Molecular regulatory mechanisms of Populus deltoides under combined high temperature and short-photoperiod stress associated with DNA methylation.

Populus deltoides is a key species for industrial timber and ecological construction in temperate regions, where increasingly frequent and persistent heat waves pose serious challenges to its survival. However, the epigenetic mechanisms by which DNA methylation regulates environmental responses remain poorly understood. Here, whole-genome bisulfite sequencing and RNA-seq were performed on five P. deltoides genotypes grown in temperate and tropical regions. Results revealed that CG/CHG methylation stability is closely correlated with environmental sensitivity. Significant CG/CHG methylation var

Conserved 4-coumarate 3-hydroxylase/ascorbate peroxidase bifunctionality coordinates lignin deposition and plant growth in Brachypodium and Populus.

Cytosolic ascorbate peroxidases (APXs) have been proposed to have bifunctional 4-coumarate 3-hydroxylase (C3H) activity, linking redox regulation to lignin biosynthesis in plants. Although this dual role has been shown in vitro, in vivo validation remains limited. Here, we used CRISPR/Cas9 gene editing to knock out cytosolic C3H/APX genes in Brachypodium distachyon and poplar (Populus tremula × Populus alba). In Brachypodium, BdC3H/APX1 catalyzed the ascorbate-dependent hydroxylation of 4-coumarate to caffeate in vitro. Loss of BdC3H/APX1 function led to reduced lignin content, altered monomer

Elevated CO2 Shifts Photosynthetic Constraint from Stomatal to Biochemical Limitations During Induction in Populus tomentosa and Eucalyptus robusta.

The relative impacts of biochemical and stomatal limitations on photosynthesis during photosynthetic induction have been well studied for diverse plants under ambient CO2 concentration (Ca). However, a knowledge gap remains regarding how the various photosynthetic components limit duction efficiency under elevated CO2. In this study, we experimentally investigated the influence of elevated CO2 (from 400 to 800 μmol mol-1) on photosynthetic induction dynamics and its associated limitation components in two broadleaved tree species, Populus tomentosa and Eucalyptus robusta. The results show that

Cell death induction and nitric oxide biosynthesis in white poplar (Populus alba) suspension cultures exposed to alfalfa saponins.

The present work reports on the biological activity of alfalfa (Medicago sativa) saponins on white poplar (Populus alba, cultivar 'Villafranca') cell suspension cultures. The extracts from alfalfa roots, aerial parts and seeds were characterized for their saponin content by means of thin layer chromatography (TLC) and electrospray ionisation coupled to mass spectrometry. The quantitative saponin composition from the different plant extracts was determined considering the aglycone moieties and determined by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS) analyses. Acclimation and Compensating Metabolite Responses to UV-B Radiation in Natural and Transgenic Populus spp. Defective in Lignin Biosynthesis.

Plants have evolved to protect leaf mesophyll tissue from damage caused by UV-B radiation by producing an array of UV-absorbing secondary metabolites. Flavonoids (phenolic glycosides) and sinapate esters (hydroxycinnamates) have been implicated as UV-B protective compounds because of the accumulation in the leaf epidermis and the strong absorption in the wavelengths corresponding to UV. Environmental adaptations by plants also generate a suite of responses for protection against damage caused by UV-B radiation, with plants from high elevations or low latitudes generally displaying greater adap

Sex-related differences in growth and carbon allocation to defence in Populus tremula as explained by current plant defence theories.

Plant defence theories have recently evolved in such a way that not only the quantity but also the quality of mineral nutrients is expected to influence plant constitutive defence. Recently, an extended prediction derived from the protein competition model (PCM) suggested that nitrogen (N) limitation is more important for the production of phenolic compounds than phosphorus (P). We aimed at studying sexual differences in the patterns of carbon allocation to growth and constitutive defence in relation to N and P availability in Populus tremula L. seedlings. We compared the gender responses in p

E3 Ubiquitin Ligase PdSINA2D in Populus deltoides Positively Regulates ABA-Dependent Stress Tolerance through Ubiquitination of FREE1 in Poplar.

Populus deltoides "Shalinyang" (PdS) displays reduced tolerance to abiotic stresses. SINA (seven in absentia) proteins, a subtype of E3 ubiquitin ligases, play crucial roles in regulating plant growth and responses to abiotic stresses. However, the characteristics and potential functions of SINA proteins in PdS remain unclear. We identified 12 SINA genes via comprehensive bioinformatic characterization. Overexpression of PdSINA2D improved drought and saline-alkali tolerance in PdS, whereas gene silencing exacerbated plant sensitivity to these stresses. We also found that the abscisic acid (ABA

Graphene nanomaterials and copper co-exposure in Populus nigra L.: ultrastructural, biochemical, and nutritional impact under in vitro conditions.

The wide application of graphene nanomaterials has led to their release into the environment, raising ecological risk concerns, especially when co-existing with other pollutants like copper (Cu), one of the most ubiquitous environmental metals. The impact of co-presence of these nanomaterials and Cu on woody plants remains unstudied and, in this regard, callus culture represents a reliable tool for toxicological studies. In this work, we investigated the effects of Cu in combination with two different graphene nanomaterials, graphene oxide (GO) and graphene nanoplatelets (GNP), on the cell ult

Soil cadmium pollution elicits sex-specific plant volatile emissions in response to insect herbivory in eastern cottonwood Populus deltoides.

Soil heavy metal pollution is a major abiotic stressor frequently encountered by plants in conjunction with other biotic stresses like insect herbivory. Yet, it remains largely unexplored how soil metal pollution and insect herbivory act together to influence emissions of plant volatile organic compounds (VOCs), which mediate multiple ecological functions and play crucial roles in atmospheric processes. Here, we assessed the individual and combined effects of soil cadium (Cd) pollution and insect herbivory by Clostera anachoreta on VOC emissions from the seedlings of eastern cottonwood Populus

Comparative study of volatile components from male and female flower buds of Populus × tomentosa by HS-SPME-GC-MS.

The differences of volatile components in male (MFB) and female flower buds (FFB) of Populus × tomentosa were analysed and compared by HS-SPME with GC-MS for the first time. A total of 34 compounds were identified. Two clusters were clearly divided into male and female by hierarchical clustering analysis. Both the male and female flower buds showed methyl salicylate (22.83 and 24.09%, respectively) and 2-hydroxy-benzaldehyde (10.05 and 12.41%, respectively) as the main volatile constituents. The content of 2-cyclohexen-1-one, benzyl benzoate, and methyl benzoate in FFB was remarkably higher th

Ploidy and hybridity effects on leaf size, cell size and related genes expression in triploids, diploids and their parents in Populus.

Cell-size enlargement plays a pivotal role in increasing the leaf size of triploid poplar, and polyploidization could change leaf shape. ABP1 was highly expressed in triploid plants and positively related to cell size. In the plant kingdom, the leaf is the most important energy production organ, and polyploidy often exhibits a "Gigas" effect on leaf size, which benefits agriculture and forestry. However, little is known regarding the cellular and molecular mechanisms underlying the leaf size superiority of polyploid woody plants. In the present study, the leaf area and abaxial epidermal cells

Integrating dendrometric traits and wood density to predict biomass productivity, bioenergy yield, and carbon sequestration in hybrid Populus clones.

Accurate prediction of biomass productivity and bioenergy potential remains a major challenge in plantation forestry, particularly for fast-growing species such as hybrid Populus where tree architecture, wood properties, and genetic variability interact in complex ways. This study evaluates the relationships between dendrometric traits and biomass-based bioenergy indicators across 19 hybrid poplar clones cultivated under northern European conditions. Diameter at breast height (DBH), tree height, stem volume, stem dry mass, branch dry mass, and wood density were used to estimate above-ground bi

Sex-related responses of European aspen (Populus tremula L.) to combined stress: TiO2 nanoparticles, elevated temperature and CO2 concentration.

The combined effects of climate change and chemical contaminants on plant performance are still not well understood. Especially, whether different sexes of dioecious plants respond differently to combined stresses is unknown. In order to study the sex-related responses of European aspen to soil nTiO2 contamination (0, 50, 300 mg kg-1) under elevated temperature (+1.6 °C) and CO2 (730 ppm), we conducted a study in greenhouses. Ti accumulated in roots exposed to nTiO2 (1.1-3.3 and 2.7-21.1 mg kg-1 in 50 and 300 mg kg-1 treatments, respectively). Elevated CO2 had no effects on Ti uptake, while el

Chromosome-scale assemblies of the male and female Populus euphratica genomes reveal the molecular basis of sex determination and sexual dimorphism.

Reference-quality genomes of both sexes are essential for studying sex determination and sex-chromosome evolution, as their gene contents and expression profiles differ. Here, we present independent chromosome-level genome assemblies for the female (XX) and male (XY) genomes of desert poplar (Populus euphratica), resolving a 22.7-Mb X and 24.8-Mb Y chromosome. We also identified a relatively complete 761-kb sex-linked region (SLR) in the peritelomeric region on chromosome 14 (Y). Within the SLR, recombination around the partial repeats for the feminizing factor ARR17 (ARABIDOPSIS RESPONSE REGU

Decoupling photo- and thermoperiod by projected climate change perturbs bud development, dormancy establishment and vernalization in the model tree Populus.

BACKGROUND: The performance and survival of deciduous trees depends on their innate ability to anticipate seasonal change. A key event is the timely production of short photoperiod-induced terminal and axillary buds that are dormant and freezing-tolerant. Some observations suggest that low temperature contributes to terminal bud initiation and dormancy. This is puzzling because low temperatures in the chilling range universally release dormancy. It also raises the broader question if the projected climate instabilities, as well as the northward migration of trees, will affect winter preparatio

Morphometric analysis of young petiole galls on the narrow-leaf cottonwood, Populus angustifolia, by the sugarbeet root aphid, Pemphigus betae.

An insect-induced gall is a highly specialized structure resulting from atypical development of plant tissue induced by a reaction to the presence and activity of an insect. The insect induces a differentiation of tissues with features and functions of an ectopic organ, providing nutrition and protection to the galling insect from natural enemies and environmental stresses. In this anatomical and cytological study, we characterized how the gall-inducing aphid Pemphigus betae reshapes the leaf morphology of the narrow-leaf cottonwood Populus angustifolia to form a leaf fold gall. Genetic variation in NIN1 and C/VIF1 genes is significantly associated with Populus angustifolia resistance to a galling herbivore, Pemphigus betae.

The identification of genes associated with ecologically important traits provides information on the potential genetic mechanisms underlying the responses of an organism to its natural environment. In this study, we investigated the genetic basis of host plant resistance to the gall-inducing aphid, Pemphigus betae, in a natural population of 154 narrowleaf cottonwoods (Populus angustifolia). We surveyed genetic variation in two genes putatively involved in sink-source relations and a phenology gene that co-located in a previously identified quantitative trait locus for resistance to galling.

Genome-wide identification and characterization of the Populus WRKY transcription factor family and analysis of their expression in response to biotic and abiotic stresses.

WRKY proteins are a large family of regulators involved in various developmental and physiological processes, especially in coping with diverse biotic and abiotic stresses. In this study, 100 putative PtrWRKY genes encoded the proteins contained in the complete WRKY domain in Populus. Phylogenetic analysis revealed that the members of this superfamily among poplar, Arabidopsis, and other species were divided into three groups with several subgroups based on the structures of the WRKY protein sequences. Various cis-acting elements related to stress and defence responses were found in the promot
