Beta-lactamase production in Prevotella and in vitro susceptibilities to selected beta-lactam antibiotics [corrected].

This study looked for beta-lactamase production in 100 Prevotella isolates. MICs were determined for amoxycillin, ticarcillin, amoxycillin+clavulanate, cephalothin, cefuroxime, cefixime, cefpodoxime and cefotaxime using the reference agar dilution method (standard M11 A4, NCCLS). Beta-lactamase activity was detected in 58 of the 100 isolates, 24 of 46 black-pigmented Provotella and 34 of 54 non-pigmented Prevotella. All beta-lactamase-negative strains were susceptible to all beta-lactam antibiotics with the exception of cefuroxime and cefixime. Overall, resistance rates of Prevotella strains w

Subgingival distribution of periodontopathic bacteria in adult periodontitis and their susceptibility to minocycline-HCl.

The purpose of this study was to investigate the distribution of several periodontopathic bacteria in adult periodontitis, their in vitro susceptibility to minocycline-HCl, and whether the efficacy of the drug changes with a decrease in bacterial susceptibility. Twenty-one patients (43 to 75 years old) with 62 periodontal lesions from pockets > or =4 mm participated in the study. After subgingival sampling, an ointment containing 2% minocycline-HCl was applied locally to the selected pockets once a week for 4 weeks. The lesions were clinically examined after 1 and 4 weeks of administration. Th

Azithromycin and erythromycin susceptibility and macrolide resistance genes in Prevotella from patients with periodontal disease.

OBJECTIVES: To study oral Prevotella spp. isolated from patients with chronic periodontitis, to determine their susceptibility to azithromycin and erythromycin and to screen the presence of macrolide resistance genes therein. MATERIAL AND METHODS: Isolates with a Prevotella-like morphology were obtained from subgingival samples of 52 patients with chronic periodontitis. Each isolate was identified to the species level by sequencing of the 16S rRNA gene. In 100 Prevotella spp. isolates, azithromycin and erythromycin susceptibility was determined using the E test method, and the screening of er

Early detection of oral bacteria causing gum infections and dental caries in children.

BACKGROUND AND OBJECTIVES: Periodontal diseases are resulted from gum infections and dental plaques, which are mainly caused by the bacterial agents. Since dental monitoring includes important prognostic roles, the aim of this study was to detect the most common periodontal pathogenic bacteria in children. MATERIALS AND METHODS: A total of 200 clinical samples were collected from dental plaques and gingival grooves. Target-specific primers were designed for hbpA in Aggregatibacter actinomycetemcomitans, fimA in Porphyromonas gingivalis and 16S rRNA in Prevotella intermedia, Tannerella forsyth

The Role of the Oral Microbiome in Periodontal Disease: A Systematic Review of Microbial Associations and Therapeutic Implications.

Periodontitis is a chronic inflammatory disease driven by bacterial pathogens. This meta-analysis aimed to quantitatively assess the association between nine bacterial strains and periodontitis across 11 studies. We extracted odds ratios (ORs) and 95% confidence intervals (CIs) from studies evaluating Streptococcus mutans, Lactobacillus, Fusobacterium nucleatum, Prevotella intermedia, Staphylococcus aureus, Porphyromonas gingivalis, Aggregatibacter actinomycetemcomitans, Tannerella forsythia, and Treponema denticola. Heterogeneity was evaluated using Cochran's Q and I2 tests. We systematically

Periodontal Microbial Profiles Across Periodontal Conditions in Pediatric Subjects: A Narrative Review.

Periodontal diseases in pediatric subjects represent a challenging and relatively underexplored area compared to the extensive data available about periodontal diseases in adults. The present narrative review aims to explore the periodontal status and the related subgingival and/or salivary microbial profiles in pediatric subjects (≤18 years), focusing also on the state of health or systemic diseases. In healthy periodontium, early colonizers, such as Streptococcus and Actinomyces spp., dominate the subgingival microbiota, supporting an eubiosis state. Low levels of Candida albicans and latent

Phototargeting oral black-pigmented bacteria.

We have found that broadband light (380 to 520 nm) rapidly and selectively kills oral black-pigmented bacteria (BPB) in pure cultures and in dental plaque samples obtained from human subjects with chronic periodontitis. We hypothesize that this killing effect is a result of light excitation of their endogenous porphyrins. Cultures of Prevotella intermedia and P. nigrescens were killed by 4.2 J/cm2, whereas P. melaninogenica required 21 J/cm2. Exposure to light with a fluence of 42 J/cm2 produced 99% killing of P. gingivalis. High-performance liquid chromatography demonstrated the presence of v

[Research progress of correlation between periodontal pathogens and systemic diseases].

Periodontal pathogens are the main pathogenic factor of periodontitis. Periodontal pathogens have a large variety of virulence factors such as lipopolysaccharide, fimbriae and proteases, which enables the pathogens to infect periodontal tissues and stimulate the secretion of inflammatory cytokines, causing chronic systemic inflammation. Periodontal pathogens may invade multiple systems such as the circulatory system, immune system, respiratory system and digestive system to cause systematic diseases. Recent studies have shown that periodontal pathogens may have close relations with systemic di

[Study of adherence of periodontal pathogens to collagen-treated hydroxyapatite].

OBJECTIVE: The ability of oral bacteria to adhere to tooth surface is associated with their pathogenicity. The objective of this study was to compare the ability of 4 strains of periodontal pathogens attaching to collagen-treated hydroxyapatite (C-HA) beads in order to evaluate the ability of the main periodontal pathogens to form the biofilm on root surface. METHODS: The binding amount and binding percentage of 4 strains to C-HA were measured and compared by 3H-labeled binding assay. 4 strains of periodontal pathogens were Fusobacterium nucleatum (F. nucleatum) ATCC 10953, Porphyrin gingival

Periodontitis, periodontopathic bacteria and lactoferrin.

Lactoferrin (LF) is a component of saliva and is suspected to be a defense factor against oral pathogens including Streptococcus mutans and Candida albicans. Periodontitis is a very common oral disease caused by periodontopathic bacteria. Antimicrobial activities and other biological effects of LF against representative periodontopathic bacteria, Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis, and Prevotella intermedia, have been widely studied. Association of polymorphisms in LF with incidence of aggressive periodontitis and the role of LF in the gingival crevicular fluid as

[Clinical and microbiological effects of the initial periodontal therapy].

INTRODUCTION: Periodontitis is a destructive inflammatory disease of the tooth-supporting tissues, primarily caused by Gram-negative microorganisms. Thus, the primary objective of cause-related initial periodontal therapy is disruption and removal of the subgingival biofilm. OBJECTIVE: The aim of this study was to evaluate the clinical and microbiological effects of the initial therapy in patients diagnosed with chronic periodontitis. METHODS: Forty patients with chronic periodontitis were included in the study. As a part of the clinical assessment undertaken prior to the initial therapy, as

Culture medium influences detectability of antimicrobial effects of autologous platelet concentrates against anaerobic periodontal pathogens.

BACKGROUND: Autologous platelet concentrates (PRP, PRF, i-PRF) represent blood-derived biomaterials that have been reported to exhibit antimicrobial activity against oral microorganisms in vitro. In diffusion-based assays, visualization of inhibitory effects depends not only on bacterial growth but also on matrix-dependent diffusion behavior within the surrounding culture medium, particularly those complex media used for fastidious anaerobic species. However, it remains unclear whether commonly used agar media provide comparable assay environments for evaluating APC-derived antimicrobial effec

The association of carious dentin microflora with tissue changes in human pulpitis.

Correlations were sought between the number and type of microorganisms recovered from sampled carious dentin and the cellular responses observed in dental pulpal tissues from a sample of 65 vital, carious teeth extracted from adult humans who had declined the option of tooth restoration. Standardized homogenates of carious dentin were plated using selective and nonselective media under anaerobic and microaerophilic conditions. Pulpal responses were characterized histopathologically on the basis of the nature of the cellular infiltrate and the extent of degenerative changes. A significant assoc

Evaluation of the Antimicrobial Efficacy of Ethanolic and Aqueous Extracts of Licorice (Glycyrrhiza glabra) Against a Periodontal Pathogen: An In Vitro Study.

BACKGROUND: Periodontal disease is a major contributing factor to tooth loss and can significantly impact overall health. Traditional periodontal therapy typically involves mechanical debridement to remove plaque and calculus, along with the use of antimicrobial agents to reduce bacterial load. Licorice (Glycyrrhiza glabra) has a long history of use in traditional medicine for treating various ailments since it contains glycyrrhizin, glabridin, liquiritin, and flavonoids, which possess anti-inflammatory, antioxidant, and antimicrobial activities. Hence, licorice can be considered a viable alte

[The significance of endocrine factors and microorganisms in the development of gingivitis in pregnant women].

40-100% of pregnant women suffer from the co-called pregnancy gingivitis. The cause of pregnancy gingivitis is possible multicausal: increased plasma female sex-hormones, alteration in dental plague and perhaps Prevotella intermedia in the subgingival plague, together with alteration of immunoresponse. Increasing levels of progesterone in the gingiva as well as estrogens due to specific receptors affect vascular permeability and exudation, provoke stasis of microcirculation, increase prostaglandine E2 formation in human gingiva. Decreased gingival keratinization and capability of cell regenera
