Increased asymmetric pulvinar magnetic resonance imaging signals in Creutzfeldt-Jakob disease with florid plaques following a cadaveric dura mater graft.

A 9-year-old Japanese girl received a cadaveric dura mater graft during surgery following a head injury with brain contusion. She continued to do well, but when she became 19-years-old, she gradually showed a violent character and was treated in a psychiatric hospital. Another 6 years later, 200 months after the procedure, she developed a progressive gait ataxia, which subsequently led to her death within 10 months of onset. An autopsy showed she had CJD. This patient represents an atypical case of dura-associated CJD (dCJD) with unusual clinicopathological features including the late occurren

Transient Subplate Sublayer Forms Unique Corridor for Differential Ingrowth of Associative Pulvinar and Primary Visual Projection in the Prospective Visual Cortical Areas of the Human Fetal Occipital Lobe.

Cytoarchitectonical parcellation of the visual cortex into the striate and extrastriate cortex requires complex histogenetic events within a precise spatio-temporal frame to attain the specification of areal domains and associated thalamocortical connections during the fetal brain development. We analyzed a deep subplate cellular monolayer (subplate "corridor" cells) present during a restricted period of 13-15 postconceptional weeks, showing the 3D caudo-ventro-medial position in the human fetal occipital lobe, corresponding to the segregation point of pulvinocortical and geniculocortical fibe

Thalamic projections to the posterior suprasylvian gyrus and the ventrally adjacent cortex in the cat traced with horseradish peroxidase.

The present study has been undertaken to elucidate the thalamic projections to the posterior suprasylvian gyrus (PSG) and the ventrally adjacent areas in the cat using retrograde axonal transport of horseradish peroxidase. The posterior part of the PSG, area 19 b, receives fibers mainly from the dorsolateral part of the medial pulvinar nucleus, and a lesser number from the lateral pulvinar nucleus. It receives a few fibers from the inferior pulvinar nucleus and the medial interlaminar nucleus (NIM) of the lateral geniculate. The anterior part of the PSG, area 21 b, receives a large number of f

Complementary Organization of Mouse Driver and Modulator Cortico-thalamo-cortical Circuits.

Corticocortical (CC) projections in the visual system facilitate hierarchical processing of sensory information. In addition to direct CC connections, indirect cortico-thalamo-cortical (CTC) pathways through the pulvinar nucleus of the thalamus can relay sensory signals and mediate cortical interactions according to behavioral demands. While the pulvinar connects extensively to the entire visual cortex, it is unknown whether transthalamic pathways link all cortical areas or whether they follow systematic organizational rules. Because mouse pulvinar neurons projecting to different areas are spa

Bilateral and ipsilateral ascending tectopulvinar pathways in mammals: a study in the squirrel (Spermophilus beecheyi).

The mammalian pulvinar complex is a collection of dorsal thalamic nuclei related to several visual and integrative processes. Previous studies have shown that the superficial layers of the superior colliculus project to multiple divisions of the pulvinar complex. Although most of these works agree about the existence of an ipsilateral tectopulvinar projection arising from the stratum griseum superficialis, some others report a bilateral projection originating from this same tectal layer. We investigated the organization of the tectopulvinar projections in the Californian ground squirrel using

Thalamic lesions revealed by MR associated with periventricular leukomalacia and clinical profiles of subjects.

Magnetic resonance (MR) findings at the cerebral white matter and the thalamus in 44 children with spastic cerebral palsy born at preterm were analysed. Periventricular leukomalacia (PVL) was found in all of the children. Lesions of the thalamus were revealed in 22 children, 19 of which were in the anterior part of the pulvinar and 3 of which were in other areas. Gestational ages and birthweights of the children with a lesion of the pulvinar were significantly greater than those without lesions of the thalamus. Mental retardation and paroxysmal ocular downward deviation were more frequently se

Thalamic connections of the caudal part of the posterior parietal cortex differ from the rostral part in galagos (Otolemur garnettii).

In this study, thalamic connections of the caudal part of the posterior parietal cortex (PPCc) are described and compared to connections of the rostral part of PPC (PPCr) in strepsirrhine galagos. PPC of galagos is divided into two parts, PPCr and PPCc, based on the responsiveness to electrical stimulation. Stimulation of PPC with long trains of electrical pulses evokes different types of ethologically relevant movements from different subregions ("domains") of PPCr, while it fails to evoke any movements from PPCc. Anatomical tracers were placed in both dorsal and ventral divisions of PPCc to

Vestibular nucleus-thalamus-cortex pathway abnormalities persist in patients with chronic unilateral vestibulopathy after the establishment of vestibular static compensation.

BACKGROUND: After vestibular impairment, the body undergoes static and dynamic compensation. Static compensation is characterized by the resolution of spontaneous nystagmus, indicating rebalanced bilateral vestibular nuclei activity. It remains unclear whether ascending vestibular projections remain abnormal thereafter. This study investigated abnormal functional activity along vestibular nucleus-thalamus-cortex projection pathways in chronic unilateral vestibulopathy (CUVP) after static compensation using degree centrality (DC) and functional connectivity (FC) analyses. METHODS: In total, 25

Abnormal microstructure of the atrophic thalamus in preterm survivors with periventricular leukomalacia.

BACKGROUND AND PURPOSE: The neuroanatomic substrate of cognitive deficits in long-term survivors of prematurity with PVL is poorly understood. The thalamus is critically involved in cognition via extensive interconnections with the cerebral cortex. We hypothesized that the thalamus is atrophic (reduced in volume) in childhood survivors of prematurity with neuroimaging evidence of PVL and that the atrophy is associated with selective microstructural abnormalities within its subdivisions. MATERIALS AND METHODS: We performed quantitative volumetric and DTI measurements of the thalamus in 17 chil

Specificities of arterial spin labeling (ASL) abnormalities in acute seizure.

PURPOSE: Arterial spin labeling (ASL) is a non-invasive tool measuring cerebral blood flow (CBF) and is useful to assess acute neurological deficit. While acute stroke presents as hypoperfused vascular territory, epileptic activity causes cortical hyperperfusion. Other neurological conditions exhibit hyperperfusion, like migraine or secondary "luxury perfusion" in strokes. Our objectives were to evaluate the usefulness and potential specificities of ASL in acute seizure and correlate it with electroencephalogram. MATERIALS AND METHODS: Amongst a cohort of patients with neurological deficit, a

Do Subsets of Widefield Vertical Cells Exist in the Mouse Superior Colliculus?

Projections from widefield vertical (WFV) cells of the superior colliculus to the pulvinar nucleus form a conserved pathway for motion detection across mammalian species. While subtypes of tectopulvinar cells have been identified in the ground squirrel, it is unclear whether subtypes can be identified in mice. Using viral tracing, immunohistochemistry, and confocal and electron microscopy in Tac1, Ntsr1-GN209, and C57BL/6J mice, we attempted to characterize WFV cells based on their morphology, pulvinar projection patterns, expression of substance P, parvalbumin, or hyperpolarization-activated

Transient visual pathway critical for normal development of primate grasping behavior.

An evolutionary hallmark of anthropoid primates, including humans, is the use of vision to guide precise manual movements. These behaviors are reliant on a specialized visual input to the posterior parietal cortex. Here, we show that normal primate reaching-and-grasping behavior depends critically on a visual pathway through the thalamic pulvinar, which is thought to relay information to the middle temporal (MT) area during early life and then swiftly withdraws. Small MRI-guided lesions to a subdivision of the inferior pulvinar subnucleus (PIm) in the infant marmoset monkey led to permanent de

Dissociable Thalamocortical Circuit Disruptions During Contextual Fear Renewal in PTSD.

OBJECTIVE: Post-traumatic stress disorder (PTSD) is marked by impaired contextual modulation of fear, leaving patients vulnerable to symptom return after extinction-based therapy. The thalamus is theorized to coordinate hippocampal-prefrontal circuits during contextual updating. Yet, its role in PTSD remains uncharacterized. We examined the contribution of the medial mediodorsal thalamus (MDm) to extinction-recall and fear renewal and its association with symptom severity. METHODS: 425 participants completed threat renewal and 524 extinction-recall paradigms during fMRI (threat renewal: 189 h

Magnetic resonance imaging fingerprints of status epilepticus: A case-control study.

OBJECTIVE: Status epilepticus (SE) is frequently associated with peri-ictal magnetic resonance imaging (MRI) abnormalities (PMA). However, the anatomical distribution of these alterations has not been systematically studied. The aim of this study was to assess the localization patterns of PMA in patients with SE. METHODS: In this prospective case-control study, we compared the distribution and combinations of diffusion-restricted PMA to diffusion-restricted lesions caused by other neurological conditions. All patients of the SE group and the control group underwent MRI including a diffusion-w

Post-mortem magnetic resonance imaging in patients with suspected prion disease: Pathological confirmation, sensitivity, specificity and observer reliability. A national registry.

The relationship between magnetic resonance imaging (MRI) and clinical variables in patients suspected to have Creutzfeldt-Jakob Disease (CJD) is uncertain. We aimed to determine which MRI features of CJD (positive or negative), previously described in vivo, accurately identify CJD, are most reliably detected, vary with disease duration, and whether combined clinical and imaging features increase diagnostic accuracy for CJD. Prospective patients suspected of having CJD were referred to the National CJD Research and Surveillance Unit between 1994-2004; post-mortem, brains were sent for MRI and

Inter- and intrahemispheric sources of vestibular signals to V1.

Head movements are sensed by the vestibular organs. Unlike classical senses, signals from vestibular organs are not selectively conveyed to a dedicated cortical area but are broadcast throughout the cortex. This distributed processing pattern reflects the fundamental role of vestibular information in contextual modulation across diverse cortical computations. Surprisingly, the routes taken by vestibular signals to reach the cortex are still largely uncharted. Here, we show that the primary visual cortex (V1) receives real-time head movement signals-direction, velocity, and acceleration-from th

Weakening of Subcortical and Strengthening of Cortical Visual Pathways Across Early Adolescence.

Mounting evidence suggests that amygdalar nuclei receive visual information via both a well-characterized cortical pathway through the inferior temporal cortex and a subcortical route through the superior colliculus and pulvinar. This subcortical pathway may facilitate rapid responses to salient visual stimuli and could explain phenomena such as blindsight. However, controversies remain about the organization of the subcortical pathway, its role in visual processing, and how the cortical and subcortical pathways mature across development. To address these questions we used longitudinal, minima

Thalamus involvement in genetic frontotemporal dementia assessed using structural and diffusion MRI: a GENFI study.

Thalamic subregions are commonly, but variably, affected by different forms of frontotemporal dementia. We aimed to better characterize thalamic subregional involvement in genetic frontotemporal dementia with a recently published thalamus segmentation tool that utilizes structural and diffusion MRI, offering additional assessment of mean diffusivity and a more fine-grained analysis of the pulvinar specifically compared to previous studies. Using this tool, we performed thalamus segmentations in MRI scans from C9orf72, GRN and MAPT mutation carriers and mutation non-carriers with suitable 3-Tes

Arousal dependent modulation of thalamo-cortical functional interaction.

Ongoing changes in arousal influence sensory processing and behavioral performance. Yet the circuit-level correlates for this influence remain poorly understood. Here, we investigate how functional interaction between posterior parietal cortex (PPC) and lateral posterior (LP)/Pulvinar is influenced by ongoing fluctuations in pupil-linked arousal, which is a non-invasive measure of neuromodulatory tone in the brain. We find that fluctuations in pupil-linked arousal correlate with changes to PPC to LP/Pulvinar oscillatory interaction, with cortical alpha oscillations driving activity during low
