AIM: To study the blood schizontocidal effect of oral artesunate on P berghei in mice and P knowlesi in monkey.
METHODS: Effects of artesunate and chloroquine were detected with "4-day test" and "28-day test" on P berghei in mice and "7-day test" on P knowlesi in Macaca mudatta.
RESULTS: The suppressive efficacy of oral artesunate was inferior to chloroquine on P berghei K173 strain but the time for 50% and 90% reduction and the time of clearance of parasitemia was 10-15 h shorter than that of chloroquine. Its curative effect on RC/K173 line was markedly superior to that of chloroquine. More

Mice infected with Plasmodium berghei, during the ten days of infection, showed significant pulmonary oedema, delayed activated partial thromboplastin time, augmented plasma fibrinolytic activity, oscillations in fibrinogen and unchanged plasminogen levels. These alterations can be expected to cause the release of inflammatory peptides, leading to increased vascular permeability and the generation and/or maintenance of the pronounced pulmonary oedema affecting these animals.

BACKGROUND: Malaria is one of the most prevalent infectious disease in the world with 3.2 billion humans at risk. Malaria causes splenomegaly and damage in other organs including skeletal muscles. Skeletal muscles comprise nearly 50% of the human body and are largely responsible for the regulation and modulation of overall metabolism. It is essential to understand how malaria damages muscles in order to develop effective preventive measures and/or treatments. Using a pre-clinical animal model, the potential molecular mechanisms of Plasmodium infection affecting skeletal muscles of mice were in

Apicomplexan parasites such as Toxoplasma and Plasmodium spp. rely on the sequential secretion of parasite apical organelles, called micronemes and rhoptries, to invade host cells. The claudin-like apicomplexan microneme protein (CLAMP) is a conserved protein that plays an essential role during host cell invasion in Toxoplasma and Plasmodium zoites. Previous studies have shown that CLAMP is essential in Plasmodium merozoites for erythrocyte invasion and also in sporozoites for the invasion of the mosquito vector salivary glands and of mammalian host hepatocytes. In Toxoplasma gondii tachyzoite

Fluorescent proteins are widely used as markers to differentiate genetically modified cells from their wild-type counterparts. In malaria research, the prevalent fluorescent markers include red fluorescent proteins (RFPs) and their derivatives, such as mCherry, along with green fluorescent proteins (GFPs) and their derivatives. Recognizing the need for additional fluorescent markers to facilitate multiplexed imaging, this study introduced parasite lines expressing blue fluorescent protein (BFP). These lines enable simultaneous microscopy studies of proteins tagged with GFP, RFP, or detected by

BACKGROUND: The invasion of Plasmodium merozoites into host erythrocytes is initiated through specific ligand-receptor interactions. This interaction results in subsequent invasion events, facilitated by the formation of a moving junction via AMA-1 and associated molecular complexes. Previous studies have implicated erythrocyte surface glycosaminoglycans, particularly heparan sulfate proteoglycans, as critical receptor components in this invasion process.
METHODS: The binding affinity of the PbGAC protein to heparin and erythrocytes was assessed through western blotting, immunofluorescence, f

Malaria remains a critical global health issue, particularly in tropical and subtropical regions. The disease, caused by Plasmodium parasites, is transmitted by Anopheles mosquitoes and can lead to severe complications and death if untreated. The emergence of drug-resistant strains highlights the urgent need for new antimalarial agents. Gnetum gnemon, a plant native to Southeast Asia, has shown promise due to its rich bioactive compounds. This study aims to evaluate the suppressive, curative, and prophylactic antimalarial potential of Gnetum gnemon leaf extract (GGE) against Plasmodium berghei

BACKGROUND: Different strategies for improvement of malaria control and elimination are based on the blockage of malaria parasite transmission to the mosquito vector. These strategies include the drugs that target the plasmodial sexual stages in humans and the early developmental stages inside mosquitoes.
OBJECTIVES: Here we tested Malaria Box compounds in order to evaluate their activity against male and female gametocytes in Plasmodium berghei, mosquito infection in P. vivax and ookinete formation in both species.
METHODS/FINDINGS: The membrane feeding assay and the development of ookinete

Substrate-dependent gliding motility is key to malaria transmission. It mediates host cell traversal, invasion and infection by Plasmodium and related apicomplexan parasites. The 110 amino acid-long cell surface protein LIMP is essential for P. berghei sporozoites where it is required for the invasion of the mosquito's salivary glands and the liver cells of the rodent host. Here we define an additional role for LIMP during mosquito invasion by the ookinete. limp mRNA is provided as a translationally repressed mRNP (messenger ribonucleoprotein) by the female gametocyte and the protein translate

Malaria and lymphatic filariasis (LF) are two leading and common mosquito-borne parasitic diseases worldwide. These two diseases are co-endemic in many tropical and sub-tropical regions and are known to share vectors. The interactions between malaria and filarial parasites are poorly understood. Thus, this study aimed at establishing the interactions that occur between Brugia pahangi and Plasmodium berghei ANKA (PbA) co-infection in gerbils. Briefly, the gerbils were matched according to age, sex, and weight and grouped into filarial-only infection, PbA-only infection, co-infection, and contro

BACKGROUND: The search for new antimalarial drugs has become progressively urgent due to plasmodial resistance to most of the commercially available antimalarial drugs. As part of this effort, the study evaluated the antimalarial activity of Cucumis metuliferus and Lippia kituiensis, which are traditionally used in Tanzania for the treatment of malaria.
MATERIALS AND METHODS: In vivo antimalarial activity was assessed using the 4-day suppressive antimalarial assay. Mice were infected by injecting via tail vein 1×107 erythrocytes infected by Plasmodium berghei ANKA. Extracts were administered

Malaria is caused by mosquito-borne Plasmodium spp. parasites that must infect and survive within mosquito salivary glands (SGs) prior to host transmission. Recent advances in transcriptomics and the complete genome sequencing of mosquito vectors have increased our knowledge of the SG genes and proteins involved in pathogen infection and transmission. Membrane solute carriers are key proteins involved in drug transport and are useful in the development of new interventions for transmission blocking. Herein, we applied transcriptomics analysis to compare SGs mRNA levels in Anopheles stephensi f

Actin has important roles in Plasmodium parasites but its exact function in different life stages is not yet fully elucidated. Here we report the localization of ubiquitous actin I in gametocytes of the rodent model parasite P. berghei. Using an antibody specifically recognizing F-actin and deconvolution microscopy we detected actin I in a punctate pattern in gametocytes. 3D-Structured Illumination Microscopy which allows sub-diffraction limit imaging resolved the signal into structures of less than 130 nm length. A portion of actin I was soluble, but the protein was also found complexed in a

BACKGROUND: Artemisinin-based combination therapy (ACT) remains the most effective chemotherapeutic strategy in the management of malaria. However, reports of reduced susceptibility of Plasmodium falciparum to the ACT justify the need for continued search for alternative anti-malarial drugs. The use of antibiotics with anti-malarial properties represents a potentially valuable chemotherapeutic option for the management of drug resistant infections. Thus, the intrinsic anti-malarial activity of the combination of clinical doses of rifampicin with amodiaquine and artemether was evaluated in an a

The Anopheles pseudopunctipennis nitric oxide synthase gene (ApNOS) was identified and its partial sequence showed high homology with NOS from A. stephensi, A. gambiae (putative sequence), and Drosophila melanogaster. ApNOS was mainly expressed in male and female adult mosquitoes and was induced by a blood meal. Nitric oxide (NO) was produced by in vitro-cultured mosquito midguts inoculated by enema with Plasmodium berghei ookinetes, Saccharomyces cerevisiae, Gram-positive bacteria (Micrococcus luteus), but not with Gram-negative bacteria (Klebsiella pneumoniae, Escherichia coli or Serratia ma

BACKGROUND: Plasmodium falciparum and Plasmodium vivax account for >90% global malaria burden. Transmission intervention strategies encompassing transmission-blocking vaccines (TBV) and drugs represent ideal public health tools to eliminate malaria at the population level. The availability of mature P. falciparum gametocytes through in vitro culture has facilitated development of a standard membrane feeding assay to assess efficacy of transmission interventions against P. falciparum. The lack of in vitro culture for P. vivax has significantly hampered similar progress on P. vivax and limited s

RTS,S is the first malaria vaccine recommended for implementation among young children at risk. However, vaccine efficacy is modest and short-lived. To mitigate the risk of cerebral malaria (CM) among children under the age of 5, it is imperative to develop new vaccines. EVs are potential vaccine candidates as they obtain the ability of brain-targeted delivery and transfer plasmodium antigens and immunomodulators during infections. This study extracted EVs from BALB/c mice infected with Plasmodium yoelii 17XNL (P.y17XNL). C57BL/6J mice were intravenously immunized with EVs (EV-I.V. + CM group)

Apicomplexan parasites possess specialized structures for cell invasion and replication, notably the inner membrane complex (IMC) and the subpellicular microtubules (SPMTs) situated beneath it. SPMTs are thought to interact with proteins embedded in or associated with the IMC. The cytosolic side of the IMC contains a network of proteins known as alveolins. This study focuses on a specific alveolar protein, Plasmodium berghei IMC1j (PbIMC1j). We found that PbIMC1j undergoes palmitoylation and is predominantly expressed in ookinetes. Gene knockout analysis revealed that the absence of PbIMC1j le
