The cerebellar nodulus/uvula integrates otolith signals for the translational vestibulo-ocular reflex.

BACKGROUND: The otolith-driven translational vestibulo-ocular reflex (tVOR) generates compensatory eye movements to linear head accelerations. Studies in humans indicate that the cerebellum plays a critical role in the neural control of the tVOR, but little is known about mechanisms of this control or the functions of specific cerebellar structures. Here, we chose to investigate the contribution of the nodulus and uvula, which have been shown by prior studies to be involved in the processing of otolith signals in other contexts. METHODOLOGY/PRINCIPAL FINDINGS: We recorded eye movements in two

Eye-head movement coordination: vestibulo-ocular reflex suppression with head-fixed target fixation.

To maintain clear vision, the images on the retina must remain reasonably stable. Head movements are generally dealt with successfully by counter-rotation of the eyes induced by the combined actions of the vestibulo-ocular reflex (VOR) and the optokinetic reflex. A problem of importance relates to the value of the so-called intrinsic gain of the VOR (VORG) in man, and how this gain is modulated to provide appropriate eye movements. We have studied these problems in two situations: 1. fixation of a stationary object of the visual space while the head moves; 2. fixation of an object moving with

Visual oculomotor abnormalities and vestibulo‑ocular reflex dynamics in polyglutamine spinocerebellar ataxias (Review).

Spinocerebellar ataxias (SCAs) are a group of autosomal dominant neurodegenerative diseases that are characterized by cerebellar ataxia. The most common types of SCAs are caused by polyglutamine (polyQ)-encoding cytosine-adenine-guanine repeat expansions. Autosomal dominant SCAs share similar pathophysiological mechanisms. The cerebellum plays an important role in the generation and control of eye movement, and neuropathological findings indicate that cerebellar degeneration is commonly present in polyQ-SCAs. As a result, various patterns of oculomotor impairment are present in most SCA subtyp

The Effect of Different Auditory Stimuli and Optokinetic Stimuli on Vestibulo-Ocular Reflex Function.

This study investigated how auditory stimuli and optokinetic stimulation modulate functional Head Impulse Test (fHIT) correct response rate (CR%) across semicircular canals (SCCs) and the associated cognitive load in healthy adults. Fifty participants (20-57 years) completed repeated-measures fHIT under four conditions: silence, music, white noise, and optokinetic stimulation. The CR% from all SCCs were recorded and workload was assessed with the NASA Task Load Index (NASA-TLX). Optokinetic stimulation produced the most significant CR% decrement (e.g., right lateral 90 vs. 100 in silence; p<0.

Origin of orientation-dependent asymmetries in vestibulo-ocular reflexes evoked by caloric stimulation.

A caloric stimulus evokes primarily a horizontal vestibulo-ocular reflex (VOR) when subjects are in a supine or prone orientation with the horizontal semicircular canal plane oriented vertically. In both monkeys and humans, the magnitude of VOR eye movements is greater in the supine than in the prone orientation, indicating that some factor or factors, other than the conventionally accepted convective stimulation of the horizontal canals, contributes to the generation of the VOR. We used long-duration caloric irrigations and mathematical models of canal-otolith interactions to investigate fact

Control of spatial orientation of the angular vestibulo-ocular reflex by the nodulus and uvula of the vestibulocerebellum.

Eye velocity produced by the angular vestibulo-ocular reflex (aVOR) tends to align with the summed vector of gravity and other linear accelerations [gravito-inertial acceleration (GIA)]. Defined as "spatial orientation of the aVOR," we propose that it is controlled by the nodulus and uvula of the vestibulocerebellum. Here, electrical stimulation, injections of the GABAA agonist, muscimol, and single-cell recordings were utilized to investigate this spatial orientation. Stimulation, injection, and recording sites in the nodulus were determined in vivo by MRI and verified in histological section

Autorotation test abnormalities of the horizontal and vertical vestibulo-ocular reflexes in panic disorder.

Patients with panic disorder often describe dizziness as a disturbing symptom, with more severe episodes reported than in other psychiatric populations. Nineteen patients diagnosed as having a panic disorder were tested for vestibulo-ocular (VOR) abnormalities with the Vestibular Autorotation Test (VAT), a computerized test of the high-frequency (2 to 6 Hz) VOR. The patients were unselected for the presence or absence of balance disorders. Results showed VOR abnormalities, relative to a normal population, in the horizontal and/or vertical VORs of all 19 patients. Vestibulo-ocular reflex asymme

Evaluating the Effects of Levodopa on the Vestibulo-Ocular Reflex in Parkinson's Disease: A Case Series.

PURPOSE: Parkinson's disease (PD) is a neurodegenerative disorder primarily affecting individuals over 60 years old. The disorder is characterized by motor symptoms due in large part to the loss of dopamine-producing neurons. The vestibular system, crucial for maintaining balance, may be impacted in PD, contributing to balance issues through dysfunction in the vestibulo-ocular reflex. In this study, we aim to describe differences in vestibular responses before and after use of levodopa (L-DOPA) in patients with PD. METHOD: The study evaluated nine PD patients who underwent videonystagmography

No Gain No Pain: Relations Between Vestibulo-Ocular Reflexes and Motion Sickness in Mice.

Motion sickness occurs when the vestibular system is subjected to conflicting sensory information or overstimulation. Despite the lack of knowledge about the actual underlying mechanisms, several drugs, among which scopolamine, are known to prevent or alleviate the symptoms. Here, we aim at better understanding how motion sickness affects the vestibular system, as well as how scopolamine prevents motion sickness at the behavioral and cellular levels. We induced motion sickness in adult mice and tested the vestibulo-ocular responses to specific stimulations of the semi-circular canals and of th

Influence of stimulus interval on the habituation of vestibulo-ocular reflex and sensation of rotation in humans.

Previous studies in cats revealed that vestibular habituation of the vestibulo-ocular reflex (VOR) only occurs when velocity steps are delivered during the secondary phase nystagmus, suggesting that the presence of anti-compensatory slow phases may trigger the habituation process. We verified this property in humans by comparing vestibular habituation of VOR and sensation of rotation when steps were delivered either immediately after the perception of self-rotation had stopped, which is shortly before the nystagmus reverses direction; or when steps were delivered 60s later, i.e. during the sec

Do agility and skull architecture influence the geometry of the mammalian vestibulo-ocular reflex?

The spatial arrangement of the semicircular canals and extraocular muscles of the eye has been of considerable interest, particularly to researchers working on adaptations of the vestibulo-ocular reflex. Here we offer the first, extensive comparative analysis of the spatial relationships between each extraocular muscle and the canal providing its primary excitatory stimulus. The sample consisted of 113 specimens, representing 51 extant mammalian species. Hypotheses tested included that variations in the spatial alignments are linked with differences of skull morphology and with differences of

Adaptation of the angular vestibulo-ocular reflex to head movements in rotating frames of reference.

Head movements in a rotating frame of reference are commonly encountered, but their long term effects on the angular vestibulo-ocular reflex (aVOR) are not well understood. To study this, monkeys were oscillated about a naso-occipital (roll) axis for several hours while rotating about a spatial vertical axis (roll-while-rotating, RWR). This induced oscillations in roll and pitch eye velocity and continuous horizontal (yaw) nystagmus. For several hours thereafter, simple roll in darkness induced horizontal nystagmus and pitch and roll oscillations. The rising and falling time constants of the h

The effect of unilateral posterior semicircular canal inactivation on the human vestibulo-ocular reflex.

The responses to rapid, passive, unpredictable, low amplitude (10-20 degrees), high acceleration (3,000-4,000 degrees/s2) head rotations were used to study the human vestibulo-ocular reflex (VOR) in pitch and yaw plane after unilateral posterior semicircular canal occlusion (uPCO) in 10 subjects. The results from these 10 uPCO subjects were compared with those from 18 normal subjects. The VOR gains at a head velocity of 200 degrees/s in the uPCO subjects were: pitch upward = 0.62 +/- 0.06, pitch downward = 0.87 +/- 0.11, yew ipsilesion = 0.78 +/- 0.06, yaw contralesion = 0.79 +/- 0.10 and in n

Effects of unilateral loss of vestibular function on the vestibulo-ocular reflex and postural control.

Long-term recovery from surgically induced unilateral loss of vestibular function was studied in 14 patients. Seven patients underwent surgical extirpation or section of the vestibular nerve, and seven patients underwent labyrinthectomy without vestibular nerve section. The vestibulo-ocular reflex (VOR) and postural control were evaluated preoperatively and monitored for up to 4 years postoperatively with use of pseudorandom rotation (combined sinusoidal frequencies from 0.009 to 1.5 Hz) and moving platform posturography. Immediately following surgery all patients showed minimal reductions in

Combining Multi-Canal Vestibular Implant Stimulation to Restore the Vestibulo-Ocular Reflex in 3D.

INTRODUCTION: Vestibular implants targeting the semi-circular canals might offer a potential treatment approach for vestibulopathy patients. Electrical stimulation is provided to all three canals to restore gaze stabilization in all three planes of space. This study evaluated the effect of electrically stimulating multiple canals at the same time. It was hypothesized that eye responses of single-canal stimulation could predict eye responses of multi-canal stimulation, using vector summation. This might provide valuable information for future vestibular implant fitting strategies. METHODS: Nin
