Diurnal variation and binding characteristics of melatonin in the mouse brain and gastrointestinal tissues.

1. Compared to night time values, levels of melatonin (M), determined by specific RIA, were lower in the brain and the duodenum-jejunum segment of mice sacrificed in mid-photophase. In other parts of the gut no significant diurnal difference was observed. 2. Highest daytime levels were detected in the stomach (569 pg/ml), lowest in the brain (62 pg/ml). 3. Preliminary studies indicate the presence of specific binding sites for [125I]iodo-melatonin in the colon, ileum, jejunum, stomach and brain. These sites, which exhibit an affinity of about 1 nM, may be involved in mediating the gastrointest

A computer controlled device to facilitate studies of the kinetics of ligand-binding: binding of diazepam to bovine brain membranes.

A device to facilitate kinetic receptor filtration assays is described. The receptor containing membranes and the labeled ligand are placed in two separate syringes and are rapidly mixed into a collecting syringe using a pneumatic ram. Shortly after the start of mixing, a pneumatically controlled valve switches the collecting syringe containing the receptor-ligand mixture to the filtration unit. Filtration is performed on glass/microfiber filters or equivalent by pushing the plunger of the collecting syringe by a stepper motor. A valve positioner controlling several valves allows the filtered

Neuroleptic blood levels and clinical response.

Gas chromatography and radioreceptor assay technologies made possible the measurement of nanogram quantities of neuroleptic in the blood. They revealed large differences in blood concentrations among patients receiving the same oral dose. This led to the hope that measuring neuroleptic concentrations would allow more effective oral dosing for individual psychotic patients. Early attempts to study neuroleptic concentration/clinical response patterns, using chlorpromazine, produced confusing results. Many of the studies had serious design flaws, and were complicated by the many active metabolite

Somatomedins in aging and dementia disorders of the Alzheimer type.

Serum levels of somatomedins were determined in apparently healthy aged individuals and dementia patients primarily with clinically suspected Alzheimer type disorder. Serum somatomedin values, determined by radioreceptorassay and radioimmunoassay, fell with advancing age in normal subjects. A significant elevation in serum somatomedins was observed in dementia patients. CSF somatomedin levels were also increased in the only two patients with suspected Alzheimer type disease examined. Since somatomedins are believed to act as anabolic hormones, it was suggested that the elevated levels represen

Free thyroxine and free thyroxine index in women taking oral contraceptives.

This study compares the accuracy of the free thyroxine index and free serum thyroxine radioassay in evaluating the metabolic status of 36 euthyroid women, 17 of whom were taking oral contraceptive drugs. Results indicate that the free thyroxine index does not uniformly compensate for estrogen-induced elevations in total serum thyroxine concentrations and that the free serum thyroxine is the test of choice in assessing the functional thyroid status of women taking oral contraceptive drugs.

An improved method for the measurement of 1,25-(OH)2D3 in human plasma.

Here we report a highly sensitive and convenient ligand binding assay for the determination of 1,25(OH)2D3 in small volumes of human plasma. This method involves: (1) extraction of vitamin D3 and its metabolites using methanol-methylene chloride with separation of phases by centrifugation; (2) gel chromatography and high pressure liquid chromatography for the quantitative isolation of 1,25-(OH)2D3; and (3) a sensitive ligand binding assay for 1,25-(OH)2D3 employing cytosol receptor from the intestinal mucosa of rachitic chicks. Using modified rachitogenic chick diets allows early (less than 4

Use of protein containing magnetic microparticles in radioassays.

We describe a radioassay method that involves the use of magnetic protein microparticles composed of a water-insoluble protein matrix containing magnetically responsive material. We define two different types of particles according to the mechanism of action: (a) The substrate is sorbed nonspecifically by the protein matrix of the particle or by a second substance such as charcoal or ion-exchange resin incorporated within the protein matrix of the particle. These particles are useful for separating free from bound substrate. Examples of these are albumin magnetic microparticles for use in a to

Machine Learning for Discovery of New ADORA Modulators.

Adenosine (ADO) is an extracellular signaling molecule generated locally under conditions that produce ischemia, hypoxia, or inflammation. It is involved in modulating a range of physiological functions throughout the brain and periphery through the membrane-bound G protein-coupled receptors, called adenosine receptors (ARs) A1AR, A2AAR, A2BAR, and A3AR. These are therefore important targets for neurological, cardiovascular, inflammatory, and autoimmune diseases and are the subject of drug development directed toward the cyclic adenosine monophosphate and other signaling pathways. Initially us

Dopamine receptor binding of a novel dibenzodioxazocine derivative, EGYT-2509.

The binding of novel dibenzodioxazocine derivatives to rat striatal dopamine receptors was studied in vitro, using 3H-spiperone as radioligand. The biochemical-pharmacological characteristics of the effect of a selected representative, EGYT-2509 are discussed in details. The parameters of specific spiperone binding to rat striatal membrane preparation (KD = 0.550 nM, Bmax = 465 fmole/mg protein) as well as the displacing potencies of known dopamine receptor ligands matched closely the corresponding values in the literature. Using 0.4 nM radioligand, a Ki value of 404 nM was obtained for EGYT-2

Is the Neuropeptide PEN a Ligand of GPR83?

G protein-coupled receptor 83 (GPR83) is a class A G protein-coupled receptor with predominant expression in the cerebellum and proposed function in the regulation of food intake and in anxiety-like behavior. The neuropeptide PEN has been suggested as a specific GPR83 ligand. However, conflicting reports exist about whether PEN is indeed able to bind and activate GPR83. This study was initiated to evaluate PEN as a potential ligand of GPR83. Employing several second messenger and other GPCR activation assays as well as a radioligand binding assay, and using multiple GPR83 plasmids and PEN pept

Radioimmunoassay of tricyclic antidepressant and some phenothiazine drugs in forensic toxicology.

A radioimmunoassay suitable for screening blood and urine samples for tricyclic antidepressant and some phenothiazine drugs in forensic toxicology is described. The assay displays a remarkably broad spectrum of cross-reactivity that enables therapeutic or subtherapeutic levels of most tricyclic antidepressants and a number of phenothiazines in blood to be detected. The antiserum is commercially available and the radioligand, a radioiodinated conjugate of N-acetyl-L-histidine and nortriptyline, is easily prepared. Blood samples require prior extraction to reduce the background but urine may be

Increased insulin-like growth factor-1 receptors in thyroid tissues of Graves' disease.

Insulin-like growth factor-1 (IGF-1) plays an important role in thyroid cell growth and human thyroid diseases. In order to investigate the involvement of IGF-1 in abnormal thyroid growth in Graves' disease, 20 patients with Graves' disease were enrolled in this study. Thirty euthyroid subjects were used as normal controls for the radioimmunoassay study of circulating IGF-1. Seven normal thyroid tissue fragments from 3 patients with papillary carcinoma and 4 patients with benign thyroid nodule were used as controls in the radioligand binding assay. The concentration of serum IGF-1 in the disea

Effect of general anaesthetics and organic solvents on alpha 1-adrenoceptors in the myometrium.

The alpha 1 selective radioligand 3H-prazosin was used to assay alpha 1 receptors in membranes prepared from the rabbit myometrium. 3H-Prazosin was found to bind to a single high affinity site in these membranes which was the presumed alpha 1 receptor. A series of general anaesthetics and organic solvents were tested for their ability to inhibit 3H-prazosin binding. The order of potency of the tested agents to inhibit the binding was: chloroform=halothane=trichloroethylene greater than carbon tetrachloride greater than dichloromethane. The depression of 3H-prazosin binding seemed to be induced

Characterization of [3H]AZ12464237 as a high affinity, non-nucleotide antagonist radioligand for the P2Y12 receptor.

The purinergic receptor P2Y12 (P2Y12R) is a well-recognized target for anti-thrombotic agents. This receptor is also expressed in microglia, the main immune cells of the brain, where it modulates microglial activation states and inflammatory responses. To investigate P2Y12R-mediated actions in the central nervous system (CNS), developing novel brain-penetrant ligands and further in vitro studies on brain tissues are essential. A radiolabeled, easily accessible tool compound would significantly advance such drug discovery efforts. Herein, we describe the 3H-labeling of a non-nucleotide P2Y12R a

The development of a PET radiotracer for imaging alpha synuclein aggregates in Parkinson's disease.

M503-1619 was identified as a promising ligand for positron emission tomography (PET) imaging of α-synuclein (α-Syn) pathology in Parkinson's disease (PD). An exemplar for binding site 9 (residues GLY-86, ILE-88, PHE-94 and LYS-96) of α-Syn fibrils was generated. An in silico ultrahigh throughput screening campaign was conducted using a 42 million compound library. Secondary in silico methods followed by visual inspection were used to select 6 compounds as candidates for in vitro binding studies. M503-1619 was found to have a high binding affinity (K i = 6.5 nM versus the site 9 radioligand [3

Altered β-Adrenergic System, Cardiac Dysfunction, and Lethal Arrhythmia in a Rat Model of Metabolic Syndrome.

Metabolic syndrome (MetS) is a worldwide problem affecting at least one-third of the population. MetS patients have increased cardiovascular risk associated with an abnormal β-adrenergic response; however, it is not clear how MetS affects the cardiac β-adrenergic system. We analyzed cardiac function and the β-adrenergic response in an experimental model of MetS in rats by recording pressure-volume (PV) loops via an open-chest approach and performed a biochemical characterization of the cardiac β-adrenergic system through ELISA, radioligand binding assays, and Western blotting. Microscopy was e

Use of Amplified Lewy Body Dementia Fibrils and Autoradiography to Characterize Binding of Radioligand Tg-1-90B to Alpha-Synuclein Fibrils in Postmortem Brain Tissue.

Parkinson's disease (PD) and Lewy Body Dementia (LBD) are defined by accumulation of alpha-synuclein (Asyn) fibrils within Lewy bodies (LBs) and Lewy neurites (LNs). The development of a Positron Emission Tomography (PET) tracer for quantifying Asyn fibrils would improve diagnostic accuracy and provide a biomarker for disease progression. We previously described radioligand [3H]Tg-1-90B, which binds to in vitro Asyn fibrils (PDB 2N0A) via interactions with residues Y39, S42 and K44. Here, we performed molecular docking studies with Tg1-90B and PD/LBD Asyn fibrils (PDB 8A9L), which predicts int
