Re-analysis of narcotic critical body residue data using the equilibrium distribution concept and refined partition coefficients.

Narcosis occurs as a result of the accumulation of chemicals in the phospholipid membrane. The toxic threshold concentration in the membrane is thought to be relatively constant across different chemicals and species. Hence, estimating chemical concentrations in the membrane is expected to reduce the variability of narcotic critical body residue (CBR) data. In this study, a high quality CBR dataset for three aquatic species reported recently in the literature was evaluated with the internal equilibrium distribution concept. The raw wet-weight-based CBR values were converted to membrane-weight-

Mitochondrial-targeted H2S delivery elicits broad transcriptional responses in sulfide-adapted fish.

Hydrogen sulfide (H2S) is an environmental toxicant that inhibits cellular energy production in mitochondria. Despite its toxicity, multiple lineages of the freshwater fish Poecilia mexicana (Poeciliidae) have independently colonized and adapted to H2S-rich springs. Prior studies indicated that gene expression responses to H2S exposure in both adapted and non-adapted populations extend beyond traditional toxicity targets and enzymatic detoxification pathways, suggesting a complex molecular response. Here, we investigated whether mitochondrial H2S exposure is sufficient to generate broad transc

Exposure to Predators Induces Phenotypic Plasticity in Cone Opsin Expression Profiles but not in Eye Size of Female Trinidad Guppies.

Teleost fish display very diverse visual systems through evolutionary adaptation and phenotypic plasticity to the variable light of aquatic environments. Whether predator presence influences eye size in prey is controversial because though larger eyes endow better optics, they also consume more energy and are more conspicuous. Moreover, whether predation alters the visual spectra of prey is unclear. Predation profoundly affects inter-populational divergence of male coloration and female mating preference in the Trinidad guppy (Poecilia reticulata). In this study, newborn guppies were raised un

Microplastic exposure and consumption increases susceptibility to gyrodactylosis and host mortality for a freshwater fish.

Microplastics have been found in all surveyed ecosystems and in the diet of multiple species. Detrimental health impacts of microplastic consumption include reduced growth and fecundity, metabolic stress and immune alterations for both invertebrates and vertebrates. Limited information exists, however, on how disease resistance may be affected by microplastic exposure and consumption. Here, the impact of microplastic (0.01 and 0.05 mg l-1 of polypropylene) on fish host susceptibility to disease and mortality was assessed using the guppy Poecilia reticulata-gyrodactylid Gyrodactylus turnbulli s

Sex chromosome heteromorphism and the Fast-X effect in poeciliids.

Fast-X evolution has been observed in a range of heteromorphic sex chromosomes. However, it remains unclear how early in the process of sex chromosome differentiation the Fast-X effect becomes detectible. Recently, we uncovered an extreme variation in sex chromosome heteromorphism across poeciliid fish species. The common guppy, Poecilia reticulata, Endler's guppy, P. wingei, swamp guppy, P. picta and para guppy, P. parae, appear to share the same XY system and exhibit a remarkable range of heteromorphism. Species outside this group lack this sex chromosome system. We combined analyses of sequ

First integrative study of the diversity and specificity of metacercariae of Posthodiplostomum Dubois, 1936 from native and introduced fishes in the Caribbean.

Metacercariae of the genus Posthodiplostomum are often recorded in freshwater fish hosts. While the diversity and taxonomy of this genus are receiving increasing attention in molecular phylogenetic studies, available data remain geographically biased. Most molecular studies of Posthodiplostomum and morphologically similar (neascus) worms originate in North America and Europe and Asia (more than 60% of DNA sequences are from USA and Canada), with few data currently available from the Neotropics, where high host diversity suggests high and under-sampled parasite diversity. In this study, we repo

Host heterogeneity affects both parasite transmission to and fitness on subsequent hosts.

Infectious disease dynamics depend on the speed, number and fitness of parasites transmitting from infected hosts ('donors') to parasite-naive 'recipients'. Donor heterogeneity likely affects these three parameters, and may arise from variation between donors in traits including: (i) infection load, (ii) resistance, (iii) stage of infection, and (iv) previous experience of transmission. We used the Trinidadian guppy, Poecilia reticulata, and a directly transmitted monogenean ectoparasite, Gyrodactylus turnbulli, to experimentally explore how these sources of donor heterogeneity affect the thre

Production of F1 interspecies hybrid offspring with cryopreserved sperm from a live-bearing fish, the swordtail Xiphophorus helleri.

Despite study of sperm cryopreservation in more than 200 fish species, production of broods from cryopreserved sperm in live-bearing fish has not been demonstrated. This has not been due to a lack of effort, but instead is a result of the unique morphology, biology, and biochemistry of reproduction in viviparous fishes. For example, sperm of Xiphophorus helleri have a cylindrical nucleus, can swim for days after being activated, have glycolytic capabilities, and can reside in the female reproduction tract for months before fertilization. These traits are not found in fishes with external ferti

Exploring the use of fatty acid ethyl esters as a potential natural solution for the treatment of fish parasitic diseases.

Alternatives to conventional chemical treatments for parasitic diseases in fish are needed. Microalgal-sourced fatty acid ethyl esters (FAEEs) have shown an antiparasitic effect against Gyrodactylus turnbulli infection in guppies. Here, we tested a range of commercial FAEEs of various carbon chain lengths and unsaturation levels against two fish parasites. Guppies and barramundi infected with G. turnbulli and Trichodina sp., respectively, were used. The most effective FAEE, after excluding those toxic to fish, was ethyl laurate (12:0). For both parasites, the LD50 was 18.75 μM within 250 min o

Remembering Molly: Immediate and delayed false memory formation after acute MDMA exposure.

The entactogen 3,4-Methylenedioxymethamphetamine (MDMA) is increasingly being recognized for its therapeutic potential but is also widespread in nightlife settings where it may co-occur with crime. Since previous research detected impaired verbal memory during acute MDMA intoxication, understanding the drug's ramifications in an applied legal context becomes crucial. We conducted a double-blind, placebo-controlled trial to examine acute and delayed effects of MDMA (75 mg) on false memory in 60 healthy volunteers with a history of MDMA use, using three well-established false memory methods: a b

Evenings with Molly: Adult Couples' Use of MDMA for Relationship Enhancement.

Within the modern resurgence of psychedelics as medicinal agents for a range of conditions, the story of MDMA (Ecstasy, Molly) has been re-narrated from a dangerous street drug to a breakthrough mental health therapy. Even still, the story of MDMA remains incomplete within a binary discourse of deviant recreational use versus psychotherapeutic-medical use. The present research aimed to uncover an emerging model of MDMA use grounded in the experiences of adult couples using MDMA privately and in the context of their committed relationships. Eight adult couples who self-reported active MDMA use

Convergent evolution of conserved mitochondrial pathways underlies repeated adaptation to extreme environments.

Extreme environments test the limits of life; yet, some organisms thrive in harsh conditions. Extremophile lineages inspire questions about how organisms can tolerate physiochemical stressors and whether the repeated colonization of extreme environments is facilitated by predictable and repeatable evolutionary innovations. We identified the mechanistic basis underlying convergent evolution of tolerance to hydrogen sulfide (H2S)-a toxicant that impairs mitochondrial function-across evolutionarily independent lineages of a fish (Poecilia mexicana, Poeciliidae) from H2S-rich springs. Using compar

Triple trouble? Invasive poeciliid fishes carry the introduced tilapia pathogen Gyrodactylus cichlidarum in the Mexican highlands.

As part of ongoing surveys of the gyrodactylid parasite fauna of freshwater fishes in Mexico, we recorded the infection of three species of poeciliids (Poecilia mexicana, Poeciliopsis gracilis, and Pseudoxiphophorus bimaculatus [syn.=Heterandria bimaculata]) with Gyrodactylus cichlidarum, a monogenean parasite of cichlid fishes, which has been co-introduced globally with its translocated, African "tilapia" hosts. This tilapia pathogen was found on poeciliid fishes both within their native distribution range in the Gulf of Mexico slope, as well as on invasive species artificially introduced to

Impact of host sex and group composition on parasite dynamics in experimental populations.

To better understand the spread of disease in nature, it is fundamentally important to have broadly applicable model systems with readily available species which can be replicated and controlled in the laboratory. Here we used an experimental model system of fish hosts and monogenean parasites to determine whether host sex, group size and group composition (single-sex or mixed-sex) influenced host-parasite dynamics at an individual and group level. Parasite populations reached higher densities and persisted longer in groups of fish compared with isolated hosts and reached higher densities on i

Acid-fast bacterial infection and its control in guppies (Lebistes reticulatus) reared on an ornamental fish farm in Venezuela.

There was a spontaneous outbreak of mycobacteriosis in fancy veiltail guppies, Lebistes reticulatus, raised on an ornamental fish farm in Venezuela. The clinical signs included listlessness, emaciation, spinal curvature, sunken eyes and loss of colour. Numerous acid-fast bacteria, identified as Mycobacterium species, were detected in smears from the kidneys, liver, mesentery and spleen of the fish, from fresh faecal material, and from the unborn embryos of infected gravid females. The bacteria were eradicated by the addition of kanamycin sulphate to the water at a concentration of 50 ppm, the

Experimental and molecular study of cercariae of Clinostomum sp. (Trematoda: Clinostomidae) from Biomphalaria spp. (Mollusca: Planorbidae) in Brazil.

Despite the large number of reports of species of Clinostomum from vertebrate hosts in South America, studies evaluating the molluscan transmitters of these parasites are scarce. In the present study, clinostomatoid cercariae shed from 0.02% (4/17,485) specimens of Biomphalaria spp., collected at the Pampulha reservoir, Belo Horizonte, Minas Gerais, Brazil, were used for experimental infection of Poecilia reticulata . Samples of cercariae from molluscs and metacercariae experimentally obtained from fish were subjected to morphological and molecular analyses and compared with species of Clinost

Prevalence of Centrocestus formosanus Metacercariae in Ornamental Fish from Chiang Mai, Thailand, with Molecular Approach Using ITS2.

The prevalence of Centrocestus formosanus metacercariae was investigated in ornamental fish purchased from a pet shop in Chiang Mai, Thailand, including Carassius auratus (goldfish), Cyprinus carpio (Koi), Poecilia latipinna (Sailfin Molly), Danio rerio (Zebrafish), and Puntigrus tetrazona (Tiger barb). The parasite species was identified by the morphology of worms as well as by a molecular approach using ITS2. The results showed that 50 (33.3%) of 150 fish examined were infected with the metacercariae. The highest prevalence was found in C. auratus (83.3%), and the highest intensity was noted

The Impacts of Pregnancy on Cognition and Cell Proliferation in a Live-Bearing Fish (Poeciliopsis gracilis).

Pregnancy is a high-energy process which temporarily decreases cognitive function and affects the neurogenic capacity of the maternal brain. Pregnant females exhibit decreased performance in spatial memory tasks which has been linked to altered neurogenesis in the dentate gyrus of the hippocampus. While these processes are well-resolved for mammals, whether they are conserved across other, nonmammalian, live-bearing animal lineages without placentation remains enigmatic. Here, we test the relationship between pregnancy and cognition in the live-bearing fish Poecilipsis gracilis. Female P. grac

Has recombination changed during the recent evolution of the guppy Y chromosome?

Genome sequencing and genetic mapping of molecular markers have demonstrated nearly complete Y-linkage across much of the guppy (Poecilia reticulata) XY chromosome pair. Predominant Y-linkage of factors controlling visible male-specific coloration traits also suggested that these polymorphisms are sexually antagonistic (SA). However, occasional exchanges with the X are detected, and recombination patterns also appear to differ between natural guppy populations, suggesting ongoing evolution of recombination suppression under selection created by partially sex-linked SA polymorphisms. We used mo

Context is Key: Social Environment Mediates the Impacts of a Psychoactive Pollutant on Shoaling Behavior in Fish.

Behavior-modifying drugs, such as antidepressants, are increasingly being detected in waterways and aquatic wildlife around the globe. Typically, behavioral effects of these contaminants are assessed using animals tested in social isolation. However, for group-living species, effects seen in isolation may not reflect those occurring in realistic social settings. Furthermore, interactions between chemical pollution and other stressors, such as predation risk, are seldom considered. This is true even though animals in the wild are rarely, if ever, confronted by chemical pollution as a single str
