Local and systemic consequences of acute, low-dose ultraviolet B radiation are mediated by different immune regulatory mechanisms.

Acute, low-dose ultraviolet B (UVB) radiation alters the local skin site such that epicutaneous application of hapten fails to induce contact hypersensitivity (CH), but induces tolerance in UVB-susceptible mice. Although the inability of irradiated skin to support CH induction may be a strictly local effect, there may also be systemic immune consequences of UVB radiation delivered in this manner. To examine this matter, abdominal skin of C57BL/6 mice was exposed to acute, low-dose UVB radiation. Dinitrofluorobenzene was immediately painted directly on the irradiated site, or at a distant (unir

Role of autophagy in regulating the radiosensitivity of tumor cells.

BACKGROUND: Autophagy is a metabolic response of cells to chemical and physical factors, such as nutrition or growth factor deprivation, proinflammatory state, hypoxia, accumulation of reactive oxygen species, presence of infectious agents, and DNA damage. Autophagy maintains the homeostasis of intracellular metabolism mainly by degrading cellular organelles and critical proteins. In a sense, autophagy protects cells from death. Radiotherapy is a powerful tool used to control tumor growth, and it can induce autophagy. The relationship between radiotherapy and autophagy is worthy of further inv

Medical consequences of radiation exposure of the bronchi-what can we learn from high-dose precision radiation therapy?

The bronchial tolerance to high doses of radiation is not fully understood. However, in the event of a radiological accident with unintended exposure of the central airways to high doses of radiation it would be important to be able to anticipate the clinical consequences given the magnitude of the absorbed dose to different parts of the bronchial tree. Stereotactic body radiation therapy (SBRT) is a radiation treatment technique involving a few large fractions of photon external-beam radiation delivered to a well-defined target in the body. Despite generally favourable results, with high loca

Radiation dose-volume effects of optic nerves and chiasm.

Publications relating radiation toxicity of the optic nerves and chiasm to quantitative dose and dose-volume measures were reviewed. Few studies have adequate data for dose-volume outcome modeling. The risk of toxicity increased markedly at doses >60 Gy at approximately 1.8 Gy/fraction and at >12 Gy for single-fraction radiosurgery. The evidence is strong that radiation tolerance is increased with a reduction in the dose per fraction. Models of threshold tolerance were examined.

Solar urticaria: studies on mechanisms of tolerance.

The mechanisms by which tolerance is induced in solar urticaria were investigated in two patients whose eruptions were induced by wavelengths in the range 320-455 nm in one patient and 400-495 nm in another. Tolerance to radiation was induced by repeated exposures of the skin to the eliciting wavelengths of light. Weal and flare responses to intradermal injections of histamine and the histamine-releasing agent (codeine) were unaltered in the tolerant skin when compared with adjacent normal skin. Intradermal injection of in vitro irradiated serum or plasma from the patients induced an urticaria

[The classification of radiation protective agents as the reflection of the present state and development perspective, of current radiation pharmacology].

Pharmacological drugs which can reduce the radiation damage to the organism when applied in the nearest time before or after exposure to radiation can be referred to as radiation protective agents. The classification of radiation protective agents, which has been well established to date, is a reflection of the history of the formation and current state of radiation pharmacology: the most significant historical landmarks in the screening of radiation protective agents among different groups of pharmaceutical preparations and the formation of the theoretical knowledge about the mechanism for th

Persons chronically exposed to low doses of ionizing radiation: A cytogenetic dosimetry study.

The dosimetry and control of exposure for individuals chronically exposed to ionizing radiation are important and complex issues. Assessment may be optimized by evaluating individual adaptation and radiosensitivity, but it is not possible for a single model to account for all relevant parameters. Our goal was to develop approaches for the calculation of doses for persons chronically exposed to ionizing radiation, taking their radiosensitivities into consideration. On the basis of ex vivo radiation of blood samples, dose-effect models were constructed for dose ranges 0.01-2.0 and 0.01-0.4 Gy, u

The Significance of the Response: Beyond the Mechanics of DNA Damage and Repair-Physiological, Genetic, and Systemic Aspects of Radiosensitivity in Higher Organisms.

Classical radiation biology as we understand it clearly identifies genomic DNA as the primary target of ionizing radiation. The evidence appears rock-solid: ionizing radiation typically induces DSBs with a yield of ~30 per cell per Gy, and unrepaired DSBs are a very cytotoxic lesion. We know very well the kinetics of induction and repair of different types of DNA damage in different organisms and cell lines. And yet, higher organisms differ in their radiation sensitivity-humans can be unpredictably radiosensitive during radiotherapy; this can be due to genetic defects (e.g., ataxia telangiecta

The Impact of Clinical Stage on Radiation Doses to Organs at Risk Following Intensity-modulated Radiotherapy in Nasopharyngeal Carcinoma: A Prospective Analysis.

Background: The development of intensity-modulated radiotherapy (IMRT) has revolutionized the management of nasopharyngeal carcinoma (NPC). The purpose of this study was to investigate the impact of clinical stage on radiation doses to organs at risk (OARs) in NPC. Material and Methods: One hundred and forty-eight patients with newly diagnosed and untreated NPC were prospectively enrolled. Based on the anatomic definition and pathogenesis of radiation induced injury, a total of 28 OARs surrounding the nasopharynx were contoured on axial computed tomography (CT) planning images in each patient.

Enhancing radiation sensitivity in malignant brain tumors with chloroquine: a systematic review.

PURPOSE: Chloroquine (CQ) has been studied for over 50 years as a potential adjuvant to cancer therapy. However, clinical studies investigating CQ combined with radiotherapy for brain tumors have yielded conflicting results. We aimed to synthesize existing evidence on the safety and efficacy of adjuvant CQ with radiotherapy for treatment of high-grade gliomas (HGG) and brain metastases. METHODS: We conducted a systematic review using the PubMed, Embase, Scopus, and Web of Science databases with no date restrictions. We included English-language clinical studies (n ≥ 5 patients) that evaluated

[Radiogenic effects on liver tissue].

In a review of literature the presented studies on radiogenic liver injuries are summarized. Whereas in the past the liver tissue was supposed to be radioresistant nearly completely, to newer knowledge this organ has to be classed with most radiosensitive tissues at all. The early picture of a liver injury in man is characterized by the veno-occlusive syndrome. Like in Budd-Chiari-Syndrome a congestion begins in the central veins and the centrolobular sinusoids, that has severest threatening consequences for the patients with extensive liver irradiation. The clinic picture is demonstrated. At

Questioning the radiation limits of life: Ignicoccus hospitalis between replication and VBNC.

Radiation of ionizing or non-ionizing nature has harmful effects on cellular components like DNA as radiation can compromise its proper integrity. To cope with damages caused by external stimuli including radiation, within living cells, several fast and efficient repair mechanisms have evolved. Previous studies addressing organismic radiation tolerance have shown that radiotolerance is a predominant property among extremophilic microorganisms including (hyper-) thermophilic archaea. The analysis of the ionizing radiation tolerance of the chemolithoautotrophic, obligate anaerobic, hyperthermoph

A sense of danger from radiation.

Tissue damage caused by exposure to pathogens, chemicals and physical agents such as ionizing radiation triggers production of generic "danger" signals that mobilize the innate and acquired immune system to deal with the intrusion and effect tissue repair with the goal of maintaining the integrity of the tissue and the body. Ionizing radiation appears to do the same, but less is known about the role of "danger" signals in tissue responses to this agent. This review deals with the nature of putative "danger" signals that may be generated by exposure to ionizing radiation and their significance.

[Dosage values for characterization of patient exposure in computerized tomography and their significance for risk assessment].

In order to evaluate patients' exposure to radiation in computed tomography, dose quantities such as the computed tomography dose index (CTDI), multiple scan average dose (MSAD) and dose free in air on the axis of rotation are used. The CTDI and the MSAD, derivable from the CTDI, are a good measure for the absorbed doses in the examined volume, but they do not take the biological sensitivity of the organs into account and do not describe the radiogenic risk that is actually relevant for patients and doctors. The dose on the axis of rotation is not an accurate measure of the effective dose and

Induced radioresistance: an overview and historical perspective.

There is now considerable evidence that cellular radioprotective mechanisms exist that are upregulated in response to exposure to small doses of ionizing radiation and other DNA-damaging agents. There appear to be two ways in which these 'induced' mechanisms operate: either protecting against a subsequent exposure to radiation that may be substantially larger than the initial 'conditioning' dose, or by influencing the response to single doses so that small acute radiation exposures, or exposures at very low dose-rates, are more effective per unit dose than larger exposures above a threshold wh

Tardigrades as a potential model organism in space research.

Exposure of living organisms to open space requires a high level of tolerance to desiccation, cold, and radiation. Among animals, only anhydrobiotic species can fulfill these requirements. The invertebrate phylum Tardigrada includes many anhydrobiotic species, which are adapted to survive in very dry or cold environmental conditions. As a likely by-product of the adaptations for desiccation and freezing, tardigrades also show a very high tolerance to a number of other, unnatural conditions, including exposure to ionizing radiation. This makes tardigrades an interesting candidate for experiment

A role for inflammatory mediators in the induction of immunoregulatory B cells.

UV exposure suppresses the immune response to a variety of microbial, fungal, and viral Ags. In addition, UV radiation is a complete carcinogen and the immune suppression induced by UV radiation is a major risk factor for skin cancer induction. In this study, we examined the mechanisms underlying the induction of immune suppression and tolerance induction by UV radiation. Transferring lymph nodes cells from UV-irradiated, FITC-sensitized mice into normal recipients transferred immune tolerance. Contrary to expectations, the cell responsible was an FITC(+), IL-10-secreting, CD19(+), B220(+) B c
