Progesterone-induced progesterone receptor membrane component 1 rise-to-decline changes are essential for decidualization.

BACKGROUND: Decidualization of endometrial cells is the prerequisite for embryo implantation and subsequent placenta formation and is induced by rising progesterone levels following ovulation. One of the hormone receptors contributing to endometrial homeostasis is Progesterone Receptor Membrane Component 1 (PGRMC1), a non-classical membrane-bound progesterone receptor with yet unclear function. In this study, we aimed to investigate how PGRMC1 contributes to human decidualization. METHODS: We first analyzed PGRMC1 expression profile during a regular menstrual cycle in RNA-sequencing datasets.

Endogenous Progesterone Concentrations Affect Progesterone Release from Intravaginal Devices Used for Oestrous Synchronization in Cattle.

Intravaginal progesterone-releasing devices are largely used both as contraceptives in humans and as a component of oestrous synchronization protocols in cattle. To reduce costs in large-scale timed artificial insemination, the reuse of these releasing devices is common. Passive hormone diffusion, however, depends on the concentration gradient, which could affect the amount of residual progesterone present in these devices after a first use. To evaluate the effect of the presence of a corpus luteum in the release of progesterone from intravaginal devices, three synchronization protocols were d

The Clinical Implications of Progesterone in Preeclampsia.

Preeclampsia is a severe complication affecting both maternal and neonatal health and is becoming a significant global public health issue. As a vital steroid hormone, progesterone (P4) plays a crucial role during pregnancy and in regulating various physiological processes. Recent studies have indicated that P4 is not only involved in pregnancy maintenance, but may also be closely related to preeclampsia pathogenesis and prevention. Previous research has suggested that P4 may participate in the mechanism of preeclampsia by regulating vascular function, immune responses, and placental function.

Progesterone: A Neuroprotective Steroid of the Intestine.

The enteric nervous system (ENS) is an intrinsic network of neuronal ganglia in the intestinal tube with about 100 million neurons located in the myenteric plexus and submucosal plexus. These neurons being affected in neurodegenerative diseases, such as Parkinson's disease, before pathological changes in the central nervous system (CNS) become detectable is currently a subject of discussion. Understanding how to protect these neurons is, therefore, particularly important. Since it has already been shown that the neurosteroid progesterone mediates neuroprotective effects in the CNS and PNS, it

Progesterone Effects in the Nervous System.

The sex hormone progesterone is mainly known as a key factor in establishing and maintaining pregnancy. In addition, progesterone has been shown to induce morphological changes in the central and peripheral nervous system by increasing dendrito-, spino-, and synaptogenesis in Purkinje cells (Wessel et al.: Cell Mol Life Sci (2014a) 1723-1740) and increasing axonal outgrowth in dorsal root ganglia (Olbrich et al.: Endocrinology (2013) 3784-3795). These effects mediated mainly by the classical progesterone receptors (PRs) A and B seem to be limited to young neurons. It may be assumed that microR

Progesterone in normal and pathological pregnancy.

Progesterone is an essential hormone in the process of reproduction. It is involved in the menstrual cycle, implantation and is essential for pregnancy maintenance. It has been proposed and extensively used in the treatment of different gynecological pathologies as well as in assisted reproductive technologies and in the maintenance of pregnancy. Called "the pregnancy hormone", natural progesterone is essential before pregnancy and has a crucial role in its maintenance based on different mechanisms such as: modulation of maternal immune response and suppression of inflammatory response (the pr

[Progesterone and prevention of preterm birth].

The literature confirms the interest of progesterone for prevention of preterm delivery in specific indications for patients carrying a singleton pregnancy. In contrast, randomized trials have shown no benefit using progesterone in the prevention of prematurity in twins and even an adverse effect.

Prematurity prevention: the role of progesterone.

PURPOSE OF REVIEW: This review summarizes the evidence of the effectiveness of progesterone on the rate of preterm birth and evaluates the most recent studies. RECENT FINDINGS: The incidence of preterm delivery is about 7-11% of all pregnant women and preterm birth is one of the most important causes of neonatal morbidity and mortality. Interventions to reduce such complications have been attempted for several years. Most efforts so far have been tertiary interventions, such as treatment with antenatal corticosteroids, tocolytic agents, and antibiotics. Some of these measures have reduced per

Effect of progesterone on embryo survival.

Increased genetic selection over the past 40 years has resulted in a dairy cow with an improved biological efficiency for producing milk but with an associated reduced fertility. Embryo loss is the greatest factor contributing to the failure of a cow to conceive. The extent and timing of embryo loss indicates that 70% to 80% of this loss occurs in the first 2 weeks after artificial insemination (AI). This is the period when a number of critical phases in embryo development occur and where protein accretion, substrate utilization and embryo metabolism increase dramatically. During this time the

Progesterone signaling in breast and endometrium.

In addition to transcriptional effects, steroid hormones rapidly activate cytoplasmic signaling cascades. The ultimate targets of these cascades are not well-defined and likely include transcription factors and coactivators. To better understand the role of the rapid "non-transcriptional" effects of progestins, we investigated the mechanisms leading to activation of these pathways and their relevance in the biological response, using two model systems: breast cancer and endometrial stromal cells. Our results demonstrated that progestins rapidly activate the Src/Erk1/2 and PI3K/Akt pathways in

Immunological and dermatological impact of progesterone.

C21 steroids and especially progesterone are involved in various physiological processes besides reproduction. Langerhans cells are becoming a major topic in immunology and, from this point, it is of high interest that progesterone is able to increase the number of Langerhans cells in different epithelial cells. This has an impact for a gender specificity of HIV1 infection, and also for the high premenstrual incidence of sexually transmitted diseases. Matrix metalloproteinase activity is of high physiological importance for tissue regeneration; progesterone suppresses the expression of some me

Reproductive functions of the progesterone receptor.

Progesterone plays a central coordinate role in regulating reproductive events associated with the establishment and maintenance of pregnancy including ovulation, uterine and mammary gland development and tumorigenesis, and neurobehavioral expression associated with sexual responsiveness. The effects of progesterone are mediated by two receptor proteins (PR), termed A and B, that arise from a single gene and act as ligand-activated transcription factors to regulate the expression of reproductive target genes. Null mutation of both proteins in mice leads to pleiotropic reproductive abnormalitie

Effectiveness of vaginal administration of progesterone.

Vaginal administration of 200 or 400 mg of progesterone in a wax suppository every 12 h in normal women was effective in rapidly achieving and maintaining serum progesterone concentrations at physiological levels appropriate to the midluteal phase of the ovarian cycle and early pregnancy. Given in the follicular phase for 5 days in five subjects at each of two doses, progesterone reached concentrations in serum after 4 and 12 h of 21-105 nmol/l (mean 46.4 nmol/l SD 17.2, n = 50) with 200 mg dosage and 21-101 nmol/l (mean 53.8 nmol/l SD 16.6, n = 49) with 400 mg dosage. Estrogen and progesterone receptors in meningiomas.

Two-thirds of all meningiomas and four-fifths of intraspinal and sphenoidal meningiomas occur in women. Meningiomas frequently enlarge or become symptomatic during pregnancy or during the luteal phase of the menstrual cycle. There is an increased incidence of meningiomas in women with breast carcinoma. In a series of 23 patients with meningiomas, the authors assayed biopsy specimens of the tumor for the presence of estrogen (ER) and progesterone (PR) receptors, using glycerol density gradient centrifugation and dextran-coated charcoal techniques. Significant levels of ER were found in only 17%

Vaginal absorption of oestradiol and progesterone.

The vaginal absorption of micronized oestradiol/progesterone from a hydrophilic matrix system was investigated before and after 10 days of treatment in 7 post-menopausal women with an average age of 56.1 yr. Oestradiol and progesterone levels were measured on days 1 and 11 before administration and 0.5, 1, 2, 3, 4, 6, 8, 12, and 24 h afterwards by radioimmunoassay. Peak values were seen 6-12 h after administration. This finding differs from those of other investigations. Sex hormone binding globulin (SHBG) and androgen concentrations were unchanged after 10 days of treatment.

The biological meaning of progesterone levels.

The effect of systematically delayed progesterone treatment was examined in 45 pregnant rats near term. Progesterone (P) and prostaglandin F (PGF) were measured in uterine vein plasma and uterine tissue before and during spontaneous labor or during prolonged pregnancy. Control animals exhibited the expected P-withdrawal (Pw) prior to spontaneous labor and properly timed P-treatment predictably prevented Pw and labor. However, when P was administered 11.7 +/- 2.8 hours (Mean +/- S.E.) before spontaneous labor, the animals delivered normally despite increased plasma and tissue P-levels. These ob
