Genetic Structure of Rhizobium leguminosarum biovar trifolii and viciae Populations Found in Two Oregon Soils under Different Plant Communities.

An investigation was carried out to determine the genetic structure in soil populations of Rhizobium leguminosarum bv. trifolii and viciae at each of two Oregon sites (A and C) that were 1 km apart. Although the soils were similar, the plant communities were quite different because grazing by domestic animals had been allowed (site A) or prevented (site C). Analysis of allelic variation at 13 enzyme-encoding loci by multilocus enzyme electrophoresis delineated 202 chromosomal types (ETs) among a total of 456 isolates representing two populations of R. leguminosarum bv. trifolii (AT and CT) and

Agro‑ecological and Genetic Drivers of Symbiotic Efficiency of Indigenous Rhizobium etli Strains in Common Bean in Southern Ethiopia.

A total of 36 root‑nodule bacteria were isolated from southern Ethiopia using the Red Wolaita common bean variety, and 26 were authenticated as rhizobia and then screened for symbiotic effectiveness in a greenhouse at Arba Minch University. Among the authentic isolates, strains AMU23, AMU74, and AMU3A had symbiotic effectiveness values of 138%, 117%, and 116.7%, respectively, and were further evaluated along with a commercial strain, HB429, in farmers' fields at varying altitudes with the Red Wolaita common bean variety. The selected rhizobial isolates were introduced by seed inoculation with

Strigolactones in the Rhizobium-legume symbiosis: Stimulatory effect on bacterial surface motility and down-regulation of their levels in nodulated plants.

Strigolactones (SLs) are multifunctional molecules acting as modulators of plant responses under nutrient deficient conditions. One of the roles of SLs is to promote beneficial association with arbuscular mycorrhizal (AM) fungi belowground under such stress conditions, mainly phosphorus shortage. Recently, a role of SLs in the Rhizobium-legume symbiosis has been also described. While SLs' function in AM symbiosis is well established, their role in the Rhizobium-legume interaction is still emerging. Recently, SLs have been suggested to stimulate surface motility of rhizobia, opening the possibi

Characterization of the gene encoding nitrite reductase and the physiological consequences of its expression in the nondenitrifying Rhizobium "hedysari" strain HCNT1.

Rhizobium "hedysari" HCNT1 is an unclassified rhizobium which contains a nitric oxide-producing nitrite reductase but is apparently incapable of coupling the reduction of nitrite to energy conservation. The gene encoding the nitrite reductase, nirK, has been cloned and sequenced and was found to encode a protein closely related to the copper-containing family of nitrite reductases. Unlike other members of this family, nirK expression in HCNT1 is not dependent on the presence of nitrogen oxides, being dependent only on oxygen concentration. Oxygen respiration of microaerobically grown Nir-defic

Influence of Bovine Slurry Deposition on the Structure of Nodulating Rhizobium leguminosarum bv. viciae Soil Populations in a Natural Habitat.

The population of nodulating R. leguminosarum bv. viciae in soil from a grass-covered valley area which had been used for bovine slurry deposition over a period of 5 years was analyzed. For these studies, a rapid and reproducible method based on enterobacterial repetitive intergenic consensus (ERIC)-PCR was applied to identify Rhizobium strains which had infected pea nodules. Soil samples were taken from different areas and further analyzed in plant tests to determine the impact of the application of slurry (polluted or nonpolluted), the slope position (summit or toe), and exposure (north or s

C-N exchange model of legume-Rhizobium symbiosis incorporating ATP budget constraints and energy-mass balance between the species.

We present a comparative advantage model of carbon-nitrogen exchange in legume-Rhizobium symbiosis that incorporates ATP budget constraints and the energy-mass balance between the host and symbiont. In this framework, the uptake of carbon and nitrogen is limited by the ATP available to each partner, and any imbalance in trade is compensated by adjustments in symbiont biomass. Using empirical estimates of the ATP costs of carbon and nitrogen uptake, together with data on body C:N ratios, the model generates three key predictions, and we prove that they align with empirical results. Protocol for the High-quality Plasmid Isolation from Different Recalcitrant Bacterial Species: Agrobacterium spp., Rhizobium sp., and Bacillus thuringiensis.

High yield of good quality plasmid DNA from gram -ve bacteria (Agrobacterium tumefaciens, A. rhizogenes, and Rhizobium sp.) and gram +ve bacterium (Bacillus thuringiensis) is difficult. The widely used plasmid extraction kits for Escherichia coli yield a low quantity of poor-quality plasmid DNA from these species. We have optimized an in-house modification of the QIAprep Spin Miniprep kit protocol of Qiagen, consisting of two extraction steps. In the first, the centrifugation after adding neutralization buffer is followed by ethanol (absolute) precipitation of plasmid DNA. In the second extrac

Ecotoxicological assessment of pesticides towards the plant growth promoting activities of Lentil (Lens esculentus)-specific Rhizobium sp. strain MRL3.

This study was designed to evaluate the effect of the selected pesticides [herbicides (metribuzin and glyphosate), insecticides (imidacloprid and thiamethoxam) and fungicides (hexaconazole, metalaxyl and kitazin)] at the recommended and the higher dose rates on plant growth promoting traits of Rhizobium sp. strain MRL3 isolated from lentil-nodules. Strain MRL3 was explicitly selected owing to its high pesticide-tolerance ability and substantial production of indole acetic acid, siderophores (salicylic acid and 2, 3 dihydroxy benzoic acid), exo-polysaccharides, HCN and ammonia. A simple method for creating transgenic pea hairy roots using a Japanese pea cultivar and a Japanese Rhizobium radiobacter strain.

Pea (Pisum sativum) is an agriculturally important leguminous crop cultivated worldwide. It is also the plant from which phytoalexin was isolated for the first time. Several studies have investigated gene functions using pea hairy root culture systems. However, the procedures for producing hairy roots are relatively complicated and only a few pea cultivars and Rhizobium strains have been used. In this study, we established a simple method for generating transgenic hairy roots using a pea cultivar and a Rhizobium strain available in Japan. The transformation efficiency for the transgenic hairy

The structures of the lipopolysaccharides from Rhizobium etli strains CE358 and CE359. The complete structure of the core region of R. etli lipopolysaccharides.

The structural arrangement of oligosaccharides comprising the core region of Rhizobium etli CE3 lipopolysaccharide (LPS) has been elucidated through the characterization of the LPSs from two R. etli mutants. One mutant, CE358, completely lacks the O-chain polysaccharide, while the second mutant, CE359, contains a truncated portion of this polysaccharide. This structural arrangement of the core oligosaccharides in these LPSs was determined using electrospray ionization mass spectrometry, tandem mass spectrometry, and methylation analysis. Mild acid hydrolysis of the CE359 LPS produces two major

The CcrM DNA methyltransferase is widespread in the alpha subdivision of proteobacteria, and its essential functions are conserved in Rhizobium meliloti and Caulobacter crescentus.

The Caulobacter crescentus DNA methyltransferase CcrM (M.CcrMI) methylates the adenine residue in the sequence GANTC. The CcrM DNA methyltransferase is essential for viability, but it does not appear to be part of a DNA restriction-modification system. CcrM homologs are widespread in the alpha subdivision of gram-negative bacteria. We have amplified and sequenced a 258-bp region of the cerM gene from several of these bacteria, including Rhizobium meliloti, Brucella abortus, Agrobacterium tumefaciens, and Rhodobacter capsulatus. Alignment of the deduced amino acid sequences revealed that these

Comparative Genomic Analysis of the Type VI Secretion System in Strains of the Rhizobium leguminosarum Species Complex with Variable Host Specificity.

In this study, genes encoding components of the type VI (T6SS) secretion system and putative effectors were searched in rhizobial strains belonging to the Rhizobium leguminosarum species complex. For this purpose, the genome assembly of the new Rhizobium laguerreae Wp19 strain (genospecies N) isolated from wild pea nodules was done and compared to the type strains. Comparative analysis of tss gene clusters was conducted in the new Wp19 strain and other R. laguerreae strains, as well as R. ruizarguesonis and R. johnstonii strains with available genomes. The resulting genome assemblies of R. lag

Unraveling the pleiotropic functions of Rhizobium rhizogenes rolB and rolC genes on phenotype, secondary metabolism, and drought tolerance in Atropa belladonna.

Atropa belladonna L. (Solanaceae) is a significant source of the pharmacologically important tropane alkaloids (TAs), particularly hyoscyamine and scopolamine. Although rolB and rolC from Rhizobium rhizogenes are known to enhance plant secondary metabolism, their functional specificities and combined actions in TAs biosynthesis remain unclear. Here, we generated transgenic A. belladonna overexpressing 35S::rolB, 35S::rolC, and 35S::rolB + rolC to systematically investigate their effects on TAs accumulation, plant morphology, and drought tolerance. Co-expression of rolB and rolC exhibited metab

Living without Fur: the subtlety and complexity of iron-responsive gene regulation in the symbiotic bacterium Rhizobium and other alpha-proteobacteria.

The alpha-proteobacteria include several important genera, including the symbiotic N(2)-fixing "rhizobia", the plant pathogen Agrobacterium, the mammalian pathogens Brucella, Bartonella as well as many others that are of environmental or other interest--including Rhodobacter, Caulobacter and the hugely abundant marine genus Pelagibacter. Only a few species--mainly different members of the rhizobia--have been analyzed directly for their ability to use and to respond to iron. These studies, however, have shown that at least some of the "alphas" differ fundamentally in the ways in which they regu

Haloalkanoate dehalogenase II (DehE) of a Rhizobium sp.--molecular analysis of the gene and formation of carbon monoxide from trihaloacetate by the enzyme.

A 3-kb EcoRI fragment of genomic DNA from a Rhizobium sp. cloned into pUC19 endowed Escherichia coli K-12 with the ability to grow, albeit slowly, with 2-chloropropionic acid as substrate. The construct expressed weakly a gene that encoded a non-stereospecific 2-chloropropionic acid dehalogenase (dehalogenase II; DehE). The dehE gene was not closely linked to the organism's other two dehalogenase genes, dehD and dehL. The derived amino acid sequence of DehE showed little identity with DehD or DehL, but there was significant identity to two other dehalogenases that act non-selectively on 2-chlo

The Rhizobium sp. strain NGR234 systemically suppresses arbuscular mycorrhizal root colonization in a split-root system of barley (Hordeum vulgare).

Nitrogen-fixing bacteria (rhizobia) form a nodule symbiosis with legumes, but also induce certain effects on non-host plants. Here, we used a split-root system of barley to examine whether inoculation with Rhizobium sp. strain NGR234 on one side of a split-root system systemically affects arbuscular mycorrhizal (AM) root colonization on the other side. Mutant strains of NGR234 deficient in Nod factor production (strain NGRΔnodABC), perception of flavonoids (strain NGRΔnodD1) and secretion of type 3 effector proteins (strain NGRΩrhcN) were included in this study. Inoculation resulted in a syste

Rhizobium Inoculation Enhances Drought Tolerance of Legume Species by Increasing Leaf Hydraulic Efficiency and Safety and Speeding Up Post-Drought Recovery.

The trade-off between leaf hydraulic efficiency and safety is among the principal theories explaining plant drought adaptation, but whether rhizobium inoculation modulates this trade-off to enhance plant drought adaptation remains unclear. Here, leaf water potential (Ψleaf), stomatal conductance (gs), photosynthesis (An) and leaf hydraulic conductance (Kleaf) during and after drought were measured in rhizobium-inoculated and non-inoculated Glycine max and Caragana korshinskii grown under well-watered (WW), moderate drought stress (MS) and severe drought stress (SS) conditions. Mutation of a broadly conserved operon (RL3499-RL3502) from Rhizobium leguminosarum biovar viciae causes defects in cell morphology and envelope integrity.

The bacterial cell envelope is of critical importance to the function and survival of the cell; it acts as a barrier against harmful toxins while allowing the flow of nutrients into the cell. It also serves as a point of physical contact between a bacterial cell and its host. Hence, the cell envelope of Rhizobium leguminosarum is critical to cell survival under both free-living and symbiotic conditions. Transposon mutagenesis of R. leguminosarum strain 3841 followed by a screen to isolate mutants with defective cell envelopes led to the identification of a novel conserved operon (RL3499-RL3502

Rhizobium tropici genes involved in free-living salt tolerance are required for the establishment of efficient nitrogen-fixing symbiosis with Phaseolus vulgaris.

Characterization of nine transposon-induced mutants of Rhizobium tropici with decreased salt tolerance (DST) allowed the identification of eight gene loci required for adaptation to high external NaCl. Most of the genes also were involved in adaptation to hyperosmotic media and were required to overcome the toxicity of LiCl. According to their possible functions, genes identified could be classified into three groups. The first group included two genes involved in regulation of gene expression, such as ntrY, the sensor element of the bacterial ntrY/ntrX two-component regulatory system involved

Adaptive resistance to and intracellular bioaccumulation of potentially toxic metals in a plant growth-promoting Rhizobium tropici-like isolate: implications for phytoremediation.

The accumulation of potentially toxic metals (PTMs) poses a threat to the cellular integrity of soil microorganisms as well as ecological stability, driving the selection of resistant microorganisms with biotechnological potential. This study aimed to evaluate the behavior of the Rhizobium sp. LBMP-C04 isolate under PTM stress, focusing on adaptive resistance mechanisms and intracellular bioaccumulation. The bacterium, which was isolated from Arachis pintoi root nodules and had previously undergone genomic characterization, was subjected to antibiotic-resistance assays, growth-curve analysis i
