Production and evaluation of three kinds of vaccines against largemouth bass virus, and DNA vaccines show great application prospects.

Largemouth bass virus (LMBV) infections has resulted in high mortality and economic losses to the global largemouth bass industry and has seriously restricted the healthy development of the bass aquaculture industry. There are currently no antiviral therapies available for the control of this disease. In this study, we developed three types of vaccine against LMBV; whole virus inactivated vaccine (I), a subunit vaccine composed of the major viral capsid protein MCP (S) as well as an MCP DNA vaccine(D), These were employed using differing immunization and booster strategies spaced 2 weeks apart

Systematic screen of PKR reveals genetic variants that broadly evade divergent viral pseudosubstrate inhibitors.

Evolutionary arms races can arise at the contact surfaces between host and viral proteins, producing dynamic spaces in which genetic variants are continually pursued even while protein function must be maintained. A striking case is protein kinase R (PKR), an innate immune protein that detects viral replication and halts protein synthesis by phosphorylating the initiation factor eIF2α. PKR is targeted by many viral antagonists, including pseudosubstrate inhibitors encoded by poxviruses and ranaviruses that mimic eIF2α. We previously found that the eIF2α-binding surface of human PKR is highly m

Immunoglobulin M response in different intestinal segments of largemouth bass (Micropterus salmoides) following largemouth bass virus (LMBV) infection.

In teleost fish, the gut functions not only as a primary site for nutrient digestion and absorption but also as a critical mucosal immune organ. Although the functional heterogeneity of nutrient absorption among different intestinal segments has been well characterized, the segment-specific features of intestinal immune responses remain poorly understood, particularly with regard to mucosal IgM responses. In this study, we established a virus infection model via intraperitoneal injection and demonstrated that the virus triggers robust innate and adaptive immune responses in the gut of largemou

Virus-Targeted Transcriptomic Analyses Implicate Ranaviral Interaction with Host Interferon Response in Frog Virus 3-Infected Frog Tissues.

Ranaviruses (Iridoviridae), including Frog Virus 3 (FV3), are large dsDNA viruses that cause devastating infections globally in amphibians, fish, and reptiles, and contribute to catastrophic amphibian declines. FV3's large genome (~105 kb) contains at least 98 putative open reading frames (ORFs) as annotated in its reference genome. Previous studies have classified these coding genes into temporal classes as immediate early, delayed early, and late viral transcripts based on their sequential expression during FV3 infection. To establish a high-throughput characterization of ranaviral gene expr

BURMESE ROOFED TURTLE (BATAGUR TRIVITTATA) DISEASE SCREENING IN MYANMAR.

The Burmese roofed turtle (Batagur trivittata), a critically endangered freshwater turtle, is endemic to Myanmar. Once thought to be extinct, remnant wild populations were discovered in 2001 and limited captive individuals identified in pagoda ponds or confiscated from fishers in Myanmar. These and their offspring are maintained in five facilities in Myanmar and form the basis of a conservation program (habitat protection, captive breeding, nest protection, egg collection, head-starting, and release). Prerelease health screenings were performed in 2014 and 2018 at Yadanabon Zoological Gardens,

Virus-host metabolic interaction: SGIV-induced protein lactylation suppresses viral propagation.

The prevention and cure of Singapore grouper iridovirus (SGIV) was remained a challenging issue, primarily due to our limited understanding of the interaction mechanisms between SGIV and the host. This study investigated the metabolic and protein modification changes in grouper spleen (GS) cells following SGIV infection. SGIV infection induced significant lactate accumulation and up-regulated protein lactylation levels in GS cells at 12 h post-infection. Transcriptomic analysis revealed that SGIV infection led to host cell metabolic reprogramming, characterized by marked up-regulation of oxida

An LNP-encapsulated mRNA vaccine targeting the major capsid protein of largemouth bass virus confers protective immunity and reduces RIG-I pathway activation in Micropterus salmoides.

Largemouth bass virus (LMBV) infection poses a significant threat to largemouth bass farming in China, yet effective and safe vaccines remain scarce. In this study, we designed and evaluated four mRNA vaccine candidates against LMBV, all encoding the viral major capsid protein (MCP). The fully configured construct (LMUM) contained a 5' cap1, untranslated regions (UTRs), and a poly(A) tail, and was produced by in vitro transcription. Each mRNA was encapsulated into lipid nanoparticles (LNPs), yielding homogeneous particles (∼110 nm diameter, ∼54 mV zeta potential) with >96% encapsulation effici

Differentiation-dependent antiviral capacities of amphibian (Xenopus laevis) macrophages.

Infections by ranaviruses such as Frog virus 3 (Fv3), are significantly contributing to worldwide amphibian population declines. Notably, amphibian macrophages (Mφs) are important to both the Fv3 infection strategies and the immune defense against this pathogen. However, the mechanisms underlying amphibian Mφ Fv3 susceptibility and resistance remain unknown. Mφ differentiation is mediated by signaling through the colony-stimulating factor-1 receptor (CSF-1R) which is now known to be bound not only by CSF-1, but also by the unrelated interleukin-34 (IL-34) cytokine. Pertinently, amphibian (Xeno

In situ mineralization of nano-selenium onto inactivated LMBV to enhance immune protection against viral infection.

Largemouth bass ranavirus (LMBV) is a major viral pathogen threatening the sustainable development of the largemouth bass (Micropterus salmoides) aquaculture industry. However, conventional inactivated vaccines generally exhibit limited immunogenicity and insufficient immune protection. Herein, a nano-selenium-functionalized inactivated vaccine (SeNPs/LMBV inactivated vaccine) was successfully constructed through an in situ mineralization strategy, in which selenium ions were initially assembled onto the surface of inactivated LMBV particles through electrostatic interactions and subsequently

Lactobacillus casei displaying MCP2α and FlaC delivered by PLA microspheres effectively enhances the immune protection of largemouth bass (Micropterus salmoides) against LMBV infection.

Largemouth bass ranavirus (LMBV) seriously affects the development of largemouth bass (Micropterus salmoides) industry and causes huge economic losses. Oral vaccine can be a promising method for viral disease precaution. In this study, MCP2α was identified as a valuable epitope region superior to MCP and MCP2 of LMBV by neutralizing antibody experiments. Then, recombinant Lactobacillus casei expressing the fusion protein MCP2αC (MCP2α as antigen, C represents flagellin C from Aeromonas hydrophila as adjuvant) on surface was constructed and verified. Further, PLA microsphere vaccine loading rec

Family shapes microbiome differences in Oklahoma salamanders.

INTRODUCTION: Given the role of microbiomes in promoting host health and homeostasis, understanding the factors shaping skin microbial communities in wild vertebrates has become increasingly important in conservation. This goal is even more pressing for amphibians, for which the skin has multiple critical functions, and pathogens currently decimating populations are linked to significant changes in skin microbiomes. However, because microbiomes are also shaped by environmental and ecological influences, as well as by host phylogeny, it is important to quantify these contributions to microbiome

Protective immunity induced by DNA vaccine encoding viral membrane protein against SGIV infection in grouper.

Singapore grouper iridovirus (SGIV) is the main grouper-infecting virus in southern China that causes serious economic losses. However, there is no effective way to control this viral disease. In this study, SGIV ORF19R (SGIV-19R) encoding a viral membrane protein was constructed into pcDNA3.1-HA and then was used to evaluate the immune protective effects in grouper Epinephelus coioides. Subcellular localization showed that SGIV-19R distributed in the cytoplasm and co-localization analysis indicated the protein partially co-localized with the endoplasmic reticulum (ER). RT-PCR and Western blot

Synergistic effects in the antiviral activity of the three Mx proteins from gilthead seabream (Sparus aurata).

Due to their direct antiviral activity, Mx proteins play a main role in the response mediated by type I interferon against viral infections. The study on gilthead seabream Mx proteins is especially interesting, since this species is unusually resistant to viral diseases, being asymptomatic carrier of several viruses pathogenic to other fish species. Gilthead seabream has three Mx proteins (Mx1, Mx2 and Mx3) that, separately, display antiviral activity against a wide range of viruses, showing interesting differences in their antiviral specificities. In this work, the possible synergy between th

EcSnx27 facilitates in vitro release of Singapore grouper iridovirus and interacts with its major capsid protein.

Sorting nexin 27 (Snx27), a member of the sorting nexin (SNX) family, plays crucial roles in cell signaling, organelle motility, protein sorting and membrane remodeling/trafficking. While existing studies have focused on the functions of SNXs in mammalian viral diseases and immune regulation, little is known about fish-encoded SNXs, particularly their regulatory roles in aquatic virus infection. In this study, we characterized the Snx27 from the orange-spotted grouper (Epinephelus coioides) and found that it facilitates the in vitro release of Singapore grouper iridovirus (SGIV), as evidenced

Functional characterization of the Rab11 and its response to SGIV infection in the orange-spotted grouper (Epinephelus coioides).

Rab11, a member of the Rab GTPase family, serves as a key regulator of vesicular trafficking and modulates viral infection in mammals, yet its functional role in aquatic viral infection remains largely uncharacterized. Singapore grouper iridovirus (SGIV), a member of the genus Ranavirus in the family Iridoviridae, is one of the most important viral pathogens afflicting grouper aquaculture. In this study, we investigated the influence of a novel Rab11 homolog (EcRab11) from the orange-spotted grouper (Epinephelus coioides) on SGIV infection and the role in host innate immune responses. EcRab11

Defining the Specific Pathogen-Free State of Xenopus Using TaqMan Assays.

Colonies of valuable inbred and transgenic laboratory-reared Xenopus frogs maintained for research constitute naïve populations of animals susceptible to some opportunistic infectious diseases. Therefore, it is prudent to characterize any new animal acquisitions before introduction into an existing colony as a biosecurity measure to preclude the concurrent introduction of an infectious microorganism associated with the new animal(s). In addition, some pathogens of Xenopus, such as Chlamydia and Mycobacterium spp, are zoonotic diseases, placing frog aquarists at risk for acquiring an infection.

Cutaneous Lesions in Freshwater Turtles (Emydura macquarii krefftii and Myuchelys latisternum) in a Rainforest Creek in North Queensland, Australia.

Freshwater turtles inhabit most rivers and creeks on the east coast of Australia, but some species are only found in specific catchments, which makes them vulnerable to extinction. During annual fieldtrips to Alligator Creek, North Queensland, the resident population of Myuchelys latisternum and Emydura macquarii krefftii in a natural pond, just outside Bowling Green National Park, have been surveyed for a number of years and demographic data recorded against tagged turtles. Rounded, cutaneous lesions on individual animals were first noted in August 2016, three years after the first survey of

Largemouth bass virus infection subverts immune-redox homeostasis in zebrafish and is associated with coordinated suppression of NF-κB and Nrf2 signaling.

From lower vertebrates to higher mammals, oxidative stress and inflammatory responses form a tightly regulated homeostatic network, yet the virus-specific mechanisms that coordinate these processes remain unclear. Here, we investigated this question using Largemouth bass virus (LMBV) in both zebrafish in vivo infection models and ZF4 cell in vitro infection models. In vivo, LMBV successfully infected and replicated in TU-strain zebrafish, causing significant histopathological damage, locomotor impairment, and approximately 40% mortality. Fish TRIM35 negatively regulates the interferon signaling pathway in response to grouper nodavirus infection.

Tripartite motif-containing protein 35 (TRIM35) has been demonstrated to exert critical roles in cancer, cell death and other multiple cell processes. However, the precisely roles of TRIM35 during virus infection still remained largely unknown. In the current study, we cloned a TRIM35 gene from orange spotted grouper (EcTRIM35) and uncovered its roles in response to nodavirus infection. EcTRIM35 encoded a 456-aa protein which showed 65% and 32% identity to large yellow croaker (Larimichthys crocea) and human (Homo sapiens), respectively. Structure prediction and amino acid alignment analysis i

Identification and characterization of c-raf from orange-spotted grouper (Epinephelus coioides).

C-Raf proto-oncogene serine/threonine kinase is a mitogen-activated protein kinase (MAP) kinase kinase, which can initiate a mitogen-activated protein kinase (MAPK) cascade by phosphorylating the dual-specific MAP kinase kinases (MEK1/2), and in turn activate the extracellular signal-regulated kinases (ERK1/2). To study the function of c-Raf in teleost fish, a c-Raf cDNA sequence from orange-spotted grouper (Epinephelus coioides) was cloned. Ecc-Raf shared 81%-99% amino acid identity with other vertebrate c-Raf molecules, and shared the highest amino acid identity (99%) with Lates calcarifer c
