Production of cybrids in Brassicaceae species.

This chapter describes a method of cytoplasm transfer within the brassicaceae family through Ca-PEG-mediated protoplast fusion. The method includes a protocol of nonmutagenic albinism induction based on spectinomycin-induced plastid ribosome deficiency (PRD). The proposed application of spectinomycin-mediated albinism allows speeding up creation of albino lines, as well as of hybrid production with substituted cytoplasm. According to described method cybrids between Orychophragmus violaceus and Brassica napus, O. violaceus and Lesquerella fendleri have been produced. Methods of further molecul

[The ultrastructural characteristics of Neisseria meningitidis strains isolated in different forms of infection].

The electron microscopic study of cell populations of the first generation of several N.meningitidis strains, isolated from humans and grown on artificial culture medium (Hottinger's serum agar) has made it possible to establish that these organisms are morphologically heterogeneous. It was manifested by the presence of 7 morphological variants of cells in their cultures (different strains have different proportions of these variants): (a) "normal" and similar to normal diplococcal variants; (b) atypically dividing cells; (c) spheroplastic forms; (d) protoplastic forms (typical and atypical);

Establishment of a CRISPR/Cas9-mediated system for targeted editing of the MFS gene in mint.

The key message of this study is that we established a CRISPR/Cas9-mediated genome-editing system for Mentha haplocalyx "738" by optimizing protoplast transient assay and screening effective regulatory elements. Targeted knockout of the MFS gene generated edited mint plants with reduced menthofuran content, offering a strategy for quality improvement of mint essential oil. The commercial value of mint (Mentha spp.) essential oil is often diminished by the presence of undesirable metabolites, notably menthofuran, which impairs flavor and raises safety concerns. This study aimed to develop a rob

AtPNP-A is a systemically mobile natriuretic peptide immunoanalogue with a role in Arabidopsis thaliana cell volume regulation.

Cellular and physiological evidence suggests the presence of a novel class of systemically mobile plant molecules that are recognized by antibodies against vertebrate atrial natriuretic peptides (ANPs). In order to characterize the function of these immunoanalogues we have expressed the full-length recombinant (AtPNP-A[1-126]) and demonstrate that this molecule induces osmoticum-dependent H(2)O uptake into protoplasts at nanomolar concentrations and thus affects cell volume. A similar response is also seen with a recombinant that does not contain the signal peptide (AtPNP-A[26-126]) as well as

In vitro and in vivo antipseudomonal activity, acute toxicity, and mode of action of a newly synthesized fluoroquinolonyl ampicillin derivative.

Compounds N-(6,7-difluoroquinolonyl)-ampicillin (AU-1) and N-(6-fluoroquinolonyl)-ampicillin (FQ-1), synthesized by coupling of the carboxyl group of 6,7-difluoroquinolone (FP-3) and 6-fluoroquinolone (FP4), respectively, with the alpha-amino-group of ampicillin side chain, exhibit antipseudomonal activity similar to and lower acute toxicity than that of norfloxacin, whereas neither ampicillin nor the fluoroquinolone moieties, compound FP-3 or FP4, alone have such activity. Also, AU-1 and FQ-1 are active against tested clinical isolates of Pseudomonas aeruginosa that are highly resistant to no

Cell-based CRISPRa screen identifies a papillae-specific receptor kinase mediating broad-spectrum fungal resistance in rice.

Gain-of-function screens enable the discovery of gene functions and candidate targets for molecular breeding. Here we developed a protoplast-based dCas9-TV-mediated CRISPR activation screen in rice and identified OsTV1, an epidermal papillae-specific receptor kinase, as a positive regulator of fungal resistance. OsTV1 overexpression enhanced silicon deposition, leaf rigidity and basal defence gene expression, conferring broad-spectrum fungal resistance without compromising growth. This study establishes a high-throughput gain-of-function screen adaptable for investigating diverse cellular proc

Plant Tissue Cultures.

Plant tissue cultures are an efficient system to study cell wall biosynthesis in living cells in vivo. Tissue cultures also provide cells and culture medium from which enzymes and cell wall polymers can easily be separated for further studies. Tissue cultures with tracheary element differentiation or extracellular lignin formation have provided useful information related to several aspects of xylem and lignin formation. In this chapter, methods for nutrient medium preparation and callus culture initiation and its maintenance as well as those for protoplast isolation and viability observation a

Short-term and long-term clinostat and vibration-induced biochemical changes in dwarf marigold stems.

Stems of 21-day dwarf Marigold plants cultivated on the clinostat were compared with plants cultivated on vertical axis rotators ("vibrational controls") and stationary controls for long-term changes in cell wall composition. Stems of 21-day plants grown under stationary conditions and subsequently exposed to the clinostat for 24 hours were also analyzed. Among the long-tern markers, calciun, lignin, and protein-bound hemicellulose (possibly cell wall glycoprotein) clearly differentiated the effects of vibration from those of the clinostat. Short-term differential responses included rate of et

[Ultrastructure of parasitic forms of the agent of coccidioidomycosis].

The authors studied the ultrastructure of the Coccidiodes immitis spherules in the sections of the abdominal cavity foci and pneumonic foci of Cricetus auratus W. and in albino mice infected intraperitoneally with a lethal dose of the mycelium fungus phase. It was found that the spherules of coccidiod fungus represented multinuclear cells with a well-organized protoplast included into a complicated supermembrane structure--the cell wall. Endospores (which copied the spherule structure in miniature) formed as a result of division of the cytoplasm by entire sections into polygonal segments with

Effect of oxygen on the non-photochemical quenching of vascular plants and potential oxygen deficiency in the stroma of PsbS-knock-out rice.

The excessive and harmful light energy absorbed by the photosystem (PS) II of higher plants is dissipated as heat through a protective mechanism termed non-photochemical quenching (NPQ) of chlorophyll fluorescence. PsbS-knock-out (KO) mutants lack the trans-thylakoid proton gradient (ΔpH)-dependent part of NPQ. To elucidate the molecular mechanism of NPQ, we investigated its dependency on oxygen. The development of NPQ in wild-type (WT) rice under low-oxygen (LO) conditions was reduced to more than 50% of its original value. However, under high-oxygen (HO) conditions, the NPQ of both WT and Ps

Ultrastructural characterization of stable L-form cells from Erysipelothrix rhusiopathiae and of accompanying artifacts.

The stable L-form of Erysipelothrix rhusiopathiae is a typical protoplast type L-form. Cells are surrounded by a trilamellar cytoplasmic membrane only. They grow in form of aggregations in liquid media and their diameters vary between 0.1 and 2 micrometer. Always a large portion of cells undergoes lysis. It seems to be characteristic for L-form cultures of E. rhusiopathiae that always many artifact structures are formed. The artifacts are spherical particles with diameters of 0.1 micrometer to more than 3 micrometer. They can be differentiated from L-form cells only by electron microscopy. The

Change in the surface hydrophobicity of substrate cells during bdelloplast formation by Bdellovibrio bacteriovorus 109J.

During intraperiplasmic growth of Bdellovibrio bacteriovorus 109J, the substrate cell surface becomes more hydrophobic. This was shown (i) by comparing the sensitivity to hydrophobic antibiotics of wild-type and lipopolysaccharide mutant strains of Salmonella typhimurium to that of the bdellovibrio growing on these strains and (ii) by measuring the binding efficiency of these strains, Escherichia coli, and their derived bdelloplasts to octyl Sepharose. The kinetics of increase in surface hydrophobicity was similar to the kinetics of the conversion of the substrate cell peptidoglycan to a lysoz

Phosphorylation of Arabidopsis response regulator 7 (ARR7) at the putative phospho-accepting site is required for ARR7 to act as a negative regulator of cytokinin signaling.

Cytokinins are plant hormones that regulate diverse aspects of plant growth and development. Arabidopsis cytokinin signal transduction utilizes a multi-step two-component signaling (TCS) system by histidyl-aspartidyl phosphorelays. We here show that phosphorylation of ARR7, an A-type response regulator that acts as a negative regulator of cytokinin signaling, is required for its function in plants. Phosphorylation of ARR7 is inhibited in vitro by mutation in a putative phospho-accepting Asp residue into an Asn residue (ARR7(D85N)). While ectopic expression of ARR7 decreases root-growth inhibit

Non-photoaging and eco-corona reinforce nanoplastics-root engagement via root exudates in the rhizosphere.

How non-photoaging alters the binding of rhizosphere nanoplastics to root exudates (eco-corona) and subsequent root surface interactions remains unclear. Here, polystyrene nanoplastics (PSNPs) were subjected to Fenton and sulfidation aging. Spectroscopic analyses showed that root exudates formed distinct eco-coronas on pristine and aged PSNPs via non-covalent interactions. Aging shifted the binding mode from weak hydrophobic adsorption on pristine PS to hydrogen-bonding interactions with organic acids on aged PS. Consequently, colloidal behaviors differed, with hydrodynamic sizes increasing to

Genetic tools for transformation of Xyleborus ambrosia beetle fungal symbionts.

BACKGROUND: Ambrosia beetles rely on obligate fungal partners for survival, and these associations are emerging as model systems for examining the development and evolution of fungal-animal mutualism. However, genetic tools for the manipulation of such mutualistic fungi remain largely lacking, and consequently the genetic basis employed by these fungi to establish associations with their beetle partners remains almost completely unexplored. Here, we provide methods for protoplast generation and transformation of several major filamentous fungal partners of Xyleborus ambrosia beetles including

Overproduction of rifamycin B by Amycolatopsis mediterranei and its relationship with the toxic effect of barbital on growth.

A novel method for selecting overproducing strains of rifamycin B was developed. This technique involves the use of lysozyme and the effect of barbital on the growth of A. mediterranei. Complete medium added with glycine and barbital was inoculated with mutagenized mycelium, incubated for 48 hours and treated with lysozyme. The lysozyme resistant mycelium was washed with dilute detergent. Complete medium with glycine but without barbital was inoculated with the washed mycelium. Protoplasts were obtained and regenerated and the colonies were picked and seeded on Bennet agar plates with and with

Genetic studies on Staphylococcal strains from patients with toxic shock syndrome.

Thirteen isolates of Staphylococcus aureus that produce the toxic shock syndrome exotoxin were screened to identify and characterize this specific determinant and understand its role in pathogenicity. These stains belong to phage group I, are sensitive to phage 29, and are similar with respect to their resistance to cadmium, arsenate, and penicillin. These three resistances, commonly found on plasmids in many strains of S. aureus, were not plasmid-associated in 13 toxic shock strains. The cadmium and arsenate resistances were cotransferred both in transduction and in protoplast fusion; penicil

A Rapeseed WRKY Transcription Factor Phosphorylated by CPK Modulates Cell Death and Leaf Senescence by Regulating the Expression of ROS and SA-Synthesis-Related Genes.

Salicylic acid (SA) and reactive oxygen species (ROS) are two well-defined inducers of leaf senescence. Here, we identified a novel WRKY transcription factor gene WSR1 (WRKY regulating SA and ROS 1) in Brassica napus (rapeseed) in promoting SA and ROS production, which eventually led to leaf senescence thereafter. Its expression increased in senescing leaves. Ca2+-dependent protein kinase (CPK) 5 and -6 interacted with and phosphorylated BnaWSR1. Overexpression of phosphomimic BnaWSR1 (BnaWSR1ca) in rapeseed protoplasts elicited ROS production and cell death while its ectopic expression in Ara

EVIDENCE FOR TUBULAR MATING STRUCTURES INDUCED IN EACH MATING TYPE OF HETEROTHALLIC GONIUM PECTORALE (VOLVOCALES, CHLOROPHYTA)(1).

Gametes were induced separately in cultures of each mating type of the heterothallic, isogamous colonial volvocalean Gonium pectorale O. F. Müll. to examine the tubular mating structure (TMS) of both mating types plus and minus (plus and minus), referred to as "bilateral mating papillae." Addition of dibutyryl cyclic adenosine monophosphate (DcAMP or db-cAMP) and 3-isobutyl-1-methylxanthine (IBMX) to approximately 3-week-old cultures of each mating type induced immediate release of naked gametes from the cell walls. Both plus and minus gametes formed a TMS in the anterior region of the protopl
