly d

Population structure and dynamics of Gammarus fossarum (Amphipoda) upstream and downstream from effluents of sewage treatment plants.

Two streams in Germany (Körsch and Lockwitzbach), each with two sampling sites above and below a sewage treatment plant (STP) discharging effluent, were investigated. Sampling sites were characterized, and exposure monitoring for chemicals with known or assumed endocrine disrupting potential was carried out. Both the population structure and the population dynamics of Gammarus fossarum were examined. The physicochemical parameters measured at the sampling sites of the Lockwitzbach and Körsch streams were found not to reach levels having an acute toxic impact on the development of gammarids. Th

Recent gene duplication and structural remodeling drive rapid lineage-specific gene family evolution in plants.

Gene duplication promotes the generation of novel gene functions and trait diversity across species. Here, we present DupHIST, a computational pipeline that reconstructs the hierarchical timing of gene duplications by integrating maximum likelihood (ML)-based phylogeny with substitution-derived timing via statistical smoothing. Applied to over 4.5 million genes from 114 plant genomes, we successfully inferred duplication histories across nearly 130,000 orthogroups. This large-scale analysis showed that 53.0% of genes arose from recent, lineage-specific duplications, with high concentrations in

Seasonal Effect of a Small Hydropower Plant on Benthic Biofilm Microbial Community Structure and Predicted Functional Potential.

Benthic biofilm microbial communities play a critical role in riverine primary production, nutrient cycling, and organic matter decomposition, yet their responses to hydrological alterations due to small hydropower plants (SHPs) remain poorly understood, particularly with respect to seasonal dynamics. This study conducted a one-year monitoring survey across four river sections (upstream, reservoir, dewatered reach, and downstream) in the Oujiang River Basin, Zhejiang Province, China, covering all four seasons. The results showed that the reservoir section exhibited distinct bacterial compositi

Breaking it down: Unveiling the roles of chemical structure, concentrations and technology on micropollutant biotransformation in wastewater treatment plants.

Complex mixtures of anthropogenic micropollutants (MPs) pose risks to ecosystems and health by entering the environment through multiple pathways. Microbial biotransformation is a major removal process, yet the key drivers remain poorly understood. Activated sludge (AS) treatment in wastewater treatment plants (WWTPs) acts as a partial barrier and reduces MP emissions into aquatic ecosystems, yet removal efficiencies vary widely across MPs. Here, we studied the biotransformation of 189 MPs in AS from six Swiss WWTPs operated at three treatment conditions: carbon elimination (Celim), nitrificat

MYB transcription factors in plants: A comprehensive review of their discovery, structure, classification, functional diversity and regulatory mechanism.

The MYB transcription factor (TF) family is one of the largest families in plants and performs highly diverse regulatory functions, particularly in relation to pathogen/pest resistance, nutrient/noxious substance absorption, drought/salt resistance, trichome growth, stamen development, leaf senescence, and flavonoid/terpenoid biosynthesis. Owing to their vital role in various biological regulatory processes, the mechanisms of MYB TFs have been extensively studied. Notably, MYB TFs not only directly regulate targets, such as phytohormones, reactive oxygen species signaling and secondary cell wa

Legacy Effects of Plant Community Structure Are Manifested in Microbial Biofilm Development With Consequences for Ecosystem CO2 Emissions.

To better understand linkages between hydrology and ecosystem carbon flux in northern aquatic ecosystems, we evaluated the relationship between plant communities, biofilm development, and carbon dioxide (CO2) exchange following long-term changes in hydrology in an Alaskan fen. We quantified seasonal variation in biofilm composition and CO2 exchange in response to lowered and raised water table position (relative to a control) during years with varying levels of background dissolved organic carbon (DOC). We then used nutrient-diffusing substrates (NDS) to evaluate cause-effect relationships bet

Prioritised identification of structural classes of natural products from higher plants in the expedition of antimalarial drug discovery.

The emergence and spread of drug-recalcitrant Plasmodium falciparum parasites threaten to reverse the gains made in the fight against malaria. Urgent measures need to be taken to curb this impending challenge. The higher plant-derived sesquiterpene, quinoline alkaloids, and naphthoquinone natural product classes of compounds have previously served as phenomenal chemical scaffolds from which integral antimalarial drugs were developed. Historical successes serve as an inspiration for the continued investigation of plant-derived natural products compounds in search of novel molecular templates fr

Effects of N-Glycosylation on the Structure, Function, and Stability of a Plant-Made Fc-Fusion Anthrax Decoy Protein.

Protein N-glycosylation is an important post-translational modification and has influences on a variety of biological processes at the cellular and molecular level, making glycosylation a major study aspect for glycoprotein-based therapeutics. To achieve a comprehensive understanding on how N-glycosylation impacts protein properties, an Fc-fusion anthrax decoy protein, viz rCMG2-Fc, was expressed in Nicotiana benthamiana plant with three types of N-glycosylation profiles. Three variants were produced by targeting protein to plant apoplast (APO), endoplasmic reticulum (ER) or removing the N-gly

Increasing the Size of the Microbial Biomass Altered Bacterial Community Structure which Enhances Plant Phosphorus Uptake.

Agricultural production can be limited by low phosphorus (P) availability, with soil P being constrained by sorption and precipitation reactions making it less available for plant uptake. There are strong links between carbon (C) and nitrogen (N) availability and P cycling within soil P pools, with microorganisms being an integral component of soil P cycling mediating the availability of P to plants. Here we tested a conceptual model that proposes (i) the addition of readily-available organic substrates would increase the size of the microbial biomass thus exhausting the pool of easily-availab

Cellular localization of cadmium and structural changes in maize plants grown on a cadmium contaminated soil with and without liming.

The effects of different concentrations of soil cadmium (0, 1, 3, 5, 10, and 20mgkg(-1)) on growth, structural changes and cadmium cellular localization in leaves of maize plants (Zea mays L.) were investigated in a pot experiment. The results showed that the structural changes observed in maize leaves were not only a response to the Cd-induced stress but also a cellular mechanism to reduce the free Cd(+2) in the cytoplasm. However, this mechanism seems to be efficient only up to a Cd concentration in leaves between 27 and 35mgkg(-1) for soils without and with liming, respectively. The cellula

Programmed cell death in plants: effect of protein synthesis inhibitors and structural changes in pea guard cells.

Pea leaf epidermis incubated with cyanide displayed ultrastructural changes in guard cells that are typical of apoptosis. Cycloheximide, an inhibitor of cytoplasmic protein synthesis, and lincomycin, an inhibitor of protein synthesis in chloroplasts and mitochondria, produced different effects on the dynamics of programmed death of guard cells. According to light microscopy data, cycloheximide reinforced and lincomycin suppressed the CN(-)-induced destruction of cell nuclei. Lincomycin lowered the effect of cycloheximide in the light and prevented it in the dark. According to electron microsco

Structural requirements for arthritogenicity of peptidoglycans from Staphylococcus aureus and Lactobacillus plant arum and analogous synthetic compounds.

The comparative studies on the arthritogenicity of chemically well defined peptidoglycans (PG)2 from the cell walls of Staphylococcus aureus (FDA 209P) and Lactobacillus plantarum (ATCC 8014) showed that 1) a polymer of disaccharide-peptide with or without the presence of N-acetylglucosaminyl-ribitol-teichoic acid produced severe arthritis; 2) oligosaccharide-peptides with or without the special structure (N-acetylglucosaminyl-ribitol-teichoic acid in S. aureus, a polymer of rhamnose and glucose in L. plantarum) produced severe disease; 3) disaccharide-heptapeptide-disaccharide with or without
