[The association between post anoxic encephalopathy and startle epilepsy].

INTRODUCTION: The startle is a variant of primitive physiological reflex, present in developed animals. It could be defined as a sudden defensive reaction triggered by an adequate sensorial stimulus, most effective if auditory, consisting under a clinical basis on the sudden and sequential contraction of some muscular groups, in first place affecting the facial territory (orbicularis oculi bilaterally), followed by the rest of the cranial nerves, the axial musculature and finally the limbs, predominantly proximal and flexor regions of superior extremities. The pathological startle is that whic

The gap-startle paradigm for tinnitus screening in animal models: limitations and optimization.

In 2006, Turner and colleagues (Behav. Neurosci., 120:188-195) introduced the gap-startle paradigm as a high-throughput method for tinnitus screening in rats. Under this paradigm, gap detection ability was assessed by determining the level of inhibition of the acoustic startle reflex produced by a short silent gap inserted in an otherwise continuous background sound prior to a loud startling stimulus. Animals with tinnitus were expected to show impaired gap detection ability (i.e., lack of inhibition of the acoustic startle reflex) if the background sound containing the gap was qualitatively s

Discrimination of auditory from nonauditory toxicity by reflex modulation audiometry: effects of triethyltin.

The evaluation of toxicity in sensory systems presents particular problems because of the need to distinguish specific sensory loss from other toxic effects. It has recently been shown that modulation of reflex behavior by low-intensity test stimuli can be used to provide a rapid assessment of sensory acuity and of ototoxic hearing loss. In this report the ability of the method to distinguish the known neuromuscular consequences of triethyltin (TET) from hearing loss is demonstrated. Rats treated with TET bromide (30 mg/liter) in their drinking water for 3 weeks showed a profound decrease in a

Lack of startle modulation by smoking cues in smokers.

RATIONALE: The startle reflex methodology has been used to study the effects of nicotine in humans and the motivational effects of smoking cues in smokers. However, no other studies investigate startle modulation by smoking cues in smokers compared to non-smokers. In the other studies, smoking deprivation was manipulated in smokers or smokers were not compared directly to non-smokers. OBJECTIVE: The goal of this study was to examine the temporal course of information processing following the presentation of a smoking-related cue using the startle probe methodology in smokers compared to non-s

[The sensorimotor reactivity of rats with an initially high anxiety-phobia level].

Sensorimotor response was measured by acoustic startle reflex in Wistar male rats with innate high and low levels of anxiety. The levels of anxiety were determined using a complex multiparameter method for evaluating anxiety-phobic states in rats by a ranged scale. Amplitude and prepulse inhibition of the acoustic startle response were increased, but latency of the startle reflex was decreased in rats with innate high level of anxiety as compared with those with innate low level. In rats with innate high level of anxiety injection of subconvulsive doses of pentylenetetrazol (10 and 15 mg/kg i.

Amygdala dysfunction in narcolepsy-cataplexy.

The blink reflex of acoustic startle reflex (ASR) is modulated by emotions and a loss of physiological aversive ASR potentiation is reported in humans following amygdala lesions. Patients with narcolepsy-cataplexy (NC) were found to have normal ASR, but they failed to exhibit startle potentiation during unpleasant stimuli. This absence of aversive ASR potentiation gives support to the hypothesis of an amygdala dysfunction in human NC.

Late onset startle induced tics.

Three cases of late onset Gilles de la Tourette's syndrome are presented. The motor tics were mainly induced by an unexpected startling stimulus, but the startle reflex was not exaggerated. The tics developed after physical trauma or a period of undue emotional stress. Reflex tics may occur in Gilles de la Tourette's syndrome, but have not been described in late onset Tourette's syndrome. Such tics must be distinguished from psychogenic myoclonus and the culture bound startle syndromes.

DYT-PRKRA Dystonia-Parkinsonism with Pathological Startle: Expanding the Phenotype of PRKRA-Related Disease.

BACKGROUND: DYT-PRKRA (formerly DYT16) is an autosomal recessive dystonia-parkinsonism syndrome caused by biallelic pathogenic variants in PRKRA, a gene encoding the stress-responsive protein PACT. While early-onset generalized dystonia and speech disturbance are well-recognized features, pathological startle has not previously been described. CASES: We report two siblings with genetically confirmed DYT-PRKRA (homozygous pathogenic PRKRA variant p.Pro222Leu). Case 1, a 37-year-old male, presented with childhood-onset dystonia and persistent pathological startle. Case 2, a 33-year-old female,

Measuring anxious responses to predictable and unpredictable threat in children and adolescents.

Research has highlighted the need for new methods to assess emotions in children on multiple levels to gain better insight into the complex processes of emotional development. The startle reflex is a unique translational tool that has been used to study physiological processes during fear and anxiety in rodents and in human participants. However, it has been challenging to implement developmentally appropriate startle experiments in children. This article describes a procedure that uses predictable and unpredictable aversive events to distinguish between phasic fear and sustained anxiety in ch

Covariation bias in the affect-modulated startle paradigm.

A covariation bias (CB) reflects the overestimation of an objectively random covariation between fear-relevant picture stimuli (e.g., spiders) and aversive consequences (e.g., electric shocks). The affect-modulated startle (AMS) paradigm is similar to it because aversive reflex probes are delivered in the context of unpleasant, neutral or pleasant picture stimuli. The present study explored if a covariation bias is present in the affect-modulated startle paradigm. There was a covariation bias for unpleasant and pleasant compared to neutral pictures probably due to differences in arousal. Start

Research Letter: Concussion-Related General Startle Suppression in Adolescent Athletes.

OBJECTIVES: We investigated the acoustic startle reflex in recently concussed adolescent athletes compared to healthy controls and those with concussion history (>1 year prior) but no current symptoms. We hypothesized that individuals with recent concussion would have a suppressed startle response compared to healthy controls. METHODS: We conducted a cross-sectional study on 49 adolescent athletes with a recent concussion (n = 20; age: 14.6 ± 1.6 years; 60% female), a concussion history > 1 year prior (n = 16; age: 14.8 ± 2.0 years; 44% female), and healthy controls (n = 13; age: 13.3 ± 2.8 y

Diethyldithiocarbamate depresses the acoustic startle response in rats.

The relationship between norepinephrine (NE) content in cortex and spinal cord and acoustic startle amplitude was investigated in two experiments. Administration of diethyldithiocarbamate (DDC) depressed startle amplitude at the same time and dose that it most severely depleted NE content. These results support the conclusion that NE facilitates the normal elaboration of the acoustic startle reflex and also support evidence that NE activity in the spinal cord may be of particular importance in the maintenance of normal startle amplitude.

Extreme startle and photomyoclonic response in severe hypocalcaemia.

We report the case of 62-year-old woman referred to our department because of a clinical suspicion of tonic-clonic seizures. Clinical examination revealed an exaggerated startle reflex, EEG showed a photomyoclonic response, and blood tests indicated severe hypocalcaemia. Additional clinical data, treatment strategies, and long-term follow-up visits were reported. The present report discusses the difficulties in distinguishing between epileptic and non-epileptic startles, and shows, for the first time, exaggerated startle reflex and extreme photomyoclonic response due to severe hypocalcaemia.

Intractable reflex audiogenic epilepsy successfully treated by peri-insular hemispherotomy.

We report a case of an infantile hemiplegia seizure syndrome (IHSS) that presented with intractable reflex audiogenic startle epilepsy which in itself is an uncommon form of seizure disorder. Peri-insular hemispherotomy provided complete seizure control. Also of particular interest was that this syndrome resulted from an iatrogenic brain injury sustained during the course of a caesarian section. We review the different mechanisms of birth injury reported in the literature and, discuss the physiopathogenesis of the hemispheric damage in this patient. We also review the literature on "reflex epi

Didn't Even Blink: Dissociation, Complex Trauma Exposure and Decreased Startle Reactivity.

The startle eyeblink reflex is thought to function as a means of orienting to salient stimuli, and, by proxy, sensitivity to threat cues. The absence or attenuation of this reflex may thus suggest disengagement from one's environment, potentially in circumstances when engagement is called for, and, therefore, may serve as a potential marker for dissociation as it occurs. The present study investigates whether individual differences in startle response magnitude and habituation are attributable to early and multiple trauma exposure, dissociation, and PTSD symptom severity. Augmented startle responses in opsoclonus-myoclonus syndrome.

We report a 3-year-old boy with opsoclonus-myoclonus syndrome (OMS) who presented with exaggerated startle responses to unexpected auditory stimuli during an episode of myoclonic status. An augmented blink reflex was also observed clinically and electrophysiologically. Based on the assumption that hyperexcitability in the lower pontine tegmentum may be responsible for the acoustic startle and blink reflex in OMS, we considered that increased excitability of independent but neighboring structures, including the pontine paramedian reticular formation, may cause OMS symptoms.

Clarifying domains of internalizing psychopathology using neurophysiology.

Current initiatives such as the National Institute of Mental Health's Research Domain Criteria project aim to reorganize classification of mental disorders along neurobiological lines. Here, we describe how consideration of findings from psychiatric research employing two physiological measures with distinct neural substrates--the startle blink reflex and the error-related negativity (ERN)--can help to clarify relations among disorders entailing salient anxiety or depressive symptomatology. Specifically, findings across various studies and reviews reveal distinct patterns of association for bo

Startle-triggered responses indicate reticulospinal drive is larger for voluntary shoulder versus finger movements.

Recent primate studies have implicated a substantial role of reticulospinal pathways in the production of various voluntary movements. A novel way to assess the relative reticulospinal contributions in humans is through the use of a "StartReact" paradigm where a startling acoustic stimulus (SAS) is presented during a simple reaction time (RT) task. The StartReact response is characterized by short-latency triggering of a prepared response, which is attributed to increased reticulospinal drive associated with startle reflex activation. The current study used a StartReact protocol to examine dif

Acquisition of acoustic startle response in relation to growth and thyroid function in rats.

Thiourea was administered by gavage to pregnant and lactating rats from gestational day 18 until postnatal day 10, at doses of 100 or 250 mg per day (approximately 350 or 900 mg/kg/day). From the day after birth until postnatal day 14 the neonates were weighed, and beginning on postnatal day 10 acquisition of acoustic startle reflex was assessed. Serum thyroxine and thyrotropin were measured on postnatal day 14. High dose thiourea severely inhibited thyroid function in pups, significantly depressed body growth, and retarded development of the acoustic startle response. At the low dose there wa

Evidence for Increased High Alpha Intermuscular Coherence as a Measure of Reticulospinal Motor Drive.

The reticulospinal (RS) system is a key descending motor pathway involved in human movement control. However, it remains poorly characterized, largely due to methodological challenges in assessing its anatomy and physiology. Current approaches to gauge RS drive rely on intense stimuli (auditory cues, transcranial magnetic stimulation), limiting their feasibility. Here, we examined intermuscular coherence as a potential biomarker of common neural drive related to the RS system. Intermuscular coherence was evaluated during the startle reflex and StartReact paradigm, both known to be primarily me
