The dopamine agonism on ADP-stimulated platelets is mediated through D2-like but not D1-like dopamine receptors.

Monoamines such as serotonin and epinephrine are known to be involved in platelet activation and aggregation. Dopamine is another monoamine identified in platelets, but published data about its effect on platelets and the receptors involved are controversial. In the present study, we investigated the dopamine agonism in platelets and the receptors involved in these pathways. Platelet-rich plasma (PRP) of healthy individuals was treated with agonists (ADP, epinephrine, dopamine) and various dopamine receptor and transporter antagonists such as SCH-23390, raclopride, clozapine, methylphenidate,

Bromocriptine induces marked locomotor stimulation in dopamine-depleted mice when D-1 dopamine receptors are stimulated with SKF38393.

In mice pretreated with reserpine plus alpha-methyl-p-tyrosine, neither the D-2 selective agonist bromocriptine, nor the D-1 selective agonist SKF38393, produced any measurable increase in locomotion in mice. However, the combination of the two agonists produced a marked and dose-dependent increase in co-ordinated locomotor activity. In mice with their dopamine stores and dopamine synthesis intact, SKF38393 was inactive by itself, but significantly enhanced the stimulant effect produced by bromocriptine. The data suggest that bromocriptine requires concomitant stimulation of D-1 receptors for

Targeting dopamine D2 receptors to alter fear extinction in adolescent rats.

Anxiety disorders emerging in adolescence are especially challenging to treat, partly due to adolescents having poor extinction recall and high relapse rates following exposure therapy, relative to other ages. Despite its clear clinical significance, little work has examined treatments to enhance extinction in adolescence. Dopamine, a key neurotransmitter involved in learning and memory, plays a critical role in fear extinction in adults, but its role in extinction in adolescents has not been well-explored. In a series of experiments, we investigated the effects of sulpiride, a dopamine antago

D2-type dopamine receptors are required for LPS-induced acute hematopoiesis.

Hematopoietic stem and progenitor cells (HSPCs) rapidly proliferate during infection and stress to replenish immune cells, which is essential for host defense. Dopamine, released by bone marrow (BM) nerves, regulates HSPCs via D2-type dopamine receptors. However, their role in emergency hematopoiesis across organs is unclear. We show that D2-type receptors are crucial for hematopoiesis in BM, spleen, lymph nodes, and thymus. Genetic deletion of D2-type receptors in hematopoietic cells (DKO∆HC) impairs HSPC proliferation and reduces blood cell production under lipopolysaccharide (LPS) stimulati

Oxygen chemoreceptor inhibition by dopamine D2 receptors in isolated zebrafish gills.

Dopamine is an essential modulator of oxygen sensing and control of ventilation and is the most well described and abundant neurotransmitter in the mammalian carotid body. Little is known of the evolutionary significance of dopamine in oxygen sensing, or whether it plays a similar role in anamniotes. In the model vertebrate, zebrafish (Danio rerio), presynaptic dopamine D2 receptor (D2R) expression was demonstrated in gill neuroepithelial cells (NECs), analogues of mammalian oxygen chemoreceptors; however, a mechanism for dopamine and D2R in the gills had not been defined. The present study te

The Dopamine Gene Receptors (DRD1-5) Expression Alteration in Psoriasis Patients.

BACKGROUND: Psoriasis is a chronic inflammatory autoimmune disease that is considered linked to genetic and environmental factors such as stress. Since the neurotransmitter dopamine has a close association with stress configuration, it can be a candidate for relieving psoriasis representation. In addition to the CNS, immune cells can play a decisive role in regulating immune functions through dopamine synthesis and the expression of its receptors. Altered response of immune cells to dopamine as well as a distorted expression of dopamine receptors (DRs) in immune cells have been reported in som

Dopamine receptors regulate preference between high-effort and high-risk rewards.

RATIONALE: Optimal decision-making necessitates evaluation of multiple rewards that are each offset by distinct costs, such as high effort requirement or high risk of failure. The neurotransmitter dopamine is fundamental toward these cost-benefit analyses, and D1-like and D2-like dopamine receptors differently modulate the reward-discounting effects of both effort and risk. However, measuring the role of dopamine in regulating decision-making between options associated with distinct costs exceeds the scope of traditional rodent economic decision-making paradigms. OBJECTIVES: We developed the

Epilepsy, amyloid-β, and D1 dopamine receptors: a possible pathogenetic link?

Experimental and clinical observations indicate that amyloid-β1-42 (Aβ1-42) peptide not only represents a major actor in neurodegenerative mechanisms but also induce hyperexcitation in individual neurons and neural circuits. In this abnormal excitability, possibly leading to seizures, the D1 dopamine (DA) receptors may play a role. Cerebrospinal fluid levels of Aβ1-42 were measured in patients with late-onset epilepsy of unknown etiology. Moreover, the effect of amyloid peptide on the hippocampal epileptic threshold and synaptic plasticity and its link to D1 receptor function were tested in ex

Expression of stereotyped behaviour requires stimulation of nigral D1 dopamine receptors.

Dopamine receptors in the pars reticulata of the substantia nigra were inactivated following bilateral injections of N-ethoxycarbonyl-2-ethoxy-1,2-dihydroquinoline (EEDQ). Loss of D1, but not D2, receptors was associated with a significant decrease in the incidence of stereotyped head-down sniffing elicited by the mixed D1/D2 receptor agonist, apomorphine. The results support the hypothesis that D1 receptors in the substantia nigra are necessary for the expression of dopamine mediated stereotyped behaviour.

Dopamine D1 receptors of the calf parathyroid gland: identification and characterization.

The dopamine D1 receptor was identified in the calf parathyroid gland. The binding of the selective D1 receptor antagonist [3H]SCH-23390 to membranes of calf parathyroid was specific, reversible, and saturable with a dissociation constant of approximately 200 pM and a receptor density of 30 fmol/mg of protein. Dopaminergic agonists and antagonists inhibited [3H]SCH-23390 binding in a concentration-dependent and stereoselective manner with an appropriate pharmacological specificity for D1 dopamine receptors. Moreover, potent dopaminergic agonists recognized two affinity forms of the receptor, o

Behavioural role of dopamine D1 receptors in the reserpine-treated mouse.

The effects of 2,3,4,5-tetrahydro-7,8-dihydroxy-1-phenyl-1H-3-benzazepine (SKF 38393) (D1 agonist) on the motor behaviour of mice rendered hypokinetic with reserpine, were studied in the absence and presence of additional treatment with N-n-propyl-N-phenylethyl-p(3-hydroxyphenyl)ethylamine hydrochloride (RU 24213), lisuride (D2 agonists) or apomorphine (mixed D1/D2 agonist). Three hours after reserpine (5 mg/kg) stimulating dopamine D2 receptors evoked slow, ponderous walking and head-down sniffing. SKF 38393 (1.5-15 mg/kg) had no direct effect of its own, but greatly amplified the D2 response

Modulation of dopamine D1 receptors via histamine H3 receptors is a novel therapeutic target for Huntington's disease.

Early Huntington's disease (HD) include over-activation of dopamine D1 receptors (D1R), producing an imbalance in dopaminergic neurotransmission and cell death. To reduce D1R over-activation, we present a strategy based on targeting complexes of D1R and histamine H3 receptors (H3R). Using an HD mouse striatal cell model and HD mouse organotypic brain slices we found that D1R-induced cell death signaling and neuronal degeneration, are mitigated by an H3R antagonist. We demonstrate that the D1R-H3R heteromer is expressed in HD mice at early but not late stages of HD, correlating with HD progress

Dopamine, vesicular transporters, and dopamine receptor expression in rat major salivary glands.

The localization of dopamine stores and the expression and localization of dopamine (DAT) and vesicular monoamine transporters (VMAT) type-1 and -2 and of dopamine D1-like and D2-like receptor subtypes were investigated in rat submandibular, sublingual, and parotid salivary glands by HPLC with electrochemical detection, as well as immunochemical and immunohistochemical techniques. Male Wistar rats of 2 mo of age were used. The highest dopamine levels were measured in the parotid gland, followed by the submandibular and sublingual glands. Western blot analysis revealed DAT, VMAT-1, VMAT-2, and

History of the discovery of the antipsychotic dopamine D2 receptor: a basis for the dopamine hypothesis of schizophrenia.

The 1975 publication of Seeman et al. (Proc Nat Acad Sci, USA), reporting the discovery of the antipsychotic receptor in the brain, is a classic example of translational medicine research. In searching for a pathophysiological mechanism of psychosis, the team sought to identify sites that bound the antipsychotic drug haloperidol. Their criterion was that haloperidol bound to the site at one to two nanomoles per liter, corresponding to haloperidol concentrations found in spinal fluid or plasma water in treated patients. They requested de novo synthesis of tritiated haloperidol, and it readily d

Sensitivity of binding of high-affinity dopamine receptor radioligands to increased synaptic dopamine.

PET and SPECT studies have documented that D2 radioligands of moderate affinity, but not radioligands of high affinity, are sensitive to pharmacological challenges that alter synaptic dopamine levels. The objective of this work was to determine whether the brain kinetics of high-affinity radioligands for dopamine D1 ([(3)H]SCH 23390) and D2 ([(123)I]epidepride) receptors were altered by a prolonged elevation of synaptic dopamine induced by the potent cocaine analog RTI-55. Mice were injected intravenously with radioligands either 30 min after or 4 h before intraperitoneal administration of RTI

Polydipsia and dopamine: behavioral effects of dopamine D1 and D2 receptor agonists and antagonists.

Substantial evidence implicates dopaminergic neural systems in the occurrence of polydipsia in both animals and humans. Two experiments were conducted in order to specify the behavioral mechanisms whereby manipulation of dopaminergic neural transmission can affect scheduled-induced polydipsia (SIP). The role of dopamine D1 and D2 receptors was investigated by comparing the behavioral effects of dopamine D1 agonists (SKF 38393 and SKF 82958) and antagonists (SCH 23390 and SKF 83566) to those of a dopamine D2 agonist (quinpirole) and antagonist (haloperidol) by using an animal model of excessive

Mouse spinal cord cellular mapping of dopamine D2 receptors-containing cells.

The spinal cord (SC) serves as the primary relay for sensory information originating in the periphery and transmitted to the brain for processing. Sensitive primary afferent fibers project to the dorsal horn, which contains a highly diverse array of neurons forming a complex network of excitatory and inhibitory circuits. Previous studies have indicated that this neuronal network can be modulated by the monoaminergic system, particularly through the spinal dopaminergic circuit, partly via dopamine D2 receptors (D2R). However, the identity of the cells expressing D2R within the spinal cord remai

Dopamine receptors in ocular surface biology: Current evidence and future directions.

Dopamine, a classical catecholamine neurotransmitter, exerts diverse regulatory functions in the central nervous system, retina, and intraocular signaling. Recent research has highlighted its emerging role in ocular surface biology, which includes the cornea, conjunctiva, tear film, and lacrimal glands. The ocular surface serves as a neuroepithelialimmune interface where neuromodulators such as dopamine influence epithelial homeostasis, wound repair, inflammation, and sensory signaling. Molecular and functional studies have confirmed the expression of dopamine receptor subtypes (DRD1-DRD5) and

Inhibition of Nigrostriatal Dopamine Release by Striatal GABAA and GABAB Receptors.

Nigrostriatal dopamine (DA) is critical to action selection and learning. Axonal DA release is locally influenced by striatal neurotransmitters. Striatal neurons are principally GABAergic projection neurons and interneurons, and a small minority of other neurons are cholinergic interneurons (ChIs). ChIs strongly gate striatal DA release via nicotinic receptors (nAChRs) identified on DA axons. Striatal GABA is thought to modulate DA, but GABA receptors have not been documented conclusively on DA axons. However, ChIs express GABA receptors and are therefore candidates for potential mediators of
