Propionibacterium acnes Induces Autophagy in Keratinocytes: Involvement of Multiple Mechanisms.

Propionibacterium acnes is a dominant member of the cutaneous microbiota. Herein, we evaluate the effects of different P. acnes strains and propionic acid on autophagy in keratinocytes. Our results showed that P. acnes strain 889 altered the architecture of the mitochondrial network; elevated the levels of microtubule-associated protein 1 light chain 3B-II, Beclin-1, and phospho-5'-adenosine-monophosphate-activated protein kinase α; stimulated autophagic flux; facilitated intracellular redistribution of microtubule-associated protein 1 light chain 3B; increased average number of autophagosomes

Clinical impact of positive Propionibacterium acnes cultures in orthopedic surgery.

BACKGROUND: The clinical significance of a positive culture to Propionibacterium acnes in orthopedic specimens remains unclear, whether about its role as a contaminant or a pathogen, or its impact as a coinfectant. Therefore, we performed a retrospective study to provide a more accurate description of the clinical impact of P. acnes in an orthopedic population aiming to determine: 1) if there is a clinical difference between P. acnes infection and contamination? 2) If there is a clinical difference between P. acnes monoinfection, and coinfection. HYPOTHESIS: There is a clinical difference bet

Clinical Analysis of Propionibacterium acnes Infection After Total Knee Arthroplasty.

BACKGROUND: Propionibacterium acnes is a common cause of upper extremity arthroplasty infection and usually presents in an indolent subacute fashion. It is not well described how total knee arthroplasty (TKA) patients infected with P acnes present. METHODS: We retrospectively compared patients undergoing revision TKA for infection from P acnes and methicillin-sensitive Staphylococcal aureus (MSSA) in our institutional infection database. Patients were classified as having a periprosthetic joint infection based on the Musculoskeletal Infection Society criteria and were excluded if they had a p

Propionibacterium skull base osteomyelitis complicated by internal carotid artery pseudoaneurysm.

The microbiology of skull base osteomyelitis (SBO) is evolving. We present here the first case of SBO caused by Propionibacterium acnes leading to the development of a pseudoaneurysm of the internal carotid artery. Otolaryngologists should recognize this pathogen as a potential cause of invasive temporal bone infection to optimize prompt diagnosis and treatment. Laryngoscope, 127:2337-2339, 2017.

In vitro anti-propionibacterium activity by curcumin containing vesicle system.

Propionibacterium acnes acts a critical role in the development of inflammatory acne when it overgrows in pilosebaceous units. The spread of multiple drug resistance bacteria indicates a growing need for new antimicrobial agents. The objective of this study is to develop lipid vehicles to deliver curcumin and inhibit P. acnes in the skin. The inhibitory activities of the curcumin containing vehicles against P. acnes were studied by the bioluminescence assay. Curcumin accumulation patterns in neonate pig skin were studied using Franz diffusion cells and confocal laser scanning microscopy. The p

A rare complication of Nd-YAG capsulotomy: propionibacterium acnes endopthalmitis.

Posterior capsular opacification, also known as after cataract develops in 33 % of patients after uneventful cataract surgery. Nd-YAG Laser capsulotomy is the treatment of choice for PCO and is routinely performed by the general ophthalmologist on an OPD basis. Endophthalmitis, although a rare complication after Nd-YAG Laser capsulotomy, has been reported in the literature. We report a case of Propionibacterium acnes endophthalmitis following laser capsulotomy and the possible mechanism of its occurrence.

Comparative photodynamic evaluation of new phenothiazinium derivatives against Propionibacterium acnes.

The screening of recently patented derivatives of the standard phenothiazinium agents methylene blue and toluidine blue was carried out against Propionibacterium acnes, the main bacterium associated with acne vulgaris. Comparative tests were made using the topical agent benzoyl peroxide (BPO) and standard tetracyclines. Each of the photosensitizers employed, including the lead compounds, was photobactericidal at lower concentrations than BPO, and produced a much more rapid kill than the tetracyclines. In addition, the tetracyclic and pentacyclic phenothiazinium derivatives exhibited high light

The antimicrobial activity of liposomal lauric acids against Propionibacterium acnes.

This study evaluated the antimicrobial activity of lauric acid (LA) and its liposomal derivatives against Propionibacterium acnes (P. acnes), the bacterium that promotes inflammatory acne. First, the antimicrobial study of three free fatty acids (lauric acid, palmitic acid and oleic acid) demonstrated that LA gives the strongest bactericidal activity against P. acnes. However, a setback of using LA as a potential treatment for inflammatory acne is its poor water solubility. Then the LA was incorporated into a liposome formulation to aid its delivery to P. acnes. It was demonstrated that the an

[Hypertrophic cranial pachymeningitis with propionibacterium acnes detected by dural biopsy].

A 30-year-old woman experienced recurrent dull headache and frequent partial motor seizure (Jacksonian type) that marched from right fingers. This motor seizure was uncontrollable with ordinary anticonvulsant therapy. Cerebrospinal fluid showed mild pleocytosis. Cranial CT examination was unremarkable, but MRI revealed thickened dural lesion on the left fronto-parietal site, giving diagnosis of hypertrophic cranial pachymeningitis. Dural biopsy showed nonspecific chronic granulomatous state without specific granuloma such as tuberculosis nor sarcoidosis. Anaerobic culture revealed Propionibact

[Intrascrotal and seminal vesicular granuloma probably induced by propionibacterium acnes].

A case of right intrascrotal and seminal vesicular granuloma probably induced by Propionibacterium acnes is reported. A 62-year-old man was admitted to our department because of low grade fever and the right intrascrotal and retrovesical masses without tenderness. Ultrasonography computed tomography, magnetic resonance imaging and vesiculography suggested a neoplasm. However, leukocytosis and high erythrocyte sedimentation rate (ESR) and c-reactive protein level (CRP) suggested inflammatory disease. He was treated for 1 week with parenteral cefazolin (CEZ) (4 g/day), and the intrascrotal mass

Immunopotentiation of cell-mediated hypersensitivity by Corynebacterium parvum (Propionibacterium acnes).

We have investigated the activity of local Corynebacterium parvum (Propionibacterium acnes) as an immunopotentiator of allergic contact dermatitis (ACD) and photo-ACD in rodents. C. parvum given into the sensitization site, but not at a distance, increased the delayed-type hypersensitivity to the chemical allergen. An immunoadjuvant effect could be realized even when C. parvum was given several days after allergen; it was readily obtained in B cell-deficient mice. Cyclophosphamide pretreatment heightened the immunopotentiation seen with local C. parvum. It is unlikely that the specific immunop

Propionibacterium acnes-induced immunopathology correlates with health and disease association.

Genomic studies revealed the existence of health- and acne-associated P. acnes strains and suggested novel approaches for broadening understanding of acne vulgaris. However, clinical association of P. acnes with disease or health has yet to be corroborated experimentally. Current animal models of acne do not closely mimic human disease and have unclear translational value. We have developed a murine model of acne by combining P. acnes inoculation with topical application of a synthetic human sebum. We showed that human sebum promoted persistence of intradermally injected P. acnes with little l

Histological Identification of Propionibacterium acnes in Nonpyogenic Degenerated Intervertebral Discs.

Purpose. Low-virulence anaerobic bacteria, especially the Propionibacterium acnes (P. acnes), have been thought to be a new pathogeny for a series of disc diseases. However, until now, there has been no histological evidence to confirm this link. The purpose of this study was to confirm the presence of P. acnes in nonpyogenic intervertebral discs via histological observation. Method. Degenerated intervertebral discs were harvested from 76 patients with low back pain and/or sciatica but without any symptoms of discitis or spondylodiscitis. The samples were cultured under anaerobic conditions an

Propionibacterium in Shoulder Arthroplasty: What We Think We Know Today.

➤ Propionibacterium is a slow-growing gram-positive rod that is part of the normal skin microbiome but can be found on culture of specimens from a large number of patients having revision shoulder arthroplasty performed for pain, stiffness, and component loosening. ➤ Propionibacterium infections do not present with obvious signs of infection, such as swelling, erythema, drainage, or tenderness, but rather are of the so-called stealth type, presenting with unexplained pain, stiffness, or component loosening months to years after the index arthroplasty. ➤ Not all propionibacteria are the same: c

Investigation of antibacterial activity of aspidin BB against Propionibacterium acnes.

In the present study, antibacterial activity of four kinds of phloroglucinol derivatives extracted from Dryopteris fragrans (L.) Schott against S. aureus, S. epidermidis and P. acnes has been tested. Aspidin BB exerted the strongest antibacterial activity with minimal inhibition concentration (MIC) values ranging from 7.81 to 15.63 μg/mL. The time-kill assay indicated that aspidin BB could kill P. acnes completely at 2 MIC (MBC) within 4 h. By using AFM, we demonstrated extensive cell surface alterations of aspidin BB-treated P. acnes. SDS-PAGE of supernatant proteins and lipid peroxidation re

Propionibacterium acnes infection in shoulder arthroscopy patients with postoperative pain.

BACKGROUND: Recent studies have identified Propionibacterium acnes as the causal organism in an increasing number of postoperative shoulder infections. Most reports have found a high rate of P acnes infection after open surgery, particularly shoulder arthroplasty. However, there are limited data regarding P acnes infections after shoulder arthroscopy. MATERIALS AND METHODS: We prospectively collected data on all shoulder arthroscopies performed by the senior author from January 1, 2009, until April 1, 2013. Cultures were taken in all revision shoulder arthroscopy cases performed for pain, sti

Propionibacterium acnes isolated from lymph nodes of patients with sarcoidosis.

Propionibacterium acnes has been repeatedly suggested as a candidate causative agent of sarcoidosis. It is the only microorganism that has been isolated from sarcoid lesions by bacterial culture so far and this has been described in Japanese patients only. We report two non-Japanese patients in whom mediastinoscopy was performed in order to obtain lymph node tissue for histopathology, which was suggestive for sarcoidosis. Bacterial culture of these uncontaminated mediastinal lymph nodes revealed P. acnes in both patients. As shown in these two cases, P. acnes can be isolated from sterile biops

New insights into acne pathogenesis: propionibacterium acnes activates the inflammasome.

The precise contribution of the commensal bacterium Propionibacterium acnes (P. acnes) in the inflammatory response associated with acne vulgaris remains controversial. In this issue Qin et al. show that P. acnes induces robust IL-1β secretion in monocytic cells by triggering the activation of the NLRP3 inflammasome. In vivo, the encounter of P. acnes and macrophages in the peri-follicular dermis could locally result in the release of substantial amounts of IL-1β and therefore exacerbate inflammation. Such findings suggest that molecules targeting IL-1β and/or the NLRP3 inflammasome may consti
