CKLF induces microglial activation via triggering defective mitophagy and mitochondrial dysfunction.

Although microglial activation is induced by an increase in chemokines, the role of mitophagy in this process remains unclear. This study aimed to elucidate the role of microglial mitophagy in CKLF/CKLF1 (chemokine-like factor 1)-induced microglial activation and neuroinflammation, as well as the underlying molecular mechanisms following CKLF treatment. This study determined that CKLF, an inducible chemokine in the brain, leads to an increase in mitophagy markers, such as DNM1L, PINK1 (PTEN induced putative kinase 1), PRKN, and OPTN, along with a simultaneous increase in autophagosome formatio

Aggregation-induced dual-emission carbon dots for visual discrimination of methanol from ethanol.

BACKGROUND: Discriminating methanol from ethanol is critically important for preventing fatal poisoning incidents and ensuring industrial quality control, yet it remains a significant challenge due to their nearly identical physicochemical properties. Conventional analytical techniques are time-consuming, expensive, and require sophisticated instrumentation. Fluorescent probes offer a promising alternative, but most reported sensors rely on single-emission quenching mechanisms that are susceptible to interference. Therefore, developing a simple, low-cost, yet highly selective strategy for visu

Relationship between cytosolic Ca2+ level and contractile tension in canine basilar artery of chronic vasospasm.

In order to study the role of the Ca2+/calmodulin/myosin light chain kinase system in the development of chronic vasospasm caused by subarachnoid hemorrhage, the cytosolic Ca2+ level ([Ca2+]i), measured with fura-2, a fluorescent Ca2+ indicator, and contractile tension were measured simultaneously, and their quantitative correlation was examined in basilar arterial tissue obtained from the canine "two-hemorrhage" model. Sixteen adult mongrel dogs were divided into two groups, control (n = 8) and vasospasm (n = 8), and were killed 7 days after the first experimental subarachnoid hemorrhage. In

Black Cumin Seed Oil Disrupts Structural Biomass and Attenuates Porphyrin-Associated Maturation in Mature Oral Microcosm Biofilms: An In Vitro Study.

Background/Objective: Mature oral biofilms resist conventional antiseptics via dense extracellular polymeric substance (EPS) matrices. Consequently, the prolonged use of broad-spectrum agents like chlorhexidine (CHX) raises profound ecological concerns. Using a mature oral microcosm biofilm model, we evaluated the multifaceted efficacy of 0.5% black cumin seed oil (BCSO) in disrupting structural biomass, suppressing aciduric bacterial viability, and attenuating porphyrin-associated anaerobic biofilm maturation. Methods: Saliva-derived microcosm biofilms were cultivated on hydroxyapatite discs

Quantifying Sample Collection and Processing Impacts on Fiber-Based Tear Fluid Chemical Analysis.

PURPOSE: Noninvasive analyses of tear fluid from humans and animal models in clinical and research settings most commonly use absorbent material for collection and processing. Still, the impact of these analytical techniques on tear chemical analyses remains largely unknown. The purpose of this study was to quantify the impacts of phenol red thread fiber-based tear sample collection and processing on the primary amine content. METHODS: Human tears were collected by placing the folded end of phenol red thread on the palpebral conjunctiva of the right eye for 20 seconds. The wetted thread was t

An anatomically informed computational fluid dynamics modeling approach for quantifying hemodynamics in the developing heart.

Congenital heart defects occur in approximately 1% of newborns in the US annually. Currently, less than a third of congenital heart defects can be traced to a known genetic or environmental cause, suggesting that a large proportion of disease-causing mechanisms have yet to be fully characterized. Hemodynamic forces such as wall shear stress are critical for heart development and are known to induce changes in embryonic cardiac patterning leading to malformations. However, measuring these hemodynamic factors in vivo is infeasible due to physical limitations, such as the small size and constant

Single-Molecule Visualization of Nanoscale Spatiotemporal Dynamics of Charge-Carrier-Driven Photocatalysis on 2D InSe.

The photocatalytic performance of two-dimensional (2D) semiconductors is governed by the intricate interplay between nanoscale surface structures and charge-carrier dynamics. However, ensemble-averaged and ex situ measurements obscure the spatial heterogeneity and temporal fluctuations, limiting mechanistic understanding. Here, we employed single-molecule fluorescence imaging to quantitatively visualize the photocatalytic activity of 2D layered InSe at the nanoscale, differentiating the carrier extraction positions and efficiencies of photogenerated holes and electrons for light-driven semicon

Multifunctional SnO2/SnS2/CdSe heterojunction coupled with CuO nanoenzyme-regulating synergistic strategy enable multimodal biosensing for breast cancer genes.

Multimodal detection significantly enhanced detection accuracy in complex biological matrices. In this work, a high-efficiency multimodal biosensor is proposed based on multifunctional SnO2/SnS2/CdSe QDs heterojunction and CuO nanoenzyme-regulating synergistic strategy, which achieves ultra-sensitive and specific detection of breast cancer-related BRCA1 gene. A high-performance photoelectrochemical (PEC) SnO2/SnS2/CdSe QDs heterojunction was constructed for the first time, which effectively enhances the light absorption capability and photogenerated charge separation efficiency. The heterojunc

Detection and imaging of gadolinium accumulation in human bone tissue by micro- and submicro-XRF.

Gadolinium-based contrast agents (GBCAs) are frequently used in patients undergoing magnetic resonance imaging. In GBCAs gadolinium (Gd) is present in a bound chelated form. Gadolinium is a rare-earth element, which is normally not present in human body. Though the blood elimination half-life of contrast agents is about 90 minutes, recent studies demonstrated that some tissues retain gadolinium, which might further pose a health threat due to toxic effects of free gadolinium. It is known that the bone tissue can serve as a gadolinium depot, but so far only bulk measurements were performed. Her

[Pit and fissure morphology and its implication for caries prevention and treatment].

Dental caries is a chronic and progressive destruction of dental hard tissue under the combined action of multiple factors, with the pits and fissures on the occlusal surfaces of posterior teeth being the most susceptible sites. During tooth development, fissure morphology is regulated by the enamel knot. The depth, width, and shape of fissures directly influence plaque retention and cleaning difficulty, thereby determining caries susceptibility. Deep and narrow fissures, which are difficult to clean, pose a higher risk of caries. Pit and fissure caries begin with enamel demineralization cause

The Application of Methylprednisolone Nanoscale Zirconium-Porphyrin Metal-Organic Framework (MPS-NPMOF) in the Treatment of Photoreceptor Degeneration.

BACKGROUND: Photoreceptor degeneration is one of the most refractory oculopathy in the world, leading to vision loss in severe cases. Methyprednisolone is one of the most commonly prescribed medications for the treatment of retinal degenerative diseases, either by oral administration or repeated intraocular injections. However, the efficacy was unsatisfactory due to its systemic or local side effects and short retention time within the retina. METHODS: Nanoscale zirconium-porphyrin metal-organic framework (NPMOF) was synthesized and characterized. The biotoxicity and imaging capability of NPM

EGFR targeted nanobody-photosensitizer conjugates for photodynamic therapy in a pre-clinical model of head and neck cancer.

Photodynamic therapy (PDT) induces cell death through local light activation of a photosensitizer (PS) and has been used to treat head and neck cancers. Yet, common PS lack tumor specificity, which leads to collateral damage to normal tissues. Targeted delivery of PS via antibodies has pre-clinically improved tumor selectivity. However, antibodies have long half-lives and relatively poor tissue penetration, which could limit therapeutic efficacy and lead to long photosensitivity. Here, in this feasibility study, we evaluate at the pre-clinical level a recently introduced format of targeted PDT

Development and repair of cataract induced by ultraviolet radiation.

BACKGROUND: Several epidemiological investigations show a correlation between cataract development and the dose of ultraviolet radiation (UVR) received. It is experimentally well established that exposure of animal eyes to UVR induces cataract. Most cataracts develop as a gradual increase in lens opacity. Despite this, current estimations of toxicity for cataract are based on the concept that cataract is a binary event. Moreover, current exposure limits for UVR are based on subjective inspections with slit lamp microscopy. PURPOSE: The first purpose of the present study was to determine a sta

Restoration of lysosomal function attenuates autophagic flux impairment in nucleus pulposus cells and protects against mechanical overloading-induced intervertebral disc degeneration.

Intervertebral disc degeneration (IVDD) is a leading cause of low back pain that incurs large socioeconomic burdens. Growing evidence reveals that macroautophagy/autophagy dysregulation contributes to IVDD, but the exact role of autophagy and its regulatory mechanisms remain largely unknown. Here, we found that mechanical overloading impaired the autophagic flux of nucleus pulposus (NP) cells in vivo and in vitro. Mechanistically, the impairment of autophagic flux was attributed to lysosomal dysfunction induced by overloading. Overloading could also lead to lysosomal membrane permeabilization

Three-Dimensional Light Sheet Fluorescence Microscopy of Lungs To Dissect Local Host Immune-Aspergillus fumigatus Interactions.

Aspergillus fumigatus is an opportunistic fungal pathogen that can cause life-threatening invasive lung infections in immunodeficient patients. The cellular and molecular processes of infection during onset, establishment, and progression of A. fumigatus infections are highly complex and depend on both fungal attributes and the immune status of the host. Therefore, preclinical animal models are of paramount importance to investigate and gain better insight into the infection process. Yet, despite their extensive use, commonly employed murine models of invasive pulmonary aspergillosis are not w

Repeated moderate hypothermia leads to sustained glymphatic dysfunction and loss of vascular AQP4 polarization.

Cerebrospinal fluid (CSF) enters the brain along perivascular spaces in a manner that depends on aquaporin-4 (AQP4) polarization, facilitating brain clearance and is called the glymphatic system. While physiological factors such as cardiovascular activity and respiration are known to influence glymphatic function, the effects of hypothermia remain poorly understood. In this study, we used quantitative optical and light-sheet microscopy to track fluorescent CSF tracers in mice under ketamine–xylazine (KX) anesthesia at normothermia (37 °C), mild hypothermia (33 °C), and moderate hypothermia (30

Presenilin overexpression arrests cells in the G1 phase of the cell cycle. Arrest potentiated by the Alzheimer's disease PS2(N141I)mutant.

To investigate the mechanism by which presenilin (PS) overexpression induces apoptosis, we studied the effects of these proteins on cell cycle progression. Transiently transfected HeLa cells were bromodeoxyuridine (BrdU) labeled to visualize DNA synthesis by immunofluorescence and stained with propidium iodide to measure DNA content by fluorescence-activated cell sorting (FACS). BrdU labeling was decreased in cells expressing presenilin-1 (PS1), presenilin-2 (PS2), an Alzheimer's disease-associated missense mutation PS2(N141I), and the carboxyl-terminally deleted PS2 construct PS2(166aa), comp
