Abbott ARCHITECT clinical chemistry and immunoassay systems: digoxin assays are free of interferences from spironolactone, potassium canrenoate, and their common metabolite canrenone.

Spironolactone, which is metabolized to canrenone, is often used in combination with digoxin. Potassium canrenoate is a similar drug that is also metabolized to canrenone. As a result of reported interference of spironolactone, potassium canrenoate, and their common metabolite canrenone with digoxin immunoassays, we investigated potential interference of these compounds with two relatively new digoxin assays for application on ARCHITECT clinical chemistry platforms (cDig, particle-enhanced turbidimetric inhibition immunoassay) and ARCHITECT immunoassay platforms (iDig, chemiluminescent micropa

Efficient Cr(VI) removal by pyrite/porous biochar: Critical role of potassium salt and sulphur.

The excessive accumulation of hexavalent chromium (Cr(VI)) in the environment poses a risk to environment and human health. In the present study, a potassium bicarbonate-modified pyrite/porous biochar composite (PKBC) was prepared in a one-step process and applied for the efficient removal of Cr(VI) in wastewater. The results showed that PKBC can significantly remove Cr(VI) within 4 h over a wide range of pH (2-11). Meanwhile, the PKBC demonstrated remarkable resistance towards interference from complex ions. The addition of potassium bicarbonate increased the pore structure of the material an

Protease inhibitor homologues of dendrotoxin do not bind to dendrotoxin acceptors on synaptosomal membranes or facilitate neuromuscular transmission.

The dendrotoxins are a homologous group of potassium channel-blocking polypeptides found in mamba snake venom. They are similar in sequence and structure to Kunitz-type serine protease inhibitors. Modified and native protease inhibitors were assayed for dendrotoxin-like activity using radioligand-binding and twitch tension-recording methods. Despite the large number and high concentration of compounds tested, no protease inhibitor displayed dendrotoxin-like activity. The results indicate that the protease-inhibiting and potassium channel-blocking activities of these two groups of polypeptides

Life-threatening hyperkalemia from nutritional supplements: uncommon or undiagnosed?

Potassium chloride and other potassium compounds are used by the general public as salt substitutes, muscle-building supplements, and panacea. Severe hyperkalemia from oral potassium is extremely rare if kidney function is normal because of potassium adaptation. The oral potassium dose has to be large enough to overcome the normal renal excretory mechanisms to cause severe hyperkalemia. This occurs most commonly in patients with renal impairment or those who take potassium-sparing diuretics, angiotensin receptor blockers, or angiotensin-converting enzyme inhibitors. We present two unique cases

Access to chiral β-sulfonyl carbonyl compounds via photoinduced organocatalytic asymmetric radical sulfonylation with sulfur dioxide.

An organocatalytic enantioselective radical reaction of potassium alkyltrifluoroborates, DABCO·(SO2)2 and α,β-unsaturated carbonyl compounds under photoinduced conditions is developed, which provides an efficient pathway for the synthesis of chiral β-sulfonyl carbonyl compounds in good yields with excellent enantioselectivity (up to 96% ee). Aside from α,β-unsaturated carbonyl compounds with auxiliary groups, common chalcone substrates are also well compatible with this organocatalytic system. This method proceeds through an organocatalytic enantioselective radical sulfonylation under photoind

Citrate and renal calculi: new insights and future directions.

Citrate is pathogenetically important in stone formation, because it retards the crystallization of stone-forming calcium salts and because its level in urine is low in many patients with nephrolithiasis. Potassium citrate is useful therapeutically, because it can often restore normal urinary citrate. Hypocitraturia often results from dietary aberrations, including sodium excess, and exaggerated intake of animal proteins. Hypocitraturia is frequently accompanied by a low net gastrointestinal absorption of alkali. New drugs are under development as improvements or refinements of currently avail

ACETYLCHOLINESTERASE INHIBITION ACTIVITY OF SOME QUINOLINYL SUBSTITUTED TRIAZOLOTHIADIAZOLE DERIVATIVES.

A series of aralkanoic acids was converted into aralkanoic acid hydrazides through their esters formation. The aralkanoic acid hydrazides upon treatment with carbon disulfide and methanolic potassium hydroxide yielded potassium dithiocarbazinate salts, which on refluxing with aqueous hydrazine hydrate yielded 5-aralkyl-4-amino-3-mercapto-1,2,4-triazoles. The target compounds, 3-aralkyl-6-(substitutedquinolinyl)[1,2,4]triazolo[3,4-b][1,3,4]thiadiazoles, were synthesized by condensing various quinolinyl substituted carboxylic acids with 5-aralkyl-4-amino-3-mercapto-1,2,4-triazoles in phosphorus

Genotoxic activity of different chromium compounds in larval cells of Drosophila melanogaster, as measured in the wing spot test.

Two chromium(VI) compounds (potassium chromate and potassium dichromate) and one chromium(III) compound, chromium chloride, were evaluated for genotoxic effects in the wing spot test of Drosophila melanogaster following standard procedures. This assay detects both somatic recombination and mutational events. The genotoxic effects were determined from the appearance of wing spots in flies transheterozygous for the third chromosome recessive markers multiple wing hairs (mwh) and flare-3 (flr(3)), as well as in flies heterozygous formwh and the multiply inverted TM3 balancer chromosome. The centrosymmetric metal metaborate KCdB3O6.

The title compound, potassium cadmium metaborate, crystallizes in a monoclinic cell, featuring infinite one-dimensional CdO(5) chains and trigonal planar B atoms in hexagonal B(3)O(6) metaborate ions. The trigonal bipyramidal CdO(5) chains and metaborate ions are interlinked to form a three-dimensional framework, creating channels running parallel to the [101] direction in which the potassium ions reside.

[Effect of malic acid salts on physical work capacity and its recovery after exhausting muscular activity].

The effect of malic acid salts of iron, potassium, copper, chromium, lithium and zinc on the motor activity and electroencephalographic indices in rats was studied under the course of oral treatment (250 mg/kg). Potassium malate was found to exert the most pronounced stimulating effect on the motor activity and excitatory processes in the sensory motor brain areas. The study of the influence of this compound on total physical working capacity and its restoration revealed a significant dose-dependent stimulating action. It was shown that the effects of potassium malate are related to its abilit

Antispasmodic effects of flavoxate, MFCA, and REC 15/2053 on smooth muscle of human prostate and urinary bladder.

The antispasmodic effects of the flavone compounds flavoxate hydrochloride, 3-methylflavone carboxylic acid (MFCA), and REC 15/2053 (and in the case of the detrusor, oxybutynin), on the human detrusor, prostatic adenoma, prostatic capsule, and bladder neck, were studied by the in vitro isometric method. All the compounds inhibited, in different orders of potency, potassium-induced contractions of the tissues. Flavoxate showed a slightly greater activity than the other two compounds in the prostatic and bladder neck tissues. However, REC 15/2053 displayed greater activity in the detrusor than i

Toxicity and mutagenicity of hexavalent chromium on Salmonella typhimurium.

Four hexavalent and two trivalent chromium compounds were tested for toxicity and mutagenicity by means of the Salmonella typhimurium/mammalian-microsome test. All hexavalent compounds yielded a complete inhibition of bacterial growth at doses of 400 to 800 mug/plate, a significant increase of his(+) revertant colonies at doses ranging from 10 to 200 mug, and no effect at doses of less than 10 mug. The distinctive sensitivity of the four Salmonella strains tested (TA1535, TA1537, TA98, and TA100) suggested that hexavalent chromium directly interacts with bacterial deoxyribonucleic acid by caus

Cytochrome P450 metabolites of arachidonic acid in the control of renal function.

Recent studies indicate that arachidonic acid is primarily metabolized by cytochrome P450 enzymes of the 4A and 2C families in the kidney to 20-hydroxyeicosatetraenoic acid (HETE), epoxyeicosatrienoic acids (EETs) and dihydroxyeicosatrienoic acids. These compounds play central roles in the regulation of renal tubular and vascular function. 20-HETE is produced by renal vascular smooth muscle (VSM) cells and is a potent constrictor that depolarizes VSM cells by blocking the calcium-activated potassium channel. Inhibition of the formation of 20-HETE blocks the myogenic response of isolated renal

Toxicity of lapachol and isolapachol and their potassium salts against Biomphalaria glabrata, Schistosoma mansoni cercariae, Artemia salina and Tilapia nilotica.

The toxicity of soluble derivatives (potassium salt) of lapachol and isolapachol in different stages of the life cycle of Schistosoma mansoni is evaluated. The potassium salts of isolapachol and lapachol showed significant molluscicidal activity against the adult snail (LC90<7 ppm) and snail egg masses (LC90<3 ppm). Cercaricidal assays revealed strong activities for both compounds. Lethality assays against brine shrimp eggs (Artemia salina Leach) indicated very high toxicity for the potassium salt of isolapachol (LC90=1.54 ppm), differently from the potassium salt of lapachol that can be consi

Regulation of ionic currents by protein kinase A and intracellular calcium in outer hair cells isolated from the guinea-pig cochlea.

Two prominent potassium currents, termed IK and IK,n, and a cation current are found in outer hair cells (OHCs) of the guinea-pig cochlea. We report here whole-cell recordings which indicate that the currents are regulated by intracellular factors. 8-bromo-cAMP (500 microM), a membrane-permeable cAMP analogue, activated potassium currents in OHCs in both apical and basal turns of the cochlea. In OHCs from the cochlear apex, the drug effect was largest at potentials positive to -40 mV, indicating IK as the target. In short cells from the cochlear base, both IK and IK,n were affected. The effect

New mixed aluminium-chromium diarsenate.

Potassium chromium aluminium diarsenate, KCr1/4Al3/4As2O7, was prepared by solid-state reaction. The structure consists of (Cr1/4/Al3/4)O6 octa-hedra and As2O7 diarsenate groups sharing corners to build up a three-dimensional anionic framework. The potassium cations are located in wide channels running along the c-axis direction. The crystal structure is isostructural with the triclinic AIMIIIX2O7 (AI = alkali metal; MIII = Al, Cr, Fe; X = As, P) compounds. However, the MIII octa-hedrally coordinated site is 25% partially occupied by chromium and 75% by aluminium.

Suppression of sensitivity to drugs and antibiotics by high external cation concentrations in fission yeast.

BACKGROUND: Potassium ion homeostasis plays an important role in regulating membrane potential and therefore resistance to cations, antibiotics and chemotherapeutic agents in Schizosaccharomyces pombe and other yeasts. However, the precise relationship between drug resistance in S. pombe and external potassium concentrations (particularly in its natural habitats) remains unclear. S. pombe can tolerate a wide range of external potassium concentrations which in turn affect plasma membrane polarization. We thus hypothesized that high external potassium concentrations suppress the sensitivity of t

Enhancement effect of potassium ferrate on self-activation: Significant improvement in pore structure and adsorption performance of activated carbon.

This work developed a method using bamboo shoot shells as raw material to produce Fe-modified ACs combining self-activation and chemical modification. Adding small amounts (0.5-5 %) of K2FeO4 accelerated the pyrolysis process and CO2 release, and reduced the activation energy and temperature of self-activation reaction. This increased the reaction rate and activation efficiency, ultimately significantly improving the pore structure of ACs. The addition of 3 % K2FeO4 resulted in a substantial increase in specific surface area and pore volume of AC, from 1340 m2/g and 0.72 cm3/g to 2184 m2/g and
