Picolinic Acid Derivatives and Biosynthetic Pathway from Epicoccum sorghinum SDU-F549.

Two new picolinic acid derivatives, ethylfusaric acid A (1) and ethylfusaric acid B (2), along with two known picolinic acid derivatives (3-4), were isolated from the culture of Rehmannia glutinosa endophytic fungus Epicoccum sorghinum SDU-F549. The chemical structures of the compounds were elucidated by the combination of 1D and 2D nuclear magnetic resonance spectroscopy and electronic circular dichroism calculations. Compounds 1-4 exhibited antibacterial activity against a panel of Staphylococcus aureus strains, with MIC values ranging from 16 to 32 μg/mL. In contrast, they showed no signifi

Effects of Kampo herbal medicine on plasma melatonin concentration in patients.

There are studies reporting that some natural products containing melatonin. Sleep disturbance and nocturnal polyuria are related to abnormal biorhythm. It is possible that kampo medicine may influence biorhythm. We therefore examined the symptoms and plasma melatonin of patients treated with rokumi-gan, hachimi-jio-gan, gosha-jinki-gan and seishinrenshi-in. Furthermore, the concentration of four kampo medicines and crude drugs making up these formulations were investigated. Diurnal plasma melatonin was not affected by Rokumi-gan, but tended to be lowered by treatment with hachimi-jio-gan, gos

Exploring the effects of Chinese herbal ingredients on the signaling pathway of alopecia and the screening of effective Chinese herbal compounds.

ETHNOPHARMACOLOGICAL RELEVANCE: alopecia is a hair disorder that can add a significant medical and psychological burden to patients. Currently, the FDA-approved drugs for the treatment of androgenetic alopecia (AGA) are minoxidil and finasteride and immunosuppressives are therapeutic options for alopecia areata (AA), but the objective adverse effects and high cost of these treatments reduce patient compliance and thus the effectiveness of the drugs. Traditional Chinese medicine (TCM) has good efficacy, a high safety profile and low treatment costs, but its mechanism of action is still not full

Spiky mesoporous silica-based nanovaccines enhance mucosal immunity with improved intestinal retention and antigen accessibility.

Intestinal vaccination faces several key challenges, including limited epithelial interaction, inefficient antigen uptake, and insufficient immune stimulation. Here, we developed a virus-inspired oral nanovaccine based on spiky mesoporous silica nanoparticles (VMSNs) functionalized with Rehmannia glutinosa polysaccharide (RGP), a natural polysaccharide with favorable intestinal biocompatibility. The resulting RGP-VMSN showed enhanced intestinal retention and epithelial interaction and was associated with increased antigen accessibility within the mucosal environment, reflected by stronger anti

Reporting quality and risk of bias of randomized controlled trials of Chinese herbal medicine for multiple sclerosis.

BackgroundMultiple sclerosis (MS) is the most common non-traumatic disabling disease affecting young adults. A definitive curative treatment is currently unavailable. Many randomized controlled trials (RCTs) have reported the efficacy of Chinese herbal medicine (CHM) on MS. Because of the uncertain quality of these RCTs, the recommendations for routine use of CHM for MS remain inconclusive. The comprehensive evaluation of the quality of RCTs of CHM for MS is urgent.MethodsNine databases, namely, PubMed, Embase, Web of Science, Cochrane Library, EBSCO, Sinomed, Wanfang Database, China National

Magnetic nanoparticle carrying verbascoside suppresses oral squamous cell carcinoma progression by targeting ICAM1 via IL-6/STAT3 axis.

Oral squamous cell carcinoma (OSCC), the most common oral malignancy, requires innovative therapeutic strategies. We developed a magnetic nanoparticle carrying verbascoside (VB-Fe3O4) to enhance the bioavailability and antitumor efficacy of VB, a bioactive compound from Rehmannia glutinosa. VB-Fe3O4 demonstrated superior tumor-targeting capabilities, achieving higher drug accumulation compared with free VB. In the human OSCC cell line HSC-3, VB-Fe3O4 markedly induced apoptosis, inhibited proliferation and invasion through IL-6/STAT3 pathway. Mechanistic studies screened out intercellular cell

[Connotation of distal bleeding based on modern pathophysiological mechanism and application of Huangtu Decoction for acute coronary syndrome complicated with acute upper gastrointestinal hemorrhage in critical care medicine].

Huangtu Decoction, first recorded in Essentials from the Golden Cabinet(Jin Kui Yao Lue) from ZHANG Zhong-jing in Han dynasty, is used to treat distal bleeding. It is mainly treated for the syndrome of failing to control blood with spleen-yang deficiency. The connotation of distal bleeding is more extensive, including not only upper gastrointestinal bleeding in the traditional sense such as peptic ulcer bleeding, gastrointestinal tumors, gastric mucosal lesions, vascular dysplasia, esophagogastric variceal bleeding, and pancreatic and biliary tract injury, but also other anorectal diseases suc

Catalpol-Loaded Silk Fibroin Nanoparticle/GelMA Hydrogels Promote Autophagy-Mediated Angiogenesis for Vascular Regeneration in Limb Ischemia.

Limb ischemia is a severe manifestation of peripheral arterial disease with limited therapeutic options. Induction of therapeutic angiogenesis represents a promising strategy to restore blood perfusion. Natural bioactive compounds, particularly when integrated with biomaterial engineering, offer therapeutic potential with favorable biosafety profiles. Here, we demonstrate that catalpol (CTP), a bioactive compound from Rehmannia glutinosa, exerts pro-angiogenic effects by enhancing endothelial cell viability, proliferation, migration, and vascular sprouting in vitro. Catalpol upregulates angiog

Mechanistic insights into the neuroprotective effects of yangyin formula on cerebral ischemia-reperfusion injury via the IL-17A/MAPK/NF-κB signaling pathway.

Ischemic stroke is a leading cerebrovascular disorder frequently complicated by cerebral ischemia-reperfusion injury (CIRI), which aggravates neurological damage and worsens clinical outcomes. Yangyin Formula (YYF), a traditional Chinese medicine composed of Rehmannia glutinosa, Dendrobium officinale, and Pueraria lobata, has shown potential in mitigating CIRI. To elucidate its therapeutic mechanisms, we integrated network pharmacology, molecular docking, and molecular dynamics simulations, followed by in vivo validation in a rat model of middle cerebral artery occlusion/reperfusion (MCAO/R).

Catalpol alleviates hypoxia ischemia-induced brain damage by inhibiting ferroptosis through the PI3K/NRF2/system Xc-/GPX4 axis in neonatal rats.

Hypoxic-ischemic encephalopathy (HIE) is a brain damage caused by perinatal hypoxia and blood flow reduction. Severe HIE leads to death. Available treatments remain limited. Oxidative stress and nerve damage are major factors in brain injury caused by HIE. Catalpol, an iridoid glucoside found in the root of Rehmannia glutinosa, has antioxidant and neuroprotective effects. This study examined the neuroprotective effects of catalpol using a neonatal rat HIE model and found that catalpol might protect the brain through inhibiting neuronal ferroptosis and ameliorating oxidative stress. Catalpol suppresses osteoclastogenesis and attenuates osteoclast-derived bone resorption by modulating PTEN activity.

Excessive activation of osteoclast activity is responsible for many bone diseases, such as osteoporosis, rheumatoid arthritis, periprosthetic osteolysis, and periodontitis. Natural compounds that inhibit osteoclast formation and/or function have therapeutic potential for treating these diseases. Catalpol, a bioactive iridoid extracted from a traditional herbal medicine Rehmannia glutinosa, exhibits various pharmacological properties, including anti-inflammatory, antioxidant, antidiabetic, and antitumor effects. However, its effects on osteoclast formation and function remain unknown. In the pr

Ajugol ameliorates mitochondrial dysfunction and cognitive decline in Alzheimer's Disease via BNIP3-dependent mitophagy.

Defective mitophagy plays key roles in mitochondrial dysfunction, inflammation, and energy deprivation, and this defect can lead to synaptic loss and cognitive decline in Alzheimer's disease (AD). Although pharmacological enhancement of mitophagy has been found to ameliorate cognitive impairment in AD models, therapeutic strategies directly targeting this pathway remain limited. Ajugol, a bioactive iridoid glycoside isolated from Rehmannia glutinosa, has recently been identified as a potential metabolic regulator. In this study, we demonstrated that ajugol markedly alleviates mitochondrial dam

Catalpol ameliorates high-fat diet-induced insulin resistance and adipose tissue inflammation by suppressing the JNK and NF-κB pathways.

Catalpol, a bioactive component from the root of Rehmannia glutinosa, has been shown to possess hypoglycemic effects in type 2 diabetic animal models, however, the underlying mechanisms remain poorly understood. Here we investigated the effect of catalpol on high-fat diet (HFD)-induced insulin resistance and adipose tissue inflammation in mice. Oral administration of catalpol at 100 mg/kg for 4 weeks had no effect on body weight of HFD-induced obese mice, but it significantly improved fasting glucose and insulin levels, glucose tolerance and insulin tolerance. Moreover, macrophage infiltration

Catalpol protects mesencephalic neurons against MPTP induced neurotoxicity via attenuation of mitochondrial dysfunction and MAO-B activity.

Catalpol, an iridoid glucoside, separated from the root of Rehmannia glutinosa Libosch, has been known to show various neuroprotective effects. In humans and rodents, MPTP is well known to produce clinical, biochemical and neurochemical changes similar to those which occur in Parkinson's disease (PD). Furthermore, the accumulated evidence suggests that MPP(+), conversed by monoamine oxidase type B (MAO-B) in astrocytes principally, is the active metabolite of MPTP and the major cause to PD associated with mitochondrial dysfunction. In this study, we treated mesencephalic neuron-astrocyte and a
