Diverse thalamic projections to the prefrontal cortex in the rhesus monkey.

We studied the sources of thalamic projections to prefrontal areas of nine rhesus monkeys with the aid of retrograde tracers (horseradish peroxidase or fluorescent dyes). Our goal was to determine the proportion of labeled neurons contributing to this projection system by the mediodorsal (MD) nucleus compared to those distributed in other thalamic nuclei, and to investigate the relationship of thalamic projections to specific architectonic areas of the prefrontal cortex. We selected areas for study within both the basoventral (areas 11, 12, and ventral 46) and the mediodorsal (areas 32, 14, 46

Prefrontal cortex synchronization with the hippocampus and parietal cortex is strategy-dependent during spatial learning.

During spatial learning, subjects progressively adjust their navigation strategies as they acquire experience. The medial prefrontal cortex (mPFC) supports this operation, for which it may integrate information from distributed networks, such as the hippocampus (HPC) and the posterior parietal cortex (PPC). However, the mechanism underlying the prefrontal coordination with HPC and PPC during spatial learning is poorly understood. Here we show that during navigation trials, mice displayed two sequential behavioral stages: searching and exploration. Exclusively during searching, mice gradually i

The Role of Insular Cortex and Prefrontal Cortex in the Pathogenesis of Fibromyalgia: Biochemical and Electrophysiological Rodent Study.

Neuroimaging implicates the medial prefrontal cortex (mPFC) and insula in fibromyalgia (FM), but synaptic and network mechanisms are unclear. To verify mPFC hypersynapticity in a reserpine-induced FM model and test whether anterior insular deep brain stimulation (DBS) restores mPFC oscillations and pain behavior. Thirty-six male Sprague-Dawley rats received reserpine (1 mg·kg⁻¹ i.p., days 1-3) or vehicle. For Objective 1, mPFC tissue was analyzed by ELISA for glutamate, γ-aminobutyric acid (GABA), C-FOS, nerve growth factor (NGF), synaptophysin, and postsynaptic density protein-95 (PSD-95). Fo

Ventral Hippocampal-Prefrontal Interaction Affects Social Behavior via Parvalbumin Positive Neurons in the Medial Prefrontal Cortex.

Ventral hippocampus (vHIP) and medial prefrontal cortex (mPFC) are both critical regions for social behaviors. However, how their interactions affect social behavior is not well understood. By viral tracing, optogenetics, chemogenetics, and fiber photometry, we demonstrated that inhibition of vHIP or direct projections from vHIP to mPFC impaired social memory expression. Via rabies retrograde tracing, we found that all three major GABAergic neurons in mPFC received direct inputs from vHIP. Activation of parvalbumin positive (PV+) neurons in mPFC but not somatostatin positive (SST+) neurons can

Prefrontal cortex hemodynamics and marksmanship accuracy following cold stress exposure.

INTRODUCTION: Military operations require performance in extreme environments, where cold stress exposure (CSE) may compromise safety and mission success. Using functional near-infrared spectroscopy (fNIRS), we examined the effects of CSE on marksmanship performance and prefrontal cortex (PFC) cerebral hemodynamics. We hypothesized that CSE will decrease performance and increase oxygenated (O2Hb) and deoxygenated (HHb) hemoglobin under cold stress. MATERIALS AND METHODS: Thirteen participants (24.3 ± 3.2 years) completed thermoneutral (TN, 24 °C air, no water immersion) and CSE (0 °C air, 15

Scn1a-mediated developmental regulation of prefrontal cortex plasticity and cognition.

OBJECTIVE: The voltage-gated sodium channel Nav1.1, encoded by Scn1a, is essential for γ-aminobutyric acid (GABA)ergic function, and its alteration is associated with neurological disorders such as Dravet syndrome and Alzheimer's disease. We previously demonstrated that local Nav1.1 dysfunction in the medial prefrontal cortex (mPFC) during adolescence causes epilepsy, cognitive deficits, and depressive-like behaviors. Here, to investigate putative developmental differences, we analyzed whether Nav1.1 dysfunction in the adult mPFC produces comparable phenotypic outcomes. METHODS: Nav1.1 dysfun

Sex-dependent effects of neonatal hyperoxia on prefrontal cortex development.

Premature infants often require supplemental oxygen therapy, a major risk factor for bronchopulmonary dysplasia and subsequent neurodevelopmental impairment. To determine how neonatal hyperoxia affects prefrontal cortex (PFC) development, we exposed neonatal mice to 85% oxygen (O₂) from postnatal day (P)1-P14 and performed integrated single-nucleus transcriptomic and chromatin accessibility profiling with in vivo and in vitro validation. Hyperoxia induced sex-dependent cellular remodeling, reducing L4/5 intratelencephalic projecting glutamatergic neurons in females and mature oligodendrocytes

Attentional selection drives information convergence within macaque lateral prefrontal cortex.

Attention is a coherent state, in which multiple brain regions converge to represent selected features of a focal object or event. Lateral prefrontal cortex (LPFC), with its flexible coding of multiple task features and their conjunctions, is widely believed to play a key role in this process. While much research has investigated biased competition between features within a brain region, and cofluctuations of activity between regions, less is known about the representational dynamics through which a coherent neural state emerges. Here, we examine directed mutual information concerning multiple

Neural Stimulation of Prefrontal Cortex Improves Food-Related Memory Suppression.

OBJECTIVE: Individuals with obesity often struggle to resist the temptation of palatable food, which may be linked to impaired inhibitory control-a crucial cognitive function for regulating eating behavior. Enhancing inhibitory control over food-related memories has been proposed as a potential intervention strategy for obesity. The right dorsolateral prefrontal cortex (dlPFC) plays a key role in regulating reward-driven impulses. However, it remains unclear whether neuromodulating the right dlPFC can selectively strengthen inhibitory control over food-related memories, particularly in individ

Comprehensive analysis of prefrontal cortex-directional rhythms categorization for rehabilitation.

Prefrontal Cortex-Directional Rhythms (PFC-DR) classification plays a significant role in Brain-Computer Interface (BCI) research since it is crucial for the effective rehabilitation of injured voluntary movements. The primary aims of this study are to conduct a thorough examination of traditional classification techniques, while emphasizing the significance of radial basis functions within support vector machine (RBF-SVM) based approaches in the context of BCI systems. Consequently, in contrast to existing machine learning-based approaches, this generalized RBF-SVM classifier effectively iden

Medial prefrontal cortex circuit dynamics involved in stage-specific addiction.

The medial prefrontal cortex (mPFC) serves as a critical hub in addiction pathology across binge/intoxication, withdrawal/negative affect, and preoccupation/anticipation/craving stages. This review provides the roles of the mPFC in different stages of addiction, and a focus on the mPFC neurotransmitter systems, neural circuits, molecules and synaptic adaptations on the regulation of addictive behaviors. Neurotransmitter systems of dopaminergic, glutamatergic, and GABAergic imbalances are related to pathological addiction. Circuits of dynamic dysregulation in the mPFC interaction with the stria

Stimulating prefrontal cortex facilitates training transfer by increasing representational overlap.

A recent hypothesis characterizes difficulties in multitasking as being the price humans pay for our ability to generalize learning across tasks. The mitigation of these costs through training has been associated with reduced overlap of constituent task representations within frontal, parietal, and subcortical regions. Transcranial direct current stimulation, which can modulate functional brain activity, has shown promise in generalizing performance gains when combined with multitasking training. However, the relationship between combined transcranial direct current stimulation and training pr

Behavioral State-Dependent Modulation of Prefrontal Cortex Activity by Respiration.

Respiration-rhythmic oscillations in the local field potential emerge in the mPFC, a cortical region with a key role in the regulation of cognitive and emotional behavior. Respiration-driven rhythms coordinate local activity by entraining fast γ oscillations as well as single-unit discharges. To what extent respiration entrainment differently engages the mPFC network in a behavioral state-dependent manner, however, is not known. Here, we compared the respiration entrainment of mouse PFC local field potential and spiking activity (23 male and 2 female mice) across distinct behavioral states: du

Rejection Distress Suppresses Medial Prefrontal Cortex in Borderline Personality Disorder.

BACKGROUND: Borderline personality disorder (BPD) is characterized by an elevated distress response to social exclusion (i.e., rejection distress), the neural mechanisms of which remain unclear. Functional magnetic resonance imaging studies of social exclusion have relied on the classic version of the Cyberball task, which is not optimized for functional magnetic resonance imaging. Our goal was to clarify the neural substrates of rejection distress in BPD using a modified version of Cyberball, which allowed us to dissociate the neural response to exclusion events from its modulation by exclusi

Fixation target representation in prefrontal cortex during the antisaccade task.

Neurons that discharge strongly during the time period of fixation of a visual target and cease to discharge before saccade initiation have been described in the brain stem, superior colliculus, and cortical areas. In subcortical structures, fixation neurons play a reciprocal role with saccadic neurons during the generation of eye movements. Their role in the dorsolateral prefrontal cortex is less obvious, and it is not known if they are activated by fixation, inhibit saccade generation, or play a role in more complex functions such as the inhibition of inappropriate responses. We examined the

Synaptic Cytoskeletal Plasticity in the Prefrontal Cortex Following Psychostimulant Exposure.

Addiction is characterized by maladaptive decision-making, a loss of control over drug consumption and habit-like drug seeking despite adverse consequences. These cognitive changes may reflect the effects of drugs of abuse on prefrontal cortical neurobiology. Here, we review evidence that amphetamine and cocaine fundamentally remodel the structure of excitatory neurons in the prefrontal cortex. We summarize evidence in particular that these psychostimulants have opposing effects in the medial and orbital prefrontal cortices ('mPFC' and 'oPFC', respectively). For example, amphetamine and cocain

Visual space is compressed in prefrontal cortex before eye movements.

We experience the visual world through a series of saccadic eye movements, each one shifting our gaze to bring objects of interest to the fovea for further processing. Although such movements lead to frequent and substantial displacements of the retinal image, these displacements go unnoticed. It is widely assumed that a primary mechanism underlying this apparent stability is an anticipatory shifting of visual receptive fields (RFs) from their presaccadic to their postsaccadic locations before movement onset. Evidence of this predictive 'remapping' of RFs has been particularly apparent within

Diffusion weighted imaging of prefrontal cortex in prodromal Huntington's disease.

Huntington's disease (HD) is a devastating neurodegenerative disease with no effective disease-modifying treatments. There is considerable interest in finding reliable indicators of disease progression to judge the efficacy of novel treatments that slow or stop disease onset before debilitating signs appear. Diffusion-weighted imaging (DWI) may provide a reliable marker of disease progression by characterizing diffusivity changes in white matter (WM) in individuals with prodromal HD. The prefrontal cortex (PFC) may play a role in HD progression due to its prominent striatal connections and doc
