Quantitative autoradiographic analysis of muscarinic cholinergic and GABAA (benzodiazepine) receptors in the forebrain of rats flown on the Soviet Biosatellite COSMOS 2044.

The quantitative autoradiographic analysis of muscarinic cholinergic and GABAA (benzodiazepine) receptors was performed on selected regions of the cerebral cortex and striatum of rats flown in the Soviet Biosatellite COSMOS 2044. An age- and strain-matched synchronous ground-based control group was employed for comparison. Muscarinic cholinergic receptor density was found to be significantly lower in the striatum of the flight animals as compared with that in the synchronous control group. No significant differences between flight and synchronous control groups were found in the other regions

Loss of muscarinic cholinergic receptors from the temporal cortex of alcohol abusers.

It is currently controversial whether all the brain damage in alcohol abusers in the result of thiamine deficiency (Wernicke-Korsakoff's disease) or whether, in addition, alcohol abuse may affect the brain by other mechanisms as well. The purpose of this study was to determine if alcohol abuse affects muscarinic cholinergic and benzodiazepine receptors in histologically normal brains obtained at autopsy in a general hospital population. Patients were excluded from this study if they had clinical brain diseases (including Wernicke's disease), died in coma, or had liver disease, significant brai

Piracetam elevates muscarinic cholinergic receptor density in the frontal cortex of aged but not of young mice.

Chronic treatment (2 weeks) with piracetam (500 mg/kg, once daily PO) elevated m-cholinoceptor density in the frontal cortex of aged (18 months) female mice by about 30-40%, but had no effect on m-cholinoceptor density in the frontal cortex of young (4 weeks) mice. The effect of piracetam on m-cholinoceptor density as determined by the specific binding of tritiated QNB was not affected by concomitant daily treatment with either choline (200 mg/kg) or scopolamine (4 mg/kg). It is concluded that the effect of piracetam on m-cholinoceptor density could explain the positive effects which have been

Comparison of muscarinic acetylcholine binding in the urinary bladder and submandibular gland of the rabbit.

In order to explore the possibility of heterogeneity in peripheral muscarine receptors, receptors were characterized in membrane fractions isolated from rabbit urinary bladder and submandibular gland. With [3H]QNB as radioligand, specific binding with very high affinity was found in both preparations. Although the BmaxS for binding in the two preparations were very similar, the mean KD value in the submandibular gland was significantly higher (P less than 0.005) than that in the bladder. Among the anticholinergic drugs, oxybutynin had a significantly lower value for the inhibition constant (Ki

Muscarinic receptors in diabetic rat prostate.

To investigate the effects of experimentally-induced diabetes on prostatic muscarinic cholinergic receptors, the binding characteristics of [3H]quinuclidinyl benzilate ([3H]QNB) to prostatic membrane particulates were examined in four groups of rats: control, diabetic, diabetic insulin treated, and diabetic myo-inositol treated. Diabetes was induced by i.v. injection of streptozotocin (STZ), 65 mg/kg. Diabetic and diabetic myo-inositol-treated rats had hyperglycemia, hypoinsulinemia, glucosuria, polydipsia, and polyuria as well as significantly smaller prostates and lower body weights compared

Release of digestive enzymes from isolated rat pancreatic acini following muscarinic stimulation: a comparative study with enzyme release and receptor binding.

Effect of cholinergic stimulation on the release of digestive enzymes from isolated rat pancreatic acini prepared by collagenase digestion was investigated. The release of enzymes (amylase, chymotrypsinogen, lipase) increased linearly with the time of incubation in the absence of secretagogues. Carbachol, a muscarinic agonist, induced a remarkable increase in the release of these enzymes. This carbachol-stimulated amylase release showed a biphasic curve, and its maximal response was observed at 10(-5) M. The release patterns of chymotrypsinogen and lipase were similar to that of amylase. This

Effects of acute and chronic administration of antidepressant drugs on the central cholinergic nervous system. Comparison with anticholinergic drugs.

The purpose of this investigation was to study the effects of antidepressant drugs on the central cholinergic system of the rat after acute and chronic administration. Drugs (antidepressants and non-antidepressants) were first divided into highly potent, moderately potent or weak anticholinergic categories based upon the ability of each compound to displace [3H]-QNB [( 3H]quinuclidinyl benzilate from synaptosomal membranes. One antidepressant drug and one non-antidepressant drug, with similar anticholinergic properties, were chosen as representative agents of each category of anticholinergic p

Structure of the human M2 muscarinic acetylcholine receptor bound to an antagonist.

The parasympathetic branch of the autonomic nervous system regulates the activity of multiple organ systems. Muscarinic receptors are G-protein-coupled receptors that mediate the response to acetylcholine released from parasympathetic nerves. Their role in the unconscious regulation of organ and central nervous system function makes them potential therapeutic targets for a broad spectrum of diseases. The M2 muscarinic acetylcholine receptor (M2 receptor) is essential for the physiological control of cardiovascular function through activation of G-protein-coupled inwardly rectifying potassium c

Muscarinic acetylcholine receptor binding in the guinea pig cochlea.

Because of the possibility that olivocochlear bundles may use acetylcholine as an inhibitory transmitter, we examined the cochlea of the guinea pig for cholinergic receptor binding sites. Binding of the potent muscarinic antagonist [3H]quinuclidinyl benzilate (3H-QNB) showed that the cochlea has sites with the pharmacological specificity of muscarinic cholinergic receptors. Specific 3H-QNB binding was saturated at 0.8 nM and was half-saturated at 0.03 nM concentrations. Total 3H-QNB binding was reduced 70% in the presence of saturating doses of acetylcholine, oxotremorine and atropine, and hal

Acute and chronic effects of barbiturates on regional turnover of acetylcholine in brain.

Data concerning both the acute and chronic effects of barbiturates on acetylcholine [ACh] turnover in mouse and rat brain are presented. A single anaesthetic dose of pentobarbital [60 mg/kg, i.p.] to mice increased the endogenous content of ACh and decreased the turnover of ACh in the hippo-campus and cortex. No effect was found in the striatum. At sacrifice after chronic treatment with barbital to rats for about 30 weeks no effect was found on the endogenous ACh content. 3 days after the barbital solution was replaced by water [at the day with maximal number of abstinence convulsions] the amo

The rabbit cornea lacks cholinergic receptors.

Cholinergic receptors were studied in membranes prepared from rabbit cornea, iris-ciliary body, and retina, using 3H-quinuclidinyl benzilate (3H-QNB) to identify muscarinic receptors and 125I-alpha-bungarotoxin (125I-BGT) to identify nicotinic receptors. Muscarinic cholinergic receptors were not found in the cornea. As a positive control, muscarinic cholinergic receptors were characterized in preparations of the iris-ciliary body. Specific binding of 3H-QNB to iris-ciliary body membrane preparations was saturable, with a Kd of 1.3 nM QNB. Specificity of the assay for muscarinic receptors was c

Characterization of muscarinic receptors in cultured human iris sphincter and ciliary smooth muscle cells.

Muscarinic receptors present in cultured human iris sphincter and ciliary smooth muscle cells were characterized by both ligand ([3H]QNB binding) and functional (phosphoinositide hydrolysis) studies. Ligand binding studies showed that [3H]QNB represented a single population of binding sites with KD values of 4.02 x 10(-11) M in the ciliary and 5.6 x 10(-11) M in the iris sphincter cells. In competition studies, the selective antagonist, 4-diphenylacetoxy-N-methylpiperidine-methobromide (4-DAMP) was the most potent in displacing [3H]QNB with selectivity of 150-350-fold over pirenzepine (M1) and

Development of simultaneous determination of dopamine 2, histamine 1, and muscarinic acetylcholine receptor occupancies by antipsychotics using liquid chromatography with tandem mass spectrometry.

Receptor occupancy is an indicator of antipsychotic efficacy and safety. It is desirable to simultaneously determine the occupancy of multiple brain receptors as an indicator of the efficacy and central side effects of antipsychotics because many of these drugs have binding affinities for various receptors, such as dopamine 2 (D2), histamine 1 (H1), and muscarinic acetylcholine (mACh) receptors. The purpose of this study was to develop a method for the simultaneous measurement of multiple receptor occupancies in the brain by the simultaneous quantification of unlabeled tracer levels using liqu

Muscarinic binding sites on bovine pulmonary arterial endothelial cells in culture.

We have investigated the presence and nature of muscarinic binding sites on membranes from cultured bovine pulmonary arterial endothelial cells (BPAE). BPAE were harvested and subcultured nonenzymatically; experiments were performed 3-5 days postconfluence and between 10 and 25 passage numbers. Utilizing radioligand binding techniques with the muscarinic receptor antagonists [3H]3-quinuclidinyl benzilate ([3H]QNB) and [3H]N-methylscopolamine ([3H]MS) as probes, we identified a small population of atropine-sensitive muscarinic sites (1,800-2,000 sites/cell or 7-8 fmol/mg protein). Regulation of atrial autonomic receptors in experimental cyanotic heart disease.

During chronic hypoxemia, left ventricular beta-adrenergic receptor density is decreased and a dissociation occurs between increased chronotropic and decreased inotropic responses to chronically elevated sympathetic tone. To determine whether this dissociation was related to alterations in autonomic receptor populations in the right atrium, we studied right atrial cholinergic and beta-adrenergic receptors in chronically hypoxemic newborn lambs and in normoxemic controls. Heart rate response was determined by infusing isoproterenol at 0.1 or 0.5 microgram.kg-1.min-1. Muscarinic receptors were q

Muscarinic receptor binding in central airway musculature in chronic airflow obstruction.

The hypothesis of an increased muscarinic receptor sensitivity in airway musculature of patients with chronic airflow obstruction (CAO) has been investigated by in vitro radioligand binding studies. The receptor binding profiles were determined in membrane homogenates of tracheal and main bronchial smooth muscle, as well as in segmental bronchial tissue preparations. The number of receptors was measured by binding of the radioactive muscarinic antagonist [3H]-(-)-quinuclidinyl benzilate ([3H]-(-)-QNB). The affinity for agonists was investigated by studying the inhibition of [3H]-(-)-QNB bindin
