Long noncoding RNA KCNQ1OT1 aggravates cerebral infarction by regulating PTBT1/SIRT1 via miR-16-5p.

Cerebral infarction (CI) is one of the leading causes of disability and death. LncRNAs are key factors in CI progression. Herein, we studied the function of long noncoding RNA KCNQ1OT1 in CI patient plasma samples and in CI models. Quantitative real-time PCR and Western blotting tested gene and protein expressions. The interactions of KCNQ1OT1/PTBP1 and miR-16-5p were analyzed using dual-luciferase reporter and RNA immunoprecipitation assays; MTT assays measured cell viability. Cell migration and angiogenesis were tested by wound healing and tube formation assays. Pathological changes were ana

High-throughput mutagenesis identifies mutations and RNA-binding proteins controlling CD19 splicing and CART-19 therapy resistance.

Following CART-19 immunotherapy for B-cell acute lymphoblastic leukaemia (B-ALL), many patients relapse due to loss of the cognate CD19 epitope. Since epitope loss can be caused by aberrant CD19 exon 2 processing, we herein investigate the regulatory code that controls CD19 splicing. We combine high-throughput mutagenesis with mathematical modelling to quantitatively disentangle the effects of all mutations in the region comprising CD19 exons 1-3. Thereupon, we identify ~200 single point mutations that alter CD19 splicing and thus could predispose B-ALL patients to developing CART-19 resistanc

The post-transcriptional regulatory RNA-binding protein PTBP1 functions as an inhibitor in hepatic stellate cell activation and liver fibrosis.

BACKGROUND: Hepatic stellate cell (HSC) activation is the key step in liver fibrogenesis characterized by excessive extracellular matrix (ECM) accumulation, which is based on fibrogenic factors-induced changes in gene expression in HSCs. The post-transcriptional gene-expression regulation plays a crucial role in regulation of gene expression by controlling mRNA fate through RNA binding proteins (RBPs), RBPs are essentially involved in the process of HSC activation and liver fibrogenesis. Since the activated HSCs possess the characteristics of neuron and the roles of multifunctional polypyrimid

HnRNPA1 and HuR, but not PTB, modulate cap-independent translation initiation from sense and antisense human T-cell lymphotropic virus type 1 mRNAs.

The human T-cell lymphotropic virus type 1 (HTLV-1) causes adult T-cell leukemia and HTLV-associated myelopathy/tropical spastic paraparesis. HTLV-1 transcribes sense and two antisense messenger RNAs (mRNAs). The spliced and unspliced antisense mRNA encode the HTLV-1 basic zipper factor (HBZ). The sense mRNA can initiate translation using a cap- or internal ribosomal entry site (IRES)-mediated mechanism (HTLV-1 IRES). The spliced antisense mRNA initiates translation via the sHBZ IRES. Using bicistronic vectors harboring the 5'-untranslated region of the HTLV-1 sense or spliced antisense mRNAs

The lncRNA SNHG9, an oncogenic regulator, stabilizes PRMT7 by binding to PTBP1 in the progression of colorectal cancer.

Long noncoding RNAs (lncRNAs), widely regarded as nonprotein-coding transcripts longer than 200 nucleotides in length, play active roles in tumorigenesis, including in colorectal cancer (CRC). The abnormally elevated lncRNA small nucleolar RNA host gene 9 (SNHG9) in CRC samples was observed in The Cancer Genome Atlas (TCGA) database. However, the biological role and potential mechanism of SNHG9 in CRC development remain elusive. Herein, 49 paired CRC tissues and matched adjacent normal tissues were obtained to examine SNHG9 levels. Biological functions related to cell proliferation, migration

Insulin-like growth factor-I activates extracellularly regulated kinase to regulate the p450 side-chain cleavage insulin-like response element in granulosa cells.

IGF regulates steroidogenesis in granulosa cells through expression of the cytochrome P450 side-chain cleavage enzyme (P450scc) (CYP11A1), the rate-limiting enzyme in this biosynthetic process. We showed previously that the polypyrimidine tract-binding protein-associated splicing factor (PSF) acts as a repressor, whereas Sp1 is an activator, of P450 gene expression. The aim of the present study was to investigate IGF-stimulated ERK signaling regulating P450scc gene expression in the immortalized porcine granulosa cell line JC-410. We used a reporter gene under control of the IGF response eleme

Betulonic acid, a bioactive triterpenoid from Celastrus orbiculatus extract, reverses chemoresistance in gastric cancer via CD44v6-mediated actin cytoskeleton remodeling.

Chemoresistance remains a major obstacle limiting the therapeutic efficacy of gastric cancer treatments, and cytoskeleton remodeling together with the abnormal membrane localization of drug efflux proteins may contribute to resistance phenotypes. This study aimed to investigate the role of CD44v6 in gastric cancer chemoresistance and to clarify whether betulonic acid, an active monomeric component of Celastrus orbiculatus extract, can reverse drug resistance via CD44v6. Analysis of public databases and validation in clinical tissue samples revealed that CD44v6 was highly expressed in gastric c

SPSB1 inhibition induces cancer immune evasion by modulating the KLF6/PD-L1 axis in non-small cell lung cancer cells.

Non-small cell lung cancer (NSCLC) remains among the most lethal malignancies, with aggressive progression and limited survival. The binding of programmed cell death 1 ligand 1 (PD-L1) to its receptor PD-1, which is expressed on T cells, functions as a critical checkpoint that undermines anti-tumor immune defenses. Despite encouraging anti-tumor efficacy, PD-1 inhibition yields only modest objective response rates in clinical practice. This limitation stems, in part, from an incomplete elucidation of the molecular circuitry controlling PD-L1 expression within the tumor microenvironment. In thi

The therapeutic potential and mechanisms of targeting the PTBP1/Nogo-A/NgR axis in PTSD induced by single prolonged stress in mice.

The pathophysiology of post-traumatic stress disorder (PTSD) shows notable associations with compromised hippocampal neurophysiology. Notwithstanding ongoing debates, PTBP1 knockdown (KD) demonstrates the capacity to drive glia-to-neuron reprogramming, potentially offering therapeutic benefits for some neurodegenerative pathologies. However, PTBP1 KD can upregulate the expression of Nogo-A by alternative splicing, triggering the inhibition of nerve regeneration. Currently, the role of PTBP1 in PTSD remains unknown. Here we sought to elucidate the neurorestorative effects of modulating the PTBP

Direct glia-to-neuron conversion mitigates hippocampal damage in a vascular dementia mouse model.

Vascular dementia, which is considered the second most common cause of dementia, has consistently remained a worldwide health concern. One possible approach to improving vascular dementia is by inducing neurogenesis. The asymmetric carotid artery stenosis (ACAS) model was employed and viral vectors were administered to induce an astrocytes-to-neuron conversion, including the Ptbp1 knockdown (KD) or transduction of Ascl1, NeuroD1, and Sox2 (ANS), with the aim of assessing their therapeutic effects. Mice were sacrificed 2 months after ACAS surgery and administration of the viral vector, followed

Functional validation of tensin2 SH2-PTB domain by CRISPR/Cas9-mediated genome editing.

Podocytes are terminally differentiated and highly specialized cells in the glomerulus, and they form a crucial component of the glomerular filtration barrier. The ICGN mouse is a model of glomerular dysfunction that shows gross morphological changes in the podocyte foot process, accompanied by proteinuria. Previously, we demonstrated that proteinuria in ICR-derived glomerulonephritis mouse ICGN mice might be caused by a deletion mutation in the tensin2 (Tns2) gene (designated Tns2nph). To test whether this mutation causes the mutant phenotype, we created knockout (KO) mice carrying a Tns2 pro

PTBP1 mediates Sertoli cell actin cytoskeleton organization by regulating alternative splicing of actin regulators.

Spermatogenesis is a biological process within the testis that produces haploid spermatozoa for the continuity of species. Sertoli cells are somatic cells in the seminiferous epithelium that orchestrate spermatogenesis. Cyclic reorganization of the Sertoli cell actin cytoskeleton is vital for spermatogenesis, but the underlying mechanism remains largely unclear. Here, we report that the RNA-binding protein PTBP1 controls Sertoli cell actin cytoskeleton reorganization by programming alternative splicing of actin cytoskeleton regulators. This splicing control enables ectoplasmic specializations,

Ptbp1 knockdown induces conversion of rat spinal cord astrocytes into neuron like cells.

Polypyrimidine tract-binding protein 1 (Ptbp1) is crucial in regulating neuronal differentiation and maturation through alternative splicing. Direct neuronal reprogramming via Ptbp1 knockdown has shown therapeutic effects in various neurodegenerative disease models. However, these findings are not reproducible when stringent lineage-tracing methods are used, and few studies have strictly examined the process of Ptbp1-mediated conversion in in vitro models. In this study, we knocked down Ptbp1 in rat spinal cord astrocytes using two different methods, i.e., short hairpin RNAs (shRNAs) and small

Endothelial PTBP1 Deletion in Transplanted Cardiac Tissue Limits Cardiac Allograft Vasculopathy.

BACKGROUND: Cardiac allograft vasculopathy (CAV) is a leading cause of late graft failure and mortality following heart transplantation, with limited therapeutic options. Endothelial cells (ECs), at the interface between the donor graft and host immune system, play a central role in CAV development. However, the molecular mechanisms driving endothelial dysfunction and vascular remodeling in chronic heart transplant rejection remain poorly understood. METHODS: To characterize endothelial alterations associated with CAV, we isolated nuclei from cardiac tissues of four human donor groups: (1) ea

A conserved 3'UTR short motif regulates gene expression in vertebrates.

The messenger RNA (mRNA) 3' untranslated region (3'UTR) contains important regulatory sequences, including upstream sequence elements (USEs), which regulate gene expression. One well-characterised USE in the 3'UTR of the Drosophila polo gene affects adult fly phenotypes when disrupted. We have now identified a highly conserved sequence within this USE (DplUSE) in the 3'UTR of several vertebrate genes, including in zebrafish, mouse, and human genomes and show that DplUSE enhances gene expression in human cells and zebrafish embryos. We show that, in humans, DplUSE-containing genes are associate

Lactylation of PTBP1 drives a pro-apoptotic positive feedback loop in microglia following oxygen-glucose deprivation/reoxygenation-induced injury.

Cerebral hypoxia-ischemia disrupts cellular energy metabolism and exacerbates microglial apoptosis through mechanisms that remain elusive. In this study, given the substantial lactate accumulation under hypoxic-ischemic conditions, we explored how lactylation, a lactate-derived post-translational modification, drives apoptotic signaling to identify potential therapeutic targets. Global lactylome profiling of microglia subjected to oxygen-glucose deprivation/reoxygenation (OGD/R) revealed widespread protein hyperlactylation, involving 2 555 lactylated sites across 1 071 proteins. Notably, we id

MiR-10b-5p attenuates spinal cord injury and alleviates LPS-induced PC12 cells injury by inhibiting TGF-β1 decay and activating TGF-β1/Smad3 pathway through PTBP1.

Spinal cord injury (SCI) is a debilitating condition characterized by significant sensory, motor, and autonomic dysfunctions, leading to severe physical, psychological, and financial burdens. The current therapeutic approaches for SCI show limited effectiveness, highlighting the urgent need for innovative treatments. MicroRNAs (miRNAs) like miR-10b-5p are known to play pivotal roles in gene expression regulation and have been implicated in various neurodegenerative diseases, including SCI. Polypyrimidine tract binding protein 1 (PTBP1) has also been associated with neural injury responses and

Preferential integration of adeno-associated virus type 2 into a polypyrimidine/polypurine-rich region within AAVS1.

Adeno-associated virus type 2 (AAV2) preferentially integrates its genome into the AAVS1 locus on human chromosome 19. Preferential integration requires the AAV2 Rep68 or Rep78 protein (Rep68/78), a Rep68/78 binding site (RBS), and a nicking site within AAVS1 and may also require an RBS within the virus genome. To obtain further information that might help to elucidate the mechanism and preferred substrate configurations of preferential integration, we amplified junctions between AAV2 DNA and AAVS1 from AAV2-infected HeLaJW cells and cells with defective Artemis or xeroderma pigmentosum group
