Experimental infection models of ticks of the Rhipicephalus sanguineus group with Rickettsia conorii.

Rhipicephalus sanguineus group ticks collected on dogs in Thailand were maintained in the laboratory over several generations to test methods to infect these ticks with Rickettsia conorii, the agent of Mediterranean spotted fever. Three methods were tested: (1) infection of adults and nymphs through artificially induced bacteremic rabbit, (2) capillary feeding of solution containing 5 x 10(3) and 5 x 10(5)pfu/mL of R. conorii to adult female ticks, and (3) immersion of engorged nymphs which were "one leg-cut," "two leg-cut," or "cuticle cut" in solution containing R. conorii. The most efficien

Prolonged Attachment of Rhipicephalus microplus Tick Without Systemic Manifestations: A Case Report.

Rhipicephalus microplus, commonly known as the Asian blue tick, is a cattle parasite rarely reported to infest humans. We describe a 70-year-old Saudi man who presented with a month-long, asymptomatic tick attachment on his left flank after a camping trip. Examination revealed a live, engorged tick with mild surrounding erythema and edema but no systemic symptoms. Laboratory investigations were unremarkable, and histopathology of a punch biopsy showed eosinophilic dermatitis consistent with a localized hypersensitivity reaction. The tick was removed intact and identified as R. microplus by the

Coxiella burnetii detected in Rhipicephalus sanguineus ticks collected in Portuguese autochthonous livestock.

Coxiella burnetii is a bacterium that causes coxiellosis in animals and Q fever in humans. While ruminants are the main reservoirs, the role of ticks in their transmission is still uncertain. This study looked for C. burnetii in ticks collected from two autochthonous breeds in Portugal: Churra Galega Mirandesa sheep and Garrano horses. A total of 555 ticks were tested, including 100 Rhipicephalus sanguineus sensu (s.) stricto (s.) from sheep and 455 Rhipicephalus bursa from horses. Coxiella burnetii was found in 7 % of R. sanguineus s.s., but not in R. bursa. Phylogenetic analysis confirmed th

Partial DnaK protein expression from Coxiella-like endosymbiont of Rhipicephalus annulatus tick.

Q fever is one of the most important zoonotic diseases caused by the obligate intracellular bacteria, Coxiella burnetii. This bacterial infection has been frequently reported in both humans and animals, especially ruminants. Ticks are important ectoparasite and serve as reservoir hosts of Coxiella-like endosymbionts (CLEs). In this study, we have attempted to express chaperone-coding genes from CLEs of Rhipicephalus annulatus ticks collected fromcow path. The partial DnaK coding sequence has been amplified and expressed by Escherichia coli. Amino acid sequences have been analyzed by MS-MS spec

Repellent properties of Rotheca glabrum plant extracts against adults of Rhipicephalus appendiculatus.

BACKGROUND: Rotheca glabrum (formerly known as Clerodendrum glabrum [Verbenaceae]) is used by local communities in the Limpopo Province of South Africa to control ticks on livestock and was selected from the database of the ARC-Onderstepoort Veterinary Institute. Its leaves were extracted using organic solvents ranging from polar to non-polar solvents (methanol, acetone and dichloromethane (DCM)). In addition, the traditional soap-water (infusion) and water-based (decoction) methods were used. The tick repelling activity was determined against the adult stage of the livestock tick Rhipicephalu

Mitochondrial DNA analysis of Rhipicephalus sanguineus s.l. from the western Iberian peninsula.

Rhipicephalus sanguineus Latreille (1806) (Ixodida: Ixodidae) is considered to be the most widely distributed tick and to have a vast range of habitats and hosts, including livestock, pets and wildlife. In addition to morphological differences, recent investigations using approaches based on molecular genetic markers have revealed the existence of different R. sanguineus lineages in different geographic regions. In this study, 475 ticks collected from dogs in the western Iberian peninsula were studied both morphologically and genetically, using 12S and 16S rDNA and COI gene markers in order to

Comparative microarray analyses of adult female midgut tissues from feeding Rhipicephalus species.

The cattle tick, Rhipicephalus microplus, has a debilitating effect on the livestock industry worldwide, owing to its being a vector of the causative agents of bovine babesiosis and anaplasmosis. In South Africa, co-infestation with R. microplus and R. decoloratus, a common vector species on local livestock, occurs widely in the northern and eastern parts of the country. An alternative to chemical control methods is sought in the form of a tick vaccine to control these tick species. However, sequence information and transcriptional data for R. decoloratus is currently lacking. Therefore, this

Central nervous system of Rhipicephalus sanguineus ticks (Acari: Ixodidae): an ultrastructural study.

This study performed the ultrastructural description of the synganglion of Rhipicephalus sanguineus males and females, aiming to contribute to the understanding of the cellular organization of this organ. The results show that the central nervous system of these individuals consists of a mass of fused nerves, named synganglion, from where nerves emerge towards several parts of the body. It is surrounded by the neural lamella, a uniform and acellular layer, constituted by repeated layers of homogeneous and finely granular material. The perineurium is just below, composed of glial cells, which e

Synergistic activity of carvacrol in combination with permethrin against permethrin resistant Rhipicephalus annulatus.

The rapid escalation of acaricide resistance in Rhipicephalus annulatus poses a significant threat to livestock health and productivity, necessitating the development of effective resistance-breaking tools. This study characterized the high-level resistance status of R. annulatus populations in Mazandaran Province, Iran, and evaluated the synergistic potential of carvacrol in combination with permethrin. Field-collected ticks and a susceptible reference strain were evaluated in triplicate using adult immersion tests (AIT), ovicidal assays, larval immersion tests (LIT), and larval packet tests

Acaricidal Activity of Liquid Culture Filtrates from Nematophagous Fungi Against Rhipicephalus microplus Ticks.

Rhipicephalus microplus ticks harm cattle health and productivity. Frequent use of ixodicides can result in resistant tick populations and environmental contamination, as residues enter soil. Nematophagous (NF) and entomopathogenic fungi (EF) are natural enemies of nematodes and insect pests. This study tested the in vitro lethal effects of liquid culture filtrates (LCFs) from selected NF on R. microplus. Fungi were grown in Czapek-Dox broth for 30 days at room temperature to obtain LCFs, which were tested using the immersion method. Experimental groups included: (1) Arthrobotrys musiformis, (

Babesia bovis infection alters the composition and assembly of Rhipicephalus microplus midgut microbiota.

INTRODUCTION: Babesia bovis is one of the main causative agents of bovine babesiosis. Livestock farmers are constantly struggling to control the population of the tick vector and reduce babesiosis outbreaks. For this reason, the development of new control strategies is necessary. Tick microbiota consists of a diverse group of symbiotic, commensal, and pathogenic microorganisms. It has been shown that altering the microbiota population prevents the transmission of apicomplexan pathogens. This work represents a primary exploratory approach to determine the changes B. bovis infection causes in th

Transovarial transmission of Babesia ovis in Rhipicephalus bursa, confirmed by multi-generational experiments.

Babesia ovis is a tick-transmitted intraerythrocytic apicomplexan parasite that causes ovine babesiosis, a disease with high economic impact in endemic regions. Although Rhipicephalus bursa is a known biological vector of B. ovis, the dynamics of vertical (transovarial) transmission are poorly understood. In the present study, the transovarial transmission of B. ovis was investigated experimentally in four continuous generations of R. bursa under laboratory controlled conditions. A total of 11 sheep were used during the study. An initial stabilate co-infected with B. ovis and Theileria ovis wa

Mutations in Rhipicephalus microplus GABA gated chloride channel gene associated with fipronil resistance.

The tropical cattle tick, Rhipicephalus microplus, is one of the most damaging parasites that affects cattle in tropical and subtropical regions in the world. Tick resistance to acaricides is dispersed worldwide and a number of associated mutations in target site genes have been described. Phenylpyrazole (e.g. fipronil) and cyclodiene (e.g. lindane, dieldrin) insecticides both have the same mode of action blocking the GABA-gated chloride channel encoded by the GABA-Cl gene. A conserved mutation, rdl (resistance to dieldrin) is found across a number of arthropods resistant to cyclodienes and ph

Assessment and partial purification of serine protease inhibitors from Rhipicephalus (Boophilus) annulatus larvae.

Ticks are rich sources of serine protease inhibitors, particularly those that prevent blood clotting and inflammatory responses during blood feeding. The tick Rhipicephalus (Boophlus) annulatus is an important ectoparasite of cattle. The aims of this study were to characterize and purify the serine protease inhibitors present in R. (B.) annulatus larval extract. The inhibitors were characterized by means of one and two-dimensional reverse zymography, and purified using affinity chromatography on a trypsin-Sepharose column. The analysis on one and two-dimensional reverse zymography of the larva

Differential immunoproteomics enables identification of Metarhizium anisopliae proteins related to Rhipicephalus microplus infection.

Differential immunoproteomics was applied to identify proteins secreted by Metarhizium anisopliae induced by the Rhipicephalus microplus cuticle. In addition, IgG anti-spore surface proteins were used for searching for proteins possibly involved in early stages of fungus versus tick infection. LC-MS/MS of differentially secreted proteins led to the identification of proteases (carboxypeptidase and Pr1A), chitinase, carboxylic acid transport and proline-rich protein. Differential immunoproteomics strategy facilitated the detection and the identification of new proteins related to M. anisopliae

Proteomic profiling of Rhipicephalus (Boophilus) microplus midgut responses to infection with Babesia bovis.

Differences in protein expression in midgut tissue of uninfected and Babesia bovis-infected southern cattle ticks, Rhipicephalus (Boophilus) microplus, were investigated in an effort to establish a proteome database containing proteins involved in successful pathogen transmission. The electrophoretic separation of midgut membrane proteins was greatly improved by using liquid-phase isoelectric focusing combined with one-dimensional or two-dimensional (2-D) gel electrophoresis. A selection of differentially expressed proteins were subjected to analysis by capillary-HPLC-electrospray tandem mass

A hard tick relapsing fever group spirochete in a Brazilian Rhipicephalus (Boophilus) microplus.

Tick-borne diseases usually comprise a complex epidemiological and ecological network connecting the vector, pathogen, and a group of host species. Symptoms associated with Lyme disease have been reported in Brazil, but no Borrelia sp. has been definitively related to these events. Here we have identified a B. lonestari/B. theileri-related spirochete DNA in the cattle tick Rhipicephalus (Boophilus) microplus from Brazil. Four hundred R. microplus and 80 Amblyomma cajennense ticks were screened, and only 1 horse-fed R. microplus was infected. A Borrelia sp. 16S rDNA sequence was amplified by po
