Primary cultures of human vestibular schwannoma: selective growth of schwannoma cells.

OBJECTIVE: To establish primary vestibular schwannoma (VS) cultures that selectively favor the growth of schwannoma cells. BACKGROUND: The lack of a suitable in vitro model of human VS cells has directly limited the progress of research on tumorigenesis and therapy. The problems of establishing pure VS culture include control of fibroblast proliferation. Current efforts to extend VS cell life span using viral oncogenes, by conferring the ability to proliferate in vitro, will yield cells intrinsically different from in vivo VS tumors. Much more desirable is the ability to culture VS cells with

A comparison of the effects of verocytotoxin-1 on primary human renal cell cultures.

Infection with verocytotoxin-producing Escherichia coli causes haemolytic uraemic syndrome (HUS). Verocytotoxin-1 (VT1) is cytopathic to renal microvascular endothelial cells in culture, supporting the hypothesis that the vasculopathy of HUS is caused directly by the toxic action of VT1 on cells. We provide evidence that VT1 inhibits protein synthesis in primary cultures of glomerular epithelial cells (GE), cortical tubular epithelial cells (CTE) and mesangial cells (MC). Using 100 pg/ml of VT1 we saw a decrease in protein synthesis to 14.3+/-1.9% in vero cells (a primate cell line), 1.7+/-0.3

Testosterone metabolism in primary cultures of human prostate epithelial cells and fibroblasts.

We compare testosterone (T) metabolism in primary cultures of epithelial cells and fibroblasts separated from benign prostate hypertrophy (BPH) and prostate cancer tissues. In all cultures, androstenedione (delta 4) formed by oxidation of T by 17 beta-hydroxysteroid dehydrogenase (17 beta-HSD) represented 80% of the metabolites recovered. The amounts of 5 alpha-dihydrotestosterone (DHT), formed by reduction of T by 5 alpha-reductase (5 alpha-R), were small: 5 and 2% (BPH) and 8 and 15% (adenocarcinoma) for epithelial cells and fibroblasts, respectively. Northern blot analysis of total RNA from

Intracellular pH measurement in primary monolayer cultures of rat epididymal cells.

Intracellular pH (pHi) of primary monolayer cultures of the rat epididymal cells has been measured using fluorescent dye 2',7'-bis(carboxyethyl)-5(6)-carboxyfluorescein (BCECF). When incubated in normal Krebs-Henseleit (K-H) solution containing 25 mM HCO3, cell monolayers exhibited a basal pHi of 7.17 +/- 0.08. Amiloride (0.5 mM) added to the cell monolayers caused an immediate and sustained fall in pHi by 0.43 +/- 0.05 pH unit. Similar effects were produced when extracellular Na ions were substituted by N-methyl-D-glucamine. Addition of SITS (0.5 mM) or DPC (0.6 mM) resulted in a transient in

22Na+ uptake and catecholamine secretion by primary cultures of adrenal medulla cells.

The uptake of 22Na+ and secretion of catecholamines by primary cultures of adrenal medulla cells under the influence of a variety of agonists and antagonists were determined. Veratridine, batrachotoxin, scorpion venom, and nicotine caused a parallel increase in 22Na+ uptake and Ca2+-dependent catecholamine secretion. Ba2+, depolarizing concentrations of K+, and the Ca2+ ionophore Ionomycin stimulated secretion of catecholamines but did not increase the uptake of 22Na+. Tetrodotoxin inhibited both 22Na+ uptake and catecholamine secretion evoked by veratridine, batrachotoxin, and scorpion venom,

[Human breast cancer cells under serum-free culture. Its hormone-dependency and application to the primary culture].

Assessment of hormone-dependency of breast cancer cells in human to estradiol (E2) and progesterone (Pg) has been made by making serum-free culture of the cells, and primary culture of breast cancer cells is made by this method. The results are as follows: 1. E2 promotes proliferation of estrogen receptor (ER)-positive cells in low concentrations but rather inhibits it in high concentrations. This inhibitory effect is also observed in ER-negative cells. If the mode of this inhibitory effect is compared with the cellular dynamics of Tamoxifen (TAM) which binds predominantly to ER, the cellular

Pre-clinical phaeochromocytoma and paraganglioma models: Cell lines, animal models, and a human primary culture model.

While the establishment of human phaeochromocytoma and paraganglioma (PPGL) cell lines has proven to be particularly difficult over several decades of research, there are other reliable pre-clinical PPGL models currently available. This review provides a summary of these models, together with our recently established personalised drug screening platform using patient-derived PPGL primary cultures. Such currently available PPGL models include murine and rat PPGL cell lines, of which only one cell line (PC12) is publicly accessible through a cell repository, and PPGL animal models, of which the

Human primary middle ear epithelial cell culture: A novel in vitro model to study otitis media.

OBJECTIVES: Otitis media (OM) is a ubiquitous pediatric disease leading to a significant health care burden. There is no medication beneficial to resolving COM fluid, highlighting the need for research in the field. Crucially, current human middle ear epithelial cell models are transformed cells not recapitulating physiological functions. Herein, we describe a new method to proliferate and differentiate pediatric primary middle ear epithelial cells (pMEEC) from patients as a physiological model for the study of OM. METHODS: We adapted a cell reprogramming protocol using irradiated fibroblast

Ischemia-induced death of astrocytes and neurons in primary culture: pitfalls in quantifying neuronal cell death.

We have demonstrated that both astrocytes and cerebellar granule cell neurons die during an ischemic insult only when there is complete loss of mitochondrial membrane potential. This was determined by comparing the ability of mitochondria to sequester rhodamine 123 with the ability of cells to exclude propidium iodide. We have also demonstrated that in astrocyte cultures cellular LDH loss correlates directly with propidium iodide uptake, i.e. cell death, and inversely with rhodamine 123 uptake. Thus, both LDH loss and rhodamine 123 uptake can be used to quantitatively measure astrocyte cell de

Single-cell transcriptomic profiling of patient-derived pancreatic ductal adenocarcinoma primary cell cultures.

Single-cell RNA sequencing is a powerful approach for characterising cellular heterogeneity and elucidating transcriptional programs that drive tumour plasticity, therapeutic resistance, and disease progression. In this study, we present a single-cell transcriptomic dataset comprising 41 patient-derived cell cultures (PDCs) established from pancreatic ductal adenocarcinoma (PDAC). The dataset was generated using Evercode™ technology, which is based on Split Pool Ligation-based Transcriptome sequencing (SPLiT-seq). This scalable method enables high-throughput profiling across multiple samples w

Establishment of epithelial and fibroblast cell cultures and cell lines from primary renal cancer nephrectomies.

Renal cell carcinoma (RCC) is one of the most lethal urogenital cancers and effective treatment of metastatic RCC remains an elusive target. Cell lines enable the in vitro investigation of molecular and genetic changes leading to renal carcinogenesis and are important for evaluating cellular drug response or toxicity. This study details a fast and easy protocol of establishing epithelial and fibroblast cell cultures or cell lines concurrently from renal cancer nephrectomy tissue. The protocol involves mechanical disaggregation, collagenase digestion and cell sieving for establishing epithelial

Helicobacter pylori-infected human antral primary cell cultures: effect on gastrin cell function.

Although Helicobacter pylori infection increases gastrin secretion, it is unknown whether this is a direct effect or requires activation of the immune system. We developed an H. pylori-infected human primary antral epithelial cell culture model to address this question. This culture protocol favors growth of H. pylori, and infected cultures could be maintained for up to 48 h. These cultures were enriched for gastrin (10-40%), somatostatin (2-5%), and gastric mucin (60-80%) cells but did not contain immunocytes. Bacterial attachment occurred in a random manner within 2 h of infection, although

Mononuclear cells and cytokines stimulate gastrin release from canine antral cells in primary culture.

Patients with Helicobacter pylori-associated gastritis have an increased release of gastrin. The mechanisms by which H. pylori affects the endocrine cells are unclear. We have used primary cultures containing canine antral G cells to examine the effects of human blood mononuclear cells, purified monocytes and lymphocytes, recombinant cytokines, and NH4Cl on gastrin release. Mononuclear cells and purified monocytes in direct contact with G cells stimulated gastrin release dose dependently. Separating mononuclear cells from G cells by Transwell filters with 0.4-micron pore size still produced a

A primary cell-based fluidic co-culture model to investigate drug transport across the human placenta.

Although women often need to take medication during pregnancy, reliable human-based models mimicking the maternal-fetal interface and allowing predictions on drug transport across the human placenta are scarce. In this study, we developed a novel microfluidic Transwell-based co-culture model consisting exclusively of primary cells (trophoblasts/endothelial cells) for assessing maternal-fetal drug transfer. We aimed to (1) investigate the effects of fluidic flow on drug transfer patterns, (2) evaluate barrier integrity and different transfer processes (diffusion, active transport) across the co

Primary cell culture systems to investigate host-pathogen interactions in bacterial respiratory tract infections of livestock.

Respiratory infections of livestock represent a major health issue for the animals and cause high economic losses for the farmers. Still, little is known about the intricate interactions between host cells and the many different pathogens that cause respiratory diseases, leaving a substantial knowledge gap to be filled in order to develop effective therapies. Immortalized cell lines and two-dimensional cultures of primary respiratory epithelial cells do not reflect the complex architecture and functionality of the respiratory tract tissues. Thus, it is essential to develop and apply appropriat

Fungal endocarditis resembling primary cardiac malignancy in a patient with B-cell ALL with culture confirmation.

Fungal endocarditis is a rare subtype of infective endocarditis that often presents with nonspecific symptoms in patients with complex medical histories, making diagnosis challenging. Patients with a history of ALL may present with congestive heart failure, chemo-induced cardiomyopathy, acute coronary syndrome, cardiac lymphomatous metastasis, or infections. We present the case of a patient with a history of ALL who presented with acute coronary syndrome and imaging concerning for primary cardiac lymphoma, when in fact the patient ended up suffering from culture proven fungal endocarditis.

An improved and easy protocol for primary epithelial cell culture from atretic tissue in biliary atresia.

Biliary atresia (BA) is a lethal disease of infancy with obscure etiology. Insight into the pathogenesis of this disorder is limited by lack of availability of adequate epithelial tissue. Primary culture of human biliary epithelium may help to provide material for diagnostic and research purposes. However, culture of these cells from atretic tissue is a challenging task. We aimed to develop a reliable and easier protocol for culture of human biliary epithelial cells from excised atretic extrahepatic bile duct. An explant culture was performed using tissue obtained from 30 children with disease
