Plant viruses represent a major threat to agriculture, affecting a wide range of crops with substantial economic losses. This study presented a novel strategy for managing plant viral diseases through the development an attenuated vaccine utilizing cucumber mosaic virus (CMV) for virus-induced gene silencing (VIGS) targeting susceptibility gene. TOBAMOVIRUS MULTIPLICATION 2A (TOM2A) gene was identified as a critical factor that enhances susceptibility to TMV infection in plants. Two vaccines were constructed based on CMV attenuated vaccine vector-CR2V. The first vaccine was constructed by inco

The ubiquitin-proteasome system (UPS) constitutes a highly conserved regulatory hub governing protein turnover and signal transduction in eukaryotes, which precisely determines the fate of substrate proteins via dynamic and reversible ubiquitination. During long-term coevolution between plants and viruses, the UPS has evolved into a critical battlefield for host-virus arms races. Plants exploit the substrate recognition specificity and proteolytic activity of the UPS to selectively eliminate essential viral proteins required for infection, thereby establishing multilayered antiviral immune bar

The metagenomic approach based on high-throughput sequencing is becoming increasingly prevalent for the detection of viral infections in plants. This method allows us to study the species composition of viruses associated with the plant, including novel species, describe their population genetic structure, and develop genetic test systems for routine diagnostics. A metagenomic approach to phytosanitary monitoring can help to determine the cause of unknown plant diseases, which is particularly important for preventing the spread of pathogens, such as viruses. Furthermore, as it is impossible to

Entomopathogenic viruses affect insect populations by reducing fitness and influencing other measures of performance such as development and longevity. Given the huge diversity of viruses, studies are needed to understand the mechanisms underlying how viruses affect their insect hosts. The Junonia coenia densovirus (JcDV) infects a wide range of Lepidoptera and can cause death in the larval and pupal stages. In this study, we used the Melissa blue butterfly (Lycaeides melissa: Lycaenidae) to describe infection progression at the individual level, the effects of infection on development, surviv

The outbreak of vector-borne plant viruses entails both efficient in planta virus infection and productive vector-mediated transmission. Yet, the factors that concurrently regulate these two key viral traits remain understudied. Here we examine the role of viral proteins and the salicylic acid (SA) signaling pathway in modulating virus infection and vector preference to virus-infected plants. Infection of plants by the bipartite begomovirus, Sri Lankan cassava mosaic virus (SLCMV), dramatically induces the accumulation of SA, a positive regulator of antiviral defense. As countermeasures, SLCMV

A previously undescribed group of related plant viruses has recently been identified within the Benyviridae family. This group is called reclovirids, named after the type species, red clover RNA virus 1 (RCRV1). The single genomic RNA of most reclovirids contains two main open reading frames (ORFs): a large, well-conserved 5′-proximal ORF1 that encodes a viral replicase and a less conserved 5′-distal ORF2 that encodes a polypeptide of unknown function with a C4-type zinc finger motif. In this study, the ORF2 proteins of RCRV1 and a Gymnadenia rhellicani virus, another reclovirid, are found to

Cross-protection is a phenomenon in which a plant prior infected with a mild viral strain can be protected from subsequent infection by severe strains of the same or closely related viruses. Since its discovery in the 1920s, this phenomenon has become an important strategy for controlling viral diseases in plants. Telosma mosaic virus (TelMV, a species of Potyvirus) is an emerging pathogen that threatens passion fruit production in subtropical and tropical regions. Here, we present a protocol for screening mild TelMV strains and their application in protecting passion fruit from severe TelMV i

Vegetatively propagated sugarcane can accumulate multiple viruses that compromise planting-material health. Here, we present a model-guided alternating high-temperature thermotherapy (AHTT) protocol to eliminate major viruses from diverse sugarcane cultivars. We describe preparation and disinfection of single-bud axillary-bud cuttings, planting, thermal preconditioning, and a 12-day AHTT regimen. We then detail post-thermotherapy recovery, health scoring, leaf sampling, total-RNA extraction, control-validated one-step RT-PCR, result interpretation, and the detection of additional sugarcane vir

Selective autophagy has emerged as a central interface in plant-virus interactions, acting at once as an antiviral defense, a proviral vulnerability, and a regulator of host immune homeostasis. In plants, autophagy can directly target viral proteins, replication complexes, and even entire virions for degradation, while viruses have evolved diverse strategies to exploit, evade, or repurpose the autophagy machinery. Recent work has revealed an expanding repertoire of plant- and virus-encoded selective autophagy receptors, as well as unexpected roles for autophagy in restraining excessive immune

Sugarcane viruses threaten yield and sucrose content, imposing substantial global economic losses. This review provides current knowledge on the major sugarcane pathogens, with a focus on virus biology, transmission, and virus-host interactions, and uses this foundation to frame practical control strategies. We first describe the epidemiology and economic impact of sugarcane viral diseases to establish the urgency of robust surveillance and management. Building on these insights, we then evaluate detection and surveillance technologies, ranging from traditional immunoassays and nucleic acid hy

Autophagy is an evolutionarily conserved degradation and recycling pathway that plays essential roles in plant development, stress adaptation, and immunity. Over the past decade, increasing evidence has revealed that autophagy also shapes the outcome of plant-virus interactions. This review summarizes current understanding of the plant autophagy machinery as well as the experimental approaches used to monitor autophagic flux during viral infection. We then explore the diverse roles of autophagy in the plant-virus interface, highlighting its function as an antiviral mechanism that targets viral

As a globally vital food crop, rice suffers severe yield losses from viral diseases. While over a dozen rice viruses have been documented in China, functional reverse genetics system remains unavailable for most, significantly limiting molecular analysis and breeding for virus resistance. This chapter presents a detailed protocol for establishing a viral reverse genetics system using rice tiller inhibition virus (RTIV) as a case. The methodology provides a robust framework for developing infectious clones of other rice viruses.

Lilium is an economically important genus widely cultivated for ornamental and commercial use. In this study, we report the identification and characterization of a novel mitovirus, designated Lily Bulb Virus-1 (LbV-1), detected in commercial lily bulb stocks. This virus represents a previously uncharacterized member of the mitochondrial RNA virus group associated with lily crops. A survey of 50 commercially cultivated lily bulbs, including Lilium longiflorum varieties, revealed LbV-1 infection in a single cultivar, indicating a low but detectable incidence under the tested conditions. To protect global strawberry production, clean plant programs rely on rigorous certification and quarantine testing to prevent the spread of disease. For decades, biological graft indexing has been the "gold standard" for virus detection; however, it often fails to identify viruses that remain latent in single infections or viruses requiring extended periods to induce symptoms. High throughput sequencing (HTS) offers an alternative that identifies all known and emerging viruses independent of the nucleotide sequences of their genomes including those undetected by traditional bioassays. This st

Pepper (Capsicum annuum L.) plants produce berry fruits that are used as spices. Here, we examined the viromes of 15 pepper cultivars through RNA sequencing. We obtained 1,325 virus-associated contigs derived from 8 virus species. Bean broad wilt virus 2 (BBWV2) and cucumber mosaic virus (CMV) were identified as the major viruses infecting pepper plants, followed by potato virus Y, bell pepper endornavirus, and hot pepper endornavirus. The proportion of viral reads in each transcriptome ranged from 0.04% to 24.5%. BBWV2 was the dominant virus in seven cultivars, whereas CMV was dominant in fiv

Recently, previously known viruses have changed their pathogenicity and encompassed new types of host populations. An example of such an infection is that caused by SARS-CoV, belonging to the "well-known" coronavirus family. Another group of viruses that are of great importance to the human population are the herpes viruses. Due to increasing viral resistance to existing antiviral drugs, plant extracts are attracting increasing interest due to their complex composition and their simultaneous attack of different viral targets. Based on the above, we tested the antiviral potential of water-alcoh

Viral infections are among the most complex medical problems and have been a major threat to the economy and global health. Several epidemics and pandemics have occurred due to viruses, which has led to a significant increase in mortality and morbidity rates. Natural products have always been an inspiration and source for new drug development because of their various uses. Among all-natural sources, plant sources are the most dominant for the discovery of new therapeutic agents due to their chemical and structural diversity. Despite the traditional use and potential source for drug development

Plant growth promoting rhizobacteria have been proposed as effective biocontrol agents against several fungal and bacterial plant pathogens. However, there is limited knowledge regarding their effect against viruses. In this study, Bacillus amyloliquefaciens strain MBI600 (MBI600), active ingredient of the biological fungicide Serifel® (BASF SE), was tested for its antiviral action in tomato plants. Drench, foliar or soil amendment applications of MBI600 reduced up to 80% the incidence of Tomato spotted wilt virus under two different sets of environmental conditions. In addition, drench applic

Plant viruses are major plant pathogens and account for nearly half of emerging plant diseases worldwide. To date, few effective management measures are available for the efficient control of plant viral diseases. Cross-protection based on attenuated vaccine is an effective strategy to prevent plant viral diseases. The primary requirement for the development of attenuated vaccines is a vector with excellent characteristics, such as low pathogenicity, genetic stability, and the capacity for stable insertion of exogenous fragments. In this study, the cucumber mosaic virus (Fny strain) was geneti
